The effect of 4-nonylphenol on semen quality, viability of gametes, fertilization success, and embryo and larvae survival in rainbow trout (Oncorhynchus mykiss).

The present study investigated in vivo and in vitro effects of environmental relevant concentrations of 4-nonylphenol (100-750 ng l(-1)) on the reproduction of rainbow trout (Oncorhynchus mykiss). To determine the effect of 4-nonylphenol on semen quality rainbow trout were exposed to three concentrations of 4-nonylphenol in a flow-through system during the spawning period (60 days). At an estimated 4-nonylphenol concentration of 750 ng l(-1) semen production was completely inhibited, at 280 and 130 ng l(-1) the semen production was significantly reduced in comparison to the control. Sperm dens…

Rainbow Trout (Oncorhynchus mykiss) as a Model Organism for Assessing Long-Term Impacts of Municipal Wastewater Pollution on Early Fish Development.

The increasing release of micropollutants from wastewater treatment plant (WWTP) effluents poses growing risks to aquatic ecosystems; however, linking environmental exposure to biological responses under environmentally realistic conditions remains challenging. This study aimed to evaluate biological responses associated with WWTP effluent exposure during the early life stages of rainbow trout (Oncorhynchus mykiss) using an integrated field-based approach. Five floating incubations containing 1000 eggs each were deployed at each of an upstream reference site and a downstream WWTP-affected site…

Hematological and biochemical alterations in rainbow trout Oncorhynchus mykiss during a natural Aeromonas sp. outbreak in high-altitude Ladakh, trans-Himalayas.

An outbreak of bacterial septicemia associated with Aeromonas sp. was investigated in intensive high-altitude aquaculture of rainbow trout, Oncorhynchus mykiss, at the Drass fish farm, Ladakh, during the seasonal transition of May-June 2025. This observational study documents physiological associations rather than experimental causality. A total of 72 fish were sampled: 36 severely diseased and 36 asymptomatic but exposed specimens from adjacent raceways of the same facility. Phenotypic, biochemical, and 16S rRNA gene sequencing of isolate SFB099 identified the agent as Aeromonas sp. with sequ…

Metal-induced inhibition of Na+/K+-ATPase and SERCA in the rainbow trout (Oncorhynchus mykiss) heart: Implications for cardiac ion regulation.

Metal ions are known to inhibit sarcolemmal ion currents and induce cardiac arrhythmias in fish, but their actions are not fully explained by ion channel blockade. To further clarify the effects of di- and trivalent cations on fish heart function, we examined the impact of eight metals on the activities of Na+/K+-ATPase and Sarco/Endoplasmic Reticulum Calcium ATPase (SERCA) in vitro in ventricular homogenates of rainbow trout (Oncorhynchus mykiss). Unfractionated ventricular homogenates were exposed in vitro to eight metals (Al3+, Mn2+, Fe2+, Co2+, Ni2+, Cu2+, Zn2+ or Hg2+) at concentrations o…

Valorization of Mushroom By-Products From Agaricus bisporus and Pleurotus ostreatus as Novel Feed Ingredients for Rainbow Trout (Oncorhynchus mykiss) Diets.

A 60-day feeding trial evaluated meals derived from discarded mushroom biomass, including stems and nonmarketable mushrooms, generated during commercial cultivation and processing of Agaricus bisporus (AB) and Pleurotus ostreatus (PO) as lower-impact alternatives to soybean meal (SBM) in rainbow trout (Oncorhynchus mykiss, BWi: 22.1 ± 0.3 g) diets. Five isoproteic (47%) and isolipidic (23%) diets were tested: a control diet and four diets replacing 25%-50% of SBM with AB or PO meals (AB25, AB50, PO25, PO50). Fish fed the AB25 diet showed growth performance, feed intake (FI), and nutrient utili…

Combined Transcriptomic and Histological Profiling Uncover Hepatic Regulatory Hierarchy of Triploid Oncorhynchus mykiss Under Interactive Salinity, Temperature and Body Weight Regimes.

Salinity, temperature and body weight dominate seawater acclimation in rainbow trout (Oncorhynchus mykiss), yet few studies simultaneously explore their main effects and potential correlative interactive patterns in hepatic responses. A 60-day L9 (33) orthogonal trial was performed on triploid rainbow trout with three gradients of body weight, temperature and salinity. Hepatic transcriptomics revealed that salinity drove global transcriptional remodeling and high salinity induced far fewer DEGs than medium salinity. WGCNA screened a salinity-positive blue module (r = 0.408, p = 0.0346), while …

Transcriptomics, antioxidant enzyme activities, and immune-associated parameter analysis reveal the molecular responses of rainbow trout (Oncorhynchus mykiss) to transportation stress.

To explore how transportation stress affects the physiology of rainbow trout, we measure antioxidant and immunity-related indicators in spleen tissues of adult fish during transportation (at 0, 2, and 6 h) and 24 h following it (r24 h). We report a significant reduction in spleen catalase activity among the 2, 6, and r24 h groups compared with the 0 h group (P < 0.05). Levels of glutathione peroxidase activity and total antioxidant capacity, and lipid peroxide, complement C4, and immunoglobulin M contents first decreased, then increased; complement C3 content and lysozyme activity first increa

Production of sterile trout (Triploids) by chromosome set manipulation using thermal shock treatment in rainbow trout (Oncorhynchus mykiss) from Kashmir Himalayas.

