Spike protein derived from an apathogenic IBV strain confers attenuated phenotype to a nephropathogenic IBV strain.

Infectious bronchitis virus (IBV), a Gammacoronavirus, causes the economically damaging disease of poultry, infectious bronchitis. Development of IBV live attenuated vaccines (LAVs) is dependent on the supply of embryonated hens' eggs and attenuation through serial passaging, upwards of 80 times. Vaccine development needs to keep pace with IBV evolution; an issue compounded as cross protection between individual IBV serotypes is unpredictable and often limited. Rational attenuation alongside the ability to propagate LAVs in cell culture offers significant advantages in terms of adaptability, c

Three Amino Acid Changes in Avian Coronavirus Spike Protein Allow Binding to Kidney Tissue.

Infectious bronchitis virus (IBV) infects ciliated epithelial cells in the chicken respiratory tract. While some IBV strains replicate locally, others can disseminate to various organs, including the kidney. Here, we elucidate the determinants for kidney tropism by studying interactions between the receptor-binding domain (RBD) of the viral attachment protein spike from two IBV strains with different tropisms. Recombinantly produced RBDs from the nephropathogenic IBV strain QX and from the nonnephropathogenic strain M41 bound to the epithelial cells of the trachea. In contrast, only QX-RBD bin

Seroepidemiologic Survey of Potential Pathogens in Obligate and Facultative Scavenging Avian Species in California.

Throughout the world, populations of scavenger birds are declining rapidly with some populations already on the brink of extinction. Much of the current research into the factors contributing to these declines has focused on exposure to drug residues, lead, and other toxins. Despite increased monitoring of these declining populations, little is known about infectious diseases affecting scavenger bird species. To assess potential infectious disease risks to both obligate and facultative scavenger bird species, we performed a serosurvey for eleven potential pathogens in three species of scavengi

Antiviral activity of a novel mixture of natural antimicrobials, in vitro, and in a chicken infection model in vivo.

The aim of this study was to test in vitro the ability of a mixture of citrus extract, maltodextrin, sodium chloride, lactic acid and citric acid (AuraShield L) to inhibit the virulence of infectious bronchitis, Newcastle disease, avian influenza, porcine reproductive and respiratory syndrome (PRRS) and bovine coronavirus viruses. Secondly, in vivo, we have investigated its efficacy against infectious bronchitis using a broiler infection model. In vitro, these antimicrobials had expressed antiviral activity against all five viruses through all phases of the infection process of the host cells.

Immune Responses to Virulent and Vaccine Strains of Infectious Bronchitis Viruses in Chickens.

Infectious bronchitis (IB) is an acute and highly contagious chicken viral disease, causing severe economic losses to poultry producers worldwide. In the last few decades, infectious bronchitis virus (IBV) has been extensively studied, but knowledge of immune responses to virulent or vaccine strains of IBVs remains limited. This review focuses on fundamental aspects of immune responses against IBV, including the role of pattern recognition receptors (PRRs) in identification of conserved viral structures and the role of different components of innate immunity (e.g., heterophils, macrophages, de

Immunity to avian infectious bronchitis.

Avian infectious bronchitis is recognized clinically as a respiratory disease in its only natural host, the chicken, but the virus is disseminated throughout other systems by a viraemia with localization especially in the kidney and oviduct. The sensitivity or instability of the antigenic and immunogenic properties of the virus under laboratory stress or natural influences complicates the selection of seed virus for vaccines. Modified or attenuated active virus vaccines induce greater protection against subsequent infection than do inactive virus vaccines. Maternal antibody is effective in prov

First Report and Molecular Confirmation of Chicken Proventricular Necrosis Virus Associated with Transmissible Viral Proventriculitis in Bangladesh.

Transmissible viral proventriculitis (TVP) is an emerging disease in chickens, linked to chicken proventricular necrosis virus (CPNV), a recently identified birnavirus. Here, we provide the first molecular confirmation of TVP in Bangladesh from a coloured meat-type parent stock (PS) flock, while documenting a contemporaneous white layer flock with consistent clinical signs and characteristic gross lesions. Affected birds exhibited growth retardation, diarrhoea, and increased mortality, alongside hallmark gross changes in proventricular enlargement and wall thickening. From the meat-type PS, pr

Avian infectious bronchitis and deep pectoral myopathy - a case control study.

Infectious bronchitis is caused by a coronavirus, infectious bronchitis virus (IBV). Infectious bronchitis is an acute and highly contagious disease of economic importance due to the reduction in weight gain observed with infected broilers and the drop in egg quality and production associated with infected laying hens. The presence of deep pectoral myopathy has been associated with IBV variants. This lesion is detected at slaughterhouses and is characterized by paleness and atrophy of the deep pectoral muscle, including necrosis of the region, leading to condemnations of the breast muscle, a v

Antiviral effects of flavonoids on animal viruses.

