Avian cholera is a bacterial disease caused by infection with Pasteurella multocida (P. multocida), which seriously threatens the poultry industry. Liver injury is a key factor in the acute death of infected chickens. This study sought to clarify how P. multocida causes liver injury in broilers, specifically examining liver pyroptosis and the involvement of oxidative stress and inflammation. Experiments with 28-day-old broilers infected with P. multocida showed that post-infection serum indicators were significantly higher for ALT, AST, and T-Bil, but lower for ALB. Histopathological evaluatio…

The bacterial agent that causes fowl cholera, Pasteurella multocida, was isolated from two deceased wild waterbirds in Victoria, Australia, in 2013. Whole genome sequence analysis placed the isolates into ST20, a subtype described in farmed chickens from Queensland, Australia and more recently in feedlot cattle and in pigs across a broader area of the continent. This study also found ST20 between 2009 and 2022 on three chicken farms and two turkey farms located in four Australian states. The sequences of 25 of these ST20 isolates were compared to 280 P. multocida genomes from 23 countries and …

The rise of antibiotic-resistant bacterial infections necessitates alternative therapeutic strategies, such as phage therapy. This study investigates the potential of phage vB_PmuM_CFP3 (CFP3) as a therapeutic agent against avian cholera caused by Pasteurella multocida (P. multocida). Phage CFP3 was isolated from the feces and wastewater of a laying hen farm and underwent comprehensive biological characterization, including host range, lytic activity, and environmental stability. Transmission electron microscopy revealed CFP3's typical myovirus morphology, with a head diameter of approximately…

Infection with Pasteurella multocida represents a significant economic threat to Australian pig producers, yet our knowledge of its antimicrobial susceptibilities is lagging, and genomic characterization of P. multocida strains associated with porcine lower respiratory disease is internationally scarce. This study utilized high-throughput robotics to phenotypically and genetically characterize an industry-wide collection of 252 clinical P. multocida isolates that were recovered between 2014 and 2019. Overall, antimicrobial resistance was found to be low, with clinical resistance below 1% for a…

Pasteurella multocida.(PM) infection is a major cause of avian cholera, but the pathogenesis of the disease is unknown. The purpose of this study was to further understand the host response to infection by using a duck model of PM, 20 female ducks were divided into two groups (n = 10). One group was infected with PM, while the other served as an uninfected control group. The ducks were observed after infection and samples were collected for testing. In this study, we report the mechanism of PM-induced inflammation to further mediate apoptosis and autophagic signaling pathways in liver cells. O…

Macrolides are commonly used to control respiratory tract infections in ruminants, but the susceptibility of Pasteurella multocida strains has shown a decrease to macrolide antibiotics in the last decade. In this work we assessed the prevalence of macrolide resistance of 100 P. multocida isolates from ruminant hosts and studied the resistance genotypes with newly designed PCRs. Susceptibility to erythromycin and tilmicosin was tested using minimal inhibitory concentration (MIC) test strips. A newly designed PCR was used for the detection of macAB genes, and a PCR plus restriction enzyme-based …

Pasteurella multocida is an important pathogenic bacterium of domestic animals. However, the mechanisms of infection are still poorly understood. Here, we found that Pm0442 was dramatically up-regulated in infected mice among 67 predicted lipoproteins of P. multocida serotype A CQ2 strain (PmCQ2). To explore the role of Pm0442 in virulence and the potential of the mutant as a vaccine, Pm0442 mutant of PmCQ2 was successfully constructed. Then, the virulence characteristics, immune/inflammatory responses, and the survival rates of challenged mice were determined. As a result, it was found that t…

Pasteurella multocida (P. multocida) is an important pathogen that causes bovine respiratory disease (BRD) in China and other countries. To investigate the antimicrobial susceptibility of P. multocida isolated from different provinces in China, we analyzed antimicrobial susceptibility phenotypes and pulsed-field gel electrophoresis (PFGE) types of P. multocida; then, we sequenced the complete genome of strain found to be multidrug-resistant. The isolates exhibited resistance to many antimicrobial agents, especially amikacin, sulfamethoxazole, sulfachloropyridazinesodium, macrolides, and fluoro…

