The active transport of fluorescein by the retinal vessels and the retina.

1. The movement of fluorescein across the retinal surface of the rabbit's eye was estimated by measuring the concentration gradient of the dye in the vitreous body. These measurements were made in vivo by means of a slit-lamp fluorophotometer, or were taken from frozen sections of enucleated eyes.2. In the normal eye, fluorescein does not pass from the blood to the vitreous body across any part of the retina. When injected into the vitreous body it passes rapidly out across the entire retinal surface, even against a very large concentration gradient.3. A variety of metabolic and competitive in…

US Retina Specialists' Preferences for Geographic Atrophy Treatment.

PURPOSE: Two complement inhibitors are US-approved for geographic atrophy (GA) and other treatments are in development. We present the first quantitative assessment of retina specialists' (RS) preferences for GA treatment. We explored how GA treatment characteristics affect RS' preferences, the trade-offs RS are willing to make between them, and whether this varies across patient profiles. METHODS: US-based RS with ≥7 years of clinical experience and ≥50 patients with GA in the previous year selected their best and second-best treatment recommendations for specific patient profiles in 12 task…

Localization of Ceramide Synthase 5 in Mouse Retina.

Ceramide synthases (CerS) are key enzymes in sphingolipid metabolism that regulate fundamental cellular processes, including apoptosis, cell growth, and homeostasis. Among the six known mammalian isoforms (CerS1-CerS6), CerS5 has been particularly well studied for its involvement in the synthesis of the sphingolipid C16-ceramide. However, its expression, localization, and functional significance of CerS5 in the retina remain unclear. In the present study, we investigated the presence, distribution, and functional role of CerS5 in mouse retina using CerS5 knockout (KO) mice. We performed quanti…

Müller Glia-Vasculature Interactions in the Developing Retina.

Coordinated signaling among neurons, glia, and the vasculature is essential for nervous system development. In the developing retina, spontaneous cholinergic retinal waves are the primary source of neural activity during the early maturation of the vasculature. Here, we test the hypothesis that retinal waves influence angiogenesis and the maturation of the glial-vascular interface. We first found that retinal vasculature grew normally in mice lacking β2-containing nicotinic acetylcholine receptor-mediated retinal waves, demonstrating that early spontaneous activity is not required for angiogen…

Endothelial Zmiz1 modulates developmental angiogenesis in the retina.

Angiogenesis is a highly coordinated process involving the control of various endothelial cell behaviors. Mechanisms for transcription factor involvement in the regulation of endothelial cell dynamics and angiogenesis have become better understood, however much remains unknown, especially the role of non-DNA binding transcriptional cofactors. Here, we show that Zmiz1, a transcription cofactor, is enriched in the endothelium and critical for embryonic vascular development, postnatal retinal angiogenesis, and pathological angiogenesis in a model of oxygen-induced retinopathy (OIR). In mice, endo…
