INTRODUCTION: Retinoblastoma, hepatoblastoma, germ cell tumors, bone tumors, and soft tissue tumors are rare types of childhood cancer. Identifying modifiable risk factors is essential to inform primary prevention. This review aimed to systematically examine literature and synthesize risk factors for childhood rare cancers.
METHODS: PubMed, Web of Science, and Embase databases were searched using lists of keywords and MeSH terms related to parental and childhood exposures. Studies were included if they were case-control or cohort studies of children under the age of 20 years at diagnosis and

Streptococcus thoraltensis is a recently described species, isolated from the intestinal and genital tracts of swine and from rabbit feces. We describe here a case of chorioamnionitis, with paternal swine exposure, potentially attributable to S. thoraltensis. To our knowledge, this is the first reported human infection by this organism.

BACKGROUND: Infertility is a common reproductive disorder, with male factor infertility accounting for approximately half of all cases. Taking a paternal perceptive, recent research has shown that sperm epigenetics, such as changes in DNA methylation, histone modification, chromatin structure, and noncoding RNA expression, can impact reproductive and offspring health. Importantly, environmental conditions during the preconception period has been demonstrated to shape sperm epigenetics.
OBJECTIVES: To provide an overview on epigenetic modifications that regulate normal gene expression and epig

This study found abnormalities in multiple reproductive stages in captive American kestrels (Falco sparverius) when exposed to polychlorinated biphenyls (PCBs) through dietary and in ovo exposure. American kestrels laid eggs with environmentally relevant total PCB levels (34.1 micrograms/g whole egg wet weight) when consuming PCB-spiked (Aroclor 1248:1254:1260) food (5-7 micrograms/g body weight per day) for 100 d only in 1998. In 1999, the same adults laid eggs with estimated total PCBs of 23 micrograms/g. Effects of maternal (only female exposed) and paternal (only male exposed) in ovo PCB e

As the evidence for an association between spina bifida and parental agricultural occupations is inconclusive, we evaluated this association and the potential associations between spina bifida and other parental occupations. Three register-based case-referent studies were conducted in Sweden, Spain, and Hungary. From the registries of congenital malformations in each country, 482, 478, and 1119 cases with spina bifida were identified, respectively. Identified as referents were 964, 434, and 1489 children without abnormalities. Information on parental occupation was available in the registries

The authors conducted a case-control study in Comunidad Valenciana, Spain, to assess the relation between occupational exposure to pesticides, mainly as a result of agricultural work, and the prevalence of congenital malformations. A total of 261 cases and 261 controls were selected from those infants born in eight public hospitals during 1993-1994. The cases were those who were diagnosed with selected defects (nervous system, cardiovascular, oral clefts, hypospadias/epispadias, musculoskeletal, and unspecified anomalies) during their first year of life. Information on occupational exposures a

BACKGROUND: Tobacco smoke exposure has been associated with early childhood asthma symptoms. We assessed the associations of tobacco smoke exposure during pregnancy and childhood with wheezing patterns, asthma, airway interrupter resistance (Rint), and fractional exhaled nitric oxide (Feno) in school-age children and whether birth characteristics explained the associations.
METHODS: This study was embedded in a population-based prospective cohort study among 6,007 children. Paternal and maternal smoking during pregnancy (never, first trimester only, continued), secondhand tobacco smoke exposu

We explored the relation between various potential sources of maternal periconceptional pregnancy exposures to pesticides and congenital anomalies in offspring. Data were derived from a case-control study of fetuses and liveborn infants with orofacial clefts, neural tube defects, conotruncal defects, or limb anomalies, among 1987-1989 California births and fetal deaths. We conducted telephone interviews with mothers of 662 (85% of eligible) orofacial cleft cases, 265 (84%) neural tube defect cases, 207 (87%) conotruncal defect cases, 165 (84%) limb cases, and 734 (78%) nonmalformed controls. To the authors' knowledge, attributable fractions for cardiac malformations have not been reported before. The Baltimore-Washington Infant Study published factors associated with several major cardiac malformations in Maryland, the District of Columbia, and adjacent counties of northern Virginia in 1981-1989. For eight of these malformations, the authors provide attributable fractions of those factors that are potentially causal. Summary attributable fractions range from 13.6% (four factors) for hypoplastic left heart to 30.2% (seven factors) for transposition of great arteries with intact ven

