Molecular Engineering at Electrode Interface Enables Efficient Catalytic Nitrogen Reduction.

Molecular catalysts are promising for nitrogen (N2) fixation due to their well-defined sites and tunable coordination, yet most molecules in homogeneous systems reside in the bulk solution, leading to poor catalyst utilization. Here, we introduced a molecular engineering strategy by assembling a functionalized molecular molybdenum complex (PyMoBr3PNP) on graphdiyne (GDY) through π-π interactions. The resulting interface-assembled molecular electrode, PyMoBr3PNP/GDY/carbon paper (CP), catalyzed N2 conversion to ammonia (NH3) using 2,4,6-trimethylpyridinium triflate ([ColH][OTf]) as the proton s

Carboxylation and Oxygenation Kinetics and Large Subunit (rbcL) DNA Sequences for Rubisco From Two Ecotypes of Plantago lanceolata L. That Are Native to Sites Differing in Atmospheric CO2 Levels.

Rubisco, the most prevalent protein on Earth, catalysers both a reaction that initiates C3 carbon fixation, and a reaction that initiates photorespiration, which stimulates protein synthesis. Regulation of the balance between these reactions under atmospheric CO2 fluctuations remains poorly understood. We have hypothesised that vascular plants maintain organic carbon-to-nitrogen homoeostasis by adjusting the relative activities of magnesium and manganese in chloroplasts to balance carbon fixation and nitrate assimilation rates. The following examined the influence of magnesium and manganese on

Nitrogen Sources and Iron Availability Affect Pigment Biosynthesis and Nutrient Consumption in Anabaena sp. UTEX 2576.

Anabaena sp. UTEX 2576 metabolizes multiple nitrogen (N) sources and is deemed a biotechnological platform for chemical production. Cyanobacteria have been identified as prolific producers of biofertilizers, biopolymers, biofuels, and other bioactive compounds. Here, we analyze the effect of different N-sources and Fe availability on the bioproduction of phycobiliproteins and β-carotene. We characterize nutrient demand in modified BG11 media, including data on CO2 fixation rates, N-source consumption, and mineral utilization (e.g., phosphorus (P), and 11 metallic elements). Results suggest tha

Boosting mechanochemical N2-to-amine conversion through nanosizing iron nitride mediators.

The direct conversion of N2 into organonitrogen compounds under mild conditions remains a grand challenge due to the inertness of N2. Mechanochemical looping using iron nitride (FexN) as the nitrogen carrier has emerged as an alternative strategy for N2-to-amine conversion, yet the overall efficiency remains limited by the weak N2 dissociation capability, low N uptake of bulk Fe and relatively slow kinetics. In this study, we employed nanostructured Fe powder as the feedstock to synthesize the FexN mediator through ball milling under a N2 atmosphere. Downsizing the Fe particle size led to a co

Cobalt: An Essential Micronutrient for Plant Growth?

Cobalt is a transition metal located in the fourth row of the periodic table and is a neighbor of iron and nickel. It has been considered an essential element for prokaryotes, human beings, and other mammals, but its essentiality for plants remains obscure. In this article, we proposed that cobalt (Co) is a potentially essential micronutrient of plants. Co is essential for the growth of many lower plants, such as marine algal species including diatoms, chrysophytes, and dinoflagellates, as well as for higher plants in the family Fabaceae or Leguminosae. The essentiality to leguminous plants is

Genetic Diversity and Symbiotic Efficiency of Indigenous Common Bean Rhizobia in Croatia.

Nodule bacteria (rhizobia) in symbiotic associations with legumes enable considerable entries of biologically fixed nitrogen into soil. Efforts are therefore made to intensify the natural process of symbiotic nitrogen fixation by legume inoculation. Studies of field populations of rhizobia open up the possibility to preserve and probably exploit some indigenous strains with hidden symbiotic or ecological potentials. The main aim of the present study is to determine genetic diversity of common bean rhizobia isolated from different field sites in central Croatia and to evaluate their symbiotic e…

High light alters biomacromolecule composition and enhances exopolysaccharide production in Porphyridium aerugineum.

