The levels of rhodopsin and opsin were investigated in relation to the maintenance of retinal structure in retinas of vitamin A--deficient rats in low levels of cyclic illumination (1.5 to 2 foot-candles). Rhodopsin levels decreased in the deficient retinas to approximately 20% of control at 9 weeks, and this level was retained through 39 weeks on the deficient diet. Opsin levels decreased at a slower rate but reached about 20% of control levels at 32 weeks. Despite the decrease in rhodopsin levels, obvious deterioration of disc structure was not observed until 16 weeks of deficiency, when ops

Retinal development was studied in eyes from fetal and neonatal human anencephalic (AnC) and normal age-matched infants to determine the time of retinal ganglion cell (GC) loss and its effect on the development of other retinal neurons. At fetal week (Fwk) 14, GC loss was evident in central retina and by Fwk 19-20 almost all GC were absent, although immunocytochemical labeling for GC markers brain 3, neurofilament M and parvalbumin detected a few GC in the AnC far periphery at older ages. The inner nuclear and inner plexiform (IPL) layers showed variable amounts of thinning but all normal bipo

Autosomal recessive childhood-onset severe retinal dystrophy (arCSRD) designates a heterogeneous group of disorders affecting rod and cone photoreceptors simultaneously. The most severe cases are termed Leber congenital amaurosis (LCA), while the less aggressive forms are usually considered juvenile retinitis pigmentosa. Recently, mutations in the retinal-specific guanylate cyclase gene were found in patients with LCA. Disease genes implicated in other forms of arCSRD are expected to encode proteins present in the neuroretina or in the retinal pigment epithelium (RPE). The RPE, a monolayer of

To explore the molecular similarities and potential evolutionary origins of vertebrate photoreceptor types, we analyzed single-cell and -nucleus transcriptomic atlases from six vertebrate species: zebrafish, chicken, lizard, opossum, ground squirrel, and human. Comparative analyses identified conserved transcriptional signatures for the five ancestral photoreceptor types: red, blue, green, and UV cones, as well as rods. We further identified and validated molecular markers of the principal and accessory members of the tetrapod double cone. Comparative transcriptomics suggests that the principa

A hallmark of inherited retinal degenerative diseases such as retinitis pigmentosa (RP) is progressive structural and functional remodeling of the remaining retinal cells as photoreceptors degenerate. Extensive remodeling of the retina stands as a barrier for the successful implementation of strategies to restore vision. To understand the molecular basis of remodeling, we performed analyses of single-cell transcriptome data from adult zebrafish retina of wild type AB strain (WT) and a P23H mutant rhodopsin transgenic model of RP with continuous degeneration and regeneration. Retinas from both

PURPOSE: To study and compare effects of syngeneic bone marrow mononuclear stem cells (BM-MNCs) transplants on inherited retinal degeneration in two animal models with different etiologies: the RCS and the P23H-1 rats. To compare the safety and efficacy of two methods of intraocular delivery: subretinal and/or intravitreal.
METHODS: A suspension of BM-MNCs was injected subretinally or intravitreally in the left eyes of P23H-1 and RCS rats at post-natal day (P) 21. At different survival intervals after the injection: 7, 15, 30 or 60 days, the retinas were cross-sectioned, and photoreceptor sur

Retinal circuit assembly relies on the precise timing and positioning of key molecules between neuronal partners to mediate proper synapse formation. In the outer retina, horizontal cells (HCs) are important interneurons that make the first contacts with photoreceptors and begin to segregate visual information into two distinct pathways by selectively forming synapses to the different types of photoreceptors. Dendrites of HCs synapse exclusively to cone photoreceptors, whereas the axon terminal synapses to rod photoreceptors. Failure to properly form these early connections disrupts the downst

It has been shown that mammalian retinal glial (Müller) cells act as living optical fibers that guide the light through the retinal tissue to the photoreceptor cells (Agte et al., 2011; Franze et al., 2007). However, for nonmammalian species it is unclear whether Müller cells also improve the transretinal light transmission. Furthermore, for nonmammalian species there is a lack of ultrastructural data of the retinal cells, which, in general, delivers fundamental information of the retinal function, i.e. the vision of the species. A detailed study of the cellular ultrastructure provides a basic

Objectives: Retinitis pigmentosa (RP) is commonly initiated by rod photoreceptor degeneration due to genetic mutations, followed by secondary cone loss and progressive blindness. Preserving rod function during the earlier stages of RP is a key therapeutic goal, as rod survival supports cone maintenance and delays vision loss. In this study, we investigated the therapeutic potential of transient knockdown of retinoid-related orphan receptor beta (RORB) using a cell-penetrating asymmetric small interfering RNA (cp-asiRORB) in RhoP23H mice, a model of autosomal dominant RP. While the role of RORB

PURPOSE: Synaptic dysfunction and neurite remodeling are early features of many inherited retinal diseases, but whether synaptic function can be restored after circuit remodeling remains unclear. We evaluated the therapeutic potential of adeno-associated virus (AAV)-mediated α2δ4 gene supplementation in a mouse model of CACNA2D4-associated retinal synaptopathy.
METHODS: AAV8-GRK1-α2δ4 was delivered by subretinal injection to α2δ4 knockout mice at neonatal, young adult, or middle-aged adult stages. Retinal function was assessed by electroretinography. Synaptic organization and circuit remodeli

