Oxidative stress and immune signatures of carbamazepine in freshwater mussels.

The persistence of pharmaceutical compounds in aquatic environments and their limited removal by conventional treatment processes necessitate evaluating organism-level effects of long-term exposure using biomarkers. To determine how carbamazepine (CBZ), commonly detected in environmental waters, modulates immune and oxidative stress responses in the freshwater mussel Unio delicatus, individuals (n = 160) were exposed to sublethal CBZ concentrations for 21 and 45 days under semi-static conditions. Exposure concentrations were set at 199 µg/L (D1), 995 µg/L (D2), and 1.99 mg/L (D3), correspondin

An LPS-induced TNF-α factor involved in immune response of oyster Crassostrea gigas by regulating haemocytes apoptosis.

LPS induced TNF-α Factor (LITAF) is a transcription factor widely involving in activation of Tumor Necrosis Factor (TNF) and other cytokines in the inflammatory response. In the present study, a homologue of LITAF with a conserved LITAF domain was identified from the Pacific oyster Crassostrea gigas. The transcripts of CgLITAF were detected in all examined tissues with highest expression in hepatopancrease. The immunofluorescence assay and Western blot showed that LPS stimulation induced an obvious nucleus translocation of CgLITAF protein in haemocytes. While the mRNA level of CgLITAF changed

Use of Galleria mellonella as a model organism to study Legionella pneumophila infection.

Legionella pneumophila, the causative agent of a severe pneumonia named Legionnaires' disease, is an important human pathogen that infects a … species in the insect and in mammalian models. A key component of the larvae's immune defenses are hemocytes, professional phagocytes, which take up and destroy invaders. L. pneumophila is able to infect, form a LCV and replicate within these cells. Here we demonstrate protocols for analyzing L. pneumophila virulence in the G. mellonella model, including h …

A flow cytometric approach to study intracellular-free Ca2+ in Crassostrea gigas haemocytes.

Bivalve haemocytes are essential in defence mechanisms including phagocytosis. They also produce molecules including hydrolytic enzymes and antimicrobial peptides that contribute to pathogen destruction. Although haemocyte activities have been extensively studied, relatively little is known about the intracellular signalling pathways that are evoked during haemocyte activation and especially the role of calcium. Flow cytometry has been used for the first time to define the effect of cell incubation in haemolymph and artificial sea water (ASW) on Pacific oyster, Crassostrea gigas, haemocytes. C

In vitro biofilm formation and virulence of Paracoccidioides lutzii (LDR2) in a Galleria mellonella larval model.

Paracoccidioidomycosis (PCM) is a systemic mycosis caused by the thermodimorphic fungi Paracoccidioides brasiliensis and the newly identified species P. lutzii. This study investigated in vitro biofilm formation by P. lutzii (LDR2) and assessed its impact on Galleria mellonella larvae. Scanning electron microscopy (SEM) revealed that P. lutzii (LDR2) forms dense biofilms composed of a complex cellular network embedded in an extracellular matrix. Biofilm-associated infection significantly increased fungal virulence, resulting in higher larval mortality, reduced hemocyte density, and enhanced me

Dietary manganese potentiates BmNPV resistance in silkworms via BmcGLR-mediated activation of BmRelish through BmSTING.

The silkworm (Bombyx mori) is a vital economic species threatened by Bombyx mori nucleopolyhedrovirus (BmNPV), causing severe losses in sericulture. Here, we demonstrate that dietary Mn2+ supplementation enhances silkworm resistance to BmNPV. Transcriptomic analysis and RT-qPCR validation revealed Mn2+-dependent upregulation of Relish-mediated antimicrobial peptides. We identified a cGAS-like receptor (BmcGLR) in B. mori, highly expressed in hemocytes and structurally poised for Mn2+ binding. Mn2+ directly stimulated BmcGLR to amplify -2',3'-cGAMP production, which activated BmSTING-dependent …

Toll-like signaling pathway in the transcriptome of Littorina littorea.

The transcriptome of the caenogastropod mollusk Littorina littorea was scanned for the presence of sequences encoding Toll-like receptors (TLRs) and corresponding proteins involved in downstream TLR signaling pathway. In the transcriptomic snapshots of hemocytes and kidney tissues, 45 complete TLRs encoded by 35 genes were identified. Out of the 59 non-TLR molecules involved in a canonical TLR signaling pathway, 35 genes were classified as homologous and could be placed within the TLR-mediated MyD88-and MAPK-dependent circuitries. No reference vertebrate adapters TIRAP, TRIF and TRAM were iden…

Cyclization of the antimicrobial peptide gomesin with native chemical ligation: influences on stability and bioactivity.

Gomesin is an 18-residue peptide originally isolated from the hemocytes of the Brazilian spider Acanthoscurria gomesiana. A broad spectrum of bioactivities have been attributed to gomesin, including in vivo and in vitro cytotoxicity against tumour cells, antimicrobial, antifungal, anti-Leishmania and antimalarial effects. Given the potential therapeutic applications of gomesin, it was of interest to determine if an engineered version with a cyclic backbone has improved stability and bioactivity. Cyclization has been shown to confer enhanced stability and activity to a range of bioactive peptid…

Gomesin, a peptide produced by the spider Acanthoscurria gomesiana, is a potent anticryptococcal agent that acts in synergism with fluconazole.

Gomesin is an 18-residue cysteine-rich antimicrobial peptide produced by hemocytes of the spider Acanthoscurria gomesiana. In the present study, the antifungal properties of gomesin against Cryptococcus neoformans, the etiologic agent of cryptococcosis, were evaluated. Gomesin bound to the cell surface of cryptococci, which resulted in cell death associated with membrane permeabilization. Antifungal concentrations of gomesin were not toxic for human brain cells. Supplementation of cryptococcal cultures with the peptide (1 microM) caused a decrease in capsule expression and rendered fungal cell…

Silica treatment increases the susceptibility of the Cabo Frio strain of Biomphalaria tenagophila to Schistosoma mansoni infection but does not alter the natural resistance of the Taim strain.

The present work demonstrates that silica treatment represents a suitable in vivo method to evaluate the role of host phagocytic hemocytes in the mechanisms of resistance to parasitic infection. Silica inoculation into Biomphalaria tenagophila snail induced a significant reduction in the circulating hemolymph granulocytes in both strains tested (Taim and Cabo Frio). The granulocyte reduction was accompanied by a significant increase in the number of circulating dead cells. In B. tenagophila Cabo Frio, silica treatment enhanced snail susceptibility to Schistosoma mansoni, shortening the intramo…
