Genetic Determinants of Statin-induced Myopathy: A Network Meta-analysis of Observational Studies.

INTRODUCTION: Statin-induced myopathy (SIM) is a prevalent adverse event impacting treatment adherence. Despite extensive exploration of single nucleotide polymorphisms (SNPs), conflicting evidence obscures their role in SIM incidence, prompting this network meta-analysis. METHODS: Observational studies meeting eligibility criteria (patients on any statin with reported SNPs and SIM details) were systematically reviewed. Severe SIM was defined as creatine kinase elevations exceeding 10 times the upper limit of normal. Mixed treatment comparison pooled estimates were generated from direct and i

Genetic determinants of cell- and egg-preferred replication in H3N2 canine influenza virus.

The H3N2 canine influenza virus (CIV), which originated from avian influenza viruses and has circulated in dogs for over two decades, remains poorly understood in terms of its host adaptation mechanisms and cellular tropism. In this study, we isolated two phenotypically distinct H3N2 CIV strains (CX3 and CX12) from a canine respiratory disease outbreak. Although both were isolated using MDCK cells, CX3 showed higher replication efficiency and plaque-forming ability in MDCK cells, whereas CX12 replicated more efficiently in chicken embryos. Using reverse genetics, we identified the PB1 gene as

Genetic and epigenetic determinants of vitamin D metabolism: nutrigenomic insights for precision nutrition.

Vitamin D plays a pivotal role in immune regulation, metabolic balance, skeletal health, and gene expression. Growing evidence indicates that genetic and epigenetic factors contribute to interindividual differences in vitamin D status and physiological responses. This review summarizes current findings on the nutrigenomic determinants of vitamin D metabolism, with emphasis on genetic polymorphisms in vitamin D receptor (VDR), GC, CYP2R1, CYP27B1, and CYP24A1, as well as epigenetic mechanisms that modulate vitamin D related gene expression. Peer-reviewed original studies and review articles pub

Genetic Determinants of Interindividual Differences in Provitamin A Carotenoid Concentrations in Human Adipose Tissue.

Adipose tissue is a major storage site for provitamin A carotenoids (proVA CAR)-mainly β-carotene (BCAR), α-carotene (ACAR), and β-cryptoxanthin (BCRY). However, the determinants of their concentrations in this tissue remain poorly understood. This study aimed to identify genetic variants associated with adipose tissue proVA CAR concentrations. Periumbilical adipose tissue samples were collected on six occasions in 43 healthy adult males and proVA CAR concentrations were quantified by HPLC. Participants were genotyped for 2,398 SNPs in 44 candidate genes involved in CAR and lipid metabolism, a

Genetic determinants of heart failure susceptibility and response in the collaborative cross mouse population.

Genetic variation and lived experiences shape how our hearts respond to chronic stress and development of heart failure, manifested as compromised pumping function and abnormal hemodynamics. The hallmark of heart failure etiology is excessive stress signals followed by maladaptive structural, electrical, and functional changes to the heart muscle, also known as cardiac remodeling. The specific genetic mechanisms which underly such phenomenon, however, are still unclear, due in part to difficulties in accounting for environmental effects in human population studies. To overcome this challenge,

Network ontology transcript annotation identifies genetic signals underlying sex determination.

Cannabis sativa L. (marijuana, hemp, cannabis) is an angiosperm species currently evolving sex chromosomes. Genetic mechanisms, primarily an XY chromosome system, dictate cannabis sex expression in dioecious populations. However, sexual expression is also governed by the interplay of hormone regulatory gene networks, influenced by both genetic and environmental factors. Within the species, some populations exhibit dioecy, monoecy, or a gradient of both. Dioecious individuals produce exclusively male or female flowers, while monoecious plants bear both male and female flowers. Remarkably, throu

Host and Pathogen Genetic Determinants of Brucellosis in Veterinary-One Health Interface: A Review.

Brucellosis remains among the most widespread global zoonoses, yet its genetic architecture-spanning host susceptibility and pathogen virulence-is increasingly being investigated. Advances in whole-genome sequencing, transposon mutagenesis, CRISPR screening, and machine learning-driven transcriptomics have accelerated marker discovery on both sides of the host-pathogen interface, although most signatures remain at discovery or early-validation stages. In hosts, polymorphisms in innate immune receptors (TLR2, TLR5, VDR), cytokine regulators (TNF, CTLA4), and novel candidates such as ASAP1 are l

From Genes to Membrane Failure: Genetic Determinants of Peritoneal Dialysis Physiology and Outcomes.

Peritoneal dialysis (PD) has long been an established modality of renal replacement therapy for patients with end-stage kidney disease (ESKD). Despite the modality's advantages, significant inter-individual variability exists in peritoneal membrane transport characteristics, ultrafiltration capacity, and long-term technique survival. While PD therapy-related factors, such as dialysis solution composition, peritonitis episodes, and duration of therapy, contribute to these outcomes, genetic factors also play important roles in peritoneal membrane biology. Genetic studies have identified polymorp

Large-Scale Mutagenesis Screening for Genetic Determinants of Plasmodium falciparum Sexual Development.

