Effects of radiation and role of plants in radioprotection: A critical review.

Radiation can be lethal at high doses, whereas controlled doses are useful in medical applications. Other applications include power generation, agriculture sterilization, nuclear weapons, and archeology. Radiation damages genetic material, which is reflected in genotoxicity and can cause hereditary damage. In the medical field, it is essential to avoid the harmful effects of radiation. Radiation countermeasures and the need for radioprotective agents have been explored in recent years. Considering plants that evolve in radiative conditions, their ability to protect organisms against radiation

Low dose or low dose rate ionizing radiation-induced health effect in the human.

The extensive literature review on human epidemiological studies suggests that low dose ionizing radiation (LDIR) (≤100 mSv) or low dose rate ionizing radiation (LDRIR) (<6mSv/H) exposure could induce either negative or positive health effects. These changes may depend on genetic background, age (prenatal day for embryo), sex, nature of radiation exposure, i.e., acute or chronic irradiation, radiation sources (such as atomic bomb attack, fallout from nuclear weapon test, nuclear power plant accidents, 60Co-contaminated building, space radiation, high background radiation, medical examinations

Signaling cascades in radiation-induced apoptosis: roles of protein kinase C in the apoptosis regulation.

Apoptosis is a biologic mechanism for eliminating damaged cells from the cell population. Apoptosis is known to be induced by irradiation and can prevent the development of disease states such as carcinogenesis or abnormal tissue formation. On the other hand, if the mechanism is properly controlled, radiotherapy can be used to kill cancer cells more efficiently. Radiation-induced apoptosis is regulated by the balance between cellular anti-apoptotic and (pro-)apoptotic signals. Many regulators of radiation-induced apoptosis have been identified and analyzed. Protein kinase C (PKC) is a family o

[Radiation effects of exposure during prenatal development].

The embryo and fetus are very radiosensitive during the total prenatal development period. The quality and extent of radiation effects depend strongly on the developmental stage at which the exposure occurs. During the preimplantation period radiation exposure can cause death of the embryo after radiation doses of 0.2 Gy and higher. Malformations are only observed in very rare cases when genetic predispositions exist. Macroscopic-anatomical malformations are induced only after irradiation during the major organogenesis. On the basis of experimental data with mammals it is assumed that a radiat

Moyamoya syndrome after proton beam therapy in a pediatric patient with a pineal germ cell tumor and a germline polymorphism in RNF213.

The effects of RNF213, which leads to moyamoya disease susceptibility, on radiation-induced moyamoya syndrome (MMS) remain unknown. We report a case of MMS after proton beam therapy (PBT) was deployed to treat a brain tumor in a patient with an RNF213 polymorphism. An 8-year-old boy underwent whole ventricular and local PBT for a pineal germ cell tumor and was diagnosed with radiation-induced MMS 9 months later. He underwent right and left revascularization surgeries for cerebral hemodynamic compromise at 17- and 18-years of age, respectively. Genetic analysis revealed a heterozygous germline

Familial Actinic Lichen Planus: Three Cases from the Same Family.

Actinic lichen planus (ALP) that affects sun-exposed areas of the skin is an uncommon variant of lichen planus. While ALP is commonly triggered by ultraviolet radiation exposure, genetic predisposition may also be important in the pathogenesis of the disease. Herein, we report three patients with ALP from the same family, which supports the genetic etiopathogenetic factors of ALP.

Radiation-Triggered Chemokine-Secreting Probiotics for Boron Neutron Capture Therapy Based Radioimmunotherapy.

An ideal radioimmunotherapy eradicates primary tumors and drives immune clearance of residual or metastatic tumor cells, but its clinical translation is severely limited by systemic immune toxicity and the immunosuppressive tumor microenvironment (TME). Here, we constructed engineered Escherichia coli Nissle 1917 with a tannic acid-mediated boronophenylalanine coating and a radiation-responsive genetic circuit, enabling TME-responsive boron delivery for boron neutron capture therapy (BNCT). BNCT-mediated selective tumor killing spared intratumoral probiotics, whose integrated genetic circuit w

Microsatellite instability in acute myelocytic leukaemia developed from A-bomb survivors.

