GyneFix levonorgestrel-releasing intrauterine system for Chinese patients: a preliminary retrospective self-controlled observational study.

BACKGROUND: Device expulsion is a common complication of the levonorgestrel-releasing intrauterine system (LNG-IUS), with a higher incidence among high-risk patients presenting with enlarged uterine cavities, cervical laxity or menorrhagia. At present, no commercially available frameless LNG-IUS is marketed in mainland China, leaving these high-risk patients without effective clinical interventions. OBJECTIVES: This study describes the assembly procedures and clinical application of a modified frameless LNG-IUS based on the GyneFix anchoring system (GyneFix-LNG-IUS), and the intrauterine rete…

Association of levonorgestrel intrauterine system and chronic kidney disease: A nationwide population-based cohort study.

OBJECTIVE: Sex-, age-related discrepancies in development of chronic kidney disease (CKD) suggest that sex hormones play important roles in the development of CKD. We aimed to investigate the effect of levonorgestrel intrauterine system (LNG-IUS), one of the progestin-only agents, on the development of CKD. MATERIALS AND METHODS: This retrospective cohort study used claims data from the Korean Health Insurance Review and Assessment Service. The study participants were women aged 30-49 years old who were diagnosed with endometriosis, uterine fibroids, or abnormal uterine bleeding (AUB) between…

Venous Thromboembolic Risk of Estradiol Valerate-Dienogest Compared with Ethinyl Estradiol-Levonorgestrel Combined Oral Contraceptives.

This pooled analysis compared the risk of venous thromboembolism (VTE) associated with combined oral contraceptives (COCs) containing estradiol (E2) valerate-dienogest with those containing ethinyl E2-levonorgestrel. Data were retrieved from two large, prospective, observational cohort studies. Propensity score subclassification was applied to balance baseline parameters between the COC user cohorts. Crude and adjusted hazard ratios (HRs) were calculated based on the extended Cox model. The pooled data set included 11,616 E2 valerate-dienogest users and 18,681 ethinyl E2-levonorgestrel users, …

Effects of levonorgestrel-quinestrol (EP-1) treatment on Mongolian gerbil wild populations: a case study.

Rodent pest population outbreaks occur frequently in grassland ecosystems in northern China. The Mongolian gerbil (Meriones unguiculatus) is a dominant pest rodent which is distributed across the semi-desert grasslands of Inner Mongolia, China. In 2009, we studied the contraceptive effect of levonorgestrel-quinestrol (EP-1), concentration 50 ppm, on a wild Mongolian gerbil population. The one-off contraceptive treatment was compared with a control group using a semi-monthly live trapping method in the Ordos Semi-desert Grassland Region of Inner Mongolia. The results show that juveniles were no…

Menstrual pattern changes from levonorgestrel subdermal implants and DMPA: systematic review and evidence-based comparisons.

BACKGROUND: Many women want a lengthy duration of contraception but are wary of the menstrual changes from depot medroxyprogesterone acetate (DMPA). A subdermal levonorgestrel (LNG) implant may be a reasonable alternative. However, information on menstrual changes from these methods has not been summarized and compared in an easy-to-understand form. STUDY DESIGN: We systematically reviewed the published literature on these contraceptives to find research that used menstrual diaries and standard World Health Organization definitions. We attempted to find information on amenorrhea, number of bl…

Effects of levonorgestrel-releasing IUS and progesterone receptor modulator PRM CDB-2914 on uterine leiomyomas.

We have found that the use of levonorgestrel-releasing IUS results in a remarkable decrease in endometrial proliferation and a remarkable increase in apoptosis in the endometrium; therefore, it is effective for long-term management of menorrhagic women with uterine myomas because of the striking reduction in menorrhagia. This prompted us to characterize the effects of progesterone (P(4)) and progesterone receptor modulator (PRM) CDB2914 on uterine myoma growth. In vitro studies with cultured uterine leiomyoma cells and normal myometrial cells revealed that P(4) stimulated the proliferative act…

Comparative effects of monophasic desogestrel plus ethinyloestradiol and triphasic levonorgestrel plus ethinyloestradiol on lipid metabolism.

The effects of the monophasic oral contraceptive preparation desogestrel plus ethinyloestradiol (EE) and triphasic levonorgestrel plus EE on lipid metabolism were compared. Sixteen young healthy female volunteers received monophasic desogestrel plus EE (21 X 150/30), whereas eighteen women received triphasic levonorgestrel plus EE (6 X 50/30 + 5 X 75/40 + 10 X 125/30) for six consecutive cycles. The lipid composition of the various lipoprotein fractions was estimated after separation in a density gradient by ultracentrifugation. In addition, the apolipoproteins A-I and B were assessed. Levonorgestrel intrauterine system: do the contraceptive benefits outweigh the risks in light of new FSRH guidance?

BACKGROUND: The levonorgestrel-releasing intrauterine system (LNG-IUS) is a long-acting reversible contraceptive. In 2023, the Faculty of Sexual and Reproductive Healthcare (FSRH) extended its recommended duration of use from 5 to 8 years. This change underlines the need to re-examine the balance of efficacy and risks associated with LNG-IUS. AIM: To systematically review evidence on contraceptive effectiveness, safety, and acceptability of LNG-IUS, and evaluate its alignment with updated FSRH guidance. METHOD: A systematic search of MEDLINE, Embase, PsycINFO, CINAHL, and Scopus (April 2025)…
