Long-term protection in children with meningococcal C conjugate vaccination: lessons learned.

Owing to an increase in group C disease, extensive prelicensure studies have been funded by both the UK Department of Health and vaccine manufacturers. These demonstrated the safety and immunogenicity of three candidate meningococcal group C conjugate (MCC) vaccines (two conjugated to CRM(197) and one to tetanus toxoid) in the targeted age groups. Induction of immunological memory in infants and young children was also demonstrated by either a low dose of polysaccharide challenge following primary immunization with MCC or by an increase in avidity indices post-primary to pre-challenge. Immune

Babesia bovis: computer simulation of the relationship between the tick vector, parasite, and bovine host.

Components contributing to the survival of Babesia bovis in its tick vector (Boophilus microplus) and bovine host were identified and their relationship simulated with an interactive BASIC computer program. Estimates for major independent variables (host-finding success, feeding success, filial infection rate, recovery rate) were derived from published laboratory and field data. Values for dependent variables (tick burden, inoculation rate, tick and bovine infection rates) were calculated during successive tick generations until equilibrium conditions were reached. The simulation was used to p

Accumulation of Immunity in Heavy-Tailed Sexual Contact Networks Shapes Mpox Outbreak Sizes.

Many countries affected by the global outbreak of mpox in 2022 have observed a decline in cases. Our mathematical model accounting for heavy-tailed sexual partnership distributions suggests that mpox epidemics can hit the infection-derived herd immunity threshold and begin to decline, with <1% of sexually active men who have sex with men infected regardless of interventions or behavioral changes. We consistently found that many countries and US states experienced an epidemic peak, with cumulative cases of around 0.1% to 0.5% among men who have sex with men. The observed decline in cases may no

Antivirals for Coexistence with COVID-19: Brief Review for General Physicians.

In order to end the coronavirus disease 2019 (COVID-19) pandemic that has lasted for nearly two years, it is most necessary to introduce antiviral drugs specific to COVID-19 along with the establishment of herd immunity by vaccination. Candidates currently being studied include nucleoside analogues that inhibit replication, protease inhibitors, and entry blockers. Not only the virus itself, but also the host protein that the virus uses in its pathogenesis is the target of treatment. Although the severe acute respiratory syndrome coronavirus 2 will not be completely eradicated, if the use of an

A game-theoretical analysis of poliomyelitis vaccination.

Poliomyelitis is a worldwide disease that has nearly been eradicated thanks to the Global Polio Eradication Initiative. Nevertheless, the disease is currently still endemic in three countries. In this paper, we incorporate the vaccination in a two age-class model of polio dynamics. Our main objective is to see whether mandatory vaccination policy is needed or if polio could be almost eradicated by a voluntary vaccination. We perform game theoretical analysis and compare the herd immunity vaccination levels with the Nash equilibrium vaccination levels. We show that the gap between two vaccinati

Controlling Measles through Politics and Policy.

Vaccination is one of history's most successful public health interventions. Since 2000, vaccination campaigns against measles, which is highly contagious but preventable through the measles-mumps-rubella (MMR) vaccine, have reduced both the global incidence of the disease and measles deaths by 80 percent. However, progress toward measles elimination has slid backward in several previously well-protected global regions. With more communities below or at risk of falling below the 95 percent immunization rates required for herd immunity-due more and more to vaccine skepticism and declination rat

[Age for HPV vaccination: recommandations from the French high council of public health].

The French high council of public health has recently amended age for HPV vaccination. The paper will describe reasons that have contributed to the development of these new guidelines. Parental and adolescent acceptance of HPV vaccination is reported as well as potential barriers among medical practices. First results of immunogenicity of vaccines using alternative dosing schedules are presented. Finally, preliminary data on clinical efficacy of the quadrivalent vaccine on genital warts and cervical precancerous lesions are presented. A decrease in the prevalence of infection with genotypes in

Optimising the use of conjugate vaccines to prevent disease caused by Haemophilus influenzae type b, Neisseria meningitidis and Streptococcus pneumoniae.

