Joint exposure to positive affect, life satisfaction, broad depression, and neuroticism and risk of cardiovascular diseases: A prospective cohort study.

BACKGROUND AND AIMS: Psychologic wellbeing can impact cardiovascular health. We aimed to evaluate the joint association of multiple psychologic wellbeing factors with cardiovascular diseases (CVD) and examine whether this association was modified by genetic susceptibility. METHODS: In the UK Biobank, 126,255 participants free of CVD (coronary heart disease [CHD], stroke, and heart failure [HF]) at baseline, who completed a questionnaire on psychological factors, were included. The psychological wellbeing score was calculated by four factors: happiness, life satisfaction, broad depression, and

Polygenic risk predicts obesity in both white and black young adults.

OBJECTIVE: To test transethnic replication of a genetic risk score for obesity in white and black young adults using a national sample with longitudinal data. DESIGN AND METHODS: A prospective longitudinal study using the National Longitudinal Study of Adolescent Health Sibling Pairs (n = 1,303). Obesity phenotypes were measured from anthropometric assessments when study members were aged 18-26 and again when they were 24-32. Genetic risk scores were computed based on published genome-wide association study discoveries for obesity. Analyses tested genetic associations with body-mass index (BM

Development of an Integrated Monogenic and Polygenic Risk Assessment Tool for Coronary Artery Disease and Its Application in a Community-Based Health Care Biobank.

BACKGROUND: Current genetic testing for coronary artery disease (CAD) primarily targets monogenic variants in individuals with severe hypercholesterolemia. Whether supplementing monogenic testing with polygenic risk scores for CAD and Lp(a; lipoprotein[a]) levels [PRSLp(a)] improves identification of high-risk individuals in the general population remains understudied. METHODS: A genetic probability for CAD (GenProbCAD), incorporating monogenic pathogenic variants (PVs), polygenic risk scores for CAD, and PRSLp(a) was developed using Cox regression in 226 145 UK Biobank participants, validate

Susceptibility Loci for Clinical Coronary Artery Disease and Subclinical Coronary Atherosclerosis Throughout the Life-Course.

BACKGROUND: Recent genome-wide association studies have identified 49 single nucleotide polymorphisms associated with clinical coronary artery disease. The mechanism by which these loci influence risk remains largely unclear. METHODS AND RESULTS: We examined the association between a genetic risk score composed of high-risk alleles at the 49 single nucleotide polymorphisms and the degree of subclinical coronary atherosclerosis in 7798 participants from 6 studies stratified into 4 age groups at the time of assessment (15-34, 35-54, 55-74, and >75 years). Atherosclerosis was quantified by stain

Endothelial Susceptibility-Related Genetic Variants and Hypertensive Disorders of Pregnancy-Brief Report.

BACKGROUND: The hypertensive disorders of pregnancy (HDPs), that is, preeclampsia and gestational hypertension, are characterized by endothelial dysfunction in pregnancy and are epidemiologically and genetically associated with risk for coronary artery disease (CAD). A recent study suggested that endothelial cell (EC)-acting CAD risk variants may identify individuals who benefit from more intensive lipid-lowering treatment. The present study evaluated whether the same EC CAD genetic risk score is also associated with HDPs using comprehensive statistical genetic approaches. METHODS: We examine

Genetic variation supports a causal role for valproate in prevention of ischemic stroke.

BACKGROUND: Valproate is a candidate for ischemic stroke prevention due to its anti-atherosclerotic effects in vivo. Although valproate use is associated with decreased ischemic stroke risk in observational studies, confounding by indication precludes causal conclusions. AIMS: We applied Mendelian randomization to determine whether genetic variants that influence seizure response among valproate users associate with ischemic stroke. METHODS: We derived a genetic score for valproate response using genome-wide association data of seizure response after valproate intake from the Epilepsy Pharma

Polygenic risk score and cluster-based analysis suggests links between type 2 diabetes and vascular dementia in the KARE study.

Type 2 diabetes is an established risk factor for dementia. However, how its genetic heterogeneity affects different dementia subtypes remains unclear. In this study, we investigate the associations between genetic risk of type 2 diabetes and dementia subtypes among 33,136 older Chinese adults from the KARE cohort. We find that a higher overall polygenic risk score for type 2 diabetes is significantly associated with an increased risk of vascular dementia, but not Alzheimer's disease. Further analyses using cluster-specific partitioned polygenic score show that elevated genetic risk specific t

Association of lifestyle factors and genetic risk with incident abdominal aortic aneurysm.

OBJECTIVE: Although lifestyle factors are associated with the risk of abdominal aortic aneurysm (AAA), its relationship with genetic susceptibility in determining AAA remains unclear. We aim to investigate the association of lifestyle factors and genetic risk with the incidence of AAAs. METHODS: A prospective cohort study, including 274,755 adult participants with White European ancestry and no AAA at baseline, was recruited between 2006 and 2010. A polygenic risk score was calculated to quantify genetic susceptibility to AAA in participants, and a comprehensive healthy lifestyle score was de

Genetic predisposition for depression, psychosocial and behavioural factors and premenstrual symptoms: a cross-sectional study among young women in China.

