A Review on HIV's Effects on Exploratory Activity, Motor Functions, and Learning in Animals.

Infection with the human immunodeficiency virus (HIV) causes human neuropsychological disorders, such as apathy and hypokinesia, as well as deficits in motor skills, selective attention, and learning. Based on findings from multiple studies, similar signs have been reproduced in animal models following intracerebral injections of HIV-infected human monocytes or monocyte-derived macrophages, or exposure to gp120, Tat, or Nef. These include learning deficits in Morris, radial arm, and Barnes mazes; impairments in novel object place and shape recognition; motor coordination deficits on stationary

Chondromodulin-I maintains cardiac valvular function by preventing angiogenesis.

The avascularity of cardiac valves is abrogated in several valvular heart diseases (VHDs). This study investigated the molecular mechanisms underlying valvular avascularity and its correlation with VHD. Chondromodulin-I, an antiangiogenic factor isolated from cartilage, is abundantly expressed in cardiac valves. Gene targeting of chondromodulin-I resulted in enhanced Vegf-A expression, angiogenesis, lipid deposition and calcification in the cardiac valves of aged mice. Echocardiography showed aortic valve thickening, calcification and turbulent flow, indicative of early changes in aortic steno

Organelle contact sites as spatial regulators of lipid fate in adipose tissue metabolic disease.

Adipose tissue protects metabolic homeostasis by storing excess fatty acids, releasing fuels during energy demand and coordinating endocrine and inflammatory signals. These functions are often described as linear pathways of lipogenesis, lipolysis, fatty acid oxidation and thermogenesis. However, lipid handling in adipocytes is spatially organized by organelle contact sites. The endoplasmic reticulum (ER), lipid droplets (LDs), mitochondria, peroxisomes and lysosomes form dynamic interfaces that determine whether fatty acids are stored safely, mobilized for oxidation, processed into specialize

Electron resonance studies on the influence of anionic surfactants on human skin.

BACKGROUND: When skin is exposed to chemicals, raw materials interact with the lipid structure of the stratum corneum. At least two types of disorders can be distinguished--that of alkyl chains inside one lipid bilayer and that of lipid layer arrangement. Electron spin resonance (ESR) spectroscopy of a nitroxide spin label is a valuable method in the study of biological membranes. OBJECTIVE: These experiments define the effect of anionic surfactants on the lipid bilayer of human stratum corneum. METHODS: 5-Doxyl stearic acid (5-DSA) was used as the spin label. Sodium lauryl sulfate (SLS) and

Characteristic features of antilipolytic activity of serum in relation to obesity.

Antilipolytic activity (ALA) of serum of the obese and controls was tested on rat epididymal adipose tissue incubated in medium containing adrenaline; the results were compared with the effect of insulin. Serum of the obese displayed lower ALA than controls, in one third obese individuals serum potentiated the effect of adrenaline. After fasting, ALA of serum decreased. ALA value is related to the prevailing metabolic situation, an inverse relationship was observed between ALA and levels of NEFA and ketone bodies. This seems to suggest that antilipolytic effect of insulin, whose concentrations

S-acylation of ATGL is required for lipid droplet homoeostasis in hepatocytes.

Lipid droplets (LDs) are organelles specialized in the storage of neutral lipids, cholesterol esters and triglycerides, thereby protecting cells from the toxicity of excess lipids while allowing for the mobilization of lipids in times of nutrient deprivation. Defects in LD function are associated with many diseases. S-acylation mediated by zDHHC acyltransferases modifies thousands of proteins, yet the physiological impact of this post-translational modification on individual proteins is poorly understood. Here, we show that zDHHC11 regulates LD catabolism by modifying adipose triacylglyceride

Aluminum ions stimulate the oxidizability of low density lipoprotein by Fe2+: implication in hemodialysis mediated atherogenic LDL modification.

OBJECTIVE: Al(3+) stimulates Fe(2+) induced lipid oxidation in liposomal and cellular systems. Low-density lipoprotein (LDL) oxidation may render the particle atherogenic. As elevated levels of Al(3+) and increased lipid oxidation of LDL are found in sera of hemodialysis patients, we investigated the influence of Al(3+) on LDL oxidation. MATERIALS AND METHODS: Using different LDL modifying systems (Fe(2+), Cu(2+), free radical generating compounds, human endothelial cells, hemin/H(2)O(2) and HOCl), the influence of Al(3+) on LDL lipid and apoprotein alteration was investigated by altered elec

Impurity profiling of lipid-conjugated oligonucleotides using reversed-phase with and without ion-pair reagents and hydrophilic interaction liquid chromatography.

