Suppressive effect of vagal afferents on the activity of the trigeminal spinal neurons related to the jaw-opening reflex in rats: involvement of the endogenous opioid system.

The purpose of the present study is to test the hypothesis that via the endogenous pain control system, vagal afferent input modulates the activity of the trigeminal spinal nucleus oralis (TSNO) related to the tooth pulp (TP)-evoked jaw-opening reflex (JOR). Extracellular single-unit recordings were made from 36 TSNO units responding to TP electrical stimulation with a constant temporal relationship to a digastric electromyogram (dEMG) signal in 26 pentobarbital-anesthetized rats. The activity of 36 TSNO neurons and the amplitude of the dEMG increased proportionally during 1.0-3.5 times the th

Possible mechanism of liver necrosis caused by aromatic organic compounds.

Treatment of rats with phenobarbital, which stimulates the activity of the drug-metabolizing enzymes in the liver, potentiates hepatic necrosis elicited by bromobenzene and a number of other chemically inert halogenated aromatic hydrocarbons. Radioautographic studies indicate that [(14)C]bromobenzene is covalently bound at the sites of necrosis. From these results, it is inferred that the hepatotoxic effects of the halogenated aromatic hydrocarbons are mediated by chemically active metabolites formed in hepatocytes. In accord with this view, a number of aromatic halogenated hydrocarbons are co

Regulation of human drug metabolism by dietary factors.

Several dietary factors influence the oxidative metabolism of chemicals in humans. Increasing the ratio of protein to carbohydrate or fat in the diet, feeding cabbage and brussels sprouts or feeding charcoal-broiled beef for several days stimulates human drug metabolism. The chronic ingestion of ethanol stimulates drug metabolism whereas the chronic ingestion of methylxanthine-containing foods inhibits drug metabolism. In contrast, an increase in the ratio of fat to carbohydrate in the diet of normal subjects or the fasting of obese individuals for several days has little or no effect on drug

Inhibition of Na(+)/H(+) exchanger type 3 reduces duration of apnea induced by laryngeal stimulation in piglets.

Reflexes from the larynx induce cessation of breathing in newborn animals. The magnitude of respiratory inhibition is inversely related to the level of central chemical input. Recent studies indicate that selective inhibition of Na(+)/H(+) exchanger type 3 (NHE3) activates CO(2)/H(+)-sensitive neurons, resembling the responses evoked by hypercapnic stimuli. Hence, the use of NHE3 inhibitors may reduce reflexly mediated respiratory depression and duration of apnea in the neonatal period. This possibility was examined in decerebrate, vagotomized, ventilated, and paralyzed piglets by testing the

Time course of odorant- and trigeminal-induced activation in the human brain: an event-related functional magnetic resonance imaging study.

It is well known that most odorants stimulate the trigeminal system but the time course of the brain regions activated by these chemical stimulations remains poorly documented, especially regarding the trigeminal system. This functional magnetic resonance imaging (fMRI) study compares brain activations resulting from the contrast between two odorant conditions (one bimodal odor and one relatively pure olfactory stimulant) according to the duration of the stimulation (i.e. one inhalation, or three or six successive inhalations). The results show striking differences in the main brain regions ac

Electrical and chemical long-term depression do not attenuate low-Mg2+-induced epileptiform activity in the entorhinal cortex.

PURPOSE: Low-frequency electrical and magnetic stimulation of cortical brain regions has been shown to reduce cortical excitability and to decrease the susceptibility to seizures in humans and in vivo models of epilepsy. The induction of long-term depression (LTD) or depotentiation of a seizure-related long-term potentiation has been proposed to be part of the underlying mechanism. With the low-Mg(2+)-model of epilepsy, this study investigated the effect of electrical LTD, chemical LTD, and depotentiation on the susceptibility of the entorhinal cortex to epileptiform activity. METHODS: The ex

Use of acoustic stimulus to determine behavioral changes in zebrafish after Cd exposure in a water quality warning system.

Behavioral changes in zebrafish are an effective early warning system to determine water quality. However, only a few studies have examined the response of zebrafish to non-chemical stimulus after exposure to a contaminant. Therefore, this study investigated the differences in the behavioral responses of zebrafish to acoustic stimuli before and after exposure to cadmium (Cd). Acoustic escape response sensitivity curves were obtained and analyzed, followed by the determination of sensitive stimulus conditions at 100 Hz and 97 dB with a duration of 30 s and an interval of 30 min. Neuroexcitatory effects of morphine and hydromorphone: evidence implicating the 3-glucuronide metabolites.

1. Morphine is recommended by the World Health Organization as the drug of choice for the management of moderate to severe cancer pain. 2. Education of health professionals in the past decade has resulted in a large increase in the prescribing of opioids, such as morphine, and in the magnitude of the doses administered, resulting in an improvement in the quality of pain relief available for many cancer patients. 3. However, the reported incidence of neuroexcitatory side effects (allodynia, myoclonus, seizures) in patients administered large doses of systemic morphine or its structural analogue

[Endocrine reactions following S-(+)-ketamine].

