Placental extracts induce the expression of antioxidant enzyme genes and suppress melanogenesis in B16 melanoma cells.

One of the activities of placental extracts (PEs) is skin-whitening effect, but the physiological and genetic mechanism for this effect has not yet been clarified. Here, we focus on PE as a regulator of antioxidant enzyme genes. Porcine PE was prepared, and its activity was investigated in B16 melanoma cells. PE treatment decreased the melanin content of UV-irradiated B16 cells in a dose-dependent manner. PE directly reduced the enzyme activity of tyrosinase in a cell-free assay. In addition, PE treatment increased the gene expression of cytosolic superoxide dismutase (SOD-1), extracellular SO

The cytotoxicity and apoptosis induced by 4-tertiary butylphenol in human melanocytes are independent of tyrosinase activity.

It has been known for several decades that cutaneous depigmentation, i.e., contact/occupational vitiligo, can be caused by some phenolic derivatives that have a similar structure to tyrosine. Among these phenolic depigmenting agents, 4-tertiary butylphenol is the most potent. The cutaneous depigmentation induced by phenolic derivatives results from the loss of functional melanocytes. Tyrosinase is a melanocyte specific copper-containing enzyme that catalyzes the conversion of the amino acid tyrosine, through a complex series of intermediates, to melanin. In this study we tested the hypothesis

Qualitative and quantitative catalog of tyrosinase alternative transcripts in normal murine skin melanocytes as a basis for detecting melanoma-specific changes.

The decline in cell differentiation commonly associated with malignant progression may be due in part to an increase in alternative splicing of the pre-mRNAs of tissue-specific genes. As a necessary basis for investigating this possibility in a murine model of cutaneous melanoma, the complete qualitative and quantitative inventory of alternative transcripts was sought for the tyrosinase gene in normal mouse skin melanocytes, as this gene plays a key role in melanization. Of 111 alternative mRNAs predicted from known splice sites in the gene, 19 isoforms were detected, and their abundances dete

Phenoloxidase activity in the reproductive system of Biomphalaria glabrata: role in egg production and effect of schistosome infection.

Infection by larval trematodes often causes a cessation of egg production in its molluscan intermediate host and is referred to as parasitic castration. Because phenoloxidase (PO) has been shown to be involved in egg formation in other invertebrate species, we investigated the role of PO in normal egg production in the snail, Biomphalaria glabrata, and the effects of Schistosoma mansoni infection on the PO pathway in this snail. Our data showed that PO activity in the albumen gland (AG) is initially expressed when snails reach a size of approximately 8 mm in shell diameter and continues to inc

Zinc and copper metabolism in oculocutaneous albinism in the Caucasian.

Abnormalities in the zinc and copper metabolism have been described in some pigmentary retinopathies. In these eye disorders changes in the Retinal Pigment Epithelium are a common finding. Albinism is a manifestation of a metabolic disorder with abnormalities of pigment formation which also occurs in this mentioned retinal layer. Recently, we described abnormalities in the metabolism of these metals in black albino patients. In them, elevated values of ceruloplasmin were found. In the present study we performed a similar work among Caucasians. When both the Caucasian albino and control groups

Albinism. Recent advances.

There have recently been several fundamental advances in our understanding of albinism, both in animals and in man. These advances have been in the division of oculocutaneous albinism into tyrosinase-negative and tyrosinase-positive forms, the demonstration of a widespread disturbance in the structure of melanosomes in X-linked ocular albinism, the description of the condition now known as autosomal recessive ocular albinism, and a peculair anatomical abnormality of the visual pathways which appears to be present in all forms of oculocutaneous and ocular albinism. Evidence has been presented w

Ultrasound-assisted deep eutectic solvent extraction of tyrosinase inhibitors from lotus seed peel powder.

Lotus seed peel powder (LSP), a major by-product of lotus seed processing, is rich in bioactive constituents but remains largely underutilized. Conventional aqueous and ethanolic extraction methods for LSP are constrained by low extraction efficiency and poor selectivity. In this study, an ultrasound-assisted deep eutectic solvent (DES) extraction (UADE) strategy was developed to recover tyrosinase inhibitors from LSP. UADE significantly enhanced flavonoid and polyphenol extraction compared with traditional methods. Among the tested DESs, the L-proline/lactic acid system exhibited the highest

Metabolome Analysis and Functional Activities of Euphorbia royleana Boiss. Indicating Potential Health Benefits.

Euphorbia royleana (Euphorbiaceae) is an important succulent species and commonly known as Royle's spurge. It is used to treat inflammation, paralysis, and brain-related problems. The literature revealed that E. royleana was not well explored for phytochemical diversity and health benefits. Therefore, the current study was focused on investigating solvent-dependent chemical diversity and bioactive properties across stems, leaves, and reproductive tissue (flowers and seeds). Ethanol, 50% ethanol, and water extracts were prepared, in which the water extract of leaves showed the highest extractio

Thioxo-Based Kojic Acid Derivatives as Potent Tyrosinase Inhibitors With Antitubercular and Antidermatophytic Potential.

A novel series of sulfur-containing analogs derived from the 3-hydroxy-6-methyl-2-((4-phenylpiperazin-1-yl)methyl)-4H-pyran scaffold was synthesized. Spectroscopic techniques and elemental analysis confirmed their structures. The biological potential of these compounds, along with their previously reported bioisosteres, was assessed as tyrosinase inhibitors, as well as antitubercular and antidermatophytic agents. In all derivatives where the carbonyl group is converted to a thiocarbonyl group, tyrosinase inhibition increases significantly, with the most effective compounds being identified as

Effects of Co-Cultivation on the Phenolic Composition and Bioactive Properties of Basidiomycete Mycelia: Antioxidant, Photoprotective, Anti-Tyrosinase, and In Silico Evidence.

