Gold(III) porphyrin presents an attractive alternative to the use of, for example, cisplatin in chemotherapy. However, approaches that allow to selectively target cancer cells are highly sought. Many plant and mammalian lectins have been shown to bind oligosaccharide sequences of the aberrant glycosylation pattern found on cancerous tumors. For example human galectin-3, of the galectin family specific for β-galactoside, is overexpressed in the extracellular matrix of tumorigenous and metastatic tissues. We searched for non-carbohydrate ligands for galectin-3 that can guide a cytotoxic drug to

The phloem is a specialized tissue that facilitates systemic transport of carbohydrates and signal molecules, making it a common target for viruses, phloem-feeding insects, and cyst nematodes. In Arabidopsis, the stationary phloem-associated SIEVE ELEMENT-LINING CHAPERONE1 (SLI1) and RESTRICTED TEV MOVEMENT (RTM) proteins restrict insect phloem-feeding and potyviruses systemic transport, respectively. However, their broader roles in plant-attacker interactions remain largely unexplored. We investigated the roles of SLI1, RTM1, RTM2, and RTM3 in tobacco etch virus (TEV) infection, as well as in

Chickpea productivity is affected by various constraints that are biotic (Helicoverpa, Aphids, Callosobruchus, Bromus and Orobanche) and abiotic (drought and salinity). In addition, the grains of this legume are deficient in sulfur amino acids, methionine and cysteine. The possibilities for genetic improvement by marker-assisted breeding and selection approaches are limited in chickpeas due to their sexually incompatible gene pool. Transgenic chickpeas expressing either the cry1Ac/b or the cry2Aa gene and the bean α-amylase inhibitor gene are resistant to Helicoverpa and bruchids, respectively

All animals and plants have immune systems that protect them from the diversity of pathogens that would otherwise threaten their survival. The different components of the immune system may inactivate the pathogens themselves or promote the inactivation and clearance of toxic products produced by the pathogens. An important category of virulence factors of bacterial and prokaryotic pathogens are the proteases, which act to facilitate the invasion of the pathogens and to promote their destructive growth in the host organism. The present review concentrates on the comparative biology of an evolut

alpha-Amylases are important digestive enzymes in weevils that infest starchy seeds, and plants have evolved proteinaceous alpha-amylase inhibitors (alpha AI) for protection. To gain a better understanding of the interaction between weevil alpha-amylases and alpha AIs, we cloned the alpha-amylase cDNA of Zabrotes subfasciatus larvae. Larvae of this bruchid infest seeds of cultivated varieties of the common bean, Phaseolus vulgaris, although the seeds contain high levels of an alpha AI. The alpha-amylase cDNA, called ZsAmy, encodes a mature protein of 466 amino acids with a signal peptide of 17

The encephalopathy resulting from direct infection of the brain by human immunodeficiency virus (HIV), which correlates clinically with the AIDS dementia complex, has been reported as being localized to the white matter where it induces myelin loss, gliosis and perivascular infiltration by mononuclear macrophages and multinucleated giant cells. Damage to the cortical grey matter in HIV encephalopathy was investigated in nine randomly selected HIV-positive cases with or without clinical or morphological evidence of encephalopathy and in five age-matched controls, using routine histology and imm

BACKGROUND: Lupus nephritis (LN) is a devastating complication of systemic lupus erythematosus (SLE). Aberrant IgG glycosylation drives podocyte injury and is a robust marker for LN. The primary method for detecting glycosylation is limited to mass spectrometry, but it is not suitable for clinical applications due to its complexity and high cost.
METHODS: We developed and optimised a lectin-based enzyme-linked assay to detect IgG glycan residues. Lectins were selected based on the glycan structures detected in a SLE cohort and were differentially expressed in SLE with and without nephritis. 9

Adzuki beans have been historically utilized for both culinary and medicinal purposes in Japan. Lectin derived from Japanese red adzuki beans (JABs, Vigna angularis) exhibits several biological effects; however, to our knowledge, no detailed reports have been published on this topic. We purified lectin from adzuki beans to evaluate its impact on the proliferative activity of cancer cells and establish its biological profile. The Japanese red adzuki bean lectin (JABL) was purified using thyroglobulin-Sepharose 4B and evaluated for blood and sugar specificity and its effect on B16, LM8, Ehrlich