To develop effective malaria transmission-blocking vaccines and drugs, it is crucial to understand the genetic factors and molecular mechanisms that regulate the development of Plasmodium blood-stage sexual forms, known as gametocytes-parasite stage capable of surviving in the mosquito vector. We established a scalable forward genetic screen approach using single-insertion mutants generated by random piggyBac mutagenesis. This method identifies genes essential for asexual parasite forms survival or tolerance to critical in vivo phenotype responses, such as febrile temperature, antimalarial dru

Genetic determinants of Staphylococcus aureus adhesion shape virulence trade-offs in bacteremia.

Staphylococcus aureus is a major cause of bloodstream infections, but how variation between strains in their ability to adhere to host proteins influences disease severity remains unclear. Here we show that adhesion is a highly variable and biologically important trait in 236 phylogenetically diverse, representative bacteremia isolates profiled for binding to fibrinogen and fibronectin and analysed together with bacterial whole-genome sequences and matched patients' clinical data. Stronger fibrinogen-binding, particularly in strains lacking α-toxin, correlates with heightened systemic inflamma

Investigating microbiota differences across chronic pancreatitis, influenced by lifestyle and genetic determinants.

BACKGROUND: Chronic pancreatitis (CP) is a multifaceted and irreversible inflammatory condition that impacts both endocrine and exocrine pancreatic functions. Whilst there is evidence of a general intestinal microbial dysbiosis, differences across CP resulting from diverse triggers remains poorly elucidated. METHODS: In this study, we investigated how different CP triggers, namely alcohol-induced CP (AIP) and CFTR-related CP (CFRP), might undermine differences across microbiota structure and predicted function. Faecal samples from 25 CP participants were collected, alongside 9 healthy controls

Genetic determinants of silver nanoparticle resistance and the impact of gamma irradiation on nanoparticle stability.

BACKGROUND: One of the main issues facing public health with microbial infections is antibiotic resistance. Nanoparticles (NPs) are among the best alternatives to overcome this issue. Silver nanoparticle (AgNPs) preparations are widely applied to treat multidrug-resistant pathogens. Therefore, there is an urgent need for greater knowledge regarding the effects of improper and excessive use of these medications. The current study describes the consequences of long-term exposure to sub-lethal concentrations of AgNPs on the bacterial sensitivity to NPs and the reflection of this change on the bac

Genetic Diversity in Antimicrobial Resistance Determinants Among Pathogenic Pseudomonas aeruginosa in India.

The drastic rise in antibiotic resistance has become a global challenge, including India, due to high morbidity. The delayed identification and lack of treatment are the major causes of death. However, there is a shortage of precise information on the specific resistance pattern and sequence types of Pseudomonas aeruginosa from India that can help in diagnostics and therapy. A total of 16 clinical isolates were collected from the western region of India, along with 181 P. aeruginosa genomes of India from public database were retrieved and thoroughly analysed for antibiotics resistance determin

Importance of selected genetic determinants on endurance performance and physical strength: a narrative review.

Physical strength and endurance of an individual are vital for athletic performance, and minimizing the risk of injuries, especially during competitions. Other than training and diet, athletic performance is determined by genetic factors or heredity, which is less focused in sports science research. Genetic factors play a crucial role in greater cardiovascular endurance and muscular phenotypes, and thereby contribute to athletic success. Several genes and different polymorphisms are positively/negatively associated with athletic performance. This review delved into the intricate role of severa

Genetic determinants of efficacy of antiviral drugs revealed by genome-wide CRISPR screens.

Nucleoside and nucleobase analog antiviral drugs are pivotal in antiviral therapy, but comprehensive methods to understand their cellular response mechanisms and genetic regulators are still lacking. Here, we show that Eμ-Myc; Arf-/- mouse lymphoma cells, which are highly apoptosis-prone, enabled genome-wide CRISPR-Cas9 screening on such drugs to identify genes that modulate their efficacy. Using retroviral sgRNA libraries and MAGeCK analysis, we uncovered key regulators of drug transport, activation, and inactivation for these drugs. For ribavirin, adenosine kinase (ADK) and adenylsuccinate s

The Genetic Determinants of Listeria monocytogenes Resistance to Bacteriocins Produced by Lactic Acid Bacteria.

BACKGROUND: Listeria monocytogenes is a Gram-positive bacterium responsible for listeriosis, a serious foodborne disease that can lead to serious health complications. Pregnant women, newborns, the elderly, and patients with weakened immune systems are particularly susceptible to infection. Due to the ability of L. monocytogenes to survive in extreme environmental conditions, such as low temperatures, high salinity, and acidity, this bacterium poses a serious threat to food production plants and is particularly difficult to eliminate from these plants. One of the promising solutions to reduce
