Current and emerging therapies for Achondroplasia: The dawn of precision medicine.

Achondroplasia is a rare disease affecting bone growth and is caused by a missense mutation in the fibroblast growth factor receptor 3 (FGFR3) gene. In the past few years, there were multiple experimental drugs entering into clinical trials for treating achondroplasia including vosoritide, the first precision medicine approved for this indication. This perspective presents the mechanism of action, benefit, and potential mechanistic limitation of the drugs currently being evaluated in clinical trials for achondroplasia. This article also discusses the potential impact of those drugs not only in…

Managing sepsis in the era of precision medicine: challenges and opportunities.

INTRODUCTION: Precision medicine is a medical model in which decisions, practices, interventions and therapies are tailored to the individual patient based on their predicted response or risk of disease. Sepsis is a life-threatening condition characterized by immune system dysregulation whose pathophysiology remains incompletely understood. There is much hope that precision medicine can lead to better outcomes in patients with sepsis. AREAS COVERED: In this review from a comprehensive literature search in PubMed for English-language studies in adults, we highlight recent advances in the diagn…

Using Precision Medicine for the Diagnosis and Treatment of Viral Pneumonia.

The COVID-19 pandemic has drawn considerable attention to viral pneumonia from clinicians, public health authorities, and the general public. With dozens of viruses able to cause pneumonia in humans, differentiating viral from bacterial pneumonia can be very challenging in clinical practice using traditional diagnostic methods. Precision medicine is a medical model in which decisions, practices, interventions, and therapies are adapted to the individual patient on the basis of their predicted response or risk of disease. Precision medicine approaches hold promise as a way to improve outcomes f…

Hypereosinophilic syndrome: approach to treatment in the era of precision medicine.

Hypereosinophilic syndromes (HESs) are a heterogeneous group of rare disorders characterized by peripheral eosinophilia and eosinophilic end organ complications. Conventional therapies, including glucocorticoids and cytotoxic and immunomodulatory agents, have variable efficacy and significant toxicity. Although the recent development of agents that target eosinophils, including tyrosine kinase inhibitors and monoclonal antibodies, provides the possibility of more effective, less toxic approaches to treatment of HES, there are little available data to guide their use in these conditions. In the

Molecular testing in lung cancer in the era of precision medicine.

The clinical expectations how pathologists should submit lung cancer diagnosis have changed dramatically. Until mid 90-ties a clear separation between small cell lung carcinoma (SCLC) and non-small cell lung carcinoma (NSCLC) was mostly sufficient. With the invention of antiangiogenic treatment a differentiation between squamous and non-squamous NSCLC was requested. When epidermal growth factor receptor (EGFR) mutation was detected in patients with pulmonary adenocarcinomas and subsequent specific treatment with tyrosine kinase inhibitors (TKIs) was invented, sub-classification of NSCLC and mo

Biologics in alopecia areata: promise, paradox, and the precision medicine frontier.

INTRODUCTION: Alopecia areata (AA) is a chronic, immune-mediated non-scarring hair loss affecting approximately 2% of the population. The approval of oral JAK inhibitors has established targeted systemic therapy for moderate-to-severe AA. Incomplete response rates, relapse upon treatment discontinuation, and safety contraindications, however, leave a substantial proportion of patients requiring alternative options. AREAS COVERED: This review evaluates the immunopathogenic rationale and available clinical evidence for biologic therapies in AA, including inhibitors of tumor necrosis factor (TNF

Airway Disease in 2026: Precision Medicine Meets Environmental and Clinical Reality.

The 2025-2026 conference season in respiratory and allergy medicine has surfaced signals that the field is shifting faster than many clinical systems can absorb. The agendas converge on themes that were peripheral a decade ago: precision phenotyping, remission as a treatment target, the united airway as a clinical organizing principle, metabolic-inflammatory crossover, and the environmental forces reshaping disease burden in real time. This article traces those signals through emerging conference data, updated guidelines from GINA and ARIA-EAACI, and the growing evidence that climate-driven po

Ethical Aspects of Artificial Intelligence in Precision Medicine for Obesity Management.

Artificial intelligence (AI) has established itself as a key tool in the development of precision medicine, offering opportunities to improve risk prediction, early diagnosis, and personalized treatments for obesity. However, its implementation poses ethical challenges that cannot be ignored. The use of sensitive data such as genetic, metabolic, and behavioral information raises questions about privacy, security, and informed consent. Furthermore, the existence of algorithmic biases can reproduce inequities and limit the validity of models in diverse populations, particularly in low-resource s…

Mitochondrial-Centered Biological Networks in Metabolic Disease: Toward Precision Mitochondrial Medicine.

Obesity and type 2 diabetes (T2D) are multifactorial metabolic disorders characterized by progressive dysfunction of multiple organs and biological systems. Although mitochondrial dysfunction is a hallmark of disease progression, the mechanisms linking metabolic stress to coordinated tissue dysfunction remain incompletely understood. Comparative proteomic studies have consistently identified coordinated remodeling of oxidative phosphorylation, fatty acid oxidation, tricarboxylic acid cycle activity, redox regulation, mitochondrial proteostasis, and adaptive signaling across metabolically affec

Overcoming Chemoresistance in Glioblastoma: Mechanisms, Therapeutic Strategies, and Functional Precision Medicine.

Glioblastoma (GBM) is the most common primary malignant brain tumor in adults and remains highly lethal, with median overall survival rarely exceeding 15 months despite maximal surgical resection, radiotherapy, and temozolomide-based chemotherapy. Therapeutic resistance in GBM is driven by intrinsic tumor cell adaptations, extensive inter- and intratumoral heterogeneity, and microenvironmental constraints. Key mechanisms include enhanced DNA repair, disrupted apoptosis, pathway redundancy, altered drug metabolism, oxidative stress tolerance, and glioblastoma stem cell-mediated plasticity. In v