Mucins form a group of heavily O-glycosylated biologically important glycoproteins that are involved in a variety of biological functions, including modulating immune response, inflammation, and adhesion. Mucins are also involved in cancer and metastasis and often express diagnostic cancer antigens. Recently, a modified porcine submaxillary mucin (Tn-PSM) containing GalNAcalpha1-O-Ser/Thr residues was shown to bind to soybean agglutinin (SBA) with approximately 10(6)-fold enhanced affinity relative to GalNAcalpha1-O-Ser, the pancarcinoma carbohydrate antigen. In this study, dynamic force spect

OBJECTIVE: Radiation optic neuropathy usually occurs months to years after exposure of the anterior visual pathways to ionizing radiation. It is characterized by high signal on gadolinium-enhanced T1-weighted magnetic resonance imaging. Radiation-induced endothelial cell damage resulting in blood-nerve barrier breakdown is hypothesized to produce this pattern, but histologic evidence of this in the optic nerve is lacking. We attempted to evaluate the effect of radiation on endothelial cells in the optic nerve.
DESIGN: Case-controlled histologic study.
METHODS: We studied the optic nerves of

BACKGROUND: Glomerular deposition of IgA1 is a common feature of Henoch-Schönlein purpura, and is indistinguishable from that seen in IgA nephropathy. Serum IgA1 is abnormally O-glycosylated in IgA nephropathy, and this may contribute to mesangial IgA1 deposition and the development of glomerular injury. This altered O-glycosylation of IgA1 can be detected by its increased binding to the lectin Vicia villosa.
METHODS: To investigate whether IgA1 is abnormally glycosylated in Henoch-Schönlein purpura, the binding of Vicia villosa lectin to serum IgA1 was studied in the following subject groups

A conjugate of peanut lectin with horseradish peroxidase (PL-HRP) has been employed for ultrastructural localization of glycoprotein with presumed terminal galactose residues in mouse alimentary epithelial cells. The PL-HRP conjugate imparted electron opacity in sites that stain at the light microscopic level, as for example, Golgi cisternae in surface epithelial cells of the stomach and in superficial and deep crypt cells and goblet cells of the large intestine. Ultrastructural staining revealed that Golgi cisternae intermediate between the trans and cis faces stained selectively in these sit

Many natural product-derived lectins such as the red algal lectin griffithsin (GRFT) have potent in vitro activity against viruses that display dense clusters of oligomannose N-linked glycans (NLG) on their surface envelope glycoproteins. However, since oligomannose NLG are also found on some host proteins it is possible that treatment with antiviral lectins may trigger undesirable side effects. For other antiviral lectins such as concanavalin A, banana lectin and cyanovirin-N (CV-N), interactions between the lectin and as yet undescribed cellular moieties have been reported to induce undesira

During differentiation, megakaryocytes undergo nuclear endoreplication, an increase in cell size, cytoplasmic granulation, and release of platelets. The changes in highly lobulated nuclei with varying degree of polyploidy and increasing cell size are easily recognized morphologically. However, the actual cytoplasmic changes are more difficult to perceive morphologically. With the peroxidase-antiperoxidase (PAP) method using UEA-1 as the binding protein to the alpha-L-fucose of glycoprotein synthesized by megakaryocytes, we observed significant variation in cytoplasmic staining of megakaryocyte

The existence of non-cholinergic (GABAergic) components in the septo-hippocampal system but also in basal forebrain projections terminating in the olfactory bulb and certain cortical areas has been documented by several authors using retrograde and anterograde tracing techniques. On the other hand, the basal forebrain also contains a high number of mainly parvalbumin-positive neurons ensheathed by a lattice-like matrix of polyanionic proteoglycans forming so-called perineuronal nets of as yet unknown function. By a combination of retrograde tracing using Fluoro-Gold injection into mesocortical

Blood vascular and lymphatic tumors were evaluated immunohistochemically by studying a spectrum of endothelial associated antigens. UEA-1 lectin reacted with the tumor cells of one patient with malignant angioendothelioma in the non-metastatic stage. However, when metastasis occurred, the binding sites of this lectin completely disappeared from the surface of the tumor cells in both original and metastatic lesions, suggesting the loss of blood group H antigen from the tumor cells could be used as an indicator of metastasis in this tumor. Reaction with anti-HLA-A, B, C, intense in normal blood