BACKGROUND: Premenstrual disorders (PMDs) affect approximately one in three women of reproductive age and have a substantial impact on daily functioning and mental health. Given the challenges observed in diagnosis and clinical management, understanding whether genetic predisposition and readily assessable factors jointly mark greater symptom burden may help inform future risk-stratified monitoring and assessment. METHODS: After excluding 424 participants with missing data, we conducted a cross-sectional study of 1528 female college students from the Care of Premenstrual Emotion (COPE) cohort

Association of outdoor light exposure with bone mineral density and osteoporosis risk: A gene-environment interaction analysis.

OBJECTIVE: The association between time spent in outdoor light (TOL), genetic predisposition, and the risk of osteoporosis (OP) remains unclear. This study aimed to quantify the association between TOL and OP risk, and to evaluate whether genetic predisposition modifies this relationship. METHODS: Data from the UK Biobank on 326,216 participants based on self-reported TOL, estimated bone mineral density (eBMD), and hospital inpatient records for OP diagnosis. A polygenic risk score (PRS) for OP was calculated and categorized into tertiles. We used generalized linear models, Cox proportional h

Hyperthyroidism and bone mineral density: Dissecting the causal association with Mendelian randomization analysis.

INTRODUCTION: Untreated hyperthyroidism is associated with accelerated bone turnover, low bone mineral density (BMD) and increased susceptibility to fragility fractures. Although treatment appears to improve or even reverse some of these adverse skeletal effects, there is limited guidance on routine BMD assessment in hyperthyroid patients following treatment. By using Mendelian randomization (MR) analysis, we aimed to assess the causal association of hyperthyroid thyroid states with BMD and fractures using the UK Biobank. METHODS: This MR analysis included data from 473 818 participants (wome

Life's Essential 8, genetic susceptibility and the risk of psoriatic disease: a prospective cohort study.

BACKGROUND: Psoriatic disease (PsD) is closely associated with cardiovascular (CV) disease. The Life's Essential 8 (LE8) score is a new metric to assess CV health (CVH), where a higher score indicates better CVH. However, the longitudinal association between LE8 score and the risk of PsD remains uncertain. OBJECTIVES: To investigate, in a cohort study, the association between LE8 score, genetic susceptibility and the risk of PsD. METHODS: This cohort study included 261 642 participants in the UK Biobank without PsD at baseline. LE8 comprises eight indicators: diet, physical activity, nicotin

Polygenic Risk Profiles and the Manifestations of Alcohol Use Disorder and Related Outcomes.

BACKGROUND: Research suggests that multiple genetically influenced pathways of risk impact alcohol use disorder (AUD), including externalizing (disinhibition, behavioral undercontrol), internalizing (negative affect, depression), and alcohol-specific (physiological response to alcohol) factors. This study examined the joint effects of genetic influences representing these risk domains on AUD and their influence on comorbid phenotypes among individuals with AUD. METHODS: Participants of European-like ancestry from the Collaborative Study on the Genetics of Alcoholism (N = 6883) provided data o

Genetic investigation of non-affective psychosis and depression as causal risk factors for dementia.

BACKGROUND: Major psychiatric disorders are associated with increased risk of dementia but establishing whether psychiatric disorders causally increase dementia risk is challenging because dementia pathology can precede clinical diagnosis by decades. Prodromal psychiatric symptoms may arise long before cognitive decline, leaving open the possibility of reverse causation. OBJECTIVE: We aimed to determine whether non-affective psychosis or depression credibly causally influence dementia risk using a design robust to reverse causation. We tested whether people with psychiatric disorders who late

Validation of fracture-derived polygenic scores with FRAX for fracture risk prediction in postmenopausal women.

UNLABELLED: We evaluated whether adding a genetic risk score derived from forearm fracture improves fracture prediction beyond standard clinical tools. Incorporation of GPS resulted in a small increase in time-dependent AUC, with improvement in NRI. Decision curve analysis suggested potential clinical benefit across certain threshold ranges. BACKGROUND: Previous studies have incorporated estimated bone mineral density (eBMD)-derived polygenic scores into FRAX to improve fracture risk prediction, indirectly capturing genetic susceptibility through bone mineral density. However, the genetic arc

Genetic Contribution to Asthma Informs Acute Chest Syndrome Pathophysiology and Risk Stratification.

Acute chest syndrome (ACS) is a severe complication of sickle cell disease (SCD) occurring in ~50% of patients, some presenting frequent episodes. We lack tools to identify patients at high risk of ACS occurrence or frequent episodes. Epidemiological studies have found an association between asthma and ACS, but whether this link is causal is unclear. We used polygenic scores (PGS) to analyze whether the genetic propensity for asthma was associated with ACS and could be used to stratify the risk of ACS. We identified that PGS for asthma (PGSasthma) was associated with ACS rate (number of episod