Basidiomycetes produce a wide range of bioactive metabolites, yet the effects of fungal co-cultivation on phenolic composition and associated biological properties remain poorly understood. This study investigated the phenolic composition and antioxidant, photoprotective, and anti-tyrosinase activities of mycelial extracts obtained from four basidiomycetes cultivated either alone or in paired cultures. Phenolic compounds were characterized by HPLC, and the identified metabolites were further evaluated using target prediction, enrichment analyses, PASS prediction, ADME profiling, and toxicity a

Molecular Characterization, Activation Mechanism, and Antimelanosis Implications of Polyphenol Oxidase in Litopenaeus vannamei.

Polyphenol oxidase (PPO) plays a central role in the quality deterioration of shrimp, particularly in melanosis development. However, the mechanisms triggering melanosis in Litopenaeus vannamei (L. vannamei) remain poorly understood. This study purified natural PPO from L. vannamei and investigated its molecular characteristics and activation mechanism. PPO exhibited phenoloxidase activity in the form of a 210 kDa heterotetramer. Under heat-denaturing conditions, it dissociated into two 62 kDa PPOα chains and two 45 kDa PPOβ chains. During cold storage, the onset of melanosis was accompanied b

Tyrosinase inhibitory mechanism of pyrimidine-thiols and their potential application in the anti-browning of fresh-cut apples.

The tyrosinase inhibitory activity and inhibition mechanism of pyrimidine-2-thiol (PT) and pyrimidine-2,4-dithiol (PDT) were investigated. In the anti-tyrosinase activity evaluation assay, PT exhibited 2-fold stronger activity than kojic acid in the presence of L-Tyrosine and L-Dopa, and PDT showed 13 times higher activity than kojic acid, respectively. Kinetic analysis revealed PT and PDT acted as mixed inhibitors. Copper ions chelation, fluorescence quenching, 1H NMR titration, and molecular docking assays manifested PT and PDT could participate in chelation with copper ions in the tyrosinas

The in cellular and in vivo melanogenesis inhibitory activity of safflospermidines from Helianthus annuus L. bee pollen in B16F10 murine melanoma cells and zebrafish embryos.

Melanin, synthesized by tyrosinase (TYR), is a natural pigment essential for skin protection and pigmentation. However, excessive melanin production can cause dermatological disorders. Safflospermidines, comprised of safflospermidine A and B isomers isolated from sunflower (Helianthus annuus L.) bee pollen, were shown to exhibit a strong in vitro TYR inhibitory activity against mushroom TYR. However, their anti-melanogenesis activity in cellular and in vivo models remains unexplored. This study firstly evaluated the effects of these safflospermidines on melanogenesis in α-melanocyte stimulatin

Disrupting melanin transfer: innovative strategy for anti-pigmentation drug discovery.

The excessive synthesis of melanin leads to skin hyperpigmentation. While tyrosinase activity inhibition has demonstrated efficacy in ameliorating hyperpigmentation, its effectiveness remains limited, and tyrosinase inhibitors may induce irritant contact dermatitis. Therefore, there is an imperative need to develop safer and more potent anti-pigmentation agents. Melanin transfer inhibition represents a novel therapeutic strategy for treating hyperpigmentation. This review systematically elucidates the complete process of melanin transfer and its underlying mechanisms. Furthermore, it provides

Computer-Aided Virtual Screening and In Vitro Validation of Biomimetic Tyrosinase Inhibitory Peptides from Abalone Peptidome.

Hyperpigmentation is a medical and cosmetic problem caused by an excess accumulation of melanin or the overexpression of the enzyme tyrosinase, leading to several skin disorders, i.e., freckles, melasma, and skin cancer. Tyrosinase is a key enzyme in melanogenesis and thus a target for reducing melanin production. Although abalone is a good source of bioactive peptides that have been used for several properties including depigmentation, the available information on the anti-tyrosinase property of abalone peptides remains insufficient. This study investigated the anti-tyrosinase properties of H

Rejuvenation of iron oxides enhances carbon sequestration by the 'iron gate' and 'enzyme latch' mechanisms in a rice-wheat cropping system.

The 'enzyme latch' theory believes that oxygen constraints on phenol oxidase can restrain the activity of hydrolytic enzymes responsible for decomposition, while the 'iron (Fe) gate' theory suggests that Fe oxidation can decrease phenol oxidase activity and enhance Fe-lignin complexation under oxygen exposure. The objective of this study was to explore the roles of the 'enzyme latch' and 'Fe gate' mechanisms in regulating soil organic carbon (SOC) sequestration in a rice-wheat cropping system subjected to six fertilization treatments: control (CT), chemical fertilizer (CF), CF plus manure (CFM

Extracts from European Propolises as Potent Tyrosinase Inhibitors.

Tyrosinase is a key enzyme in the melanogenesis pathway. Melanin, the product of this process, is the main pigment of the human skin and a major protection factor against harmful ultraviolet radiation (UVR). Increased melanin synthesis due to tyrosinase hyperactivity can cause hyperpigmentation disorders, which in consequence causes freckles, age spots, melasma, or postinflammatory hyperpigmentation. Tyrosinase overproduction and hyperactivity are triggered by the ageing processes and skin inflammation as a result of oxidative stress. Therefore, the control of tyrosinase activity is the main g