Lipid-oligonucleotide conjugates (LONs) represent a next-generation class of biopharmaceuticals that couple nucleic-acid specificity with lipid-mediated delivery, enabling superior membrane affinity and cellular uptake, for highly targeted therapeutic action. Lipid type and conjugation position strongly influence hydrophobicity, retention, and interaction strength from the exposure of the lipid moiety, thereby affecting the chromatographic behavior of impurities such as shortmers and diastereomers. In this study, we systematically investigated the performance of different chromatographic modes

Gymnema saponin-induced lipid flip-flop identifies rigid membrane phenotype of methicillin resistant S. aureus and enhances it's antibiotic susceptibility.

Our previous study revealed that lipid flip-flop inducing phytochemicals from Gymnema sylvestre increase membrane permeability of antimicrobials in S. aureus. However, their lipid flipping and membrane permeabilizing effect on methicillin resistant S. aureus (MRSA) membrane that has intrinsically higher aminoacylated lipid content compared to methicillin sensitive S. aureus (MSSA) is poorly characterized. Gymnema saponins, gymnemic acid I and IV significantly increased the antibiotic susceptibility in both MSSA and MRSA. MRSA exhibited a rigid membrane with lipid diffusion coefficient 0.0002 μ

Lipidomics reveals cell specific changes during pluripotent differentiation to neural and mesodermal lineages.

Due to their self-renewal and differentiation capabilities, pluripotent stem cells hold immense potential for advancing our understanding of human disease and developing cell-based or pharmacological interventions. Realizing this potential, however, requires a thorough understanding of the basal cellular mechanisms which occur during differentiation. Lipids are critical molecules that define the morphological, biochemical, and functional role of cells. This, combined with emerging evidence linking lipids to neurodegeneration, cardiovascular health, and other diseases, makes lipids a critical c

Relationship between mitochondria and the development of specific lipid droplets in capillary endothelial cells of the respiratory tract in patients with sarcoidosis.

The aim of this study was to morphologically evaluate damage in the capillary endothelial cells of the respiratory tract in patients with sarcoidosis. We examined tissues of the bronchus and lung obtained from 16 patients with sarcoidosis consisting of 2 stage 0, 10 stage I and 4 stage II patients, and 11 controls. The morphology of capillary endothelial cells was studied using electron microscopy. In the samples from patients with sarcoidosis, lipid droplets exhibiting dark monophasic density (unsaturated fatty acids) were surrounded by abundant lysosomes in the capillary endothelial cells, a

Sertoli cell adaptation to glucose deprivation: Potential role of AMPK in the regulation of lipid metabolism.

Sertoli cells (SCs) provide an adequate environment for germ cell development. SCs possess unique features that meet germ cells' metabolic demands: they produce lactate from glucose, which is delivered as energy substrate to germ cells. SCs store fatty acids (FAs) as triacylglycerols (TAGs) in lipid droplets (LDs) and can oxidize FAs to sustain their own energetic demands. They also produce ketone bodies from FAs. It has been shown that exposure of SCs to metabolic stresses, such as glucose deprivation, triggers specific adaptive responses that sustain cell survival and preserve lactate supply

Toll-like receptor 4 signaling is required for clusterin-induced tumor necrosis factor-α secretion in macrophage.

Clusterin is a secretory glycoprotein that is up-regulated in areas of inflammation and under increased levels of oxidative stress. Previously, we demonstrated that clusterin activates NF-κB, and up-regulates the expression of MMP-9 and TNF-α. In this research, we extend our previous findings by reporting that such clusterin-induced macrophage response is mediated via TLR4 signaling. Specifically, we found that TNF-α induced by clusterin was significantly abrogated by pretreatment of TLR4-signaling inhibitors and anti-TLR4 neutralizing antibody. Additionally, a primary culture of macrophages d

Plasma parameters related to energy and lipid metabolism in periparturient Modenese and Italian Friesian cows.

The aim of this work was to compare energy and lipid metabolism during the peripartum period between Modenese (MO) and Italian Friesian (IF) … es during p.p. The differences observed between the two breeds suggest how MO cows are subjected to lipid mobilization during late gestation; on the contrary, IF cows are predisposed to mobilize their lipid reserves at the beginning of lactation to support high production. The results indicate a diverse ability to cope with metabolic stress and suggest the hypothesi …

Fasting Reveals the Effects of a Plant Extract- and Microalgae-Derived Nutraceutical on Lipid Metabolism and Hepatic Physiology in Juvenile Sparus aurata Fed a Plant-Based Diet.

This study investigated the effects of a plant-based diet supplemented with a nutraceutical derived from plant extracts and microalgae on hepatic function and lipid metabolism during a fasting challenge. Juvenile Sparus aurata were fed three experimental diets for 90 days, followed by 14 days of feed deprivation. The diets consisted of: (i) a control formulation representative of commercial feeds for the species (C+: 20% fishmeal [FM], 9% fish oil [FO]); (ii) a low-FM, predominantly plant-based diet containing 5% FM and 5% FO (C‒); and (iii) the same C‒ diet supplemented with 1% LB-LIVERprotec
