Extramedullary hematopoiesis: Elucidating the function of the hematopoietic stem cell niche (Review).

Extramedullary hematopoiesis (EMH) occurs under various circumstances, including during embryonic/developmental periods, pathological status secondary to insufficient bone marrow function or ineffective hematopoiesis, in hematological disorders, for example malignancies, as well as stromal disorders of the bone. EMH is characterized by hematopoietic cell accumulations in multiple body locations. Common EMH locations observed in clinical and pathological practice include the spleen, liver, lymph nodes and para‑vertebral regions. Among the various organs associated with EMH, the spleen offers a

Molluscicidal activity of some marine substances against the snail Biomphalaria glabrata (Mollusca, Planorbidae).

Freshwater snails of the genus Biomphalaria play a major role as intermediate hosts of Schistosoma mansoni, the etiologic agent of schistosomiasis. While Biomphalaria spp. control by molluscicides is one of the main strategies to reduce the snail population in infected areas, there are few effective molluscicides commercially available. Natural products may be considered as potentially useful and safe molluscicides. We have evaluated the molluscicidal activity of 12 extracts from ten marine organisms on adult and embryonic stages of Biomphalaria glabrata. Only extracts of the red algae Liagora

Contrast-agent-free visualization of murine embryo-placental vasculature with ultrafast plane-wave ultrasound.

BACKGROUND: In vivo assessment of the embryonic mouse placenta and vasculature is challenging, and ultrasound remains the most commonly used imaging modality. However, even with contrast agents, conventional preclinical ultrasound lacks the temporal resolution required to resolve complex vascular networks. Ultrafast Doppler methods offer a contrast-agent-free approach for visualizing embryonic placental and vascular flow. METHODS: A 15-MHz linear array was used to acquire 0.5 sec of ultrafast ultrasound data at mouse embryonic day 13.5 (E13.5) using a semi-invasive, non-destructive preparatio

Embryonal tumors in the WHO CNS5 classification: A Review.

Embryonal tumors are a heterogenous group of neoplasms mostly defined by recurrent genetic driver events. They have been, previously, broadly classified as either medulloblastoma or supratentorial primitive neuroectodermal tumors (PNETs). However, the application of DNA methylation/gene expression profiling in large series of neoplasms histologically defined as PNET, revealed tumors, which showed genetic events associated with glial tumors. These findings led to the definitive removal of the term "PNET" in the 2016 World Health Organization (WHO) classification of CNS tumors. Moreover, further

Glycogen shunt is essential for submandibular gland morphogenesis.

Glycogen metabolism is an important pathway in energy metabolism, and when cells require energy, glycogen is broken down into glucose-1-phosphate through glycogenolysis, which serves as an intracellular energy source. Recently, the concept of "glycogen shunt" has been proposed to regulate the synthesis, accumulation, and breakdown of glycogen to provide glucose at the appropriate time, particularly in organogenesis. This study demonstrated the timing of the localization of glycogen and molecules related to glycogen metabolism during submandibular gland development using embryonic and postnatal

Infrared laser-induced gene expression for tracking development and function of single C. elegans embryonic neurons.

Visualizing neural-circuit assembly in vivo requires tracking growth of optically resolvable neurites. The Caenorhabditis elegans embryonic nervous system, comprising 222 neurons and 56 glia, is attractive for comprehensive studies of development; however, embryonic reporters are broadly expressed, making single-neurite tracking/manipulation challenging. We present a method, using an infrared laser, for reproducible heat-dependent gene expression in small sublineages (one to four cells) without radiation damage. We go beyond proof-of-principle, and use our system to label and track single neur

Salivary gland gene expression atlas identifies a new regulator of branching morphogenesis.

