Cytogenetic screening of a Canadian swine breeding nucleus using a newly developed karyotyping method named oligo-banding.

BACKGROUND: The frequency of chromosomal rearrangements in Canadian breeding boars has been estimated at 0.91 to 1.64%. These abnormalities … chniques, and the need for prior cytogenetic skills. The aim of this study was to develop a new pig karyotyping method based on fluorescent banding patterns. RESULTS: The use of 207,847 specific oligonucleotides generated 96 fluorescent bands that are distributed across the 18 autosomes and the sex chromosomes. Tested alongside conventional G-banding, this o …

Chromosome aberrations detected by conventional karyotyping using novel mitogens in chronic lymphocytic leukemia: Clinical and biologic correlations.

To clarify whether karyotype aberrations (KA) involving regions not covered by the standard fluorescence in situ hybridization (FISH) panel … is was also the case for NOTCH1 and SF3B1 mutations in the VC. We suggest that in CLL, conventional karyotyping with novel mitogens could be more effective than FISH for the detection of KA allowing for a more precise refinement of prognosis.

COBRA: combined binary ratio labeling of nucleic-acid probes for multi-color fluorescence in situ hybridization karyotyping.

Combined binary ratio labeling (COBRA) is designed to increase the multiplicity of fluorescence in situ hybridization (FISH)--i.e., the number of targets that can be distinguished simultaneously. In principle, chemical (ULS), enzymatic (nick translation or random priming) or PCR-based labeling procedures of probes can be used. The method was originally designed to label chromosome-painting probes, but has also been used for probe sets specific for subtelomeric regions. COBRA imaging requires a digital fluorescence microscope equipped for sequential excitation and recording of color images. Sta

Rewriting wheat karyotypes through centromere-targeted chromosome engineering.

Engineering karyotypes could expand the genetic and breeding potential of crops, but restructuring gigabase-scale polyploid genomes remains challenging. Here we develop centromere editing-mediated chromosomal variation, a strategy using CRISPR-Cas9 to target abundant centromeric retrotransposons in hexaploid wheat. Centromere editing-mediated chromosomal variation efficiently induces simultaneous chromosome cleavage across the genome, generating diverse karyotypic variants and reconfiguring up to 11 chromosomes in one generation. Notably, it produces widespread reciprocal translocations, with

The Prognostic Utility of Normal Karyotype Fraction in Acute Myeloid Leukemia.

BACKGROUND: The prognostic significance of the proportion of normal metaphases detected at diagnosis in acute myeloid leukemia (AML) has not been systematically evaluated. PATIENTS AND METHODS: We conducted a retrospective cohort study of 893 adult AML patients treated at Cleveland Clinic between January 2015 and September 2023 who underwent conventional cytogenetic analysis. Normal karyotype fraction (NKF) was defined as the proportion of normal metaphases among all metaphases analyzed at diagnosis. NKF was evaluated as a continuous measure and categorized using three-group (NKF = 0, 0 < NKF

Pervasive chromosomal instability drives the karyotypic evolution of hypodiploid tumours.

BACKGROUND: Tumours frequently exhibit extreme levels of aneuploidy. While increases in ploidy are well-characterised, the opposite phenomenon-extensive chromosome loss leading to hypodiploidy-remains underexplored. METHODS: Here, we analyse over 17,000 cancer genomes from 34 cancer types and perform a pan-cancer analysis of karyotypic evolution in hypodiploid tumours. We develop methods to identify current and former hypodiploid tumours, analyse predictors of chromosome loss patterns across tissues, and characterise the relationship between hypodiploidy and other forms of chromosomal instabi

Acute Lymphoblastic Leukemia With Near-haploid Karyotype and Philadelphia Chromosome.

BACKGROUND/AIM: In precursor B-cell lineage acute lymphoblastic leukemia (BCP-ALL), leukemic cells harbor genetic abnormalities that play an important role in the diagnosis, prognosis, and treatment. A subgroup of BCP-ALL is characterized by the presence of a Philadelphia (Ph) chromosome and a chimeric BCR::ABL1 gene, whereas in another subgroup, leukemic cells exhibit near-haploidy with chromosome number 24-30. This study presents the third documented case of BCP-ALL in which a near haploid clone concurrently displayed a Ph chromosome/BCR::ABL1. CASE REPORT: Bone marrow cells obtained at dia

Modeling specific aneuploidies: from karyotype manipulations to biological insights.

An abnormal chromosome number, or aneuploidy, underlies developmental disorders and is a common feature of cancer, with different cancer types exhibiting distinct patterns of chromosomal gains and losses. To understand how specific aneuploidies emerge in certain tissues and how they contribute to disease development, various methods have been developed to alter the karyotype of mammalian cells and mice. In this review, we provide an overview of both classic and novel strategies for inducing or selecting specific chromosomal gains and losses in human and murine cell systems. We highlight how th

Pituitary Macroadenoma in a Girl with Male Karyotype: A Rare Case Study.

Macroadenoma is a tumor that typically develops in the epithelial cells of the pituitary gland. Patients suffering from the condition are often asymptomatic with complaints that are caused by hormonal imbalance. Therefore, chromosome analysis needs to be done to females aged >16 years presenting with amenorrhea. Karyotype 46,XY is a disorder of sex development (DSD) that is caused by the complex process of gene interactions, androgen synthesis, and hormone regulation. The patient initially came to the hospital for a scheduled transsphenoidal surgery due to pituitary macroadenoma, and later com