Microalgae of the genus Porphyridium are valuable bioresources for producing sulfated exopolysaccharides (EPS) with industrial potential, yet their productivity under high light (HL) stress is underexplored. Here, the HL acclimation of the red alga Porphyridium aerugineum (550 μmol/m2/s) was investigated through time-course physiological assays, ultrastructural imaging, and multi-omics profiling. HL significantly enhanced biomass accumulation and EPS yield (3.5-fold) despite reduced photosynthetic efficiency. These data indicate altered biomass composition, with proteins and intracellular carb

Environmental Benefit Assessment for the Carbonation Process of Petroleum Coke Fly Ash in a Rotating Packed Bed.

A high-gravity carbonation process was deployed at a petrochemical plant using petroleum coke fly ash and blowdown wastewater to simultaneously mineralized CO2 and remove nitrogen oxides and particulate matters from the flue gas. With a high-gravity carbonation process, the CO2 removal efficiency was found to be 95.6%, corresponding to a capture capacity of 600 kg CO2 per day, at a gas flow rate of 1.47 m3/min under ambient temperature and pressure. Moreover, the removal efficiency of nitrogen oxides and particulate matters was 99.1% and 83.2%, respectively. After carbonation, the reacted fly

Medicago truncatula Molybdate Transporter type 1 (MtMOT1.3) is a plasma membrane molybdenum transporter required for nitrogenase activity in root nodules under molybdenum deficiency.

Molybdenum, as a component of the iron-molybdenum cofactor of nitrogenase, is essential for symbiotic nitrogen fixation. This nutrient has to be provided by the host plant through molybdate transporters. Members of the molybdate transporter family Molybdate Transporter type 1 (MOT1) were identified in the model legume Medicago truncatula and their expression in nodules was determined. Yeast toxicity assays, confocal microscopy, and phenotypical characterization of a Transposable Element from Nicotiana tabacum (Tnt1) insertional mutant line were carried out in the one M. truncatula MOT1 family …

New insights into carbon acquisition and exchanges within the coral-dinoflagellate symbiosis under NH4+ and NO3- supply.

Anthropogenic nutrient enrichment affects the biogeochemical cycles and nutrient stoichiometry of coastal ecosystems and is often associated with coral reef decline. However, the mechanisms by which dissolved inorganic nutrients, and especially nitrogen forms (ammonium versus nitrate) can disturb the association between corals and their symbiotic algae are subject to controversial debate. Here, we investigated the coral response to varying N : P ratios, with nitrate or ammonium as a nitrogen source. We showed significant differences in the carbon acquisition by the symbionts and its allocation

Endophytic Bradyrhizobium spp. isolates from sugarcane obtained through different culture strategies.

Brazilian sugarcane has been shown to obtain part of its nitrogen via biological nitrogen fixation (BNF). Recent reports, based on the culture independent sequencing of bacterial nifH complementary DNA (cDNA) from sugarcane tissues, have suggested that members of the Bradyrhizobium genus could play a role in sugarcane-associated BNF. Here we report on the isolation of Bradyrhizobium spp. isolates and a few other species from roots of sugarcane cultivar RB867515 by two cultivation strategies: direct isolation on culture media and capture of Bradyrhizobium spp. using the promiscuous legume Vigna…

EFTEM cytochemistry and sexual dimorphism of secretory granules in male and female hamster submandibular glands.

After glutaraldehyde fixation followed by osmium tetroxide postfixing, the secretory granules of acinar cells in male hamster submandibular glands (SGs) exhibit a characteristic bipartite substructure, with an electron-lucid rim and a more electron-dense central core. In female hamsters, the reverse is seen, with the larger portion of the granules forming an electron-lucid core and an outer electron-dense crescent rim. In the present study of endogenous peroxidase (PO) activity of male and female hamster SGs, secretory granules in the acinar cells were studied by DAB cytochemical technique. In

Melatonin Alleviates Drought Stress in Sweet Sorghum Seedlings via Protection of Photosynthetic Apparatus and Carbon-Nitrogen Metabolism.

