OBJECTIVES: To directly measure the spatial mapping in the olfactory bulb by odor presentation and by direct electrical stimulation.
STUDY DESIGN: Experimental (animal).
SETTING: University research laboratory.
SUBJECTS AND METHODS: Odor (n = 8) and electrical stimulation (n = 4) of the olfactory bulb in rats were used to demonstrate the spatial mapping of neural responses in the olfactory bulb. Both multiunit responses to odor stimulation and evoked potential responses to localized electrical stimulation were measured in different regions of the olfactory bulb.
RESULTS: Responses that wer

PURPOSE: The purpose of this study was to investigate the relationship between the volume of the olfactory bulb (OB), the depth of the olfactory sulcus (OS), the depth of the fossa olfactoria (FO), and the height of the OB.
METHODS: A total of 54 patients participated (9 women and 45 men; mean [SD] age, 27 [38] y; range, 20-45 y). Magnetic resonance imaging was performed with a 1.5-T system (slice thickness, 1 mm). Measurements of the right and left OB volumes were performed through manual segmentation of the coronal slices. We measured the depth of the FO on the basis of Keros classification

Olfactory dysfunction is a common symptom reported by patients with neurodegenerative disorders such as Alzheimer's and Parkinson's diseases. Despite the knowledge gathered about the pathology of these diseases, little information has been generated regarding the ultrastructure modifications of the granule cells that regulate the information for odor identification. Swollen organelles and nuclear invaginations identified the exposed mice. Necrosis was evidenced at 4th week of exposure, whereas apoptosis arose at 8th week of exposure. A ruffled electron-dense membrane changes were also found. T

In the brain, neuronal activation triggers an increase in cerebral blood flow (CBF). Here, we use two animal models and several techniques (two-photon imaging of CBF and neuronal calcium dynamics, intracellular and extracellular recordings, local pharmacology) to analyze the relationship between neuronal activity and local CBF during odor stimulation in the rodent olfactory bulb. Application of glutamate receptor antagonists or tetrodotoxin directly into single rat olfactory glomeruli blocked postsynaptic responses but did not affect the local odor-evoked CBF increases. This suggests that in o

Periglomerular cells (PG) are interneurons of the olfactory bulb (OB) that modulate the first synaptic relay of the olfactory information from the olfactory nerve to the dendrites of the bulbar principal cells. Previous investigations have pointed to the heterogeneity of these interneurons and have demonstrated the presence of two different types of PG. In the rat OB, type 1 PG receive synaptic contacts from the olfactory axons and are gamma-aminobutyric acid (GABA)-ergic, whereas type 2 PG do not receive synaptic contacts from the olfactory axons and are GABA immunonegative. In this study, we

We analysed the ultrastructural distribution of the m2 muscarinic receptor (m2R) in the rat olfactory bulb (OB) using immunohistochemical techniques and light and electron microscopy. m2R was differentially distributed within the cellular compartments of gamma-aminobutyric acid (GABA)ergic bulbar interneurons. It is located in the gemmules of granule cells and in the synaptic loci of the interneurons of the external plexiform layer, suggesting that m2R activation could modulate the release of GABA from these interneurons onto principal cells by a presynaptic mechanism. By contrast, the recepto

A number of previous studies have indicated that lectin histochemistry is an obvious choice for characterizing the vomeronasal system. However, apparently inconsistent results have been obtained: notably, the affinity with which various lectins bind to the accessory olfactory bulb varies among taxa, even considering closely related species. In the present study, the binding patterns of seven lectins in the rat accessory olfactory bulb, vomeronasal nerves and vomeronasal duct were investigated. The Bandeiraea simplicifolia lectin bound exclusively to the vomeronasal nerve and glomerular layers

Rats were initially trained on a series of odor detection tasks and then received a unilateral olfactory bulbectomy and removal of different parts of the contralateral bulb. After postoperative recovery they were tested for detection of different concentrations of four odors, on a series of odor discrimination tasks and for their ability to acquire a relatively easy and a more difficult odor mixture discrimination task. Groups were formed based on which region of the bulb was intact (regional savings score) and on amount of bulb intact (bulbar savings score). In general, only rats with bulbar

We have investigated the membrane properties and excitatory synaptic transmission of mitral cells in a slice preparation of rat olfactory bulb. In response to intracellular injection of depolarizing current, most mitral cells showed several distinct membrane properties: (1) delayed onset of firing (suggesting the presence of a type of potassium A current); (2) subthreshold oscillation of the membrane potential; and (3) repetitive firing of clustered action potentials during prolonged threshold stimulation. Olfactory nerve (ON) stimulation evoked a long-lasting EPSP in most of the mitral cells.

