The FoxO-BNIP3 axis exerts a unique regulation of mTORC1 and cell survival under energy stress.

Normal cells possess adaptive mechanisms to couple energy availability with cell growth (cell size increase) and survival, and imbalances are associated with major diseases such as cancer. Inactivation of critical regulators involved in energy stress response, including adenosine monophosphate-activated protein kinase (AMPK), liver kinase B1 (LKB1), tuberous sclerosis complex 1 (TSC1) and tuberous sclerosis complex 2 (TSC2), leads to uncontrolled cell growth yet increased apoptosis under energy stress. These energy stress regulators are also important in tumor suppression and metabolism. Here,

GADD34 Function in Protein Trafficking Promotes Adaptation to Hyperosmotic Stress in Human Corneal Cells.

GADD34, a stress-induced regulatory subunit of the phosphatase PP1, is known to function in hyperosmotic stress through its well-known role in the integrated stress response (ISR) pathway. Adaptation to hyperosmotic stress is important for the health of corneal epithelial cells exposed to changes in extracellular osmolarity, with maladaptation leading to dry eye syndrome. This adaptation includes induction of SNAT2, an endoplasmic reticulum (ER)-Golgi-processed protein, which helps to reverse the stress-induced loss of cell volume and promote homeostasis through amino acid uptake. Here, we sho…

Monitoring Virus-Induced Stress Granule Dynamics Using Long-Term Live-Cell Imaging.

The integrated stress response is a highly regulated signaling cascade that allows cells to react to a variety of external and internal stimuli. Activation of different stress-responsive kinases leads to the phosphorylation of their common downstream target, the eukaryotic translation initiation factor 2 alpha (eIF2α), which is a critical component of functional translation preinitiation complexes. As a consequence, stalled ribonucleoprotein complexes accumulate in the cytoplasm and condense into microscopically visible cytoplasmic stress granules (SGs). Over the past years, numerous microscop…

Stress-Induced Molecular Sieving and Anisotropic Diffusion of CH4/CO2 in Deformed Nanoporous Coal Matrix.

Understanding the coupling between mechanical deformation and interfacial gas dynamics in disordered nanoporous media is fundamental to subsurface energy applications. To decouple these effects, this study integrated Molecular Dynamics (MD) and Grand Canonical Monte Carlo (GCMC) simulations to investigate the response of a coal matrix under three distinct stress paths (lateral pressure coefficients K = 0.5, 1.0, and 1.5). Structural analysis identified a critical "stress-induced molecular sieving" mechanism: high confinement stress triggers a structural reorganization, disproportionately colla

Maternal immune activation in mice disrupts proteostasis in the fetal brain.

Maternal infection and inflammation during pregnancy are associated with neurodevelopmental disorders in offspring, but little is understood about the molecular mechanisms underlying this epidemiologic phenomenon. Here, we leveraged single-cell RNA sequencing to profile transcriptional changes in the mouse fetal brain in response to maternal immune activation (MIA) and identified perturbations in cellular pathways associated with mRNA translation, ribosome biogenesis and stress signaling. We found that MIA activates the integrated stress response (ISR) in male, but not female, MIA offspring in…

Phyto-Oxylipins as Key Regulators of Plant Resilience to Abiotic and Biotic Stress.

Phyto-oxylipins, oxidized derivatives of unsaturated fatty acids, serve as crucial mediators in plant responses to biotic and abiotic stresses, which are becoming increasingly frequent and severe under changing climatic conditions. These signaling molecules, produced enzymatically or spontaneously, orchestrate a wide range of physiological and molecular responses that enhance plant resilience. By scavenging reactive oxygen species (ROS) and upregulating antioxidant enzymes, phyto-oxylipins help mitigate oxidative damage, thus preserving cellular integrity and sustaining growth under environmen

The regulation of Xrp1 expression by uORFs and main ORF sequences and its function in Drosophila disease models.

The Integrated Stress Response (ISR) mediates cellular adaptation to endoplasmic reticulum (ER) stress, amino acid deprivation, and mitochondrial dysfunction. The ISR regulates gene expression in part by preferentially translating the transcription factor ATF4, a process regulated by upstream open reading frames (uORFs) in its 5' leader. In Drosophila, Xrp1 is another transcription factor induced during the ISR, but the precise underlying mechanism remains unclear. Here, we report that Xrp1 induction in response to ER stress is regulated by both its uORFs and the main ORF sequence. Xrp1 has se…

Beyond hypoxia: Context-dependent transcriptional reprogramming by ERFVIIs.

