Syndrome of osteopetrosis and muscular degeneration associated with cerebro-oculo-facio-skeletal changes.

We describe an infant with cerebro-oculo-facio-skeletal manifestations, radiologic and pathologic findings of osteopetrosis, and severe myopathic degeneration proven on histopathologic study of muscles. The muscle changes appear to be part of the pathogenetic process in this syndrome and the cause of the flexion contractures present at birth. Real-time ultrasonography may prove a useful tool in prenatal diagnosis of this syndrome.

[Changes in skeletal muscles in experimental thyrotoxic myopathy].

In an experimental model of thyrotoxic myopathy in mice certain decrease in average diameter of muscle fibers (MF) has been revealed (by 16%). In 1-20% of the MF various types of focal pathologic reactions (loss of cross and longitudinal striation, glial, glomerular and adipose degeneration, Zenker's necrosis) increasing number, structural changes and position of nuclei are observed. Degree of atrophy and part of the altered fibers depend on duration and severity of thyrotoxicosis. Morphologic disorders localize focally and are not so vast as to be the cause of muscle weakness. The main cause

Experimental (-) emetine myopathy. Ultrastructural and morphometric observations.

The ultrastructural changes induced by (-) emetine hydrochloride were studied in two skeletal muscles of the rat. (-) Emetine was administered at a dose level of 2 mg/kg for periods ranging from one to four weeks. Changes were noted after the one week stage and were progressive with all muscle components appearing to be affected. Myofibrillar changes included Z line streaming, rod formation, myofilament loss and contraction clumping. Mitochondrial degeneration was detected, but the most striking feature of the induced myopathy was the extensive membrane proliferation. The membranes were consid

Generation of a NRAP-overexpressing mutant from a human iPSC line.

Cardiomyopathies are a major contributor to cardiovascular mortality and are frequently linked to abnormalities in intercalated discs, which coordinate mechanical and electrical signaling between cardiomyocytes. The Nebulin-Related Anchoring Protein (NRAP), a key component of these structures, is essential for myofibril formation and force transmission. In various cardiac diseases such as cardiomyopathies with differing genetic mutations, NRAP protein abundance is increased, yet the functional consequences of this expression change remain insufficiently characterized. To investigate the outcom

Genetic resiliency associated with dominant lethal TPM1 mutation causing atrial septal defect with high heritability.

Analysis of large-scale human genomic data has yielded unexplained mutations known to cause severe disease in healthy individuals. Here, we report the unexpected recovery of a rare dominant lethal mutation in TPM1, a sarcomeric actin-binding protein, in eight individuals with large atrial septal defect (ASD) in a five-generation pedigree. Mice with Tpm1 mutation exhibit early embryonic lethality with disrupted myofibril assembly and no heartbeat. However, patient-induced pluripotent-stem-cell-derived cardiomyocytes show normal beating with mild myofilament defect, indicating disease suppressio

The spatial distribution of actin and mechanical cycle of myosin are different in right and left ventricles of healthy mouse hearts.

The contraction of the right ventricle (RV) expels blood into the pulmonary circulation, and the contraction of the left ventricle (LV) pumps blood into the systemic circulation through the aorta. The respective afterloads imposed on the LV and RV by aortic and pulmonary artery pressures create very different mechanical requirements for the two ventricles. Indeed, differences have been observed in the contractile performance between left and right ventricular myocytes in dilated cardiomyopathy, in congestive heart failure, and in energy usage and speed of contraction at light loads in healthy

[Protein-energy malnutrition and hypobaric stress effects on ultrastructure of the heart muscle in rats].

A model that simulated nutritional and hypoxic stress is discussed. Wistar male adult rats were used in four groups: A1, A2, B1 and B2. A1 and A2 groups were fed with a diet whose composition was: 16% protein, 43% carbohydrate, and 6% lipids. Group B1 and B2 received the same type of diet but was restricted to 70% the amount consumed by groups A1 and A2. All groups were fed for 15 days. After these 15 days, the half of both groups were introduced into an hypobaric chamber simulating an altitude of 3,500 m for two hours. Pieces of the rats hearts were used for biochemical and ultrastructural st

Cardiac mitochondrial abnormalities in a mouse model of the fetal alcohol syndrome.

Mitochondrial enzymes and respiration were studied in the hearts of mice exposed to ethanol in utero from gestational Day 8 to parturition. This treatment had previously been shown by electron microscopy to result in myofibril loss and mitochondrial abnormalities. Ethanol was administered to pregnant mice by a liquid diet paradigm and pair-fed dams were used as controls. Ethanol exposure in utero reduced the activities of two mitochondrial inner membrane enzymes, cytochrome c oxidase and succinate dehydrogenase, in the hearts of perinatal mice. Secondly, mitochondrial respiration under both St

An examination of heart proteins by two-dimensional electrophoresis.

