Neurogenic arthrogryposis multiplex congenita and velopharyngeal incompetence associated with chromosome 22q11.2 deletion.

We report a case of neurogenic arthrogryposis multiplex congenita and velopharyngeal incompetence in association with a chromosome 22q11.2 deletion in a 5-month-old boy, the only child of a non-consanguineous couple without relevant antecedents. Specifically, polymerase chain reaction amplification of microsatellite markers revealed a noninherited microdeletion in position D22S306. This phenotype has not been reported previously in association with chromosome 22q11.2 deletions, and these findings raise the possibility that at least some cases of neurogenic arthrogryposis multiplex congenita mi

Multiplex PCR to Diagnose Gastrointestinal Infections: Key Limitations in Interpretation.

Multiplex polymerase chain reaction (PCR) panels are increasingly used for the evaluation of acute diarrhea, providing rapid and sensitive detection of a broad range of enteric pathogens. However, their widespread adoption has created important interpretive challenges for clinicians. Positive results may reflect colonization, prolonged shedding, or organisms of uncertain pathogenicity rather than active infection, and current panels do not provide antimicrobial susceptibility or quantitative information. Incomplete pathogen coverage and false-positive detections may further complicate result i…

Optimization of a multiplex RT-PCR assay for the detection of human parechovirus, enterovirus, adenovirus type 14, and respiratory syncytial virus using the panther fusion system.

Influenza-like illnesses (ILI) are a significant cause of morbidity and mortality for children, elderly, and immunocompromised individuals. Several ILIs are caused by viruses of clinical and epidemiological importance, including human parechovirus (HPeV), human enterovirus (EV), human adenovirus 14 (HAdV14), and respiratory syncytial virus (RSV). The Panther Fusion System provides the capability for laboratories to perform fully automated nucleic acid extraction and RT-PCR through design and optimization of a laboratory developed test (LDT) using their Open Access feature. This study reports t

Quantitative Real-Time PCR for Trypanosoma cruzi Infection Diagnosis and Treatment Response Monitoring of Patients with Chagas Disease.

The development of accurate diagnostic tools and surrogate markers of parasitological response to treatment are priorities in Chagas disease (CD) research. For years, the detection of Trypanosoma cruzi DNA by polymerase chain reaction (PCR) has proved to be useful in some clinical scenarios such as acute CD, including cases of vertical transmission and oral outbreaks, CD reactivation in immunosuppressed patients, and post-treatment follow-up. In that sense, the implementation of quantitative real-time PCR (qPCR) assays was an important step in the development of more reliable tools for T. cruz

A Multiplex TaqMan-MGB qPCR Assay for Rapid and Accurate Identification of Four Waterfowl Parvoviruses (cGPV, MDPV, MDGPV, and SBDSV).

Waterfowl parvoviruses (WPVs), including classical goose parvovirus (cGPV), Muscovy duck parvovirus (MDPV), Muscovy duck-origin goose parvovirus (MDGPV), and short beak and dwarfism syndrome virus (SBDSV), are significant pathogens that affect waterfowl flocks worldwide. Due to their high genetic similarity and frequent coinfections, rapid and accurate differentiation of these viruses remains challenging. In this study, we developed a multiplex TaqMan-minor groove binder (MGB) real-time PCR assay for the simultaneous detection and differentiation of cGPV, MDPV, MDGPV, and SBDSV. Specific prime

Rapid diagnosis of viral infections and application to infection control.

BACKGROUND: Rapid and accurate detection of viral infections is crucial in clinical settings, yet traditional quantitative polymerase chain reaction (qPCR) methods, though highly accurate, are typically laboratory-based and require significant time and resources, limiting their effectiveness in point-of-care environments. In this study, we applied a rapid qPCR method, originally developed for severe acute respiratory syndrome coronavirus 2, to a range of viral infections. The primary aim was to establish a novel diagnostic platform that combines mobile qPCR with multiplex detection for rapid,

A rapid and highly sensitive multiple detection of human adenovirus type 3, type 7 and respiratory syncytial virus by recombinase-aided reverse transcription PCR.