Conjugate vaccines exist that offer protection against disease caused by Haemophilus influenzae type b (Hib), and selected serogroups/serotypes of Neisseria meningitidis and Streptococcus pneumoniae. These vaccines are not only able to prevent serious disease, but they also provide protection against asymptomatic carriage. The resulting herd immunity effects have been striking, and have played an important role in the public health success of conjugate vaccination programmes. The aim of this paper is to review the state of the current evidence on conjugate vaccines and to identify important ar

Pneumococcal vaccine development.

A 7-valent pneumococcal conjugate vaccine has demonstrated an impact on pneumococcal bacteremia in the immunized pediatric population, extending to nonimmunized adults via herd immunity. A considerable reduction of all-cause pediatric pneumonia has also been found. The impact on all-cause pediatric otitis is limited, but postlicensure data suggests stronger reductions. Higher valency conjugate vaccines are now under development (11V or more). Licensed 23-valent pneumococcal polysaccharide vaccines have been available since 1983 with a demonstrated impact on adult pneumococcal bacteremia. The b

Epidemiological impact and cost-effectiveness of universal vaccination with Bexsero(®) to reduce meningococcal group B disease in Germany.

Bexsero, a new vaccine against serogroup B meningococcal disease (MenB), was licensed in Europe in January 2013. In Germany, Bexsero is recommended for persons at increased risk of invasive meningococcal disease, but not for universal childhood vaccination. To support decision making we adapted the independently developed model for England to the German setting to predict the potential health impact and cost-effectiveness of universal vaccination with Bexsero(®) against MenB disease. We used both cohort and transmission dynamic mathematical models, the latter allowing for herd effects, to cons

Evaluation of serological assays for intra vitam diagnosis of bovine tuberculosis in water buffalo (Bubalus bubalis).

INTRODUCTION: Bovine tuberculosis (TB) in water buffalo (Bubalus bubalis) is primarily diagnosed using intra vitam tests, as the intradermal tuberculin test (IDT) and the interferon-gamma release assay (IGRA), both of which detect cell-mediated immunity (CMI). However, a subset of infected animals fails to mount a detectable CMI response, posing a significant risk of undetected transmission. Serological tests assessing the humoral immune response could provide a valuable complementary tool for identifying infected animals that escape detection through traditional CMI-based assays. METHODS: Th

Productivity and health effects of anaplasmosis and babesiosis on Bos indicus cattle and their crosses, and the effects of differing intensity of tick control in Australia.

Tick fever is an important disease of cattle where Rhipicephalus (Boophilus) microplus acts as a vector for the three causal organisms Babesia bovis, Babesia bigemina and Anaplasma marginale. Bos indicus cattle and their crosses are more resistant to the clinical effects of infection with B. bovis and B. bigemina than are Bos taurus cattle. Resistance is not complete, however, and herds of B. indicus-cross cattle are still at risk of babesiosis in environments where exposure to B. bovis is light in most years but occasionally high. The susceptibility of B. indicus cattle and their crosses to i

Controlling bovine leukemia virus in dairy herds by identifying and removing cows with the highest proviral load and lymphocyte counts.

The objective of this field trial was to reduce bovine leukemia virus (BLV) transmission and prevalence in commercial dairy herds using proviral load (PVL) and lymphocyte count (LC) measurements as indicators of the most infectious animals for culling or segregation. Bovine leukemia virus causes lymphoma in <5% of infected cattle, and increased lymphocyte counts (lymphocytosis) in about one-third. Recent research has shown that dairy cows infected with BLV have altered immune function associated with decreases in milk production and lifespan. Recent findings show that a minority of infected ca

Progression of Mycoplasma hyosynoviae infection in three pig herds. Development of tonsillar carrier state, arthritis and antibodies in serum and synovial fluid in pigs from birth to slaughter.

