Effects of hypervitaminosis of vitamin B3 on silkworm biology.

A high-dose of vitamin B(3) in silkworm diet interrupts larval feeding and normal growth. High mortality of larvae occurs during molting and they cannot complete this process normally. Also the larvae exhibit nicotinamide hypervitaminosis symptoms such as immobility, dyspepsia, darkening of the skin, inability to excrete normally, exerting brownish fluid from anus and swelling of rectal muscles. Maximum larval weights in 1, 2 and 3 g/l treatments were 2.9, 1.6 and 1.2 g respectively, while maximum larval weight in the control was 5.6 g. Larval stage compared to control had increased 18, 26 and

Induction of the prophenoloxidase-activating system of Simulium (Diptera: Simuliidae) following Onchocerca (Nematoda: Filarioidea) infection.

Trials were carried out to study the humoral immune response of blackflies to filariae following infection using the intrathoracic injection technique. An induced 66 kDa protein was abundant in the haemolymph of the European species Simulium ornatum following infection with bovine Onchocerca lienalis. This protein was apparently at higher concentrations in the haemolymph of sham-inoculated flies, i.e. flies that received sterile medium without the parasites. A molecule of the same size was also observed in the haemolymph of infected S. damnosum s.l. following infection with human O. volvulus o

Evidence of spotted fever group rickettsiae in state of Rio de Janeiro, Brazil.

Ticks were obtained from dogs from February to September of 1999 at weekly intervals, in the County of Piraí, State of Rio de Janeiro. Four hundred seventy four ixodids were taxonomically identified, 103 Amblyomma cajennense, seven Amblyomma ovale, 209 Rhipicephalus sanguineus, and 155 Amblyomma sp. An hemolymph test associated with Giemsa's stain revealed two specimens in 163 ticks tested (R. sanguineus and Amblyomma sp), containing rickettsia-like organisms. Direct immunofluorescence verified the presence of spotted fever group rickettsia in one specimen of R. sanguineus. Considering the lim…

The intestinal gland of Trichinella spiralis with emphasis on morphology and antigenicity.

The intestinal gland of muscle larvae and adult worms of Trichinella spiralis was investigated with emphasis on its morphology and antigenic … ominent. Glycogen granules were absent. The gland was surrounded externally by the basal lamina and hemolymph. The cell membrane was extensively invaginated, and coated pits and vesicles were often observed. The cytoplasm was rather eosinophilic, PAS-negative, and stained red or yellow by AZAN. The cytoplasmic granules were antigenic against trichinosis ser …

Peptidomics of Acanthoscurria gomesiana spider venom reveals new toxins with potential antimicrobial activity.

UNLABELLED: Acanthoscurria gomesiana is a Brazilian spider from the Theraphosidae family inhabiting regions of Southeastern Brazil. Potent antimicrobial peptides as gomesin and acanthoscurrin have been discovered from the spider hemolymph in previous works. Spider venoms are also recognized as sources of biologically active peptides, however the venom peptidome of A. gomesiana remained unexplored to date. In this work, a MS-based workflow was applied to the investigation of the spider venom peptidome. Data-independent and data-dependent LC-MS/MS acquisitions of intact peptides and of peptides …

Self-toxicity and tolerance mechanisms of honeybee venom in honeybees.

Honeybee venom (apitoxin) is a potent mixture of biologically active peptides and enzymes, primarily evolved as a defence against insect predators - including other bees. Recent evidence suggests that honeybees also employ components of apitoxin for both external and internal defence against parasites and microbial infections. Consequently, they are predicted to exhibit a remarkable resistance to their own venom, which they frequently encounter within colonies. To investigate this phenomenon, we examined the physiological responses of honeybee workers and drones to envenomation. Individuals we

Histopathological analysis of filament formation of Nocardia farcinica in a silkworm infection model.

The silkworm Nocardia infection model has been established as a useful animal model for screening the pathogenicity of Nocardia and evaluating the therapeutic effects of antimicrobial agents against Nocardia infection. No histopathological analysis of silkworms infected with Nocardia farcinica has yet been performed. In this study, we performed histological analyses on organs of silkworms infected with N. farcinica. One day after infection with N. farcinica, the organism developed a branching filamentous form from coccid cells in the hemolymph. In addition, we evaluated effective doses (ED50) …

Toxicological effects and bioaccumulation of fullerene C60 (FC60) in the marine bivalve Ruditapes philippinarum.

