Limited Effects of Type I Interferons on Kyasanur Forest Disease Virus in Cell Culture.

BACKGROUND: The tick-borne flavivirus, Kyasanur Forest disease virus (KFDV) causes seasonal infections and periodic outbreaks in south-west India. The current vaccine offers poor protection with reported issues of coverage and immunogenicity. Since there are no approved prophylactic therapeutics for KFDV, type I IFN-α/β subtypes were assessed for antiviral potency against KFDV in cell culture. METHODOLOGY/PRINCIPAL FINDINGS: The continued passage of KFDV-infected cells with re-administered IFN-α2a treatment did not eliminate KFDV and had little effect on infectious particle production whereas

Internalization and TLR-dependent type I interferon production by monocytes in response to Toxoplasma gondii.

The classic anti-viral cytokine interferon (IFN)-β can be induced during parasitic infection, but relatively little is know about the cell types and signaling pathways involved. Here we show that inflammatory monocytes (IMs), but not neutrophils, produce IFN-β in response to T. gondii infection. This difference correlated with the mode of parasite entry into host cells, with phagocytic uptake predominating in IMs and active invasion predominating in neutrophils. We also show that expression of IFN-β requires phagocytic uptake of the parasite by IMs, and signaling through Toll-like receptors (T

Type-I interferon is critical for FasL expression on lung cells to determine the severity of influenza.

Infection of influenza A virus in mammals induces hyper lung pneumonia, which often causes lethal diseases. FasL is a specific ligand of Fas, which is a type-I transmembrane protein to induce cell death. Previously, it has been reported that the hyper induction of gene expression associated with Fas signal is observed in lethal influenza A virus infection. More importantly, it was also reported that functional mutation of the FasL gene protects the host against influenza A virus infection. These observations suggest that induction of FasL signal is functionally associated with the severity of

Route of Infection Determines the Impact of Type I Interferons on Innate Immunity to Listeria monocytogenes.

Listeria monocytogenes is a food-borne pathogen which causes mild to life threatening disease in humans. Ingestion of contaminated food delivers the pathogen to the gastrointestinal tract, where it crosses the epithelial barrier and spreads to internal organs. Type I interferons (IFN-I) are produced during infection and decrease host resistance after systemic delivery of L. monocytogenes. Here we show that mice benefit from IFN-I production following infection with L. monocytogenes via the gastrointestinal route. Intragastric infection lead to increased lethality of IFN-I receptor chain 1-defi

Induction and inhibition of type I interferon responses by distinct components of lymphocytic choriomeningitis virus.

Type I interferons (IFNs) play a critical role in the host defense against viruses. Lymphocytic choriomeningitis virus (LCMV) infection induces robust type I IFN production in its natural host, the mouse. However, the mechanisms underlying the induction of type I IFNs in response to LCMV infection have not yet been clearly defined. In the present study, we demonstrate that IRF7 is required for both the early phase (day 1 postinfection) and the late phase (day 2 postinfection) of the type I IFN response to LCMV, and melanoma differentiation-associated gene 5 (MDA5)/mitochondrial antiviral signa

Induction of spermidine/spermine N1-acetyltransferase by interferon type I in cells of hematopoietic origin.

A number of novel genes activated by type I interferons (IFNs) were identified by differential display. Of five induced genes examined, four were of unknown function. However, one gene sequence was identical to the human spermidine/spermine N1-acetyltransferase, the rate-limiting enzyme in polyamine catabolism. This enzyme was induced by type I IFNs in a series of hematopoietic cell lines, including Daudi, HL-60, HPBMa, and Wil-2. No induction above constitutive levels occurred in a cell line of epithelial (ME180) or liver (HepG2) origin following treatment with type I IFN. Spermidine/spermine

Mechanism of Nucleic Acid Sensing in Retinal Pigment Epithelium (RPE): RIG-I Mediates Type I Interferon Response in Human RPE.

