Nur77, together with Nurr1 and NOR-1, constitutes the NR4A subgroup of orphan nuclear receptors and plays critical roles in cell proliferation, differentiation, migration, and apoptosis. Among them, Nur77 is universally well known to contribute to neurite outgrowth. However, information regarding its regulation and possible function in the peripheral nervous system is still limited. In this study, we performed a sciatic nerve injury model in adult rats and detected an increased expression of Nur77 in the sciatic nerve, which was similar to the expression of Oct-6. Immunofluorescence indicated

Mutations in presenilins are the major cause of early onset familial Alzheimer disease. It has recently been argued that clinical presenilin mutations work as loss-of-function but not toxic gain-of-function. To investigate whether presenilins are involved in the regulation of the distribution of neuronal membrane lipids, we treated neuronally differentiated PC12 cells with DAPT, an inhibitor of presenilin-dependent γ-secretase, and performed lipid analyses of neuritic terminals, which is an initial site of Aβ deposition in brains, using liquid chromatography/electrospray ionization tandem mass

Our goal is to understand the pathogenesis of amyloid-beta (Abeta) deposition in the Alzheimer's disease (AD) brain. We established a cell culture system where central nervous system-derived neuronal cells (CAD cells) produce and accumulate within their processes large amounts of Abeta peptide, similar to what is believed to occur in brain neurons, in the initial phases of AD. Using this system, we show that accumulation of Abeta begins within neurites, prior to any detectable signs of neurodegeneration or abnormal vesicular transport. Neuritic accumulation of Abeta is restricted to a small po…

Microglia in the pineal gland of 1-day-old Sprague-Dawley rats were examined by OX-42 immunocytochemistry and DiI-acetylated-LDL uptake in pineal cell suspension and were found to comprise 3-5% of the total cells in the pineal gland of the neonates. In order to investigate the effects of microglia on pinealocyte structure and function, microglia-depleted and microglia-enriched pineal cell cultures were generated from 1-day-old neonate by fluorescence activated cell sorting (FACS). After 7 days of culture, tissues were processed for either immunocytochemistry for pinealocyte S-antigen and serot

DM-GRASP (GRASP) is an integral membrane glycoprotein expressed on a restricted set of axons in the developing chick nervous system. We demonstrate that purified GRASP supports neurite extension from the subpopulation of neurons that express GRASP. Sensory, sympathetic, and ciliary neurons express GRASP and extend neurites on a GRASP substrate. In contrast, tectal, diencephalic, and retinal cells express GRASP at a very low level, if at all, and do not extend neurites on a GRASP substrate. Moreover, as GRASP is downregulated on sensory neurons during development, they lose the capacity to exte…

Cholinesterases present homologies with some cell adhesion molecules; however, it is unclear whether and how they perform adhesive functions. Here, we provide the first direct evidence showing that neurite growth in vitro from various neuronal tissues of the chick embryo can be modified by some, but not all, anticholinesterase agents. By quantifying the neuritic G4 antigen in tectal cell cultures, the effect of anticholinesterases on neurite growth is directly compared with their cholinesterase inhibitory action. Neurite outgrowth from chick embryonic sensory ganglia explants was induced by activators of protein kinase C and other compounds known to stimulate the hydrolysis of inositol phospholipids. The addition of diacylglycerols, phospholipase C and muscarine chloride to a defined growth medium promoted the outgrowth of dense neurites from ganglia explants which were morphologically distinct from those induced by the phorbol ester TPA. Moreover, these neurite-promoting agents did not enhance non-neuronal cell proliferation and were ineffective in the absence of insulin and/or progesterone.