BACKGROUND: Effective prevention of cardiac malformations is constrained by limited understanding of etiology. We used 2011-2021 MarketScan US insurance claims data to identify and characterize associations between maternal and paternal characteristics and non-chromosomal cardiac malformations.
METHODS: Among 693,483 singleton live-birth pregnancies of women linked to infants (of which 488,146 linked to fathers), odds ratios were estimated between 2000 clinical diagnostic and medication codes (500 clinical and 500 medication codes each for mothers and fathers) and cardiac malformations (n = 7

Background/Objectives: Recurrent pregnancy loss (RPL) has traditionally been investigated predominantly through maternal factors, while the clinical role of paternal assessment remains inconsistently defined. Current guidelines differ substantially regarding semen analysis, sperm DNA fragmentation (SDF), genetic testing, and referral for andrological evaluation. This review aimed to compare contemporary guideline recommendations, critically appraise the directness, prognostic value, and clinical utility of the supporting evidence, and classify paternal assessment strategies as routine, selecti

BACKGROUND: Family clusters and epidemics of toxoplasmosis in North, Central, and South America led us to determine whether fathers of congenitally infected infants in the National Collaborative Chicago-Based Congenital Toxoplasmosis Study (NCCCTS) have a high incidence of Toxoplasma gondii infection.
METHODS: We analyzed serum samples collected from NCCCTS families between 1981 and 2013. Paternal serum samples were tested for T. gondii antibodies with immunoglobulin (Ig) G dye test and IgM enzyme-linked immunosorbent assay. Additional testing of paternal serum samples was performed with diff

The objective of this study was to examine the effect of paternal heat stress on the in vivo development of preimplantation embryos in the mouse. Synchronised B6CBF1 female mice were mated either to a control male mouse or to one that had been exposed at 7, 21 or 35 days previously, for 24 h to an ambient temperature of 36+/-0.3 degrees C and 66+/-5.6% relative humidity. Embryos were collected from the oviducts of mice at 14-16 h, 34-39 h or 61-65 h after mating or from the uterus at 85-90 h after mating and their developmental status was evaluated morphologically. The number of cells within b

BACKGROUND AND AIM: Pregnancy-related exposures have been proposed as potential risk factors for breast cancer later in life, but findings remain inconclusive. This study aimed to update evidence on the associations between the exposure to pregnancy-related factors occurring up to birth-maternal and paternal age, gestational age at birth, twin status, and maternal preeclampsia-and breast cancer risk in daughters.
MATERIAL AND METHODS: A systematic review and meta-analysis were conducted. Searches were performed in MEDLINE (via PubMed), Web of Science, and Scopus. We included observational ana

Paternal use of valproate during spermatogenesis has been associated with increased risk of neurodevelopmental disorders (NDDs) in offspring, yet the role of genetic confounding is unclear. Using data from the Norwegian Mother, Father, and Child Cohort Study (MoBa), we assessed genetic susceptibility to epilepsy, ADHD and autism spectrum disorders (ASD) in fathers with epilepsy treated with valproate (n = 41), lamotrigine or levetiracetam (n = 37), other anti-seizure medications (ASMs; n = 80), and healthy controls (n = 54,752). Fathers using valproate had significantly higher polygenic risk s

OBJECTIVE: To investigate relationships among family risk factors, childhood firesetting and animal cruelty, and adolescent delinquency.
METHOD: In 1990, mothers and children participating in a 10-year prospective study provided information about family risk factors and childhood problem behavior. Subsequent interviews with 86% of the sample in 1996 and 1998 and court record reviews in 2000 provided information about juvenile delinquency.
RESULTS: Marital violence (odds ratio [OR] 2.4, 95% confidence interval [CI] = 1.1-5.4), paternal pet abuse (OR 2.4, CI = 1.0-5.6), and paternal drinking (

7949 male doctors in the U.K. were surveyed to determine whether there was a relation between operating-theatre work (exposure) and abnormalities in the obstetric history of their marriages. Paternal exposure did not appear to influence the overall abortion-rate or the frequency of major congenital abnormality and involuntary infertility. The frequency of minor congenital abnormality in the children of exposed fathers was 3.09% compared with 2.35% (non-exposed). Maternal exposure was associated with a 15.5% frequency of abortion. Matching of exposed and non-exposed pregnancies with respect to …
