Aphicidal efficacy of scorpion- and spider-derived neurotoxins.

Insect-specific neurotoxins that act within the insect hemocoel (body cavity) represent an untapped resource for insect pest management. On the basis of recent advances made in development of appropriate delivery systems for transport of these toxins from the insect gut, across the gut epithelium to their target site, we screened neurotoxins derived from scorpion or spider venom for efficacy against the pea aphid, Acyrthosiphon pisum, and the green peach aphid, Myzus persicae. Toxins were selected to represent different modes of electrophysiological action, including activity on voltage-gated

Scorpion venom peptides with no disulfide bridges: a review.

Scorpion venoms are rich sources of biologically active peptides that are classified into disulfide-bridged peptides (DBPs) and non-disulfide-bridged peptides (NDBPs). DBPs are the main scorpion venom components responsible for the neurotoxic effects observed during scorpion envenomation as they usually target membrane bound ion channels of excitable and non-excitable cells. Several hundred DBPs have been identified and functionally characterized in the past two decades. The NDBPs represent a novel group of molecules that have gained great interest only recently due to their high diversity bot

Fatal intracerebral haemorrhage following scorpion sting.

Though uncommon, scorpion stings can cause cerebrovascular accidents by various mechanisms such as venom induced autonomic storm leading to hypertension, hypotension, myocarditis, DIC or vasculitis by direct actions. We present a case of intracerebral bleed following scorpion sting, which is a rare presentation and seldom reported.

Scorpion antivenom effect of micropropagated Aristolochia elegans.

Aristolochia elegans Mast. (Aristolochiaceae) has been used to treat scorpion envenoming in Mexican traditional medicine. In vitro studies of the pharmacological activity of raw extracts from A. elegans roots have shown activity against scorpion bite. The aim of the present study was to determine for the first time the antagonistic effect of hexane and methanol extracts of the aerial parts and roots from micropropagated A. elegans plants in a model of isolated guinea-pig ileum contracted by scorpion bite. Results showed that the methanol extracts of aerial organs (74%) and roots (65%) of micro

Reproductive system of female scorpion: a partial review.

The female scorpion ovariuterus was examined in 10 scorpion species belonging to five families: Buthidae, Vaejovidae, Scorpionidae, Urodacidae, and Diplocentridae. Two main patterns of development are known in scorpions: (1) The apoikogenic type with an ovariuterus containing yolk-rich eggs housed in follicles. This type is found in many scorpion taxa (largely buthids). A peculiar case of apoikogenic ovariuterus is a "beaded" ovariuterus where most of the ova's embryogenesis takes place inside the ovariuterus rather than on pedicels situated on the external wall of the ovariuterus as in most b…

A scorpion neurotoxin increases the potency of a fungal insecticide.

The low virulence of the insecticidal fungus Metarhizium anisopliae has stymied its widespread use in controlling insect pests. We show that high-level expression of an insect-specific neurotoxin from the scorpion Androctonus australis in hemolymph by M. anisopliae increases fungal toxicity 22-fold against tobacco hornworm (Manduca sexta) caterpillars and ninefold against adult yellow fever mosquitoes (Aedes aegypti) without compromising host specificity. Prelethal effects include reduced mobility and feeding of the insects targeted.

Genetic mechanisms of scorpion venom peptide diversification.

The diversity of scorpion venom peptides is well shown by the presence of about 400 such polypeptides with or without disulfide bonds. Scorpion toxins with disulfide bonds present a variety of sequence features and pharmacological functions by affecting different ion channels, while the venom peptides without disulfide bonds represent a new subfamily, having much lower sequence homology among each other and different functions (e.g. bradykinin-potentiating, antimicrobial, molecular cell signal initiating and immune modulating). Interestingly, all scorpion venom peptides with divergent function

[Recombinant antibodies: a new application in scorpion envenomation?].

Serotherapy is the only specific treatment for envenomation. The antibodies are obtained after the purification of serum from hyperimmunised horses and are used after fragmentation in the form of polyclonal Fab or F(ab)'2. The anti-venom sera are heterogeneous, and their protective effect is often weak. The administration of these preparations induces risks of immediate or delayed side effects: hypersensitivity reactions, anaphylactic shock and serum sickness. This observation led us to develop new forms of antibodies produced by molecular engineering, capable of specifically neutralizing the

Adaptive evolution of scorpion sodium channel toxins.

Gene duplication followed by positive Darwinian selection is an important evolutionary event at the molecular level, by which a gene can gain new functions. Such an event might have occurred in the evolution of scorpion sodium channel toxin genes (alpha- and beta-groups). To test this hypothesis, a robust statistical method from Yang and co-workers based on the estimation of the nonsynonymous-to-synonymous rate ratio (omega = d(N)/ d(S)) was performed. The results provide clear statistical evidence for adaptive molecular evolution of scorpion alpha- and beta-toxin genes. A good match between t

[Theoretical and experimental bases for treatment of scorpion envenomations].

Improving the efficacy of envenomation treatment depends on what is known about active molecules present in venoms. Regarding scorpions, studies carried out mainly on the most poisonous species have shown that the toxicity and mortality were due to small proteins-toxins--able to interfere with the normal process of the ionic channels. In certain cases, using the mouse model, it has been shown that over 90% of mortality was due to toxins operating on the sodium channels implicated in the action potential of the excitable cells. Pharmacokinetic studies have shown the diversity of their mode of a…

Serotherapy in scorpion envenomation: a randomised controlled trial.

BACKGROUND: Evidence for the benefit of scorpion antivenom, the only specific treatment for scorpion envenomation, is scarce, despite its common use. We did a prospective, randomised, controlled trial to assess the efficacy of routine administration of scorpion antivenom to scorpion-stung patients, irrespective of clinical severity. METHODS: We included 825 consecutive patients older than 10 years, who presented to the accident and emergency department of the hospital in Tozeur, Tunisia. We graded severity by absence (grade I) or presence (grade II) of systemic manifestations of scorpion enve

Scorpion toxins as tools for studying potassium channels.

The search for peptidyl inhibitors of K+ channels is a very active area of investigation. In addition to scorpion venoms, other venom sources have been investigated; all of these sources have yielded novel peptides with interesting properties. For instance, spider venoms have provided peptides that block other families of K+ channels (e.g., Kv2 and Kv4) that act via mechanisms which modify the gating properties of these channels. Such inhibitors bind to a receptor on the channel that is different from the pore region in which the peptides discussed in this chapter bind. In fact, it is possible

Scorpion toxins: tools for studying K+ channels.

Over the last period of time, a large number of scorpion toxins have been characterized. These peptidyl inhibitors of K+ channels have been very useful as probes for determining the molecular architecture of these channels, for purifying channels from native tissue and determining their subunit composition, for developing the pharmacology of K+ channels, and for determining the physiologic role that K+ channels play in target tissues. The large knowledge that we have developed regarding K+ channel function would not have been possible without the discovery of these peptidyl inhibitors. It is e

Clinical description of Parabuthus transvaalicus scorpionism in Zimbabwe.

An epidemiological and clinical study of Parabuthus transvaalicus scorpionism was conducted in Zimbabwe. Ten per cent of stings resulted in severe scorpionism. The clinical features of 17 patients with severe envenomation were primarily neuromuscular, with significant parasympathetic nervous system and cardiac involvement. The clinical course was prolonged compared to other scorpion syndromes, and significant therapeutic benefit was demonstrated in terms of hospital stay in response to species specific antivenom. The case fatality rate was 0.3%, with deaths in children below 10 years and adult