ANAESTHETICS, ENDOCRINE SYSTEM, AND STRESS: The effects of anaesthetics on the nervous system are invariably associated with endocrine reactions, which are of great importance for the general characterization of anaesthetics or anaesthetic regimens. In this context, the endocrine stress response is mainly represented by adrenaline (A), noradrenaline (NA), antidiuretic hormone/vasopressin (ADH), adrenocorticotropic hormone (ACTH), and cortisol. PHARMACOLOGICAL PROFILE AND ANAESTHETIC ACTION OF KETAMINE: The pharmacological profile of ketamine is characterized by the term "dissociative anaesthes

Ventilatory response to keto-doxapram in intact and carotid body denervated lambs.

We aimed to investigate the role of the carotid bodies in the ventilatory response to keto-doxapram, and whether this response was dose-dependent; we studied two group (n = 5 per group) of awake, intact and carotid-body denervated (CBD) lambs, aged 10-15 days. At 20 min intervals, they received 0.5, 1.0, 2.5 and 5.0 m g/kg mg/kg of keto-doxapram as an intravenous bolus infusion. Plasma keto-doxapram was measured (HPLC). Ventilation was recorded via a face mask and a pneumotachograph. In intact lambs, an immediate dose-dependent increase in minute ventilation (V1) was observed. At 2 min, VI inc

Stimulation of renin secretion by non-diuretic sulfhydryl reagents.

Organomercurial diuretics stimulate renin secretion although the underlying cellular mechanisms remain undefined. Since organomercurials are also known to react with sulfhydryl groups, the present studies determined the effects of sulfhydryl reagents on renin secretion. The effects of the non-diuretic mercurial agent, parachloromercuriphenyl-sulfonate (PCMPS), as well as that of other sulfhydryl reagents, N-ethylmaleimide (NEM), N-phenylmaleimide (NPM) and monobromotrimethylammoniobimane (qBBR), on renin secretion were determined in rabbit renal cortical slices. All four reagents stimulated re

[Mechanism of rhythmic breathing during hypocapnia].

It has been established that passive hyperventilation does not evoke apnoea if the excitability of the respiratory center is preliminarily increased. Comparison of the experimental results with the literature data suggests that rhythmical respiration during hyperventilation hypocapnia is consequent on the increased sensitivity of the respiratory center to CO2 as a result of the development of hypocapnia.

Effect of prolonged low-dose infusions of ile5-angiotensin ii on blood pressure, aldosterone and electrolyte excretion in sodium replete man.

Two healthy young males on a constant normal sodium diet (135 mM/day) were infused for 132 h with 3 ng/kg/min of angiotensin II. Plasma angiotensin II levels were therby raised to the range of moderate sodium depletion. Plasma aldosterone and the urinary excretion rate of aldosterone-18-glucuronide were markedly increased during the whole infusion period and returned to control levels after the infusion was stopped. A slight tendency of aldosterone secretion to decrease towards the end of infusion was probably due to sodium retention (appr. 200 mM and 350 mM respectively) and to a fall in plas

Lymphokine stimulation of collagen accumulation.

Lymphokine-rich supernates from normal human peripheral blood mononuclear cells, stimulated by the mitogen phytohemagglutinin, have been shown to cause enhanced collagen accumulation by human embryonic lung fibroblasts (WI-38), as measured by hydroxyproline content of fibroblast monolayers, [14C] proline incorporation into soluble collagen and collagenase release of radioactivity in supernates and monolayers of cultures incubated with [14C] proline. This fibroblast-stimulating activity, demonstrable by suitable dilutions of the supernates, coexisted with a number of other lymphokine activities

Non-contact radiofrequency stimulation to the olfactory nerve of human subjects.

Damage to the olfactory nerve, caused by aging, trauma, or neurological disorders, can lead to smell loss, negatively impacting quality of life, taste perception, safety, and emotional well-being. Currently, olfactory training, which involves exposure to aromatic odorants, is the most widely used treatment. While this method provides some benefit, it does not offer a fundamental cure. Developing an approach that directly stimulates the olfactory nerves to restore neural activity is critical for effectively addressing smell loss. This study investigates the effects of radiofrequency (RF) stimul

Inflammation increases the distribution of dorsal horn neurons that internalize the neurokinin-1 receptor in response to noxious and non-noxious stimulation.

Although the neurokinin-1 (NK-1)/substance P (SP) receptor is expressed by neurons throughout the spinal dorsal horn, noxious chemical stimulation in the normal rat only induces internalization of the receptor in cell bodies and dendrites of lamina I. Here we compared the effects of mechanical and thermal stimulation in normal rats and in rats with persistent hindpaw inflammation. Electron microscopic analysis confirmed the upregulation of receptor that occurs with inflammation and demonstrated that in the absence of superimposed stimulation, the increased receptor was, as in normal rats, conc

Effects of Asian sand dust on mucin gene expression and activation of nasal polyp epithelial cells.

BACKGROUND: Asian sand dust (ASD) contains a variety of chemical and microbiological materials such as lipopolysaccharide (LPS) and beta-glucan that can cause inflammation. The increase in the atmospheric concentration of ASD has been associated with asthma severity and adverse effects on respiratory function. The aim of this study was to evaluate the effect of ASD on the inflammatory process and mucin gene expression in nasal epithelial cells. METHODS: Primary nasal polyp epithelial cells were exposed to ASD for 72 hours, and then the supernatants were collected. To determine the activation