During organ development, local changes in gene expression govern morphogenesis and cell fate. We have generated a microanatomical atlas of epithelial gene expression of embryonic salivary glands. The mouse submandibular salivary gland first appears as a single mass of epithelial cells surrounded by mesenchyme, and it undergoes rapid branching morphogenesis to form a complex secretory organ with acini connected to an extensive ductal system. Using laser capture microdissection, we collected samples from 14 distinct epithelial locations at embryonic days 12.5, 13.5, 14, and 15, and characterize

Perinatal ablation of the mouse lens causes multiple anterior chamber defects.

PURPOSE: The purpose of this study was to reassess the role of the lens as an "embryonic organizer" of ocular tissues. METHODS: We ablated the lens in mice by lens-specific expression of an attenuated version of diphtheria toxin A subunit(Tox176) driven by a modified crystallin promoter. Alterations in the differentiation programs of ocular tissues were examined by hematoxylin and eosin staining, in situ hybridization, and immunohistochemistry. RESULTS: Transgenic mice in the family OVE1757 exhibited severe microphakia. Apoptotic lens fibers were seen by embryonic day 15 (E15) and the lenses

[Correction of age-related skin changes by methods of cellular and bioregulation therapy].

In the recent years the development of methods of skin revitalization (remodeling)--i.e. of its restoration after damage of any nature (including aging)--attracted the attention of not only cosmetologists and dermatologists, but also of endocrinologists, physiotherapists, surgeons and geriatrists. The skin grows old together with the whole organism, and it is impossible to deter this process only in the skin. Nowadays a series of methods is offered in the esthetic medicine, using embryonic cells, regulatory peptides and placenta suspensions to influence the aging processes both in the skin its

Induction of immunological tolerance to the chemical carcinogens in early ontogenesis.

It is postulated that the induction of immunological tolerance to chemical carcinogens is due to the penetration of carcinogens and its albumin and DNA adducts from the maternal organism into the embryo. Maternal serum lgG-antibodies to carcinogens promote the transportation of carcinogens and adducts into the embryo through placenta and thus stimulate the tolerance induction. On the contrary, maternal secretory lgA-antibodies to carcinogens protect maternal, embryonal and new-born organisms from environmental carcinogens and thus inhibit the tolerance induction. The low individual maternal se

Developmental and Cell Fate Analyses Support a Postnatal Origin for the Cortical Collecting System in the Mouse Kidney.

KEY POINTS: An adult-like corticomedullary organization underlying kidney function is established 10 days after birth in the mouse kidney. Genetic lineage tracing demonstrates the cortical collecting duct network is generated from progenitors after birth. Mature cell types of the nephron progenitor–derived connecting tubule and ureteric progenitor–derived collecting epithelium are established by P15. BACKGROUND: Structure and function in the mammalian kidney are organized along a radial axis highlighted by the corticomedullary organization and regional patterning of the collecting system. The

Extensive cervical spine and foregut anomaly in 'serpentine syndrome'.

INTRODUCTION: We report an extremely rare and challenging combination of congenital anomalies. Only five similar cases have been described in the English language medical literature to date. PRESENTATION OF CASE: A male infant was born at 30(+5) weeks gestation by emergency caesarian section. Cervical spine rachischisis, shortened oesophagus, intrathoracic stomach, atretic duodenum and absent spleen were noted, in addition to respiratory insufficiency. Gastrointestinal re-anastomosis, particularly oesophageal lengthening, was not feasible at the initial thoracotomy. Surgical stabilization of

Neonatally induced mild diabetes in rats and its effect on maternal, placental, and fetal parameters.

The aim of this study was to assess placental changes and reproductive outcomes in neonatally induced mild diabetic dams and fetal development in their offspring. At birth, female rats were assigned either to control or diabetic group (100 mg of streptozotocin/Kg, subcutaneously). At adulthood, the female rats were mated. During pregnancy, the blood glucose levels and glucose and insulin tolerance tests were performed. At term, maternal reproductive outcomes, fetal and placental weight, and placental morphology were analyzed. Diabetic rats had smaller number of living fetuses, implantations an

How do you get six meters of epididymis inside a human scrotum?