In this investigation, natural infection with Mycoplasma hyosynoviae was followed in groups of individual pigs in three different herds with regard to occurrence of tonsillar carrier state, clinical arthritis and development of antibodies in serum and in synovial fluid. Antibodies were detected by a polyclonal enzyme-linked immunosorbent assay (ELISA) developed for experimental use. The infection with M. hyosynoviae progressed very differently in the three herds investigated. In one herd, the infection was apparently limited to adult pigs. In a second herd, all pigs became tonsillar carriers o

Enhancing immune resilience in swine: a comprehensive meta-review of preventive and host-directed strategies against African swine fever and swine influenza.

African swine fever (ASF) and swine influenza (SI) are among the most consequential infectious threats to global swine production because they combine major economic burden with fundamentally different transmission ecology, tissue tropism, and immune-pathological behavior. ASF is a highly lethal systemic disease characterized by macrophage tropism, immune dysregulation, environmental persistence, and severe trade and production consequences, whereas SI is a highly transmissible respiratory infection associated with recurrent herd-level morbidity, viral evolution, and zoonotic significance. IgM rheumatoid factor in cattle immunized against babesiosis.

Polyclonal bovine IgM-rheumatoid factors (IgM-RFs) were examined in sera of cattle immunized against babesiosis. An enzyme-linked immunosorbent assay (ELISA) was used enabling rapid screening of serum samples. Results obtained indicate a rise of serum IgM-RF levels with age in healthy bovines. However when animals of similar age and pertaining to the same herd were examined, levels of serum IgM-RF exhibited a wide distribution range. Mean values in 60 sera of 2 yrs old clinically healthy heifers originating from a single herd were of 452.60 +/- 201.26 e.u. at 1 in 1,000 serum dilution and of 2

Ebola could be eradicated through voluntary vaccination.

Ebola virus disease (EVD) is a severe infection with an extremely high fatality rate spread through direct contact with body fluids. A promising Ebola vaccine (rVSV-ZEBOV) may soon become universally available. We constructed a game-theoretic model of Ebola incorporating individual decisions to vaccinate. We found that if a population adopts selfishly optimal vaccination strategies, then the population vaccination coverage falls negligibly short of the herd immunity level. We concluded that eradication of Ebola is feasible if voluntary vaccination programmes are coupled with focused public edu

[Laboratory-confirmed cases of hemorrhagic fever with renal syndrome which occurred in Breclav 1989-1990].

The authors describe the first three cases with a serologically confirmed hantaviral aetiology in the Czech Republic. It is the causal agent of haemorrhagic fever with renal syndrome, western type (strain CG 18-20, identical with the Puumala prototype). All patients come from a locality where since 1984 very actively a natural focus of hantaviral infection was investigated in small mammals, and where formerly groups of farmers were found with a high herd immunity with hantavirus. The disease was present in a mild to medium severe form with some sequelae. In the discussion the authors draw atte

Analysis of management factors influencing Anaplasma marginale transmission.

The aim of this study was to identify factors that play a key role in the epidemiology of bovine anaplasmosis by adapting a model primarily developed for cattle babesiosis. A cross-sectional observational study was conducted to study the proportion of calf herds in endemic stability/instability for A. marginale in a semi-arid area of Argentina. The A. marginale inoculation rate (h) was calculated from age-specific seroprevalence using double-antigen sandwich ELISA in 58 herds of 4.5-8.5-month-old calves. Herds were considered to be in endemic instability (EI) at h < 0.005 and, therefore, at ri

Tick-borne diseases in subtropical dairy farming: The role of calf rearing strategies in antibody and antigen prevalence.

Tick-borne diseases (TBDs), such as anaplasmosis and babesiosis, pose significant challenges to dairy farming in subtropical regions, where balancing tick control with immune development is essential to maintaining herd health. This study evaluated the immune responses of calves raised under confined and traditional systems in Ecuador. Serum samples from 124 cows and their calves were analysed using ELISA assays to detect Anaplasma marginale antibodies and Babesia bovis antigens. The results showed a significantly higher seroprevalence of A. marginale antibodies in calves reared under traditio
