Advanced Glycation End Products-Induced Alzheimer's Disease and Its Novel Therapeutic Approaches: A Comprehensive Review.

Advanced glycation end products (AGEs) accumulate in the brain, leading to neurodegenerative conditions such as Alzheimer's disease (AD). The pathophysiology of AD is influenced by receptors for AGEs and toll-like receptor 4 (TLR4). Protein glycation results in irreversible AGEs through a complicated series of reactions involving the formation of Schiff's base, the Amadori reaction, followed by the Maillard reaction, which causes abnormal brain glucose metabolism, oxidative stress, malfunctioning mitochondria, plaque deposition, and neuronal death. Amyloid plaque and other stimuli activate mac…

Dietary acrylamide exposure increases susceptibility to ulcerative colitis: A comprehensive analysis from network toxicology to in vivo experimental validation.

Acrylamide (AA) is a contaminant present in foods cooked at high temperatures, and its formation is associated with the Maillard reaction. Ulcerative colitis (UC) is a chronic inflammatory disease of the colon mucosa with a complex pathogenesis. This study investigated the effects of AA exposure on UC and its potential mechanisms. First, network toxicology was employed to identify genes that are targets associated with both the toxicity of AA and the pathogenesis of UC, which are enriched in pathways associated with inflammatory cascades and immune cell recruitment. Then, Mendelian randomizati…

Effect of different preparation methods on the quality of carrot preserves: Chemical composition, textural properties, and volatile compounds.

Systematic comparisons of quality impacts of various carrot preserve preparation techniques remain scarce. We compared candied carrot preser … while NEBCP showed increased levels of acids, aldehydes, lactones, and heterocyclic compounds from Maillard reactions and carotenoid degradation. Sensory evaluation indicated that NEBCP scored higher than CCP and SSCP. These findings provide insights for developing low-sugar, low-salt nutritious carrot preserves.

Rapid increase in human life expectancy: will it soon be limited by the aging of elastin?

The postponement of the most frequent age-related diseases stimulated speculations of the possibility of "dying of old age". The selective d … y system-is a cautious yes, with however the caveat that other, independent mechanisms, such as the Maillard reaction, can also interfere with and limit further the functional value of such vital physiological functions.

Effects of drying time on physicochemical properties and volatile organic compounds of pork jerky revealed by GC-IMS and interpretable machine learning.

Drying time affects dehydration, color development, Maillard reactions, and volatile organic compounds (VOCs) in pork jerky. Pork jerky was dried at 80 °C for 4-6 h, with marinated undried samples as the RAW control. Physicochemical properties, free amino acids, and volatile fingerprints were evaluated. Gas chromatography-ion mobility spectrometry (GC-IMS) detected 53 volatile signals, whose profiles changed markedly during drying. Support vector regression (SVR) and extra trees regression (ETR) were developed to predict moisture content, absorbance at 420 nm, fluorescence intensity, and L*, a…

Trade-off between chemical safety and bioactivity during cocoa roasting revealed by multivariate modeling.

Cocoa roasting triggers coupled thermal reactions that shape bioactive composition and promote neoformed contaminants (NFCs), but frameworks linking precursor dynamics and bioactivity remain limited. Here, multivariate relationships among Maillard precursors, physicochemical variables, bioactive descriptors and NFCs were evaluated during cocoa roasting at 110-130 °C across six roast progression (RP) stages. RP was the main driver of chemical change: sucrose decreased by >95%, while glucose and fructose peaked at RP1-RP2 (up to ∼2500 mg/g) before declining, alongside a decrease in asparagine to

Differentiating the processing degree of animal material by mass spectrometry: A feasibility study on porcine and bovine blood-derived feed ingredients.

Processing food and feed sets off a variety of reactions (Maillard, (lipid) oxidation), which may be traced by covalent changes to e.g. proteins. Whereas these have been previously exploited to differentiate raw from processed milk, this proof-of-principle study aims to distinguish between differently processed blood (fractions). According to European legislation, these may enter the feed chain as typically spray-dried 'blood product' (BP) or as 'blood meal' (BM) when pressure-sterilized prior to drying. Therefore, porcine (BM: 9, BP: 19) and bovine materials (BM: 2, BP: 16) were analyzed in d

Formulation of Emulsion Gels Stabilized by Soy Protein Isolate and Gellan Gum Conjugates for 3D Printing and β-Carotene Encapsulation Applications.

Soy protein isolate-gellan gum (SPI-GG) conjugates were prepared at different mass ratios to investigate how protein-polysaccharide composition affects the physicochemical, emulsion, rheological, and functional properties of CEGs. Increasing the GG proportion enhanced the degree of glycation and Maillard browning while producing opposite trends in solubility and surface hydrophobicity. All SPI-GG CEGs exhibited smaller droplets, more negative ζ-potential values, and higher emulsifying activity and stability than the SPI control. Among the formulations, S1G1 showed the smallest and most uniform

A Comparative Analysis of Maillard-Derived α-Dicarbonyl Compounds and Advanced Glycation End Products in Fresh, Wet, Kibble, and Freeze-Dried Dog Foods.