Age-related macular degeneration (AMD), a degenerative disease of the outer retina, is the leading cause of blindness among the elderly. A hallmark of geographic atrophy (GA), an advanced type of nonneovascular AMD (dry AMD), is photoreceptor and retinal pigment epithelium (RPE) cell death. Currently, there are no FDA-approved therapies for GA due to a lack of understanding of the disease-causing mechanisms. Increasing evidence suggests that chronic inflammation plays a predominant role in the pathogenesis of dry AMD. Dead or stressed cells release danger signals and inflammatory factors, whic

Role of Toll-like receptors and retinoic acid inducible gene I in endogenous production of type I interferon in dermatomyositis.

To explore the possible mechanisms implicated in the endogenous production of type I interferons within the muscle tissue of dermatomyositis (DM) patients. We detected the co-localization of plasmacytoid dendritic cells (pDCs) with Toll-like receptors (TLRs) and retinoic acid inducible gene (RIG)-I by immunohistochemistry and immunofluorescence. Western blotting confirmed the expression of TLRs and RIG-I. TLR-3 and RIG-I was preferentially expressed in the perifascicular atrophy fibers of DM. TLR-7 was only in inflammatory infiltrates of a few DM patients. TLR-4 and TLR-9 was expressed mainly

CD73 controls neutrophil responsiveness to type I interferon impairing antibacterial responses during secondary pneumococcal pneumonia.

Streptococcus pneumoniae (pneumococcus) are asymptomatic colonizers of the nasopharynx but can progress to pulmonary and systemic pathogens upon influenza A virus (IAV) infection. Polymorphonuclear cells (PMNs) are key for controlling S. pneumoniae , but their antibacterial function is impaired by IAV. Using a mouse model that allows transition of pneumococci from colonizers to disease-causing pathogens upon IAV co-infection, we examined the signaling pathways impairing PMN responses. We found that type I interferons (IFN) produced upon IAV infection drives impairment of PMN antibacterial resp

Specific bile acids can elicit the type-I interferon response through the cGAS-STING pathway.

Bile acids are metabolites crucial to lipid metabolism and immune regulation, yet their biological functions and mechanistic underpinnings remain largely elusive. In this study, we demonstrate that specific bile acids DCA, CDCA and LCA can trigger the type-I interferon response (IFN-I) in various cells through the cytosolic DNA-sensing cGAS-STING pathway. Phosphoproteomics indicates that bile acids can elicit a wide array of changes across numerous signaling pathways, culminating in the downregulation of Bcl-2 and p-BAD, resulting in the formation of Bax/Bak pore for the cytosolic release of m

Serum type I interferon promotes AIM2 inflammasome dysregulation in lupus patients through STAT1 and STAT2.

OBJECTIVE: The Absent in Melanoma 2 (AIM2) inflammasome is a crucial producer of IL-1β and IL-18 upon sensing double-stranded (ds)DNA but its role in SLE remains ambiguous. The study aims to investigate the involvement and underlying mechanism leading to AIM2 inflammasome dysregulation in lupus patients. METHODS: Expression and functional response of AIM2 inflammasome in monocytes were compared between SLE patients and healthy controls. Serological factors affecting AIM2 inflammasome response were tested by in vitro culture of primary monocytes. RNA sequencing, bioinformatics analyses and chr

Type I interferon-dependent and -independent signaling underlie autoantibody production in a murine lupus model.

Systemic lupus erythematosus (SLE) is characterized by the development of autoantibodies against diverse self-antigens and damage to multiple organs. Immunization with the SLE autoantigen β2-glycoprotein I and lipopolysaccharide (LPS) induces a murine model of SLE with the sequential emergence of multiple hallmark SLE autoantibodies. LPS is a TLR4 trigger of innate immunity and inflammation that signals through adaptor proteins MyD88 and TRIF, resulting in the production of both inflammatory cytokines and type I interferons (IFN-I). We hypothesized that both IFN-I-dependent and -independent me

Anti-Type I Interferon Autoantibodies in COVID-19 and Systemic Lupus Erythematosus: A Comparative Review.

