Chlorpromazine inhibits vesiculation, alters phosphoinositide turnover and changes deformability of ATP-depleted RBCs.

To delineate further the underlying mechanism by which amphiphilic drugs can modulate vesicle release from human RBCs, we studied the effect … addition, chlorpromazine also inhibited the decreased in RBC deformability as well as the shift of osmotic fragility that occurs during ATP depletion of erythrocytes.

Hematologic data of selenium-deficient and selenium-supplemented rats.

Effect of dietary selenium as sodium selenite on in vivo hematological parameters of Sprague-Dawley rats was examined over a 7-month period. Dietary selenium did not alter total hemoglobin, hematocrits, erythrocyte counts, or the osmotic fragility pattern of rat blood. Selenium-excessive (1.0 ppm) rats showed slightly lower but not significantly lower methemoglobin levels than selenium-adequate (0.1 ppm) or selenium-deficient rats. Platelet counts tended to be higher in selenium-excessive rats and lower in selenium-deficient rats than in selenium-adequate rats, but the differences were not sta…

[Hereditary spherocytosis: clinical characteristics and treatment with splenectomy].

PURPOSE: To analyse the clinico-biological characteristics at diagnosis, the clinical course, and the response to splenectomy of a series of patients with hereditary spherocytosis (HS). PATIENTS AND METHODS: The clinical records of 61 patients diagnosed of HS along 30 years were reviewed. The diagnosis was based upon the existence of family history, physical findings, blood cell examination, reticulocyte count, peripheral blood spherocytes, red-cell osmotic fragility, auto-haemolysis, serum haptoglobin, LDH, non-conjugated bilirubin and direct anti-human globulin test. Data regarding the time…

Genetic control of erythrocyte volume regulation: effect of a single gene (rol) on cation metabolism.

In laboratory mice we previously defined a gene, rol (resistance to osmotic lysis), based on its effect on erythrocyte osmotic fragility. Here we report a physiological characterization of rol gene action utilizing congenic strains developed for the purpose; these two strains have a common genetic background and differ only by the two alleles of rol, susceptible (rols) or resistant (rolr). In comparison to rols/s erythrocytes, rolr/r cells have a reduced mean cell volume, a higher mean corpuscular hemoglobin concentration and hemolytic volume, and respond differently to swelling induced by ion…

Integration of Trehalose Lipids with Dissociative Trehalose Enables Cryopreservation of Human RBCs.

Cryopreservation of red blood cells (RBCs) is imperative for transfusion therapy, while cryoprotectants are essential to protect RBCs from c … ell morphology and cell functions. It was further confirmed by isothermal titration calorimetry and osmotic fragility tests that the moderate membrane-binding activity of trehalose lipids exerted cell stabilization for high cryosurvival. The aforementioned study is likely to provide an alternative way for glycerol-free cryopreservation of human RBCs and other types …

Ginkgo biloba Supplement Reverses Lead (II) Acetate-Induced Haematological Imbalances, Hepatic and Renal Dysfunctions in Male Wistar Rat.

Lead is a heavy metal abundant in nature that causes haematological imbalances, and hepatic and renal dysfunction, and this imbalance has be … t enzyme activity (CAT, SOD, GSH, and MDA), organ function indices, and a lower rate of erythrocyte osmotic fragility. Conclusively, Ginkgo biloba supplementation attenuated lead toxicity by normalization of haematological imbalances, and hepatic and renal dysfunction as well as maintaining erythrocyte membrane integrity.

Identification of a novel ANK1 mutation in a Chinese family with hereditary spherocytosis: A case report.

The present study describes the clinical profile and ankyrin 1 (ANK1) mutation status of a Chinese family with hereditary spherocytosis (HS). A young male patient (proband) was diagnosed with HS after presenting with anaemia and jaundice. The Coombs test was negative and spherocytes were found in peripheral blood smears. Magnetic resonance imaging showed splenomegaly and splenic iron depositions. The red blood cell osmotic fragility test was positive. The eosin-5'-maleimide binding test showed reduced mean channel fluorescence. Whole-exome sequencing revealed a novel ANK1 mutation (c.4707G>A),…

Defects of copper deficiency in rats are modified by dietary treatments that affect glycation.

We examined the hypothesis that nonenzymatic glycosylatin of proteins (glycation) contributes to the defects of copper deficiency. We studied copper-adequate and -deficient rats while altering two factors known to affect glycation: type of dietary carbohydrate and amount of food intake. Copper deficiency caused cardiac enlargement and anemia, decreased erythrocyte osmotic fragility, enhanced heart lipid peroxidation, increased the percentage of glycated hemoglobin (Hb A1) and reduced staining of lens crystallins on SDS-PAGE gels (suggestive of glycation). Increasing dietary sucrose reduced org…

Erythrocyte abnormalities in a hypoalphalipoproteinemia syndrome resembling fish eye disease.

Erythrocyte membrane (EM) abnormalities in a 16-yr-old boy with hypoalphalipoproteinemia resembling fish eye disease (FED-LS) were investigated. The proband's erythrocytes had markedly decreased osmotic fragility with target cells observed in the peripheral film. Analysis of his EM lipids revealed normal cholesterol and phospholipid content but a marked increase in phosphatidylcholine with concomitant decreases in phosphatidylethanolamine and sphingomyelin. Of the EM enzymes examined, acetylcholinesterase and superoxide dismutase activities were decreased while those of Na+-K+ ATPase, catalase…

Hyperintense lesions of the middle cerebellar peduncle and beyond: a pictorial essay.

The middle cerebellar peduncle (MCP) is the largest afferent system of the cerebellum and consists of fibres from the cortico-ponto-cerebellar tract. Specifically, several relevant diseases can present with hyperintensity in the MCP on T2-weighted/fluid-attenuated inversion recovery (T2/FLAIR) magnetic resonance imaging sequences, including multiple sclerosis; acute disseminated encephalomyelitis; neuromyelitis optica spectrum disorder; progressive multifocal leucoencephalopathy; hepatic encephalopathy; osmotic demyelination syndrome; multiple system atrophy; fragile X-associated tremor/ataxia

Recognition and laboratory characteristics of an atypical oocyst of Cryptosporidium.

Feces from some patients with clinically unremarkable cryptosporidiosis contained an unusual variety of oocyst not previously recognized. These atypical oocysts were shown by electron microscopy to have a distinctive three-layered outer coat and, by immunofluorescence with a monoclonal antiserum, to lack an antigen present on the surface of typical oocysts. In contrast to typical oocysts, the atypical variety is very fragile and quickly collapses when suspended in solutions of high osmotic pressure or in lipid solvents. Atypical oocysts cannot be stained by methods used to stain typical oocyst

[Aplastic crises in hereditary spherocytosis].

Parvovirus B19-infections are diagnosed with increasing frequency. In addition to erythema infectiosum they cause a temporary red cell aplasia of 5-10 days duration, in patients with hemolytic anemia an aplastic crisis as demonstrated in 5 patients with hereditary spherocytosis. In every case an acute infection by parvovirus B19 could be diagnosed. Hemoglobin concentrations dropped to 4.0-6.2 g/dl, reticulocytes were diminished to 0-25,000/microliters. In every case there followed a complete recovery of erythropoiesis after 2-3 days.
