The glutamate and carbachol effects on the early post-denervation depolarization in rat diaphragm are directed towards furosemide-sensitive chloride transport.

The membrane potentials of denervated muscle fibres of the rat diaphragm kept in a tissue culture medium are depolarized by about 8-10 mV (1 … elease, from nerve endings leads to the activation of the furosemide-sensitive Cl- transport in the sarcolemma, which is responsible for the early post-denervation depolarization.

[Clinico-immunologic characteristics of complicated and uncomplicated yersiniosis].

Asymmetric affection of the major lower limb joints is a characteristic feature of the joint syndrome in yersiniosis-associated arthritis. T … e with the cardiac interstitial tissue, while in authentically diagnosed myocarditis cases with the sarcolemma. The investigation findings strongly suggest a high degree of involvement of immune and autoimmune processes in the pathogenesis of arthritides secondary to Yersinia infection.

Skeletal, cardiac and tongue muscle pathology, defective retinal transmission, and neuronal migration defects in the Large(myd) mouse defines a natural model for glycosylation-deficient muscle - eye - brain disorders.

We have recently shown that a deletion in the Large gene, encoding a putative glycosyltransferase, is the molecular defect underlying the myodystrophy (previously myd; now Large(myd)) mouse. Here we show that the muscular dystrophy phenotype is not confined to skeletal muscle, but is also present in the heart and tongue. Immunohistochemistry indicates disruption of the dystrophin-associated glycoprotein complex (DGC) in skeletal and cardiac muscle. Quantitative western blotting shows a general increase in the expression of DGC proteins and of dysferlin and caveolin-3 in mutant skeletal muscle.

Effects of adenosine on the contractility of normoxic rainbow trout heart.

Temperature strongly affects oxygen solubility in water, oxygen convection in the blood and locomotor activity of the fish. Since oxygen supply and demand are temperature dependent, it was hypothesized that the purinergic control of the heart, one of the most important mediators in oxygen-limited conditions, might also show temperature dependence. Therefore, the present study examines the effects of adenosine (Ado), a purinergic agonist, on the contractile and electrical activity of the thermally acclimated trout ( Oncorhynchus mykiss Walbaum) heart. The fish were acclimated to either 4 degree

Isoform-specific alterations in cardiac and erythrocyte Na+,K+-ATPase activity induced by norepinephrine.

BACKGROUND: Myocardial Na+,K+-ATPase activities are decreased in congestive heart failure because of an increase in plasma norepinephrine levels, but it is difficult to monitor the activities in the clinical setting. METHODS AND RESULTS: This study investigated whether erythrocyte Na+,K+-ATPase activity can reflect myocardial enzyme activity and whether isoform-specific alterations occur in the presence of catecholamine. Na+,K+-ATPase activity was measured by the colorimetric method by using the left ventricular myocardium and erythrocytes prepared from eight rabbits given norepinephrine for

Tubular aggregates and partial cytochrome c oxidase deficiency in skeletal muscle of patients with AIDS treated with zidovudine.

We report on two patients, who had myalgias while receiving long-term zidovudine treatment for an HIV infection, in whom muscle biopsy findings included a partial cytochrome c oxidase (CCO) deficiency, a feature of zidovudine myopathy, and tubular aggregates, a finding hitherto unreported in HIV-infected patients. The CCO deficit was observed in 28% and 24% of muscle fibers, respectively. Tubular aggregates were the prominent histopathological feature in patient 1, and were detected by systematic electron microscopy in patient 2. Inflammation and myonecrosis were not detected. In patient 1, th

A morphological control for ventricular pathology in man: a morphometric and morphologic assessment of LV myofibres in secundum ASD.

Ethical considerations preclude the biopsy of normal human myocardium. As a consequence, morphological investigations of diseased human heart muscle are hampered by a lack of suitable normal control tissue. The left ventricular (LV) myocardium of patients with isolated secundum atrial septal defect (ASD) is considered to be normal. This study was designed to investigate the possibility that the fine-structure of LV myofibres in hearts with ASD could be used as normal controls for myofibre pathomorphology. Wedge biopsies from the LV of four adults undergoing elective surgery for the repair of A

Sarcolemmal dependence of isosorbide dinitrate-induced relaxation of aortic smooth muscle.

Studies were conducted to determine if ISDN antagonizes the contractile responses to calcium of chemically-skinned aortic smooth muscle preparations. Isosorbide dinitrate relaxed intact aortic smooth muscle segments in a dose-dependent manner (0.01-100 microM). At all concentrations tested, ISDN was unable to relax triton-skinned smooth muscle preparations. However, the tension development of skinned preparations was inhibited by trifluoperazine (10 microM), a calmodulin antagonist. These results suggest that the relaxation of intact aortic smooth muscle preparations induced by ISDN is not rel

Junctophilin-2 abundance is unaltered in human heart failure samples with disrupted T-tubules and contractility.

