Wnt/β-catenin signaling controls intrahepatic biliary network formation in zebrafish by regulating notch activity.

UNLABELLED: Malformations of the intrahepatic biliary structure cause cholestasis, a liver pathology that corresponds to poor bile flow, which leads to inflammation, fibrosis, and cirrhosis. Although the specification of biliary epithelial cells (BECs) that line the bile ducts is fairly well understood, the molecular mechanisms underlying intrahepatic biliary morphogenesis remain largely unknown. Wnt/β-catenin signaling plays multiple roles in liver biology; however, its role in intrahepatic biliary morphogenesis remains unclear. Using pharmacological and genetic tools that allow one to manipu

Morphogen pathways in systemic sclerosis.

The morphogen pathways Wnt, hedgehog, and Notch are key regulators of organ development and tissue homeostasis. In adults, the tightly regulated activity of morphogen pathways is essential for cell renewal and tissue regeneration. Loss of control and persistent activation of morphogen pathways, however, can lead to a variety of diseases, including malignancy and fibrotic disorders. In recent years, pathological activation of Wnt, hedgehog, and Notch pathways have been described in systemic sclerosis (SSc) and other fibrotic diseases. Experimental models reveal that morphogen pathways drive fib

Novel In-Frame Deletion Mutation in NOTCH1 in a Chinese Sporadic Case of Adams-Oliver Syndrome.

Adams-Oliver syndrome (AOS) is a rare hereditary disorder characterized by aplasia cutis congenita (ACC) and terminal transverse limb defects. The etiology of AOS has remained largely unknown, although mutations in the notch receptor 1 (NOTCH1) gene are most common genetic alteration associated with this disease. In this study, we aimed to identify the case of a 6-year-old boy, who presented with large ACC of the scalp and aortic valve stenosis, suggesting the possibility of AOS. Whole-exome sequencing identified a novel, de novo, in-frame deletion in the NOTCH1 gene (NOTCH1 c.1292_1294del, p.

The expanding landscape of Deltex: signaling crosstalk and clinical implications.

The deltex (DTX) gene family encodes RING-type E3 ubiquitin ligases that are central to the regulation of the Notch signaling pathway, a mechanism governing cell fate, differentiation, proliferation, and apoptosis during development and adult tissue homeostasis. In Drosophila melanogaster, the Deltex (Dx) fine-tunes Notch signaling by modulating receptor endocytosis and degradation, acting as either an enhancer or a suppressor depending on the context. Insights from flies have revealed the evolutionary conservation of the Dx-Notch interaction across species, including humans. However, recent s

A Drosophila Su(H) model of Adams-Oliver Syndrome reveals cofactor titration as a mechanism underlying developmental defects.

Notch signaling is a conserved pathway that converts extracellular receptor-ligand interactions into changes in gene expression via a single transcription factor (CBF1/RBPJ in mammals; Su(H) in Drosophila). In humans, RBPJ variants have been linked to Adams-Oliver syndrome (AOS), a rare autosomal dominant disorder characterized by scalp, cranium, and limb defects. Here, we found that a previously described Drosophila Su(H) allele encodes a missense mutation that alters an analogous residue found in an AOS-associated RBPJ variant. Importantly, genetic studies support a model that heterozygous D

The mechanism of gamma-secretase activities through high molecular weight complex formation of presenilins is conserved in Drosophila melanogaster and mammals.

Mutations in presenilin 1 (PS1) and PS2 genes contribute to the pathogenesis of early onset familial Alzheimer's disease by increasing secretion of the pathologically relevant Abeta42 polypeptides. PS genes are also implicated in Notch signaling through proteolytic processing of the Notch receptor in Caenorhabditis elegans, Drosophila melanogaster, and mammals. Here we show that Drosophila PS (Psn) protein undergoes endoproteolytic cleavage and forms a stable high molecular weight (HMW) complex in Drosophila S2 or mouse neuro2a (N2a) cells in a similar manner to mammalian PS. The loss-of-funct

Triphenyl phosphate accelerates collagen-rich ECM remodeling and proliferation in gastric cancer via partially TGFβ/NOTCH-dependent transcription Programs.

Organophosphate esters are increasingly used as replacements for legacy halogenated flame retardants, yet their tumor-promoting potential remains poorly defined. Here, we identify triphenyl phosphate (TPP) as a potent environmental driver of extracellular-matrix (ECM) remodeling and growth in gastric cancer (GC). Chemical-gene signatures for phosphate flame retardants (PFRs) from the Comparative Toxicogenomics Database were projected onto TCGA-STAD using single-sample GSEA and compared with a curated proliferation signature, nominating TPP as the top correlate. In AGS cell line, TPP increased

Genome-Wide Scan for Visceral Leishmaniasis in Mixed-Breed Dogs Identifies Candidate Genes Involved in T Helper Cells and Macrophage Signaling.

We conducted a genome-wide scan for visceral leishmaniasis in mixed-breed dogs from a highly endemic area in Brazil using 149,648 single nucleotide polymorphism (SNP) markers genotyped in 20 cases and 28 controls. Using a mixed model approach, we found two candidate loci on canine autosomes 1 and 2. The positional association on chromosome 2 mapped to a predicted DNAse sensitive site in CD14+ monocytes that serve as a cis-regulatory element for the expression of interleukin alpha receptors 2 (IL2RA) and 15 (IL15RA). Both interleukins were previously found to lead to protective T helper 1 cell

Opposing role of Notch1 and Notch2 in a Kras(G12D)-driven murine non-small cell lung cancer model.