It is very clear that the epididymis plays a crucial role in the maturation of spermatozoa, and without a fully developed and functional epididymis, male infertility will result. We are especially interested in understanding the mechanisms that regulate the development of this important organ because disruptions to epididymal function will also arise as a consequence of abnormal development. Very little is known either of the process of epididymal development or the nature and causes of congenital defects that lead to male infertility. A major event during Wolffian/epididymal duct embryonic de

Developmental functions for the Caenorhabditis elegans Sp protein SPTF-3.

Sp factors are important for animal development and the transcriptional regulation of a wide variety of genes. How they influence the developmental decisions of individual cells within the organism, however, is poorly understood. To better understand the developmental functions for Sp transcription factors, we have characterized the functions of Caenorhabditis elegans SPTF-3 using RNAi knockdown and a non-null, hypomorphic mutant allele. We find that disruption of sptf-3 confers a variety of developmental defects, including defects in development of the egg-laying system, oocyte production, an

The RNA-binding protein Elavl1/HuR is essential for placental branching morphogenesis and embryonic development.

HuR is an RNA-binding protein implicated in a diverse array of pathophysiological processes due to its effects on the posttranscriptional regulation of AU- and U-rich mRNAs. Here we reveal HuR's requirement in embryonic development through its genetic ablation. Obligatory HuR-null embryos exhibited a stage retardation phenotype and failed to survive beyond midgestation. By means of conditional transgenesis, we restricted HuR's mutation in either embryonic or endothelial compartments to demonstrate that embryonic lethality is consequent to defects in extraembryonic placenta. HuR's absence impai

The association between autism and errors in early embryogenesis: what is the causal mechanism?

The association between embryonic errors and the development of autism has been recognized in the literature, but the mechanism underlying this association remains unknown. We propose that pleiotropic effects during a very early and specific stage of embryonic development-early organogenesis-can explain this association. In humans early organogenesis is an embryonic stage, spanning Day 20 to Day 40 after fertilization, which is characterized by intense interactivity among body parts of the embryo. This implies that a single mutation or environmental disturbance affecting development at this st

Placental failure and impaired vasculogenesis result in embryonic lethality for neuropathy target esterase-deficient mice.

Age-dependent neurodegeneration resulting from widespread apoptosis of neurons and glia characterize the Drosophila Swiss Cheese (SWS) mutant. Neuropathy target esterase (NTE), the vertebrate homologue of SWS, reacts with organophosphates which initiate a syndrome of axonal degeneration. NTE is expressed in neurons and a variety of nonneuronal cell types in adults and fetal mice. To investigate the physiological functions of NTE, we inactivated its gene by targeted mutagenesis in embryonic stem cells. Heterozygous NTE(+/-) mice displayed a 50% reduction in NTE activity but underwent normal org

Basement membrane distortions impair lung lobation and capillary organization in the mouse model for fraser syndrome.

Fras1 is a putative extracellular matrix protein that has been implicated in the structural adhesion of embryonic epidermis to dermis. Moreover, mutations in Fras1/FRAS1 have been associated with the mouse blebbed phenotype and the human rare genetic disorder Fraser syndrome, respectively. Here we report the mapping of Fras1 within the extracellular space and evaluate the effects of Fras1 deficiency on lung development in the mouse. Expression of Fras1 was detected in the mesothelial cells of the visceral pleura and in the conducting airway epithelia. Immunogold histochemistry identified Fras1

Imaging of fetal tumors and other dysplastic lesions: A review with emphasis on MR imaging.

Fetal tumors and other dysplastic masses are relatively rare. They are usually the result of failure of differentiation and maturation during embryonic or fetal life; dysplastic lesions may be the consequence of an obstruction sequence. In this review, we present the most commonly encountered tumors and masses seen during fetal life. Imaging characteristics, tumoral organ of origin, and its effect on the surrounding organs and overall fetal hemodynamics are descriptors that must be relayed to the fetal surgeon and maternal fetal medicine expert, in order to institute most accurate parental cou