Type I interferons (IFN-I), including IFN-α, IFN-β, and IFN-ω, are central to antiviral defence and immune regulation. Autoantibodies targeting IFN-I (anti-IFN-I AAbs) have emerged as key pathogenic factors in severe coronavirus disease 2019 (COVID-19) and are detectable in systemic lupus erythematosus (SLE), a prototypic IFN-driven autoimmune disease. Here we compare the prevalence and clinical impact of anti-IFN-I autoantibodies (Aabs) in COVID-19 and SLE based on a structured review of 53 studies from 2014 to 2025 and highlight the clinical associations and therapeutic opportunities present

Type I interferon signaling defines a novel disease signature in xeroderma pigmentosum C human keratinocytes.

Xeroderma Pigmentosum C (XPC) is a DNA damage recognition protein central to the global genome nucleotide excision repair (GG-NER) pathway, where it acts as a primary sensor of UV-induced DNA lesions. Loss-of-function mutations in the XPC gene lead to a photosensitive phenotype, with marked accumulation of unrepaired DNA damage and a dramatically elevated risk (10,000-fold) of skin cancer. However, understanding the molecular signaling mechanisms associated with XP-C has been hindered by the lack of reproducible disease models. Here, we overcome this challenge using our genetically engineered

Keratin 16 inhibits type I interferon responses in differentiating keratinocytes of stressed and diseased skin.

The stress-induced keratin 16 intermediate filament gene, KRT16, and K16 protein are spatially restricted to the suprabasal layers of the epidermis and widely used as biomarkers in psoriasis, hidradenitis suppurativa, atopic dermatitis, and other inflammatory disorders. Pathogenic variants in KRT16 cause pachyonychia congenita (PC), a rare genetic condition in which tissue homeostasis is disrupted in palmoplantar epidermis, nail, oral mucosa, and other KRT16-expressing epithelial appendages. How K16 may regulate the molecular mechanisms underlying these conditions is poorly understood. Here, w

Type I interferon drives T cell responses to amyloid beta in the central nervous system.

Amyloid beta (Aβ) plaque deposition in the central nervous system (CNS) is a hallmark of Alzheimer's disease (AD) and cerebral amyloid angiopathy (CAA), triggering robust innate immune responses. However, the role of the adaptive immune system remains less well understood. Here we show the immune microenvironment dynamics in APP23 transgenic (APP23-tg) mice modelling CNS amyloid pathology, using single-cell transcriptomics. We observed a marked increase in T-cell populations during late disease stages, particularly CD8⁺ T-cells that clustered around Aβ plaques, suggesting a targeted immune res

HCMV encoded UL84 hijacks FHL2 to suppress type I interferon production and enhance viral replication.

Virus infection activates the host's innate immune responses, which is a very precise and complex biological process and will lead to the immediate transcription of type I interferon. The general transcriptional activator proteins such as IRF3, ATF2/c-Jun, and NF-κB can be induced to form a stable enhanceosome in the transcriptional regulatory region of IFN-β promoter. Several cellular factors have recently been reported to be involved in the transcriptional regulation of IFN-β under certain physiological conditions. Here, we identified four and a half LIM domains protein 2 (FHL2) as an intera

Cell-Intrinsic Type I Interferon Signaling as a Pleiotropic Orchestrator of CD4 T Cell Immunity.

Type I interferons (IFN-I) are pleiotropic cytokines best known for their antiviral impacts. However, they are known to also impact immune responses outside of viral infection through directly signaling many populations of innate and adaptive immune cells. Here, we focus on the complex body of findings from viral, bacterial, and parasitic infection models, cancer and autoimmunity studies, as well as in vitro experiments using human and murine T cells, demonstrating that IFN-I can be directly sensed by CD4 T cells. Such signaling has been shown to influence many central aspects of antigen-speci
