Engineering a chitosan-based nanoemulsion for non-invasive CNS-targeting approach: dual neuroprotection against epileptic and depressive disorders.

The frequent comorbidity of epilepsy and depression presents a major therapeutic challenge, with current therapies limited by systemic side effects and poor blood-brain barrier penetration. This study aimed to develop a non-invasive, dual-therapy topical nanoemulsion for combined (additive), rather than formally demonstrated synergistic, neuroprotection. A Quality-by-Design optimized chitosan-lipid nanoemulsion co-encapsulating ashwagandha extract and citicoline was engineered. The formulation exhibited favorable characteristics: nanoscale size (~ 145 nm), positive zeta potential (+ 35.9 mV),

An analytical strategy for the identification of carbamates, toxic alkaloids, phenobarbital and warfarin in stomach contents from suspected poisoned animals by thin-layer chromatography/ultraviolet detection.

In this study, an analytical strategy to identify brucine, strychnine, methomyl, carbofuran (alkaline compounds), phenobarbital, and warfarin (acid compounds) using thin-layer chromatography (TLC) screening with ultraviolet (UV) detection at 254 nm in stomach content is shown. The optimum mobile phase was found to be a chloroform: ethyl acetate: diethylamine (0.5:8.5:1) mixture for alkaline substances while a mixture of chloroform: acetone (9:1) has given better results for acidic substances. As for extraction, an equal proportion between distillated water and crude material (1:1) is required.

Some aspects of the neuropharmacology of phenylurea.

The neuropharmacology of phenylurea was studied. The compound is a potent sedative hypnotic agent with a fairly good margin of safety. It produced significant motor incoordination and behavioral changes in sub-sedative doses, thus suggesting that it acts on the areas of the brain which control motor coordination (neurological deficit) and spontaneous motor movements (awareness of environment). The activity pattern of phenylurea as measured on the rotarod treadmill was identical with that of an ataractic drug, meprobamate, and different from that of the sedative drug, 2-ethylcrotonylurea (ectyl

Control of retinal information coding by GABAB receptors.

The directional selectivity of amacrine and ganglion cells was studied using conventional intracellular recording techniques and drug application in the superfused retina-eyecup preparation of the tiger salamander. Baclofen, a GABAB receptor agonist, enhanced normal directional responses in some directionally selective third-order neurons. In about 30% of the cells that were not normally directional, baclofen induced direction-selective responses. This effect was particularly marked when 2-amino-4-phosphonobutyrate (APB) was used to isolate the OFF pathway. Comparisons of the effects of APB an

GlyT1 (SLC6A9) inhibition in neurological and psychiatric disorders.

Glycine is a fundamental neuroactive amino acid that serves dual roles in the central nervous system: acting as a primary inhibitory neurotransmitter via strychnine-sensitive glycine receptors and as an essential co-agonist at the N-methyl-D-aspartate (NMDA) receptor. This dual functionality is important for maintaining the excitation-inhibition balance, synaptic plasticity, and network stability. The spatial and temporal availability of glycine is strictly regulated by two high-affinity, Na+/Cl--dependent transporters: GlyT1 (SLC6A9) and GlyT2 (SLC6A5). These transporters exhibit distinct cel

Discovery of novel N-(alkylpiperidinyl) substituted benzamides targeting the glycine transporter 2 with anti-nociceptive effects in neuropathic pain.

Neuropathic pain is a global health problem, affecting approximately 7%-10% of the population worldwide. In recent years, targeting glycine transporter 2 (GlyT2) has emerged as a promising strategy for the treatment of neuropathic pain, as it enhances glycinergic neurotransmission and strengthens inhibitory control of nociceptive signal transmission. A series of N-(alkylpiperidinyl) substituted benzamides were designed, synthesized, and evaluated as potential GlyT2 inhibitors, and their binding affinities were assessed in vitro by surface plasmon resonance assay. Compound 14 was found to have

Brucine Sulfate, a Novel Bacteriostatic Agent in 3D Printed Bone Scaffold Systems.

Bacterial infection is a common complication in bone defect surgery, in which infection by clinically resistant bacteria has been a challenge for the medical community. Given this emerging problem, the discovery of novel natural-type inhibitors of drug-resistant bacteria has become imperative. Brucine, present in the traditional Chinese herb Strychnine semen, is reported to exert analgesic and anti-inflammatory effects. Brucine's clinical application was limited because of its water solubility. We extracted high-purity BS by employing reflux extraction and crystallization, greatly improved its

Antiepileptic profile of Parawixin-11, purified from Parawixia bistriata spider venom (Araneae, Araneidae), in Wistar rats.

