A child with acute pancreatitis and recurrent hypoglycemia due to 3-hydroxy-3-methylglutaryl-CoA lyase deficiency.

A five-year-old-girl with a history of recurrent hypoglycemia presented with acidosis, intractable vomiting, and abdominal tenderness; the diagnosis of acute pancreatitis was made by abdominal ultrasonography and supportive biochemical studies. Urinary organic acid analysis revealed metabolites suggestive of HMG-CoA lyase deficiency, and subsequent enzyme assays of lymphocytes and fibroblasts confirmed this diagnosis. Acute pancreatitis, an uncommon condition in childhood, is seen with increased frequency in patients with Reye syndrome, a metabolic disorder with which HMG-CoA lyase deficiency

Bipartite Anchoring at the +3 Subsite Governs Trisaccharide Specificity in a PL7 Alginate Lyase and Is Transferable to a Homologous Enzyme.

Alginate lyases produce alginate oligosaccharides (AOS) whose bioactivities depend on the degree of polymerization (DP). Loop regions and individual residues influence product DP. However, how residues from loops cooperate at subsites to enforce a defined DP remains unresolved. Here, we identified ALG4, a PL7_5 alginate lyase specifically producing trisaccharides (ΔDP3), and ALG3, a same-subfamily homologue generating ΔDP2-ΔDP4 mixtures. Action-pattern analysis showed that ALG4 accommodates three sugar residues at the +1/+2/+3 subsites without extension toward +4. Structurally, the Orange Loop…

An engineered disulfide staple restricts lid loop dynamics and alters substrate specificity of phenylalanine ammonia-lyase.

In Anabaena variabilis (Trichormus variabilis) phenylalanine ammonia-lyase (AvPAL), a conserved lid-like loop sits over the active site and has been studied both for its role in positioning a catalytic tyrosine and for its contribution to phenylalanine aminomutase (PAM) activity. While the active site architecture and substrate specificity of AvPAL have been extensively characterized, the dynamic behavior of this unstructured loop beyond its role in catalysis remains poorly understood. Here, we investigate the functional role of this loop by restricting its mobility through targeted interchain

Multi-biomarker models based on the androgen backdoor pathway for diagnosing 17α-hydroxylase/17,20-lyase deficiency.

OBJECTIVES: 17α-hydroxylase/17,20-lyase deficiency (17OHD) is a rare form of congenital adrenal hyperplasia (CAH) that is frequently underdiagnosed, particularly in partial deficiency. We evaluated androgen backdoor pathway steroids as diagnostic biomarkers for 17OHD. METHODS: We included 50 confirmed 17OHD patients and 266 healthy controls. Six androgen backdoor pathway steroids-5α-dihydroprogesterone (5α-DHP), 17OH-dihydroprogesterone (17OH-DHP), 17OH-allopregnanolone (17OH-Allo), androsterone (AN), androstanediol (3α-diol), and 5α-dihydrotestosterone (DHT)-were quantified in plasma using u

Gut Microbial Choline TMA-Lyase CutC: From Metabolic Mechanism to a Novel Therapeutic Target for Diseases.

In recent years, the pivotal role of the gut microbiota and its metabolites in host health and disease has garnered increasing attention. Dietary phosphatidylcholine and choline are metabolized by gut bacteria to generate trimethylamine (TMA). Upon entering the bloodstream, TMA is oxidized by host liver enzymes to trimethylamine N-oxide (TMAO), a known independent risk factor for various systemic diseases, including atherosclerosis, thrombosis, and chronic kidney disease. Within this complex "diet-gut-host" metabolic axis, the microbial choline TMA-lyase (CutC) acts as the key rate-limiting en

Immunoresponsive Gene 1-Itaconate Exacerbates Hypertension by Inhibiting the Cystathionine Gamma-Lyase/Hydrogen Sulfide Pathway.

The role of immunoresponsive gene 1 (IRG1)-itaconate (ITA) in hypertension remains poorly understood. This study aimed to investigate this role and the underlying mechanisms. IRG1 expression and ITA levels were assessed by Western blotting, targeted metabolomics, and ELISA. We employed global Irg1 (mouse gene coding IRG1)-knockout mice, smooth muscle cell (SMC)-specific Irg1 overexpression mice, and mice receiving intraperitoneal ITA injection. Hypertension was induced by angiotensin II (Ang II) infusion. Blood pressure was measured by tail-cuff method and radiotelemetry, while vascular struct

Prenatal-Onset Lethal Adenylosuccinate Lyase Deficiency Diagnosed by Rapid Whole Genome Sequencing: A Case Report.

Adenylosuccinate lyase deficiency (ADSLD) is an ultra-rare autosomal recessive disorder of purine metabolism. The most severe neonatal phenotype presents with profound encephalopathy from birth, hypotonia, respiratory insufficiency, and early seizures. A preterm infant was delivered at 34+0 weeks with antenatal progressive microcephaly and mild polyhydramnios. At birth, the neonate exhibited absent spontaneous respiration and marked hypotonia, necessitating ventilation and intubation. Respiratory distress syndrome (RDS) was managed with surfactant, but two extubation attempts with non-invasive

A selenomethionine deficient, high-fructose diet does not lead to cardiometabolic disorder in the selenocysteine lyase knockout mice.