Although much work has been directed recently towards unravelling the protein chemistry of neurofibrillary tangle (NFT) and senile plaque (SP) components in Alzheimer's disease, the pathogeneses of these lesions remains largely unknown and the problem of their relationship is unresolved. In particular, although paired helical filaments (PHF) have long been documented in SP neurites, we do not know if they are of pathogenetic relevance for the formation of the SP. To investigate the relationship between NFT and SP, we examined antigenic properties of proteins in SP neurites in neocortical tissu…

Electroconvulsive therapy (ECT) is the most effective treatment for severe or intractable depression. The disrupt-potentiate-rewire framework proposes that ECT's therapeutic effects include neural remodeling. We used the neurite orientation dispersion and density imaging (NODDI) model to characterize ECT-related longitudinal white matter (WM) microstructural changes. Fifty-two adults with treatment-resistant major depressive episodes (32 female) from two cohorts underwent multi-shell diffusion MRI and clinical assessment before the first ECT session and after completion of the index course. In this study, we discovered compounds comprising a piperidine skeleton in Piper retrofractum; the compounds exhibit PC12 dendrite elongation activity. Furthermore, we examined the structure-activity relationship by comparing the activities of the isolated compounds 1, 3-7. Among the isolated compounds, two alkaloids (6 and 7) displayed significant neurite outgrowth (NOG) effects in the presence of 5 ng/mL nerve growth factor (NGF) in a dose-dependent manner. Comparison of the activity of 3 (Pipercide; 1.8% NOG positive at 100 µM), 6 (Dehydropipernonaline; 21.1% NOG positive at 100 µM), and 7

Neurite outgrowth is essential for neural circuit formation and is tightly regulated by secreted factors and their receptors. The secreted extracellular domain of the amyloid precursor protein (sAPPα) has been shown to modulate neurite outgrowth. Recently, the gamma amino butyric acid receptor type-B subunit 1a (GABABR1a) was identified as an sAPPα binding partner that mediates its effects on synaptic transmission. Here, we investigated whether this interaction also regulates neurite outgrowth. In mouse primary hippocampal neurons of either sex, the GABABR agonist baclofen reduced axon length;

Duplication of the SNCA gene (SNCADupl), linked to elevated levels of α-synuclein (aSyn), is a genetic cause of Parkinson's disease (PD). Our prior work with human-induced pluripotent stem cell (hiPSC)-derived midbrain neurons generated from patients with PD SNCADupl identified neuritic deficits, accompanied by decreased levels of cytoskeletal element β-tubulin-III (bTubIII). To explore mechanisms underlying these effects in SNCADupl neurons, we used CRISPR-Cas9 to generate isogenic control hiPSCs. Isogenic correction of SNCA dosage restored SNCADupl-induced neurite defects and bTubIII levels.

Neuroinflammation plays an important role in the pathological cascade of Alzheimer's disease (AD) along with aggregation of extracellular amyloid-β (Aβ) plaques and intracellular aggregates of tau protein. In animal models of amyloidosis, local immune activation is centered around Aβ plaques, which are usually of uniform morphology, dependent on the transgenic model used. In postmortem human brains a diversity of Aβ plaque morphologies is seen including diffuse plaques (non-neuritic plaques, non-NP), dense-core plaques, cotton-wool plaques, and NP. In a recent study, we demonstrated that durin

Sensory neurons in the dorsal root ganglia transmit sensory signals from the periphery to the central nervous system. Induced pluripotent stem cell derived models of sensory neurons and dorsal root ganglia are among the most advanced available tools for the study of sensory neuron activity and development in human genetic backgrounds. However, few available reagents modify sensory neuron growth with disease or other model-relevant outcomes. Small molecules, peptides, or oligonucleotides that predictably alter sensory neuron behavior in these contexts would be valuable tools with potentially wi

Hyperbaric oxygen therapy (HBOT) is a promising approach to promote peripheral nerve regeneration. However, the underlying mechanisms of action are poorly understood resulting in inconsistent treatment protocols and unclear indications. The present study evaluated the effects of single or repeated HBOT at different atmospheric pressures, with or without nerve growth factor (NGF) on PC12-cells. Cell proliferation, survival, and neurite outgrowth were analyzed. PC12-cells were exposed to a single session of HBOT or daily sessions over 4 consecutive days. Four groups were studied with or without