We examined specimens from explanted human hearts by two-dimensional electrophoresis. The protocol selected includes: (a) solubilization of the sample in a urea-detergent mix; (b) charge fractionation in the presence of urea and nonionic detergent on a pH 4-10 immobilized pH gradient; (c) size fractionation on a polyacrylamide concentration gradient in the presence of sodium dodecyl sulfate; and (d) staining with silver nitrate. The method is sensitive enough for analysis of biopsies in the 1-3 mg range (wet tissue). We saw, for explanted hearts, variations in the protein pattern with the site

Contributions of electron microscopy to the study of myocardial ischaemia.

Electron-microscope studies of experimental models of myocardial ischaemia have provided basic information on the pathogenesis of hypoxic heart injury. Correlation of ultrastructural changes with biochemical data confirms the importance of catecholamine release and ionic shifts in the early evolution of ischaemic injury. An altered cellular metabolism induced by ischaemia causes rapid depletion of glycogen and is followed quickly by alterations in the nucleus, the mitochondria and the sarcotubular system; the myofibril is the organelle most resistant to hypoxia.Postmortem autolysis mimics earl

Mechanism of enhanced salt taste perception in magnetic field-mediated/gum arabic-induced low-salt myofibrillar protein gel.

This study employed the magnetic field-mediated/gum arabic to prepare low-salt myofibrillar protein (MP) gel and simulated the oral processing of interactions between MP and mucin on the perception of saltiness. The results indicated that the amalgamation of magnetic fields and gum arabic effectively promoted the release of sodium ions, enhanced the apparent viscosity of MP, and increased its binding affinity with mucin. Consequently, this resulted in the prolongation in the retention time of MP on the tongue surface and ultimately improved the perception of saltiness. Besides, spontaneous end

Mechanistic modulation of macromolecular interactions between modified cellulose and myofibrillar protein under low salt conditions: effects on gel properties.

The effects and mechanisms of methylcellulose (MC), carboxymethylcellulose (CMC), hydroxypropyl methylcellulose (HPMC), and microcrystalline cellulose (MCC) on myofibrillar protein (MP) gelation at 0.3 M NaCl were investigated. Results showed that all celluloses enhanced the hardness, springiness, and water-holding capacity of low salt MP gel in a dose-dependent manner. Specifically, MC and HPMC reached optimal performance at 0.9% concentration, whereas CMC and MCC reached their peak at 0.3% addition. In particular, 0.9% MC addition gave the low-salt MP gel its highest hardness (502.98 g), wat

Functional evaluation of a Staphylococcus vitulinus protease: Biochemical properties, structural adaptation, and application in myofibrillar proteins.

An extracellular protease was obtained from Staphylococcus vitulinus and exhibited optimal activity at pH 8.0 and 50 °C. Mn2+ (10 mmol/L) significantly enhanced its activity, while Zn2+, Cu2+, Fe2+, and Fe3+ exhibited inhibitory effects (P < 0.05). The Km and Vmax values of the protease were 9.67 mg/mL and 27.5 U/mL·min, respectively, and the Ea was 16.5 kJ/mol. The protease from S. vitulinus exhibited ΔH* of 13.94 kJ/mol, ΔG* of 39.18 kJ/mol, and ΔS* of -70.9 J/(mol·K). Fluorescence and ultraviolet spectroscopic analyses demonstrated that variations in pH and temperature altered protease acti

Salt reduction in myofibrillar protein gel via inhomogeneous distribution of sodium-containing encapsulated fish oil coacervate: Mucopenetration ability of sodium carboxymethyl cellulose.

The potential application of fish oil microcapsules as salt reduction strategies in low-salt myofibrillar protein (MP) gel was investigated by employing soy protein isolates/carboxymethyl cellulose sodium (SPI-CMC) coacervates enriched with 25 mM sodium chloride and exploring their rheological characteristics, taste perception, and microstructure. The results revealed that the SPI-CMC coacervate phase exhibited the highest sodium content under 25 mM sodium level, albeit with uneven distribution. Notably, the hydrophilic and adhesive properties of CMC to sodium facilitated the in vitro release

Co-stabilization effects of gluten/carrageenan to the over-heated myofibrillar protein: Inhibit the undesirable gel weakening and protein over-aggregations.

High-temperature (120 °C) sterilization is an indispensable process for manufacturing ready-to-eat surimi products, yet risking the denaturation of their myofibrillar proteins (MP), thus significantly reducing the gelling properties. To resolve this problem, herein, a synergistic co-strengthening strategy was designed. The negatively charged polysaccharide carrageenan (CG) was introduced into MP simultaneously with wheat gluten, followed by 120 °C thermal treatment for 30 min. A substantial enhancement in mechanical strength, up to four times greater (from 9.86 to 42.38 g·cm), was observed for