The pharmacological treatment of epilepsy is often complex due to the lack of efficacy in many patients and profound side effects from current drugs, including sedation, motor impairment, and teratogenesis. In the quest for new antiepileptic drugs, animal venoms offer a valuable source of neuroactive molecules targeting ion channels and neurotransmitter receptors. This study investigates the antiepileptic potential of compounds isolated from the venom of the Parawixia bistriata spider. One compound, designated Parawixin-11, demonstrated significant anticonvulsant effects when injected into the

Anticonvulsant activity of methanolic extract of Withania cogulans in mice.

Mental and neurological diseases including depression, Parkinson's disease, dementia, epilepsy, anxiety disorders and bipolar disorders account for a considerable amount of the world's disease burden. Unfortunately, drugs used in the treatment of neurological diseases are expensive, symptomatic and they produce undesirable side effects. People from different cultures prefer to use medicinal plants for the treatment of various ailments ranging from plain to perplex disorders because they are most affordable, cost effective and easily accessible source of treatment in the primary healthcare syst

Assessment of a portable quadrupole-based gas chromatography mass spectrometry for seized drug analysis.

The cutting agents, classified as diluents (pharmacologically inactive) or adulterants (pharmacologically active), are substances commonly used to cut drugs of abuse to increase profits. These substances are constantly changing over time, increasing the risks to the user's health caused by the compounds' potential individual toxicities as well as their drug-drug interactions. This work aimed to develop and validate a screening method using a portable quadrupole-based gas chromatography mass spectrometer (FLIR Griffin™ G510) to identify drugs of abuse and adulterants in seized material, and com

High ethanol sensitive glycine receptors regulate firing in D1 medium spiny neurons in the nucleus accumbens.

Inhibitory glycine receptors (GlyRs) are widely expressed in spinal cord and brain stem. They are also expressed in the nucleus Accumbens (nAc) where they have been implicated in the release of dopamine from the ventral tegmental area to the nAc in the presence of ethanol. One of the major types of neurons in the nAc are the Dopamine 1 receptor-expressing (D1+) medium spiny neurons (MSNs) that are activated when addictive drugs, like ethanol, are administrated. Thus, D1(+) MSNs are a relevant target for the study of ethanol effects. Here, using electrophysiological recordings, we report that G

Tonic zinc inhibits spontaneous firing in dorsal cochlear nucleus principal neurons by enhancing glycinergic neurotransmission.

In many synapses of the CNS, mobile zinc is packaged into glutamatergic vesicles and co-released with glutamate during neurotransmission. Following synaptic release, the mobilized zinc modulates ligand- and voltage-gated channels and receptors, functioning as an inhibitory neuromodulator. However, the origin and role of tonic, as opposed to phasically released, zinc are less well understood. We investigated tonic zinc in the dorsal cochlear nucleus (DCN), a zinc-rich, auditory brainstem nucleus. Our results show that application of a high-affinity, extracellular zinc chelator (ZX1) enhances sp

Consumption of bitter alkaloids in Drosophila melanogaster in multiple-choice test conditions.

Drosophila melanogaster adapt their food consumption to their internal needs and avoid ingesting noxious molecules. Defects in the genes involved in these decisions induce behavioral alterations that are usually screened by monitoring flies feeding in 2-choice or in no-choice situations. Here, we introduce a new behavioral test in which groups of flies are given access to 6 capillary feeders (MultiCAFE) containing fructose mixed with a serial dilution of a test substance. Using quinine, we first showed that fly density, distance between capillaries, and order of presentation have a minor impac

The generation of rhythmic activity in dissociated cultures of rat spinal cord.

Locomotion in vertebrates is controlled by central pattern generators in the spinal cord. The roles of specific network architecture and neuronal properties in rhythm generation by such spinal networks are not fully understood. We have used multisite recording from dissociated cultures of embryonic rat spinal cord grown on multielectrode arrays to investigate the patterns of spontaneous activity in randomised spinal networks. We were able to induce similar patterns of rhythmic activity in dissociated cultures as in slice cultures, although not with the same reliability and not always with the

D-serine content and D-[3H]serine binding in the brain regions of the senescence-accelerated mouse.

An established senescence-accelerated model mouse strain, SAMP8, shows the deterioration of learning and memory compared with a normal control strain, SAMR1. D-Serine binds to strychnine-insensitive glycine binding sites of the N-methyl-D-aspartate (NMDA) receptor complex, and enhances glutamate binding to the receptor complex. To investigate the relationship of endogenous brain D-serine and the brain dysfunction caused by aging, the level of brain free D-serine and the D-[3H]serine binding to the brain samples were examined using the SAMP8 and SAMR1 mice. The free D-serine level was highest i
