WNT10A-SMOC2-LRP4 network affects permanent tooth development via potential tooth-bone interaction.

BACKGROUND: Permanent tooth loss is a key feature of non-syndromic tooth agenesis (NSTA). However, whether this is influenced by the mandibular environment remains unclear. We report a case of simultaneous variations in the tooth agenesis-associated genes WNT10A and SMOC2 and the bone formation-associated gene LRP4, exploring the potential impact of the mandibular environment on tooth agenesis. METHODS: A 19-year-old patient with NSTA underwent clinical examination and genetic analysis using whole-exome sequencing and Sanger sequencing. We reanalyzed single-cell RNA-seq datasets from human mo

The potential role of chromodomain helicase DNA-binding protein 3 in defining the cervical width by regulating the early growth stage of the apical papilla during tooth development.

OBJECTIVE: This study aimed to evaluate the role of the chromodomain helicase DNA-binding protein 3 (CHD3) in tooth morphogenesis in Chd3 knockout mice. METHODS: Chd3 knockout mice were generated using the CRISPR-Cas9 method. Mandibular first molars were extracted from the mice and their littermates and morphometrically analyzed. Subsequent histological and immunohistochemical analyses of teeth were performed at each developmental stage. Chd3 knockdown in mesenchymal cells from the dental papilla (mDP) and Hertwig's epithelial root sheath (HERS) was performed by Chd3 shRNA transduction or a c

The developmental relationship between teeth and dermal odontodes in the most primitive bony fish Lophosteus.

The ontogenetic trajectory of a marginal jawbone of Lophosteus superbus (Late Silurian, 422 Million years old), the phylogenetically most basal stem osteichthyan, visualized by synchrotron microtomography, reveals a developmental relationship between teeth and dermal odontodes that is not evident from the adult morphology. The earliest odontodes are two longitudinal founder ridges formed at the ossification center. Subsequent odontodes that are added lingually to the ridges turn into conical teeth and undergo cyclic replacement, while those added labially achieve a stellate appearance. Stellat

Craniofacial morphology and dental maturity in children with reduced somatic growth of different aetiology and the effect of growth hormone treatment.

Children with reduced somatic growth may present various endocrinal diseases, especially growth hormone deficiency (GHD), idiopathic short stature (ISS), chromosomal aberrations, or genetic disorders. In an attempt to normalize the short stature, growth hormone (GH) is administered to these children. The aim of this literature review was to collect information about the craniofacial morphology and dental maturity in these children and to present the existing knowledge on the effect of GH treatment on the above structures. This review demonstrated that regardless of the origin of the somatic gro

Diseases of the tooth: the genetic and molecular basis of inherited anomalies affecting the dentition.

In humans, inherited variation in the number, size, and shape of teeth within the dentitions are relatively common, while rarer defects of hard tissue formation, including amelogenesis and dentinogenesis imperfecta, and problems associated with tooth eruption are also seen. In many cases, these anomalies occur in isolation, but they can also present as a feature of numerous well-characterized developmental syndromes. Complex reiterative signaling between the epithelium and mesenchyme is a feature of normal tooth development in the embryo, occurring from early patterning through morphogenesis,

Odontogenic cell culture in PEGDA hydrogel scaffolds for use in tooth regeneration protocols.

In order to obtain a tooth-like structure, embryonic oral ectoderm cells (EOE) and bone marrow-derived stem cells (BMSC) were stratified within a synthetic hydrogel matrix (PEGDA) and implanted in the ileal mesentery of adult male Lewis rats. Whole-mount in situ hybridization was used to evaluate the expression of Pitx2, Shh and Wnt10a signals indicative of tooth initiation. In rats, expression of the three markers was present in the oral ectoderm starting at embryonic stage E12.5. which was therefore selected for cell harvesting. Embryos were obtained by controlled service of young female Lew

Cessation in root development: ramifications of 'Stevens-Johnson' syndrome.