BACKGROUND: Polymerase chain reaction (PCR) has been widely used for many pathogen detection. However, PCR technology still suffers from long detection time and insufficient sensitivity. Recombinase-aided amplification (RAA) is a powerful nucleic acid detection tool with high sensitivity and amplification efficiency, but its complex probes and inability of multiplex detection hinder the further application of this technology. METHODS: In this study, we developed and validated the multiplex reverse transcription recombinase-aided PCR (multiplex RT-RAP) assay for human adenovirus 3 (HADV3), hum

Internally Controlled, Multiplex Real-Time Reverse Transcription PCR for Dengue Virus and Yellow Fever Virus Detection.

The differential diagnosis of dengue virus (DENV) and yellow fever virus (YFV) infections in endemic areas is complicated by nonspecific early clinical manifestations. In this study, we describe an internally controlled, multiplex real-time reverse transcription polymerase chain reaction (rRT-PCR) for the detection of DENV and YFV. The DENV-YFV assay demonstrated specific detection and had a dynamic range of 2.0-8.0 log10 copies/μL of eluate for each DENV serotype and YFV. Clinical performance was similar to a published pan-DENV assay: 48/48 acute-phase samples from dengue cases were detected

Multiplex real-time PCR for detecting and typing Clostridium botulinum group III organisms and their mosaic variants.

Botulism is a neuroparalytic disease that can occur in all warm-blooded animals, birds, and fishes. The disease in animals is mainly caused by toxins produced by Clostridium botulinum strains belonging to group III, although outbreaks due to toxins produced by group I and II organisms have been recognized. Group III strains are capable of producing botulinum toxins of type C, D, and C/D and D/C mosaic variants. Definitive diagnosis of animal botulism is made by combining clinical findings with laboratory investigations. Detection of toxins in clinical specimens and feed is the gold standard fo

Alternative quadruplex real-time PCR reactions for detection and discrimination of Streptococcus pneumoniae serotypes within serogroup 6.

UNLABELLED: Streptococcus pneumoniae causes significant morbidity and mortality worldwide, and serotyping is important to assess the burden of disease that is vaccine preventable. For serotyping, the Centers for Disease Control and Prevention (CDC) use a series of 12 real-time multiplex PCRs (rmPCRs) performed in quadruplex reactions; however, rmPCR reaction 5 (rmPCR-5) for serotypes 6A, 6B, 6C, and 6D often failed at low DNA concentrations. This study investigated the cause of rmPCR-5 failure and provided alternative rmPCRs to resolve this issue. Quadruplex rmPCR target sequences were compare

Identification of enterovirus 71 hand, foot and mouth disease from other enteroviruses by microfluidic digital PCR.

Enterovirus71 (EV71) is a major etiological agent of severe hand, foot, and mouth disease (HFMD), which can lead to central nervous system damage in children. Reverse transcription quantitative polymerase chain reaction (RT-qPCR) often produces false-negative results during the early stage of EV71 infection, attributable to the low viral load. To address this limitation, we developed a rapid, precise, and highly sensitive multiplex reverse transcription digital polymerase chain reaction (RT-dPCR) assay for differential diagnosis of EV71-induced HFMD and non-EV71 pathogens. The EV71 RT-dPCR ass

Diving Into the Depths: Unveiling the Main Etiologies of Piscine Lactococcosis With a Novel Multiplex qPCR Assay.

Piscine lactococcosis poses a significant threat to a wide range of cultured and wild fish populations worldwide, typically presenting as acute haemorrhagic septicemia with high morbidity and mortality. Although Lactococcus garvieae was historically considered the sole causative agent of piscine lactococcosis, recent studies have identified L. petauri and L. formosensis as additional, highly pathogenic species. In this study, we developed a novel TaqMan-based multiplex qPCR assay for the simultaneous detection and differentiation of L. garvieae, L. petauri and L. formosensis, following a pange
