Radioimmunotherapy of solid tumors by targeting extra domain B fibronectin: identification of the best-suited radioimmunoconjugate.

PURPOSE: The expression of extra domain B (ED-B) fibronectin is always associated with angiogenic processes and can be exclusively observed in tissues undergoing growth and/or extensive remodeling. Due to this selective expression, ED-B fibronectin is an interesting target for radioimmunotherapy of malignant diseases. The aim of this study was to identify the most appropriate ED-B-targeting radioimmunoconjugate for the therapy of solid tumors. EXPERIMENTAL DESIGN: Three ED-B fibronectin-binding human antibody formats of L19 were investigated: dimeric single-chain Fv (approximately 50 kDa), "s

Identification of a monoclonal antibody, TV-1, directed against the basement membrane of tumor vessels, and its use to enhance the delivery of macromolecules to tumors after conjugation with interleukin 2.

mAbs reactive with epitopes expressed on tumor vessels were evaluated as universal delivery agents of peptides with vasoactive properties to enhance the uptake of macromolecules in tumors. Unlike other reported approaches to target tumor vessels, a mAb designated TV-1 targets a basement membrane antigen that is found in all tissues but that is accessible only in tumor vessels, making it an alternative vehicle for the delivery of biologically active peptides to tumors. A panel of 30 monoclonal and polyclonal antibodies was screened by immunohistochemistry on sections of human tumors, normal vas

The biology of MDR1-P-glycoprotein (MDR1-Pgp) in designing functional antibody drug conjugates (ADCs): the experience of gemtuzumab ozogamicin.

BACKGROUND: The treatment of cancer remains a formidable challenge owing to the difficulties in differentiating tumor cells from healthy cells to ameliorate the disease without causing intolerable toxicity to patients. In addition, the emergence of MDR1-Pgp mediated multi-drug resistance (MDR) it is a biological phenomenon that inhibits the curative potential of chemotherapeutic treatments. One way to improve the selectivity of therapeutic molecules in tumors would be to target them on the tumor site, thereby sparing normal tissues. AIMS: In this overview, we will discuss the biological facto

Labeling anti-HER2/neu monoclonal antibodies with 111In and 90Y using a bifunctional DTPA chelating agent.

The goal of this investigation was to develop stable radioimmunoconjugates (RICs) of anti-HER2/neu monoclonal antibodies (MoAbs) for imaging and therapy in an animal model bearing human breast tumor xenografts that express normal (MCF-7 cells) and increased amounts of HER2/neu receptors (HCC-1954, BT-474, SKBR-3 cells) on their cell surface membranes. Pharmacy-grade Herceptin, a murine anti-HER2/neu MoAb, and nonspecific mouse IgG protein were conjugated with the recently developed DTPA linker known as CHX-A"-DTPA. These immunoconjugates were labeled with (111)InCl(3) and (90)YCl(3). Using a m

Prophylactic vaccination protects against the development of oxycodone self-administration.

Abuse of prescription opioids is a growing public health crisis in the United States, with drug overdose deaths increasing dramatically over the past 15 years. Few preclinical studies exist on the reinforcing effects of oxycodone or on the development of therapies for oxycodone abuse. This study was conducted to determine if immunopharmacotherapy directed against oxycodone would be capable of altering oxycodone-induced antinociception and intravenous self-administration. Male Wistar rats were administered a small-molecule immunoconjugate vaccine (Oxy-TT) or the control carrier protein, tetanus

Advances in Payload and Linker Designs for ADCs.

Antibody-drug conjugates (ADCs) are a therapeutic approach that embodies the "magic bullet" concept envisioned by Paul Ehrlich. Since their initial clinical potential was recognized in the early 1980s, ADCs have undergone several generations of development to enhance their therapeutic index (Poon KA, et. al. 273(2):298-313, 2013). Recent advancements, particularly in the last third and the beginning of the fourth generation, have primarily focused on discovering new drug modalities and more potent cancer treatments, while also developing new linkers and improving site-specific and conjugation

Recent advances in Camptothecin-derived antibody-drug conjugates.

Antibody-drug conjugates (ADCs) represent a major advance in precision cancer therapy by combining the high target specificity of monoclonal antibodies with the potent cytotoxicity of small-molecule payloads through chemical linkers. Among the payload classes topoisomerase I (Topo I) inhibitors, camptothecin and its derivatives have emerged as highly promising warheads owing to their distinct mechanism of action and strong antitumor activity. The clinical success of Enhertu and Trodelvy, which employ the camptothecin-derived payloads DXd and SN-38, respectively, has validated the therapeutic p

Trial watch: antibody-drug conjugates in cancer therapy.

Antibody-drug conjugates (ADCs) combine the target specificity of monoclonal antibodies with the cytotoxic potency of small-molecule payloads. In recent years ADCs have emerged as a clinically validated component of modern precision oncology. To date, more than a dozen ADCs have received FDA approval for oncologic indications, with additional agents approved regionally and hundreds of ADC-based regimens in clinical development. Collectively, these therapies have demonstrated clinical benefit across hematologic malignancies and solid tumors, including significant overall survival improvements i

Trop2-Based Antibody-Drug Conjugates: Emerging Strategy and Progress in Triple-Negative Breast Cancer Therapy.