Vitamin A immunoreactive sites were studied in the retina and pineal organ of the frog, Rana esculenta, by the peroxidase antiperoxidase, avidin-biotinperoxidase and immunogold methods. In dark-adapted material, strong immunoreaction was found in the outer and inner segments of the photoreceptor cells of both retina and pineal organ, as well as in the pigment epithelium, retinal Müller cells and pineal ependymal cells. In light-adapted retina, cones and green (blue-sensitive) rods were immunopositive. At the electron microscopic level, immunogold particles were found on the membranes of the ph

Vision depends on the continuous recycling of the visual pigment chromophore 11-cis-retinal, which is converted by light into all-trans-retinal at the onset of the phototransduction cascade. Cellular retinaldehyde-binding protein 1 (CRALBP1) is a key component of both the canonical visual cycle in the retinal pigment epithelium (RPE) and the cone-specific recycling pathway mediated by Müller glia (MG) cells. Previous studies demonstrated that knockout of the RPE-specific gene rlbp1a (encoding CRALBP1) results in severe physiological and morphological defects that are evident already at cone do

Mutations in the photoreceptor-specific tetraspanin gene peripherin-2 (PRPH2) lead to widely varying forms of retinal degeneration ranging from retinitis pigmentosa to macular dystrophy. Both inter- and intra-familial phenotypic heterogeneity has led to much interest in uncovering the complex pathogenic mechanisms of PRPH2-associated disease. Majority of disease-causing mutations in PRPH2 reside in the second intradiscal loop, wherein seven cysteines control protein folding and oligomerization. Here, we utilize knockin models to evaluate the role of three D2 loop cysteine mutants (Y141C, C213Y

Hallmarks of Retinitis Pigmentosa (RP), a family of genetic diseases, are a typical rod-cone-degeneration with initial night blindness and loss of peripheral vision, followed by decreased daylight sight and progressive visual acuity loss up to legal blindness. Great heterogeneity in nature and function of mutated genes, variety of mutations for each of them, variability in phenotypic appearance and transmission modality contribute to make RP a still incurable disease. Translational research relies on appropriate animal models mimicking the genetic and phenotypic diversity of the human patholog

Retinitis pigmentosa (RP) is a constellation of genetically inherited diseases, most often caused initially by mutation of a rod photoreceptor gene and leading eventually to blindness.1 Although the ultimate prognosis for RP is grim, many forms of the disease are slow to progress, with patients maintaining usable vision at least until middle age (for example, Berson et al.2,3 and Hartong et al.4). Retinal circuits must therefore be able to adapt to photoreceptor loss,5 though little is presently known about the cellular mechanisms mediating these alterations. Here, we show that in the rhodopsi

Light is the visible part of the electromagnetic radiation within a range of 380-780 nm; (400-700 on primates retina). In vertebrates, the retina is adapted to capturing light photons and transmitting this information to other structures in the central nervous system. In mammals, light acts directly on the retina to fulfill two important roles: (1) the visual function through rod and cone photoreceptor cells and (2) non-image forming tasks, such as the synchronization of circadian rhythms to a 24 h solar cycle, pineal melatonin suppression and pupil light reflexes. However, the excess of illum

NADPH-dependent retinol dehydrogenase 12 (RDH12) reduces all-trans-retinal in photoreceptor inner segments to regulate the phototransduction cascade and control the concentration of retinoids in the retina. Variants in RDH12 have been reported to cause an array of retinal disease phenotypes, including autosomal recessive (AR) Leber congenital amaurosis (LCA), a severe form of childhood blindness, and autosomal dominant (AD) retinitis pigmentosa (RP), a milder late-onset rod-dominant disease. Animal models have failed to recapitulate the clinical phenotype; hence, we generated human induced plu

American Staffordshire Terriers (ASTs) with a c.296G>A variant in ARSG develop progressive ataxia, cerebellar atrophy, and neuronal accumulation of autofluorescent storage material. Human subjects with ARSG variants exhibit hearing loss and rod-cone dystrophy without apparent other neurological involvement and arsg knockout mice exhibit progressive ataxia, lysosomal storage, and photoreceptor loss. Owners of 8 of 11 affected ASTs evaluated for the ARSG risk variant reported observing visual impairment in their dogs, suggesting that the canine disease may involve retinal dysfunction consistent

This presentation will survey the retinal architecture, advantages, and limitations of several lesser-known rodent species that provide a useful diurnal complement to rats and mice. These diurnal rodents also possess unusually cone-rich photoreceptor mosaics that facilitate the study of cone cells and pathways. Species to be presented include principally the Sudanian Unstriped Grass Rat and Nile Rat (Arvicanthis spp.), the Fat Sand Rat (Psammomys obesus), the degu (Octodon degus) and the 13-lined ground squirrel (Ictidomys tridecemlineatus). The retina and optic nerve in several of these speci

Object recognition is impacted by visual crowding. This phenomenon, attributed to cortical pooling, is generally studied in the periphery. Crowding mechanisms in the foveola-the 1° retinal region with highest resolution-remain unclear due to difficulties in controlling for optical factors and incessant fixational eye motion. Using high-resolution retinal imaging and retinal-contingent stimulus rendering to overcome these limitations, we demonstrated that the critical spacing at the preferred retinal locus approximates a single cone's diameter. However, just 0.25° away, mislocalization errors s