The effects of dopamine (DA) and fluphenazine (FLU), a phenothiazine DA receptor antagonist, were examined in the salamander olfactory bulb. Field potentials were recorded in the granule cell layer of superfused hemibrain preparations, and the amplitude of the large positive peak was compared following stimulation of the olfactory nerve (ON) and lateral olfactory tract (OT). In preparations superfused with normal amphibian Ringer's solution, the large peak occurred 14-21 ms after either ON or OT stimulation. It therefore appeared to reflect the activation of granule cell synapses with mitral c

We describe the development of the olfactory bulb (OB) glomeruli, focusing on the role of purported astroglial-associated boundary molecules, as potential guidance factors in glomeruli formation. Glomeruli are unusual structures within the OB formed by extremely tight bundles of afferent fibers that fail to grow deeply into the brain. Using olfactory marker protein antibodies, we found that olfactory axons of stage E17-E18 rat embryos were organized into long strands at the outer-most edge of the bulb. By E20-E21 the axons clustered and formed small spheres and by PD1 they gathered into distin

We have investigated the effects of the phencyclidine like-compounds ketamine and MK801 on the evoked field potentials of rat olfactory bulb. Low doses of ketamine (3-6 mg/kg) blocked the inhibition of mitral cells by granule cells evoked by stimulation of lateral olfactory tract fibres or by stimulation of olfactory nerve. This blockade was not accompanied by a decrease in granule cell excitation as revealed by field potential recording. MK801 had a similar effect on the inhibition of mitral cells evoked by stimulation of the lateral olfactory tract. As ketamine does not influence the inhibit

Carnosine-immunoreactive primary olfactory nerve terminals are demonstrated in the glomerular layer of the rat olfactory bulb by immunoelectron microscopy. Asymmetrical synapses between dendrites of mitral/tufted cells and that of periglomerular cells could be observed. In the accessory olfactory system, carnosine-like immunoreactivity is also detected in the vomeronasal neurons.

A coordinated series of immunohistochemical and biochemical analyses have been conducted in the hamster to examine the dependence of substance P and tyrosine hydroxylase (TH) expression by second-order olfactory neurons, and the level of dopamine in the main olfactory bulb (MOB), on the integrity of carnosine- and olfactory marker protein (OMP)-containing primary afferent neurons. Substance P-like immunoreactivity (SPLI) is localized in external tufted cells and centrifugal afferents of the MOB; TH immunoreactivity has a wider distribution, in external tufted, middle tufted, periglomerular, an

1. The synaptic responses of mitral cells have been analysed in intracellular recordings from the isolated olfactory bulb of the turtle. 2. The response of a mitral cell to a single volley in the lateral olfactory tract consisted of and antidromic impulse and a complex hyperpolarizing potential that had the properties of an inhibitory post-synaptic potential. The inhibitory response consisted of two successive components I1 and I2, followed by a prolonged hyperpolarization. 3. High-gain recordings revealed miniature hyperpolarizing potentials during the I1 and I2 responses. Both the miniature

The terminals of centrifugal fibers to the olfactory bulbs of goldfish were studied by electron microscopy after transection of the medial, lateral or entire olfactory tract. The centrifugal fibers originate in the telencephalic hemisphere, pass through both the medial and the lateral olfactory tract, and form synaptic contacts with dendrites in the granule cell layer.

1. The effects of bilateral olfactory bulbectomy, sham-operation and inducement of peripheral anosmia were studied on locomotor activity, passive avoidance acquisition and irritability. 2. Bulbectomized rats were hyperactive, deficient at learning a step-down passive avoidance response and hyperirritable. Peripheral anosmia, induced by intranasal infusion of ZnSO4 solution resulted in no behavioural changes. 3. Chronic pretreatment with amitriptyline (3 and 10 mg/kg) and a tetracyclic antidepressant mianserin (Org GB 94, 5 and 15 mg/kg) reversed the hyperactivity and reduced the learning defic