This Commentary highlights the finding that an increased post-submergence oxidative burst reprograms plant cysteine oxidase (PCO)-group VII ethylene response factor (ERFVII) oxygen sensing, inhibiting PCO activity and transcriptionally rewiring ERFVIIs toward oxidative stress recovery. This redefines ERFVIIs as redox-responsive stress integrators and reveals targets to enhance climate-adaptive stress responses.

Transcriptomic and Physiological Responses Reveal a Time-Associated Multi-Organ Injury Pattern in European Perch (Perca fluviatilis) Under Acute Alkaline Stress.

Water alkalinization is a critical global stressor for freshwater fish, yet the systemic patterns of multi-organ responses and injury remain insufficiently understood. This study integrates histopathology, biochemistry, and multi-organ transcriptomics to provide an integrated, time-resolved assessment of stress responses in European perch (Perca fluviatilis) exposed to acute alkaline stress (20 mmol/L). The analysis indicated that alkaline stress initially causes structural disturbance of gill tissue (lamellar fusion, necrosis) within 96 h, associated with impaired osmoregulatory functions. Th

Pectin under stress: Pectin methylesterases at the core of cell wall remodeling, integrity sensing, and signaling in plants.

Plant cell walls are dynamic signaling platforms that integrate mechanical, biochemical, and immune cues during development and environmental stress responses. Among wall polysaccharides, pectin has emerged as a central regulator of cell wall integrity (CWI) and stress-responsive signaling. Here, we highlight pectin methylesterases (PMEs) and their regulators as a coordinated modulatory system that enables spatiotemporal control of pectin remodeling during biotic and abiotic stress responses. We discuss recent discoveries regarding isoform-specific PME functions, proteolytic activation, and tr

Conformational diversity of collided ribosomes determines stress signaling.

Ribosome collisions trigger pathways that clear stalled ribosomes, and when sufficiently abundant, the integrated stress response (ISR) through GCN2 and the ribotoxic stress response (RSR) through ZAK. The inhibitors anisomycin (ANS), emetine (EME), and didemnin B (DDB) are commonly used to induce collisions in studying these responses. Here, we demonstrate that these drugs induce distinct signatures: ANS and DDB potently activate ZAK whereas EME does not. We define transcriptional programs induced by these inhibitors, where collisions induce the RSR and general inhibition of translation leads…

Pathway connectivity and signaling coordination in the yeast stress-activated signaling network.

Stressed cells coordinate a multi-faceted response spanning many levels of physiology. Yet knowledge of the complete stress-activated regulatory network as well as design principles for signal integration remains incomplete. We developed an experimental and computational approach to integrate available protein interaction data with gene fitness contributions, mutant transcriptome profiles, and phospho-proteome changes in cells responding to salt stress, to infer the salt-responsive signaling network in yeast. The inferred subnetwork presented many novel predictions by implicating new regulator

Stress response mechanisms in protein misfolding diseases: Profiling a cellular model of Huntington's disease.

Stress response pathways like the integrated stress response (ISR), the mitochondrial unfolded protein response (UPRmt) and the heat shock response (HSR) have emerged as part of the pathophysiology of neurodegenerative diseases, including Huntington's disease (HD) - a currently incurable disease caused by the production of mutant huntingtin (mut-Htt). Previous data from HD patients suggest that ISR is activated while UPRmt and HSR are impaired in HD. The study of these stress response pathways as potential therapeutic targets in HD requires cellular models that mimic the activation status foun…

Integrated toxicological assessment of environmental microplastics and diuron on Mytilus galloprovincialis Larvae: Toward improved marine risk management.

Coastal marine ecosystems are increasingly threatened by complex mixtures of emerging and persistent pollutants, including environmental microplastics (EMPs) and the herbicide diuron. These contaminants, frequently detected in coastal waters, can exert harmful effects even at low, environmentally relevant concentrations, particularly during sensitive early life stages of marine invertebrates. Here, we conducted the first investigation of the combined toxicity of EMPs and diuron at realistic exposure levels (100 μg/L EMPs, 50 ng/L diuron, and their mixture), reflecting concentrations commonly f

A three-stage assembly program governing pancreatic, plasma, pituitary, and bone secretory cell differentiation: A strategy to augment protein delivery.

Defective secretory cell function underlies many diseases, and recent therapeutic strategies have focused on enhancing protein synthesis and delivery by targeting the secretory machinery of mature cells. However, mature differentiated cells appear to have intrinsic limits to their secretory capacity. In this review, we propose new strategies for engineering these cells to overcome these limits on secretion. The integrated stress response (ISR) and the related unfolded protein response (UPR) are stress adaptation systems that modulate transcriptional and translational programs of gene expressio…