Rainbow trout (Oncorhynchus mykiss) is a promising cultivable fish species with significant potential for expansion. As a cold-water fish belonging to the Salmonidae family, it requires an optimal temperature range of 10-15°C for optimal growth. This study explores a method for producing sterile rainbow trout with maximum survival rates by using heat shock treatment to enhance growth characteristics and improve aquaculture practices. A control group and four heat shock treatments were given at 26°C and 28°C for 10 min, applied 15 and 20 min after the mixing of eggs and milt, using a water bath…

Parasite abundance distribution as a model of host-parasite relationships between monogeneans Gyrodactylus spp. and cage-reared rainbow trout Oncorhynchus mykiss.

Aggregation is a fundamental feature of parasite distribution in the host population, but the biological implications of the aggregation indices used to describe the relationships between the populations of parasites and hosts are not evident. It is speculated that the form of distribution in each case is predicated on the host's varying resistance to the infection, which is hard to control, making it difficult to adequately interpret the index values. This paper examines several cases from trout farms in Russian Karelia to explore the monogenean Gyrodactylus spp. infection in rainbow trout of

Comparative transcriptomics of Atlantic Salmo salar, chum Oncorhynchus keta and pink salmon O. gorbuscha during infections with salmon lice Lepeophtheirus salmonis.

BACKGROUND: Salmon species vary in susceptibility to infections with the salmon louse (Lepeophtheirus salmonis). Comparing mechanisms underlying responses in susceptible and resistant species is important for estimating impacts of infections on wild salmon, selective breeding of farmed salmon, and expanding our knowledge of fish immune responses to ectoparasites. Herein we report three L. salmonis experimental infection trials of co-habited Atlantic Salmo salar, chum Oncorhynchus keta and pink salmon O. gorbuscha, profiling hematocrit, blood cortisol concentrations, and transcriptomic response…

Postprandial response and tissue distribution of the bile acid synthesis-related genes, cyp7a1, cyp8b1 and shp, in rainbow trout Oncorhynchus mykiss.

In mammals, cholesterol 7α-hydroxylase (CYP7A1) and sterol 12α-hydroxylase (CYP8B1) are rate-limiting enzymes in bile acid synthesis. In addition, a small heterodimer partner (SHP) is also known to inhibit bile acid synthesis via the suppression of CYP7A1 and CYP8B1 expression. However, little information is currently available regarding primary structure of the genes involved in bile acid synthesis in fish. We therefore cloned cyp7a1, cyp8b1 and shp genes from rainbow trout and obtained cDNAs encoding two isoforms each of Cyp7a1 (-1 and -2), Cyp8b1 (-1 and -2) and Shp (-1 and -2). Both cyp7a1

Changes in plasma melatonin levels and pineal organ melatonin synthesis following acclimation of rainbow trout (Oncorhynchus mykiss) to different water salinities.

Melatonin has been suggested to play a role in fish osmoregulation, and in salmonids has been related to the timing of adaptive mechanisms during smolting. It has been described that acclimation to different environmental salinities alters levels of circulating melatonin in a number of fish species, including rainbow trout. However, nothing is known regarding salinity effects on melatonin synthesis in the pineal organ, which is the main source of rhythmically produced and secreted melatonin in blood. In the present study we have evaluated, in rainbow trout, the effects of acclimation to differ

Bioavailability of co-supplemented organic and inorganic zinc and selenium sources in a white fishmeal-based rainbow trout (Oncorhynchus mykiss) diet.

The bioavailability of trace elements in fishmeal diets is influenced by their chemical forms and dietary anti-nutritional factors. In formulated fish feed, supplemented organically bound minerals may be more bioavailable than inorganic minerals. A 10-week feeding trial was undertaken with rainbow trout (Oncorhynchus mykiss) to determine whether the inclusion of organically bound selenium (Se) and zinc (Zn) improved uptake and assimilation of these elements compared to commonly used inorganic forms. The three diets tested included a control diet, no added Zn or Se; an organic Se-yeast and Zn-p…

Influence of cycle frequency, muscle strain and muscle length on work and power production of rainbow trout (Oncorhynchus mykiss) ventricular muscle.

This study investigates the effects of cycle frequency, strain and length on work and power output of isolated rainbow trout (Oncorhynchus mykiss) ventricular preparations using the work loop technique. These effects are discussed in the context of the whole heart using analogies with heart rate, stroke volume and end-diastolic volume. Power output was dependent on cycle frequency, increasing threefold beween 0.3 and 1.1 Hz. The frequency for maximum power output was approximately 1.1 Hz, corresponding to the frequency for maximum power in perfused heart experiments. The length for maximum wor…

Identification of potential virulence sites of IHNV G protein and its impact on pathogenicity and immunogenicity of IHNV in rainbow trout (Oncorhynchus mykiss).

Infectious hematopoietic necrosis virus (IHNV) is the major pathogen responsible for acute hemorrhagic disease in salmonids and causes significant economic losses worldwide. The viral glycoprotein (G protein) contains key virulence determinants that regulate pathogenicity and immunogenicity. It is therefore an important target for vaccine development. In this study, G protein sequences of J-genotype IHNV were compared with those of other genotypes to identify amino acid residues potentially associated with differences in virulence. A reverse genetics system was used to rescue recombinant virus