Fullerene C60 (FC60), with its unique physical properties, has been used in many applications in recent decades. The increased likelihood of direct release into the environment has raised interest in understanding the biological effects of FC60 to aquatic organisms. Nowadays, only few studies have analysed FC60 effects and bioaccumulation in marine organisms following in vivo exposure. To provide new data about FC60 toxicity, Ruditapes philippinarum was selected as target species to assess potential adverse effects of the contaminant. Clams were exposed for 1, 3 and 7 days to predicted environ

Staphylococcus aureus-induced sepsis in the lobster cockroach Nauphoeta cinerea.

Invertebrates have been instrumental in understanding the mechanisms involved in infectious diseases, considering the idea to replace, reduce and refine the use of mammals as well as to understand the basic principles of immune response in insect. We evaluated the consequences of Staphylococcus aureus-induced sepsis in the last instar nymphs of Nauphoeta cinerea injected with different concentrations of bacteria preserved in two culture media. Infected groups had a decrease in hemolymph metabolites (glucose, amino acids, total proteins, and cholesterol), in contrast to the proteins in the fat …

Ecdysis of decapod crustaceans is associated with a dramatic release of crustacean cardioactive peptide into the haemolymph.

On the basis of detailed analyses of morphological characteristics and behavioural events associated with ecdysis in a crab (Carcinus maenas) and a crayfish (Orconectes limosus), a comprehensive substaging system has been introduced for the ecdysis stage of the moult cycle of these decapod crustaceans. In a remarkably similar stereotyped ecdysis sequence in both species, a passive phase of water uptake starting with bulging and rupture of thoracoabdominal exoskeletal junctions is followed by an active phase showing distinct behavioural changes involved in the shedding of the head appendages, a

A differential diagnostic criterion for Babesia major and Babesia bigemina vermicules from tick haemolymph.

Babesia major mature and immature vermicules in the haemolymph of Haemaphysalis punctata were measured and found to be significantly larger than vermicules of Babesia bigemina. Mature B. major vermicules had a mean length of 15.53 micrometer and mature B. bigemina vermicules had a mean length of 11.79 micrometer. This difference provides a new criterion for the differentiation of the two species.

Natural co-infection of Babesia caballi and Encephalitozoon-like microsporidia in the tick Anocentor nitens (Acari: Ixodidae).

The present paper reports the occurrence of natural co-infection of Babesia caballi and Encephalitozoon-like microsporidia in the tick Anocentor nitens. Engorged females of ticks, collected from a naturally B. caballi-infected horse, were incubated at 27 degrees C and relative humidity over 83%. After a 6-day incubation period, Giemsa-stained smears prepared from hemolymph were examined microscopically under oil immersion. B. caballi infected ticks were dissected and samples of midgut tissue were examined by transmission electron microscopy, through which free sporokinetes were seen in the cyt…

Fat body glycolysis defects inhibit mTOR and promote distant muscle disorganization through TNF-α/egr and ImpL2 signaling in Drosophila larvae.

The fat body in Drosophila larvae functions as a reserve tissue and participates in the regulation of organismal growth and homeostasis thro … the distant disorganization of body wall muscles and the release of muscle-specific proteins in the hemolymph. Furthermore, we showed that REPTOR activity is required for fat body atrophy downstream of glycolysis inhibition, and that the effect of fat body atrophy on muscles depends on the production of TNF-α/egr and of the insulin pathway inhibitor ImpL2.

Drosophila Heart as a Model for Cardiac Development and Diseases.

The Drosophila heart, also referred to as the dorsal vessel, pumps the insect blood, the hemolymph. The bilateral heart primordia develop from the most dorsally located mesodermal cells, migrate coordinately, and fuse to form the cardiac tube. Though much simpler, the fruit fly heart displays several developmental and functional similarities to the vertebrate heart and, as we discuss here, represents an attractive model system for dissecting mechanisms of cardiac aging and heart failure and identifying genes causing congenital heart diseases. Fast imaging technologies allow for the characteriz…