Sweet sorghum, a high-quality forage and energy crop, is significantly affected by drought, the primary abiotic stress impacting its growth. Melatonin (MT) has emerged as a crucial signaling molecule in plant responses to abiotic stress. This study investigates the role of melatonin in enhancing drought tolerance in sweet sorghum, specifically using the 'Dali Shi' variety under polyethylene glycol (PEG)-induced drought conditions. Our findings demonstrate that exogenous melatonin application significantly increased proline content (by 27.76% and 5.95% under mild and moderate drought, respectiv

Bacterial and Fungal Communities Associated with the Ectomycorrhizospheric Soil and Stem Endosphere of the Mycoheterotrophic Plant Monotropa uniflora.

The mycoheterotrophic plant Monotropa uniflora relies on fungal symbionts for carbon and nutrient acquisition. However, its interactions with other microbial groups, beyond ectomycorrhizal fungi, remain unexplored. Here, we characterized bacterial and fungal communities associated with M. uniflora across two compartments: ectomycorrhizospheric soil linked to the mycorrhizal network and the surface-sterilized lower stem endosphere. Microbial community composition was assessed using high-throughput amplicon sequencing of the bacterial 16S rRNA gene and the fungal ITS region. Fungal richness was

Soil pH and Nitrogen Content Drive the Succession of RubisCO-Harboring Microbial Communities Across Picea asperata Plantation Ages.

Autotrophic carbon-fixing microbes can assimilate atmospheric carbon dioxide into biomass via the Calvin-Benson-Bassham (CBB) cycle (their primary carbon fixation pathway), thereby reinforcing soil carbon sequestration in the plantation ecosystem; however, the succession of RubisCO-harboring microbial communities across stand ages remains poorly understood. Here, we investigated the community succession of microbes carrying the gene encoding RubisCO, a key enzyme in the CBB cycle, across a stand-age chronosequence in a Picea asperata plantation ecosystem. Our results revealed a progressive dec

Physicochemical and microbial regulators of contrasting diffusive greenhouse gas fluxes in Danjiangkou Reservoir.

Reservoirs play a key role in the global greenhouse gas (GHG) balance through carbon and nitrogen cycling, yet the spatiotemporal variation in GHG fluxes from large drinking-water reservoirs and the underlying regulatory mechanisms remain unclear. This study investigated the seasonal dynamics of CO2, CH4, and N2O diffusive fluxes in Danjiangkou Reservoir and examined their physicochemical and microbial associations. In August, when water levels were low and water temperatures were high, the mean CO2 flux was close to zero (5.59 ± 10.74 mg m-2·h-1), with both source and sink fluxes observed amo

Insights into the alleviating effects of exogenous α-ketoglutaric acid on free nitrous acid stress in Tribonema minus and ammonium removal in wastewater.

α-Ketoglutaric acid (α-KG) is a key intermediate metabolite in carbon and nitrogen metabolism, playing important roles in regulating microbial stress tolerance. However, its potential to mitigate free nitrous acid (FNA) toxicity, a primary inhibitor of microalgae-nitrifier consortium, has remained unexplored. This study investigated the effects of exogenous α-KG onTribonema minusunder FNA stress and evaluated its application in wastewater treatment. Results showed that 1.0 mmol/L α-KG markedly alleviated FNA stress, but a higher concentration (2.0 mmol/L) was less effective. α-KG mitigated FNA

Unique pioneer microbial communities exposed to volcanic sulfur dioxide.

Newly exposed volcanic substrates contain negligible amounts of organic materials. Heterotrophic organisms in newly formed ecosystems require bioavailable carbon and nitrogen that are provided from CO2 and N2 fixation by pioneer microbes. However, the knowledge of initial ecosystem developmental mechanisms, especially the association between microbial succession and environmental change, is still limited. This study reports the unique process of microbial succession in fresh basaltic ash, which was affected by long-term exposure to volcanic sulfur dioxide (SO2). Here we compared the microbial
