Effects of Propylene Glycol on Negative Energy Balance of Postpartum Dairy Cows.

With the improvement in the intense genetic selection of dairy cows, advanced management strategies, and improved feed quality and disease control, milk production level has been greatly improved. However, the negative energy balance (NEB) is increasingly serious at the postpartum stage because the intake of nutrients cannot meet the demand of quickly improved milk production. The NEB leads to a large amount of body fat mobilization and consequently the elevated production of ketones, which causes metabolic diseases such as ketosis and fatty liver. The high milk production of dairy cows in ear

Lactate Buildup at the Site of Chronic Inflammation Promotes Disease by Inducing CD4+ T Cell Metabolic Rewiring.

Accumulation of lactate in the tissue microenvironment is a feature of both inflammatory disease and cancer. Here, we assess the response of immune cells to lactate in the context of chronic inflammation. We report that lactate accumulation in the inflamed tissue contributes to the upregulation of the lactate transporter SLC5A12 by human CD4+ T cells. SLC5A12-mediated lactate uptake into CD4+ T cells induces a reshaping of their effector phenotype, resulting in increased IL17 production via nuclear PKM2/STAT3 and enhanced fatty acid synthesis. It also leads to CD4+ T cell retention in the infl

Blood gas changes and hypophosphatemia in lactate-induced panic.

Alkalosis is prominent among the many physiologic and biochemical effects of sodium lactate infusion. Though this is partially due to the conversion of lactate to bicarbonate, the metabolic component, it may also be secondary to hyperventilation before and during the infusion, the respiratory component. We analyzed pH, carbon dioxide pressure, bicarbonate, and inorganic phosphate from patients with panic disorder and agoraphobia with panic attacks and from normal controls both before and during lactate infusion. Our findings extend earlier work demonstrating that many such patients are chronic

Maternal Supplementation with Goji Berry (Lycium barbarum L.) During Lactation Prevents Metabolic Syndrome in Adult Male Rats Overfed Early in Life.

BACKGROUND: Challenges during critical developmental periods, such as postnatal overfeeding, can affect an organism's development and increase its susceptibility to metabolic syndrome in adulthood. Nutritional or pharmacological interventions during this stage may mitigate this condition. OBJECTIVE: Given the wide therapeutic properties of Goji berries, we aimed to evaluate whether supplementing the diet of lactating female rats with these could prevent the development of metabolic disorders in adult offspring raised in small litters. METHODS: Pregnant Wistar rats were used to prepare the li

Computational model captures cardiac growth in hypertensive pregnancies and in the postpartum period.

Heart growth in the pregnant patient helps maintain cardiovascular function while supporting the growing fetus. However, in some cases, the cardiovascular demand of pregnancy can trigger life-threatening conditions, including hypertensive disorders of pregnancy and peripartum cardiomyopathy. The mechanisms that control heart growth throughout pregnancy are unclear, and treating these diseases remains elusive. We previously developed a computational model that accounts for hormonal and hemodynamic interactions throughout pregnancy and demonstrated its ability to capture realistic cardiac growth

Cannabidiol attenuates cognitive deficits and neuroinflammation induced by early alcohol exposure in a mice model.

Foetal alcohol spectrum disorder (FASD) is the umbrella term used to describe the physical and mental disabilities induced by alcohol exposure during development. Early alcohol exposure induces cognitive impairments resulting from damage to the central nervous system (CNS). The neuroinflammatory response accompanied by neurodegenerative mechanisms contribute to those detrimental alterations. Cannabidiol (CBD) has recently emerged as an anti-inflammatory drug that might be useful to treat several neuropsychiatric disorders. In our study, we assessed the effects of CBD on long-lasting cognitive

Maternal high-fat diet prevents developmental programming by early-life stress.

Anxiety disorders and depression are well-documented in subjects exposed to adverse childhood events. Recently, maternal obesity and/or maternal consumption of high-fat diets (HFD) have been also proposed as risk factors for offspring mental health. Here using an animal model in rats, we explored the combinatorial effects of a maternal HFD (40% of energy from fat without impact on maternal weight; during gestation and lactation) and maternal separation (MS) in offspring. In the prefrontal cortex (PFC) of pups, MS led to changes in the expression of several genes such as Bdnf (brain derived neu

[Discussion on postpartum hypogalactia treated with Shaoze (SI 1) based on the theory of "milk-blood homology"].