T-tubules are structural components of the cardiomyocyte plasma membrane that are imperative for efficient excitation-contraction coupling and cardiac contraction. Junctophilin-2 (JPH2) has been postulated in multiple preclinical models to play a key role in T-tubule structure development, maintenance, and organisation by anchoring the sarcoplasmic reticulum to the sarcolemma and maintaining the spatial distance between excitation contraction coupling proteins. Animal studies have shown that JPH2 downregulation leads to T-tubule dysfunction and impaired cardiac contractility, and upregulation …

Indirect evidence that anoxia exposure and cold acclimation alter transarcolemmal Ca2+ flux in the cardiac pacemaker, right atrium and ventricle of the red-eared slider turtle (Trachemys scripta).

We indirectly assessed if altered transarcolemmal Ca2+ flux accompanies the decreased cardiac activity displayed by Trachemys scripta with anoxia exposure and cold acclimation. Turtles were first acclimated to 21 °C or 5 °C and held under normoxic (21N; 5N) or anoxic conditions (21A; 5A). We then compared the response of intrinsic heart rate (fH) and maximal developed force of spontaneously contracting right atria (Fmax,RA), and maximal developed force of isometrically-contracting ventricular strips (Fmax,V), to Ni2+ (0.1-10 mM), which respectively blocks T-type Ca2+ channels, L-type Ca2+ chan…

Absence of synemin in mice causes structural and functional abnormalities in heart.

Cardiomyopathies have been linked to changes in structural proteins, including intermediate filament (IF) proteins located in the cytoskeleton. IFs associate with the contractile machinery and costameres of striated muscle and with intercalated disks in the heart. Synemin is a large IF protein that mediates the association of desmin with Z-disks and stabilizes intercalated disks. It also acts as an A-kinase anchoring protein (AKAP). In murine skeletal muscle, the absence of synemin causes a mild myopathy. Here, we report that the genetic silencing of synemin in mice (synm -/-) causes left vent

[Changes in hydrodynamic diameter of myometrium plasmalemma vesicles under artificial creation of transmembrane proton gradients].

A method of laser-correlation spectrophotometry was applied for finding the effect of proton transmembrane gradient on hydrodynamic diameter of plasmalemma smooth muscle vesicles. It is shown, that the building of protons gradient on the membrane of vesicles, pHi = 6.0; pHo = 7.5 (delta pH = 1.5) results in the reliable decrease of their hydrodynamic diameter (from 212 +/- 6 nm in monitoring to 174 +/- +/- 0.25 nm in experiment). The exit of K+ from vesicles (KCl concentration inside vesicles is 150 mM), on the contrary, evokes augmentation of hydrodynamic diameter. The chemical paravariation

Plectin tethers desmin intermediate filaments onto subsarcolemmal dense plaques containing dystrophin and vinculin.

Plectin is a versatile cytoskeletal linker protein that preferentially localizes at interfaces between intermediate filaments and the plasma membrane in muscle, epithelial cells, and other tissues. Its deficiency causes muscular dystrophy with epidermolysis bullosa simplex. To better understand the functional roles of plectin beneath the sarcolemma of skeletal muscles and to gain some insights into the underlying mechanism of plectin-deficient muscular dystrophy, we studied in vivo structural and molecular relationships of plectin to subsarcolemmal cytoskeletal components, such as desmin, dyst…

Glycogen consumption in hypoxic rat cardiomyocytes.

Glycogen consumption was investigated in isolated adult rat myocytes incubated for 2 h (37 degrees C) in substrate-free, hypoxic Krebs-Henseleit bicarbonate buffer. No consumption of glycogen occurred after 1 h of incubation, and the residual glycogen after 2 h was 23% despite an 89% reduction of the initial ATP content (from 27.1 +/- 1.8 to 3.1 +/- 0.5 nmol/mg dry weight, n = 12). The residual glycogen was not due to lactate inhibition of glycolytic enzymes, since myocytes incubated in the presence of 5 mM glucose maintained high energy phosphates throughout the incubation period despite a co

Utilization of myositis antibody-specific panel for diagnosis, treatment, and evaluation of disease progression.

The idiopathic inflammatory myopathies (IIM) are rare sporadic disorders with an overall annual incidence of approximately 1 in 100,000 and with a higher incidence in women. IIM is an autoimmune process leading to muscle inflammation due to a 'dysfunctional adaptive immune response evidenced by cell-mediated myocytoxicity, a high prevalence of autoantibodies and overexpression of Major Histocompatibility (MHC) I and II molecules on the muscle sarcolemma'. These autoimmune processes can be appreciated as inflammatory infiltrates in muscle biopsies. Common clinical findings in patients diagnosed…