PURPOSE: Genetic alterations, including microsatellite instability (MSI), are ultimate steps toward malignant process. To investigate MSI in A-bomb survivors, leukaemic cells were analysed from 13 acute myelocytic leukaemia patients with a history of radiation exposure and also in 12 de novo patients. MATERIALS AND METHODS: To assess the microsatellite changes, a fluorescent system in 10 loci (BAT40, D3S643, D5S107, IRF1, MYC, D9S171, WT1, TP53, DM, D17S855) was used. RESULTS: MSI analysis revealed a high frequency of multiple microsatellite changes in the exposed patients (84.6%) compared w

Acid sphingomyelinase-deficient human lymphoblasts and mice are defective in radiation-induced apoptosis.

Stress is believed to activate sphingomyelinase to generate ceramide, which serves as a second messenger in initiating the apoptotic response. Conclusive evidence for this paradigm, however, is lacking. In the present study, we used a genetic approach to address this issue directly. We show that lymphoblasts from Niemann-Pick patients, which have an inherited deficiency of acid sphingomyelinase activity, fail to respond to ionizing radiation with ceramide generation and apoptosis. These abnormalities are reversible up on restoration of acid sphingomyelinase activity by retroviral transfer of h

Retrospective biological dosimetry of absorbed radiation.

Frequencies of chromosomal translocations in human peripheral blood lymphocytes irradiated in vitro by 200 kV X rays have been estimated by the fluorescence in situ hybridisation (FISH) technique. Probes specific for whole chromosomes 1, 3 and 4 were labelled with, biotin 16 dUTP + digoxigenin 11 dUTP; biotin 16 dUTP and digoxigenin 11 dUTP respectively, to achieve three colour FISH to distinguish exchanges involving individual chromosomes. The translocation frequencies for individual chromosomes suggested a relationship between the response and the genomic content of the chromosomes studied.

[Premolecular period of the postradiation remodelling of cells].

Recovery of yeast cells after exposure to ionizing radiation was found in 1957. During the first decade, i.e. in the "premolecular period" of studying the phenomenon, its basic features were revealed: dependence on ploidy of cells, on their energy exchange, on radiation LTE, and others. A mathematical model of recovery was proposed; the damages causing death of irradiated haploid and diploid cells were shown to be double strand breaks of DNA. The concepts of universal biological importance of the cell property to repair genetic damages were formulated.

Idiopathic basal cell carcinoma in children.

Basal cell carcinoma (BCC) is the most common nonmelanoma skin cancer in humans, and is the most common malignant neoplasm among adults in the US. The peak incidence occurs in the seventh decade of life. Childhood onset of BCC is rare and usually associated with genetic disorders such as basal cell nevus syndrome, Bazex syndrome, albinism, and xeroderma pigmentosum or due to radiation therapy. Idiopathic childhood onset is less common. A girl with idiopathic onset of BCC who was treated with Mohs micrographic surgery is reported. A computerized review of the literature was performed. A total o

Diagnostic nuclear medicine and risk for the fetus.

The possible detrimental effects on the developing embryo subsequent to irradiation are discussed. The doses to the embryo or fetus encountered for the most common procedures in diagnostic nuclear medicine are evaluated with respect to the threshold doses and the risks per cGy. The threshold dose for fatal and non-fatal malformations or other defects is, at the lowest estimate, 5-10 cGy. The dose which the embryo or fetus receives from diagnostic nuclear medicine is below 1 cGy. For the induction of fatal cancer and for genetic defects no threshold dose is assumed. The risk for the induction o

Nonsolar factors in the etiology of malignant melanoma.