Based on the theory of "milk-blood homology", the evidence of Shaoze (SI 1) in treatment of postpartum hypogalactia was explored and the clinical application introduced. In traditional Chinese medicine, small intestine meridian and the functions of small intestine are extensively involved in the procedure of milk formation, i.e. transformation, transportation and connection. Shaoze (SI 1) communicates the meridians of hand-taiyang and hand-shaoyin. It enhances the connection of the small intestine with the heart, spleen and stomach. It is the jing-well point for unblocking and moistening to pr

[Evaluation of PO2, PCO2, pH, and B.E. in milking cows in different milking phases].

During one year we studied the acid-base state in the milking cows trying to solve the relationship between the secretion disorders and feeding. THe results confirm that exist a link between the acid-base state and the disorders. These disorders are connected to problems of alimentation. In our experiments it had been possible to observe a change in pH of blood versus alkalosis.

Rituximab in neurological disease: principles, evidence and practice.

Rituximab is a widely used B-cell-depleting monoclonal antibody. It is unlicensed for use in neurological disorders and there are no treatment guidelines. However, as a rapidly acting, targeted therapy with growing evidence of efficacy and tolerability in several neuroinflammatory disorders, it is an attractive alternative to conventional immunomodulatory medications. This practical review aims to explain the basic principles of B-cell depletion with therapeutic monoclonal antibodies. We present the evidence for using rituximab in neurological diseases, and describe the practical aspects of pr

Nursing sickness in the mink--a metabolic mystery or a familiar foe?

Nursing sickness, the largest single cause of mortality in adult female mink (Mustela vison), is an example of a metabolic disorder, which develops when the demands for lactation require extensive mobilization of body energy reserves. The condition is characterized by progressive weight loss, emaciation, and dehydration with high concentrations of glucose and insulin in the blood. Morbidity due to nursing sickness can be as high as 15% with mortality around 8%, but the incidence is known to vary from year to year. Stress has been shown to trigger the onset of the disease and old females and fe

Maternal Diet Enrichment in n-3 Long-Chain Polyunsaturated Fatty Acids during Pregnancy and Lactation Improves Their Transfer from the Mother to the Offspring and Counterbalances High-Fat Diet-Induced Alterations in a Sex-Dependent Manner in Rats.

BACKGROUND: Low birth weight (LBW) increases the risk of developing metabolic disorders in adulthood and reduces fatty acid (FA) brain accretion in offspring in rodent models. As milk composition is partially influenced by maternal diet, it is conceivable to act on maternal diet to improve FA brain accretion and development in offspring. OBJECTIVES: The study aimed to evaluate the impact of n-3 long-chain polyunsaturated FA (LCPUFA) deficiency or enrichment in the maternal diet during gestation and lactation on maternal diet on milk composition, offspring brain lipids, and the potential to co

Periparturient blood T-lymphocyte PD-1 and CTLA-4 expression as potential predictors of new intramammary infections in dairy cows during early lactation (short communication).

BACKGROUND: The periparturient period in dairy cows is marked by immunosuppression which increases the likelihood of infectious disorders, particularly also mastitis. An in-depth understanding of peripartum leukocyte biology is vital for the implementation of highly successful post-partum disease prevention measures. Immune checkpoint molecules, such as programmed death 1 (PD-1) and cytotoxic T-lymphocyte antigen 4 (CTLA-4), are critical inhibitory receptors expressed on immune cells, particularly T cells, that drive immunosuppressive signaling pathways. However, the potential role of immune c

Management of nipple adenomas during pregnancy: a case report.

BACKGROUND: Nipple adenoma is a very uncommon, benign neoplasm that involves the nipple. A palpable mass of the nipple associated with nipple discharge and erosion or ulceration is the common clinical presentation. Generally, complete surgical excision of the nipple is the main treatment, alternative therapeutic methods such as Mohs micrographic surgery, nipple splitting enucleation, and cryotherapy can be considered. Disorders of the breast in young women are generally benign. Even if the management during pregnancy is usually conservative and surgical excision is reserved for very strong mal

Faster lactate transport across red blood cell membrane in sickle cell trait carriers.

The physical and physiological behavior of sickle cell trait carriers (AS) is somewhat equivocal under strenuous conditions, although this genetic abnormality is generally considered to be a benign disorder. The occurrence of incidents and severe injuries in AS during exercise might be explained, in part, by the lactic acidosis due to a greater lactate influx into AS red blood cells (RBCs). In the present study, the RBC lactate transport activity via the different pathways was compared between AS and individuals with normal hemoglobin (AA). Sixteen Caribbean students, nine AS and seven AA, per