BACKGROUND/PURPOSE: High-fructose consumption is a driver of cardiometabolic disorders and metabolic syndrome, and selenium (Se) deficiency further increases the risk of developing these diseases. Consuming high amounts of fructose induces insulin resistance and oxidative stress, and alters the cardiac lipidome. Se may reduce the detrimental impacts of fructose through its incorporation into selenoproteins like the glutathione peroxidases 1 and 4, (GPX1,4) and the thioredoxin reductase 1 (TXNRD1) whose primary function is to curb oxidative stress. When Se levels are limited, selenocysteine lya

Characterization of a novel alginate lyase Alg0392 with organic solvent-tolerance from Alteromonas sp. A1-6.

Enzymatic depolymerization of seaweed polysaccharides aroused great interest in the production of functional oligosaccharides and fermentable sugars. Alginate lyase Alg0392, a potential novel member of the polysaccharide lyase PL17 family, was cloned from Alteromonas sp. A1-6. The enzymatic properties, kinetic parameters, and hydrolytic products of Alg0392 were systematically characterized. Especially, the recombinant enzyme Alg0392 showed excellent tolerance to organic reagents. When treated with 5 mmol/L of TritonX-100 or 20%(v/v) of methanol, its relative enzyme activity could be maintained

A Rare Presentation of 17α-Hydroxylase/17,20-Lyase Deficiency in a Patient with Non-Hodgkin’s Lymphoma: A Case Report.

17α‑hydroxylase/17,20‑lyase deficiency (17OHD) is a rare form of congenital adrenal hyperplasia that causes decreased cortisol and sex steroid levels and leads to high production of adrenocorticotropic hormone. Although affected patients have absolute cortisol deficiency, they do not show clinical signs of cortisol deficiency or hyperpigmentation. These patients most commonly present with delayed puberty and amenorrhea at late pubertal age. Impaired production of sex steroids leads to ambiguous or female external genitalia in affected 46, XY individuals. In this report, we describe a patient w

Differential Roles of Cystathionine Gamma-Lyase and Mercaptopyruvate Sulfurtransferase in Hapten-Induced Colitis and Contact Dermatitis in Mice.

Hydrogen sulfide (H2S) has been shown to act as both anti-inflammatory and pro-inflammatory mediators. Application of H2S donors generally protects against inflammation; however, experimental results using mice lacking endogenous H2S-producing enzymes, such as cystathionine γ-lyase (CTH) and mercaptopyruvate sulfurtransferase (MPST), are often contradictory. We herein examined two types of model hapten-induced inflammation models, colitis (an inflammatory bowel disease model of mucosal immunity) and contact dermatitis (a type IV allergic model of systemic immunity), in CTH-deficient (Cth-/-) a

Sphingosine-1-Phosphate (S1P) Lyase Inhibition Aggravates Atherosclerosis and Induces Plaque Rupture in ApoE-/-Mice.

Altered plasma sphingosine-1-phosphate (S1P) concentrations are associated with clinical manifestations of atherosclerosis. However, whether long-term elevation of endogenous S1P is pro- or anti-atherogenic remains unclear. Here, we addressed the impact of permanently high S1P levels on atherosclerosis in cholesterol-fed apolipoprotein E-deficient (ApoE-/-) mice over 12 weeks. This was achieved by pharmacological inhibition of the S1P-degrading enzyme S1P lyase with 4-deoxypyridoxine (DOP). DOP treatment dramatically accelerated atherosclerosis development, propagated predominantly unstable pl

β3 Relaxant Effect in Human Bladder Involves Cystathionine γ-Lyase-Derived Urothelial Hydrogen Sulfide.

It is now well established that the urothelium does not act as a passive barrier but contributes to bladder homeostasis by releasing several signaling molecules in response to physiological and chemical stimuli. Here, we investigated the potential contribution of the hydrogen sulfide (H2S) pathway in regulating human urothelium function in β3 adrenoceptor-mediated relaxation. The relaxant effect of BRL 37344 (0.1-300 µM), a selective β3 adrenoceptor agonist, was evaluated in isolated human bladder strips in the presence or absence of the urothelium. The relaxant effect of BRL 37344 was signifi

Inhibition of nitric oxide synthase or cystathionine gamma-lyase abolishes leptin-induced fever in male rats.

Leptin is an adipokine that regulates energy balance and immune function. Peripheral leptin administration elicits prostaglandin E₂-dependent fever in rats. The gasotransmitters nitric oxide (NO) and hydrogen sulfide (H₂S) are also involved in lipopolysaccharide (LPS)-induced fever response. However, there is no data in the literature indicating if these gasotransmitters have a role in leptin-induced fever response. Here, we investigate the inhibition of NO and H₂S enzymes neuronal nitric oxide synthase (nNOS), inducible nitric oxide synthase (iNOS), and cystathionine γ-lyase (CSE) in leptin-i