Pasteurella multocida is a gram-negative, non-motile bacterial pathogen, which is associated with chronic and acute infections as snuffles, pneumonia, atrophic rhinitis, fowl cholera and hemorrhagic septicemia. These diseases affect a wide range of domestic animals, leading to significant morbidity and mortality and causing significant economic losses worldwide. Due to the interest in deciphering the genetic diversity and process adaptive between P. multocida strains, this work aimed was to perform a pan-genome analysis to evidence horizontal gene transfer and positive selection among 23 P. mu…

A 57-year-old woman with severe bronchiectasis frequently received antibiotics, including penicillin, for acute exacerbations due to Pasteurella multocida. Although the bacteria showed a decrease in antibiotic susceptibility, her symptoms and X-ray findings became stable, and severe exacerbations were not observed for the last few years after a low-dose erythromycin treatment was started. The development of a respiratory infection with Pasteurella multocida is relatively uncommon, but it can be controlled by immunomodulation which is associated with long-term macrolide therapy.

Five-week-old turkey poults were given two consecutive intravenous injections (24 hours apart) of highly purified Pasteurella multocida lipopolysaccharide (LPS) in an effort to induce a generalized Shwartzman reaction. There were no gross lesions, and microscopic lesions were limited to focal hepatic necrosis with heterophil infiltration. Hepatic lesions did not differ qualitatively from lesions in turkeys given a single dose of lipopolysaccharide. Margination of heterophils in the pulmonary vasculature was observed in turkeys 4 hours after a single injection of LPS, but it was not present in …

During the 12-yr period, 1973-1984, 243 isolates of Pasteurella multocida were recovered or identified from specimens submitted for plague tests. Of the isolates, 79% were from rodents, 10% from lagomorphs, and 7% from carnivores; eight isolates were recovered from pools of rodent or carnivore fleas, and two were recovered from cat-bite wounds of human patients. No correlations of host or geographic sources, season, or biotypic or serotypic characteristics were found. Of the rodent strains serotyped, most were found to be serotypes 1A or 3A, which suggests a possible epizootiologic role for ro…

A live cholera vaccine was developed from a virulent avian septicemia strain of Pasteurella multocida serotype 1. The virulent parental strain was mutagenized with N-methyl-N'-nitro-N-nitroso guanidine. Mutants were selected that had either smaller colonies at 37 C or temperature sensitivity for growth at 41 C. Four small-colony mutants and 2 temperature-sensitive mutants were studied. All the mutants were avirulent for turkeys. Sixteen days after turkeys were vaccinated with each mutant, both the vaccinates and unvaccinated controls were challenge-exposed to virulent P. multocida of the homol…

CASE: A 71-year-old man with a history of multiple revision lumbar spine surgeries following rod exchange with local and systemic infection symptoms including purulent drain output. Imaging demonstrated extensive submuscular fluid collection. Subsequent surgical debridement with deep cultures demonstrated Pasturella multocida; retrospectively, the patient recalled being scratched by his cat 1 week before presentation.
CONCLUSION: Pasturella multocida is a rare cause of paravertebral abscess following spinal surgery, but prevalent in cat flora. As domesticated animals are extremely common, cli

Pasteurella multocida is a gram-negative coccobacillus that colonizes the aerodigestive tracts of cats, dogs, birds, and wild animals. This veterinary pathogen spreads to humans via licking, biting, or scratching. Pekin ducks are known carriers of multiple strains of Pasteurella bacteria that can manifest as fowl cholera, eye infection, or duck septicemia. This bacterium is a farmer's nightmare, as it is associated with high mortality rates. Herein, we report a unique case of P. multocida bacteremia in an immunocompetent host that shared a house with adopted Pekin ducks.

Pasteurella multocida is the causative agent of a wide range of diseases (pasteurellosis) and a zoonotic pathogen in humans. Recombinant subunit vaccines are hot spots in recent pasteurellosis vaccine development. A chimeric vaccine is also constructed for rabbit hemorrhagic disease virus (RHDV) protective antigen VP60 chimeric with fragments of Pasteurella multocida protective antigen PlpE. The protective efficacy of the chimeric vaccine against P. multocida is not as high as that of PlpE, and the reason is not well known. In this study, we analyzed the linear B-cell epitopes of PlpE and then…