Triple-negative breast cancer (TNBC) accounts for 15-20% of invasive breast cancers and represents a highly heterogeneous, aggressive subtype with poor prognosis and limited treatment options, necessitating the identification of novel therapeutic targets to improve clinical outcomes. Trophoblast cell-surface antigen 2 (Trop2), a calcium signal transducer, is frequently overexpressed in TNBC (approximately 88% of cases) while exhibiting minimal expression in normal tissues. Its overexpression is significantly associated with tumor invasion, metastasis, and unfavorable prognosis. Antibody-drug c

A Cathepsin B-Cleavable Self-Immolative Linker for the Delivery of Hydroxyl-Containing Payloads in Antibody-Drug Conjugates.

In development of antibody-drug conjugates (ADCs), cathepsin B-cleavable linkers have been extensively optimized for amine-containing payloads and successfully translated into clinic practice, whereas effective solutions for hydroxyl-containing payloads remain limited. Here, we report a cathepsin B-cleavable VA-PAB-2-HPCCONH self-immolative linker platform featuring three orthogonal attachment sites that enable modulation of linker-payload hydrophilicity. Using this platform, we designed and synthesized highly water-soluble linker-payload conjugates, incorporating either phenolic hydroxyl payl

Beyond Small Molecules: Applying Fragment Molecular Orbital Sygnature Platform (FMO-SP) to Emerging Modalities.

Drug discovery is rapidly expanding beyond traditional small molecules to encompass new therapeutic modalities capable of addressing previously "undruggable" targets. Between 2015 and mid-2025, FDA approvals increasingly included biologics and emerging therapeutic modalities such as antibody-drug conjugates (ADCs), nucleic acid therapeutics, and other nontraditional approaches. In parallel, PROTACs and molecular glue degraders (MGDs) have expanded rapidly in clinical development. These innovations are reshaping therapeutic strategies in oncology, metabolic disease, and rare disorders, but they

Beyond immune checkpoint blockade: T cell engagers and antibody-drug conjugates in cancer and autoimmune disease.

Immune checkpoint inhibitors have transformed cancer treatment; however, resistance, on-target toxicity, and limited efficacy in autoimmune disorders have motivated next-generation immunotherapies. T cell engagers (TCEs) redirect cytotoxic T cells to kill pathogenic cells independently of MHC restriction, whereas antibody-drug conjugates (ADCs) deliver potent payloads directly into target cells via receptor-mediated internalization. This review synthesizes preclinical and clinical data on TCEs and ADCs in oncology and immune-mediated inflammatory disorders (IMIDs). The CD47/SIRPα innate immune

Diagnosis and treatment of cutaneous adverse effects of targeted therapy, antibody-drug conjugates, and immunotherapy in cancer patients: a national consensus statement by the Spanish Society of Medical Oncology and the Spanish Academy of Dermatology and Venereology.

Cancer remains a leading cause of mortality in developed countries, with advancements in targeted therapies, antibody-drug conjugates (ADCs), and immunotherapy significantly improving patient survival. However, these treatments often result in cutaneous adverse effects (AEs), impacting the skin, mucosa, hair, and nails. This expert consensus statement provides a comprehensive overview of the mechanisms behind these skin toxicities, their clinical manifestations, and the importance of rapid diagnosis and treatment. The spectrum of skin toxicities ranges from mild reactions such as xerosis and p

Beyond approval: a mechanism-based review of novel anti-tumour agents in cervical cancer and when to use them.

Cervical cancer remains a leading cause of cancer death in women, and outcomes in recurrent or metastatic disease remain poor despite chemoradiation and bevacizumab. The HPV-driven biology that defines the disease-viral E6/E7 oncoproteins, an inflamed microenvironment and exploitable surface antigens-has enabled a rapid expansion of novel systemic therapies, yet existing reviews tend to catalogue these agents by class or report individual trials in isolation, leaving their comparative efficacy, safety and clinical positioning unresolved. Here we organise emerging anti-tumour agents by mechanis

Antibody-conjugated polymer nanoparticles for brain cancer.

Brain cancer remains a significant global health challenge, affecting over a million individuals worldwide. It includes primary tumors such as gliomas, particularly glioblastoma multiforme (GBM) being one of the most aggressive forms, and secondary brain tumors resulting from metastases of breast or lung cancer. A major obstacle in treating these malignancies is the blood-brain barrier (BBB), which limits effective drug delivery to tumor sites. Nanoparticles have long been explored as versatile drug delivery vehicles due to their potential to improve tumor specificity, penetrate the BBB, and e

Management of ocular toxicity in patients with gynecologic cancer receiving novel antibody-drug conjugates: a narrative review.

Antibody-drug conjugates (ADCs) represent a promising therapeutic approach in gynecologic cancers, particularly ovarian and cervical malignancies. Agents such as mirvetuximab soravtansine, and tisotumab vedotin, targeting folate receptor alpha and tissue factor, respectively, reported clinical efficacy in patients with limited options. However, their use is associated with ocular toxicities, including keratopathy, blurred vision, and dry eye, which may impact adherence and quality of life. This review summarizes current evidence on the incidence, pathophysiology, and clinical presentation of A

Antibody-drug conjugates for hepato-pancreato-biliary malignancies: "Magic bullets" to the rescue?

Hepato-Pancreato-Biliary (HPB) malignancies constitute a highly aggressive group of cancers that have a dismal prognosis. Patients not amenable to curative intent surgical resection are managed with systemic chemotherapy which, however, confers little survival benefit. Antibody-Drug Conjugates (ADCs) are tripartite compounds that merge the intricate selectivity and specificity of monoclonal antibodies with the cytodestructive potency of attached supertoxic payloads. In view of the unmet need for drugs that will enhance the survival rates of HPB cancer patients, the assessment of ADCs for treat