Thermal processing of pet foods can lead to the formation of harmful Maillard reaction products (MRPs), which may compromise nutritional quality and pose health risks to companion animals. This study compared the levels of MRPs in 41 commercial dog foods, including fresh (n = 15), kibble (n = 13), wet (n = 10), freeze-dried (n = 2), and cold-pressed (n = 1) types. Acrylamide was found in only two kibble samples (40 and 76 µg/kg), while 5-hydroxymethyl-2-furfural and 2-furfural were not detected in any sample. Fresh foods had significantly higher lysine content (56 ± 13 g/kg dry matter [DM]) an…

Association of triglyceride glucose-body mass index and hemoglobin glycation index with heart failure prevalence in hypertensive populations: a study across different glucose metabolism status.

BACKGROUND: The Triglyceride glucose-body mass index (TyG-BMI) and hemoglobin glycation index (HGI) are well-established surrogate markers for insulin resistance. Nevertheless, the extent to which these markers offer additive predictive value for heart failure (HF) prevalence in hypertensive populations, and their predictive utility across various diabetic statuses, remains to be clarified. Consequently, this study aimed to explore the independent and synergistic effects of TyG-BMI and HGI on HF risk among individuals with different diabetic statuses. METHODS: Data from the study population (

Serum levels of high mobility group box-1 protein (HMGB1) and soluble receptors of advanced glycation end-products (RAGE) in depressed patients treated with electroconvulsive therapy.

AIMS: High mobility group box-1 (HMGB1) is one of the damage-associated molecular patterns produced by stress and induces inflammatory responses mediated by receptors of advanced glycation end-products (RAGE) on the cell surface. Meanwhile, soluble RAGE (sRAGE) exhibits an anti-inflammatory effect by capturing HMGB1. Animal models have shown upregulation of HMGB1 and RAGE in the brain or blood, suggesting the involvement of these proteins in depression pathophysiology. However, there have been no reports using blood from depressed patients, nor ones focusing on HMGB1 and sRAGE changes associat

Mechanism of H2S in Inhibiting the Senescence and Browning of Fresh-Cut Potatoes.

The market for fresh-cut fruits and vegetables is gradually expanding and is popular among consumers, but fresh-cut fruits and vegetables are highly susceptible to browning, causing a decrease in their quality and nutrition. Although anti-browning reagents and cryopreservation methods are often used for fresh-cut fruits and vegetables, the effects are not satisfactory. In this paper, hydrogen sulfide (H2S) donor NaHS solution was used for fumigation of fresh-cut potatoes to explore the mechanism of H2S signaling on the browning of fresh-cut potatoes at the biochemical level. Fresh-cut potatoes

Advanced glycation end products overload might explain intracellular cobalamin deficiency in renal dysfunction, diabetes and aging.

Advanced glycation end products (AGEs) contribute to aging. Cobalamin (Cbl) is required for cell growth and functions, and its deficiency causes serious complications. Diabetics and renal patients show high concentrations of Cbl, but metabolic evidence of Cbl deficiency that is reversible after Cbl treatment. Cbl might be sequestered in blood and cannot be delivered to the cell. Megalin mediates the uptake of transcobalamin-Cbl complex into the proximal tubule cells. Megalin is involved in the uptake and degradation of AGEs. In aging, diabetes or renal dysfunction, AGEs might overload megalin

Lipoxidation products as biomarkers of oxidative damage to proteins during lipid peroxidation reactions.

Oxidative stress is implicated in the pathogenesis of numerous disease processes including diabetes mellitus, atherosclerosis, ischaemia reperfusion injury and rheumatoid arthritis. Chemical modification of amino acids in protein during lipid peroxidation results in the formation of lipoxidation products which may serve as indicators of oxidative stress in vivo. The focus of the studies described here was initially to identify chemical modifications of protein derived exclusively from lipids in order to assess the role of lipid peroxidative damage in the pathogenesis of disease. Effects of Coffee Extracts with Different Roasting Degrees on Antioxidant and Anti-Inflammatory Systems in Mice.

Coffee roasting affects the taste, color, and aroma of coffee. The Maillard reaction, a major reaction during the roasting process, produces melanoidin, which affects the overall antioxidant capacity and anti-inflammatory effects of coffee. In this experiment, coffee roasting was divided into four degrees: Light, Medium, City, and French. To examine the in vivo antioxidant and anti-inflammatory effects of coffee extracts with different roasting degrees, we used 10-week-old male C57BL/6 mice. Mice were pre-treated with coffee extracts for 10 days by oral gavage (300 mg/Kg.B.W). After the last p…

Dietary oxidized frying oil activates hepatic stellate cells and accelerates the severity of carbon tetrachloride- and thioacetamide-induced liver fibrosis in mice.

Deep-frying is a common cooking practice worldwide, and after repeated heating's, the oil undergoes various chemical reactions, including hydrolysis, polymerization, lipid oxidation, and the Maillard reaction. Studies have pointed out that oxidized dietary frying oil may cause teratogenesis in mice and increase cancer and cardiovascular risks. The liver is the main organ involved in dietary nutrient catabolism, detoxification, bile production, and lipid metabolism. Nevertheless, the effects of oxidized frying oil exposure on the activation of hepatic stellate cells (HSCs) and liver fibrosis ar…