The case presented here is of a 15-year-old girl in whom nearly all the teeth except for lower first molars and lower central incisors showed short roots as observed through panoramic radiograph. At the same time there was difference in the length of roots of various teeth. The patient suffered an acute attack of Stevens-Johnson syndrome (SJS) at the age of 8 years and since no other possible explanation for the short roots anomaly could be found, it could be concluded that the cessation in root development may have been caused by the destruction or damage of the epithelial root sheath during

The mixed dentition pantomogram: a valuable dental development assessment tool for the dentist.

The mixed dentition pantomogram is routinely used in paediatric patients. This paper discusses the value of the pantomogram for early identification of problems in dental development during the mixed dentition stage. Aspects regarding dental maturity, leeway space, the sequence of eruption of the permanent teeth, anomalies and the development of the canines will be reviewed.

Amphibian teeth: current knowledge, unanswered questions, and some directions for future research.

Elucidation of the mechanisms controlling early development and organogenesis is currently progressing in several model species and a new field of research, evolutionary developmental biology, which integrates developmental and comparative approaches, has emerged. Although the expression pattern of many genes during tooth development in mammals is known, data on other lineages are virtually non-existent. Comparison of tooth development, and particularly of gene expression (and function) during tooth morphogenesis and differentiation, in representative species of various vertebrate lineages is

Analysis of dental anomalies in the Siberian mole, Talpa altaica (Insectivora, Talpidae).

We re-examined tooth variation in specimens of the Siberian mole, Talpa altaica, from the collection of the Siberian Zoological Museum and discuss the mechanisms of dental evolution. The number of teeth counted in 1789 specimens ranged from 34 to 47, and supernumerary, absent, and connate teeth were observed. The most frequent tooth anomaly was an absent tooth in the premolar region (200 maxillary first premolars and 190 mandibular third premolars), which does not support Fujita and Kirino's terminal reduction hypothesis in the mandible [Fujita T, Kirino T. A permeability barrier to lanthanum and the presence of collagen between odontoblasts in pig molars.

Previous experiments in rat incisors indicate that the odontoblasts form an impermeable barrier which prevents fluid movement between pulp and dentine. The permeability of the odontoblast layer has now been investigated in pig molars which are more analogous to human teeth. The heads and necks of anesthetised piglets were perfused intra-arterially with lanthanum nitrate in Ringer's solution or with Ringer's solution alone. Molar tooth germs were removed, sliced, fixed by immersion and embedded in resin. Ultrathin sections including pulp and dentine were examined by transmission electron micros

Effect of chemotherapy on dental maturity in children with hematological malignancies.

Dental maturity or dental age was determined in 44 children with hematological malignancies treated with chemotherapy. No significant difference was observed between the chronological and dental age in children treated with chemotherapy compared to healthy controls. With regard to the number of erupted permanent teeth, no significant differences could be found between the two groups. The results indicate that chemotherapy given to children with hematological malignancies did not interfere with dental maturity or eruption of permanent teeth.

[Post-natal development of tooth structures in Didelphis albiventris].

The post-natal development of tooth germs has been studied in the opossum or South American marsupial (Didelphis albiventris) between the 2nd and 10th week. Dentinogenesis starts around the 3rd week. The enamel which begins to appear at this stage is deposited until the 8th-10th week. At 10 weeks, the enamel enters the maturation phase. It is unlikely that enamel tubules in these marsupials have a cellular origin. No specific histochemical reaction predicts their appearance which is observed in the mature tooth. This fact suggests that their origin is related either to enamel maturation or to

In vivo and in vitro study of the effects of vitamin A deficiency on rat third molar development.

We have examined the effect of in vivo vitamin A status on subsequent rat third molar formation and mineralization in an in vitro organ culture system. Vitamin A deficiency imposed during an eight-day in vitro period caused effects very similar to those of vitamin A deficiency imposed on rats in vivo. Analysis of the data also demonstrates that retinoic acid is capable of reversing the interference in mineralization of third molars induced by vitamin A deficiency in the organ culture system.