Although epidemiological evidence supports the role of solar radiation in the etiology of cutaneous malignant melanoma (MM) among Caucasians, solar exposure alone cannot explain all the epidemiological patterns of MM. This cancer can arise from both noncutaneous and cutaneous sites not exposed to the sun. Evidence is lacking for the involvement of solar exposure in the etiology of most MM among non-Caucasians. Moreover, among Caucasians, skin pigmentation and other genetically determined factors appear to be important in the risk of MM. Trauma, dietary factors (particularly retinoids), hormone

Chromosome damage and cell proliferation rates in in vitro irradiated whole blood as markers of late radiation toxicity after radiation therapy to the prostate.

PURPOSE: In vitro irradiated blood samples from prostate cancer patients showing late normal tissue damage were examined for lymphocyte response by measuring chromosomal aberrations and proliferation rate. METHODS AND MATERIALS: Patients were selected from a randomized trial evaluating the optimal timing of dose-escalated radiation and short-course androgen deprivation therapy. Of 438 patients, 3% experienced grade 3 late radiation proctitis and were considered to be radiosensitive. Blood samples were taken from 10 of these patients along with 20 matched samples from patients with grade 0 pro

The use of chromosomal aberrations in human lymphocytes for biological dosimetry.

The scoring of chromosomal aberrations in human lymphocytes provides the most sensitive method known for biological dosimetry. By scoring dicentrics in the full genome of 500 cells, average whole-body doses of about 0.1 Gy of X or gamma rays may be detected and higher doses estimated. Acute doses above about 0.2 Gy can be estimated more accurately than similar chronic doses. For radiations of higher LET, for example those encountered in the space environment, the limits of detection in grays are lower. However, expressed in sieverts, the limits of detection are more nearly independent of radia

[Report of Multidisciplinary Treatment for Sisters with Li-Fraumeni Syndrome].

Li-Fraumeni syndrome(LFS)is a hereditary cancer disorder caused by germline variant in TP53 and characterized by various malignancies. Multidisciplinary treatment is needed for tumors of LFS, however, radiation therapy is a relative contraindication because of frequent development of secondary malignancy such as sarcoma in the irradiated field. Case 1: A 22- year-old woman who was diagnosed with LFS by genetic test when she developed upper rectal cancer. Her rectal tumor with marked bilateral lateral lymph node dissection was successfully removed by low anterior resection with extensive latera

Individualized Breast Surveillance in a Young Hodgkin Lymphoma Survivor With High-Risk Features.

Survivors of pediatric Hodgkin lymphoma have an elevated lifetime risk of secondary breast malignancy, particularly those who received chest-directed radiation before age 30. This report describes a 19-year-old female patient, three years post-therapy for stage IIB Hodgkin lymphoma treated with 30 Gy mediastinal radiation, who presented within a structured survivorship program for evaluation of a self-detected, firm, mobile, non-tender left breast mass. Physical examination confirmed a 2 cm lesion in the upper outer quadrant without overlying skin changes, nipple inversion, or lymphadenopathy.

Psychological distress and the perception of radiation risks: the Fukushima health management survey.

OBJECTIVE: To assess relationships between the perception of radiation risks and psychological distress among evacuees from the Fukushima nuclear power plant disaster. METHODS: We analysed cross-sectional data from a survey of evacuees conducted in 2012. Psychological distress was classified as present or absent based on the K6 scale. Respondents recorded their views about the health risks of exposure to ionizing radiation, including immediate, delayed and genetic (inherited) health effects, on a four-point Likert scale. We examined associations between psychological distress and risk percept

The molecular basis of radiosensitivity and chemosensitivity in the treatment of breast cancer.

The molecular basis of sensitivity to therapeutic radiation and chemotherapy is a complex product of cellular and tissue responses. Certain genetic factors can be highlighted as being of special importance in the response of breast cancers to treatment. The breast cancer susceptibility genes, BRCA1 and BRCA2, determine the phenotype of the tumor, with BRCA1- or BRCA2-deficient tumors showing marked sensitivity to ionizing radiation and drugs that produce double-strand breaks. However, the extent to which loss of BRCA1 or BRCA2 function occurs in sporadic cancer has not yet been determined. The