Lung cancer is the leading cause of cancer-related deaths worldwide. Recently, we have shown that Notch1 inhibition resulted in substantial cell death of non-small cell lung cancer (NSCLC) cells in vitro. New compounds targeting Notch signal transduction have been developed and are now being tested in clinical trials. However, the tumorigenic role of individual Notch receptors in vivo remains largely unclear. Using a Kras(G12D)-driven endogenous NSCLC mouse model, we analyzed the effect of conditional Notch1 and Notch2 receptor deletion on NSCLC tumorigenesis. Notch1 deficiency led to a reduce

Plant pathogenic nematode exosomes remodel vector tracheae to enhance pathogen transmission.

Pathogens frequently employ vector-manipulation strategies to enhance their transmission efficiency. Exosomes are increasingly recognized as mediators of interspecific communication between pathogens and their vectors. However, the mechanisms by which plant pathogenic nematode exosomes mediate cross-kingdom manipulation of vector development remain largely unexplored. Here, we demonstrate that the plant pathogenic nematode (Bursaphelenchus xylophilus), transmitted by the vector beetle (Monochamus alternatus), secretes exosomes containing microRNAs (miRNAs) that remodel the tracheal development

Analysis of cancer mutations introduced into the Drosophila melanogaster Notch negative regulatory region uncovers a diversity of regulatory outcomes.

Activating mutations of Notch are drivers of the blood cell cancer, T-ALL, and some solid tumours. The negative regulatory region (NRR) of the extracellular domain (ECD) and the PEST region of the intracellular domain (ICD) are mutation hot spots which can act synergistically in T-ALL. The NRR, comprised of a heterodimerisation domain (HD) and three Lin12/Notch repeats (LNR A-C), masks the S2 cleavage site, normally only exposed following ligand binding and cleaved as the first step that ultimately leads to ICD release. Drosophila mutants have played a key role in analysing Notch structure/fun

Integrative Analysis of Key Signalling Pathways in Neural Tube Defects: From Molecular Mechanisms to Therapeutic Strategies.

Neural tube defects (NTDs), such as anencephaly and spina bifida, are prevalent congenital anomalies of the central nervous system. These defects can give rise to severe lifelong disabilities and incur substantial healthcare expenses for the affected individuals. The occurrence of NTDs is caused by multiple factors, including molecular regulatory mechanisms and environmental factors. This article comprehensively reviews the underlying mechanisms of three crucial signalling pathways associated with neural tube development: the Wnt/Planar Cell Polarity (PCP) signalling pathway, the Sonic Hedgeho

The Notch signaling pathway in regulating bone and cartilage homeostasis: novel insights into the pathogenesis and therapeutics of osteoarthritis.

Osteoarthritis (OA) is a chronic degenerative joint disorder characterized by cartilage degradation, bone hyperplasia, and synovitis. It is a primary cause of joint pain and dysfunction globally. The increasing incidence of OA, driven by an aging population, profoundly impairs patients' quality of life while imposing a growing economic burden on societies and families. Thus, in-depth investigations into the pathogenesis of OA, along with the exploration of novel therapeutic targets and strategies, hold significant clinical implications and social value. The Notch signaling pathway, a highly co

Pleiotropic Role of Notch Signaling in Human Skin Diseases.

Notch signaling orchestrates the regulation of cell proliferation, differentiation, migration and apoptosis of epidermal cells by strictly interacting with other cellular pathways. Any disruption of Notch signaling, either due to direct mutations or to an aberrant regulation of genes involved in the signaling route, might lead to both hyper- or hypo-activation of Notch signaling molecules and of target genes, ultimately inducing the onset of skin diseases. The mechanisms through which Notch contributes to the pathogenesis of skin diseases are multiple and still not fully understood. So far, No

The Role of Notch Signaling in Macrophages during Inflammation and Infection: Implication in Rheumatoid Arthritis?

Notch signaling coordinates numerous cellular processes and has been implicated in many pathological conditions, including rheumatoid arthritis (RA). Although the role of Notch signaling in development, maturation, differentiation, and activation of lymphocytes has been comprehensively reported, less is known about its role in myeloid cells. Certainly, limited data are available about the role of Notch signaling in macrophages during inflammation and infection. In this review, we discuss the recent advances pertaining to the role of Notch signaling in differentiation, activation, and metabolis

Effect of melatonin on renewal of chicken small intestinal mucosa.

A higher concentration of melatonin (MEL) in the intestine - even more than that in the plasma and pineal gland - implies its putative important role in the gastrointestinal structural or functional regulation. However, little evidence has shown that MEL can regulate the physiological functions of the intestinal mucosa. In this study, fertilized chicken eggs were treated with MEL (0.1 to 10 μg/d) from embryonic d 12 (E12) to post-hatching d 6 (D6), and the small intestine samples were collected at D6 to determine the changes in mucosal construction and function. Results of HE staining showed t

Expression of Notch signaling pathway during osteoarthritis in the temporomandibular joint.

PURPOSE: Our study aim was to characterize the expression of Notch molecules during temporomandibular joint osteoarthritis (TMJOA), thus exploring the mechanism and roles that Notch signaling possibly plays in the initiation and progression of TMJOA. MATERIALS AND METHODS: A total of 126 mice were divided randomly into experimental groups, a sham-surgery group and a normal control group. In the experimental group, total discectomy was performed in the right temporomandibular joint (TMJ) to induce TMJOA; the sham-operation group underwent the same procedure without disc removal, and the normal

Notch signaling in ocular vasculature development and diseases.

Ocular angiogenesis, characterized by the formation of new blood vessels in the avascular area in eyes, is a highly coordinated process involved in retinal vasculature formation and several ocular diseases such as age-related macular degeneration, proliferative diabetic retinopathy and retinopathy of prematurity. This process is orchestrated by complicated cellular interactions and vascular growth factors, during which endothelial cells acquire heterogeneous phenotypes and distinct cellular destinations. To date, while the vascular endothelial growth factor has been identified as the most crit