Flavonoids, plant-derived compounds widely recognized for their antioxidant, anti-inflammatory, and anticancer properties, also exhibit significant antiviral effects against various animal viruses. This review highlights the promising antiviral mechanisms of flavonoids, which include disrupting viral replication, blocking cell entry, and modulating immune responses. Notably, flavonoids like quercetin, kaempferol, and genistein have been shown to effectively inhibit viruses of veterinary importance, such as African Swine Fever Virus (ASFV), Porcine Epidemic Diarrhea Virus (PEDV), and Infectious

Insights into antiviral activity of chlorpromazine against RNA viruses: Molecular docking, ADME profile, and semi-in vivo study.

Viral entry into the host cell is a limiting step in the viral-related pathogenesis, where many viruses utilize different endocytic pathways, particularly clathrin-mediated endocytosis (CME), in cell invasion. Previously, we reclassified endocytosis inhibitors based on their mode of action, where compounds like phenothiazine derivatives inhibit viral endocytosis through different mechanisms. Also, the multifaceted therapeutic potential of these derivatives, like chlorpromazine (CPZ), is attributed to their endocytosis and non-endocytosis-related effects. Thus, this study was designed to invest

Impact on human health of climate changes.

There is increasing evidence that climate is rapidly changing. These changes, which are mainly driven by the dramatic increase of greenhouse gas emissions from anthropogenic activities, have the potential to affect human health in several ways. These include a global rise in average temperature, an increased frequency of heat waves, of weather events such as hurricanes, cyclones and drought periods, plus an altered distribution of allergens and vector-borne infectious diseases. The cardiopulmonary system and the gastrointestinal tract are particularly vulnerable to the adverse effects of globa

Negatively charged residues in the endodomain are critical for specific assembly of spike protein into murine coronavirus.

Coronavirus spike (S) protein assembles into virions via its carboxy-terminus, which is composed of a transmembrane domain and an endodomain. Here, the carboxy-terminal charge-rich motif in the endodomain was verified to be critical for the specificity of S assembly into mouse hepatitis virus (MHV). Recombinant MHVs exhibited a range of abilities to accommodate the homologous S endodomains from the betacoronaviruses bovine coronavirus and human SARS-associated coronavirus, the alphacoronavirus porcine transmissible gastroenteritis virus (TGEV), and the gammacoronavirus avian infectious bronchi

Baicalin Protects Broilers against Avian Coronavirus Infection via Regulating Respiratory Tract Microbiota and Amino Acid Metabolism.

An increasing amount of evidence indicates that Baicalin (Bai, a natural glycosyloxyflavone compound) exhibits an antiviral effect against avian viruses. However, it remains unclear if the antiviral effect of Bai against infectious bronchitis virus (IBV) is exerted indirectly by modulating respiratory tract microbiota and/or their metabolites. In this study, we investigated the protection efficacy of Bai in protecting cell cultures and broilers from IBV infection and assessed modulation of respiratory tract microbiota and metabolites during infection. Bai was administered orally to broilers by

Development and validation of a sensitive fluorescence RT-qPCR assay with TaqMan-MGB probe for detection of H6 subtype avian influenza A virus.

Avian influenza, caused by the avian influenza A virus (IAV), threatens poultry and public health. H6 subtype avian IAV is a low-pathogenic virus with hosts ranging from poultry and wild birds to mammals. H6 persists latently in poultry, which enables silent transmission and cross-species risk. The few fluorescence quantification assays that exist for H6 are mostly multiplexed. We developed a rapid, sensitive, efficient, monoplex fluorescence reverse-transcription qPCR (RT-qPCR) assay for H6 IAV. Specific primers and a TaqMan-MGB probe were designed based on the conserved hemagglutinin (HA) ge

[EBV diseases].

Epstein-Barr virus (EBV) infection in early childhood is usually asymptomatic or associated with symptoms common to many other viral infections, such as pharyngitis, bronchitis or enteritis. Typical infectious mononucleosis, as seen in young adult patients, is rare in early childhood. There is also a lower rate of heterophil antibody responses in young children. Many complications of the infection are known, and many atypical manifestations can be identified as EBV-associated using modern EBV-specific serological methods. The problems of EBV infection during gestation and the neonatal period a

Rapid and sensitive detection of H3 AIV HA1 protein using a quantum dot-labeled immunochromatographic strip.

Avian influenza virus (AIV) subtype H3 has evolved into a major zoonotic pathogen and poses a potential threat to public health. Hemagglutinin 1 region (HA1) proteins constitute the globular head region of hemagglutinin proteins, contain receptor-binding domains and esterase structural domains, and are an important molecular basis for antigenic variation in AIV. Therefore, a rapid and highly sensitive assay for the HA1 protein of this viral subtype is essential for effective epidemic control. Using quantum dots (QDs) as the core material, a fast-response fluorescent immunochromatographic test
