Synphilin-1 modulates alpha-synuclein assembly, release and uptake.

Alpha-synuclein (aSyn) is an intrinsically disordered protein involved in phase separation and several age-associated neurodegenerative disorders, including Parkinson's disease. However, its function and pathological role remain elusive. Here, we modeled different aSyn assemblies in living cells by exploiting its interaction with synphilin-1 (Sph1). We developed a model that reports on gel- and solid-like inclusions through coexpression of aSyn and Sph1. Distinct morphological differences emerged between VN-aSyn + aSyn-VC and VN-Sph1 + aSyn-VC assemblies, showing unique antibody recognition, p

Alpha synuclein overexpression can drive microbiome dysbiosis in mice.

Growing evidence indicates that persons with Parkinson disease (PD), have a unique composition of indigenous gut microbes. Given the long prodromal or pre-diagnosed period, longitudinal studies of the human and rodent gut microbiome before symptomatic onset and for the duration of the disease are currently lacking. PD is partially characterized by the accumulation of the protein α-synuclein (α-syn) into insoluble aggregates, in both the central and enteric nervous systems. As such, several experimental rodent and non-human primate models of α-syn overexpression recapitulate some of the hallmar

Tet2 loss and enhanced ciliogenesis suppress α-synuclein pathology.

There are no approved treatments that slow Parkinson's disease (PD) progression and therefore it is important to identify novel pathogenic mechanisms that can be targeted. Loss of the epigenetic marker, Tet2 appears to have some beneficial effects in PD models, but the underlying mechanism of action is not well understood. We performed an unbiased transcriptomic analysis of cortical neurons isolated from patients with PD to identify dysregulated pathways and determine their potential contributions to the disease process. We discovered that genes associated with primary cilia, non-synaptic sens

α-Synuclein Pathology Spreads in a Midbrain-Hindbrain Assembloid Model.

Understanding the progression of α-synuclein pathology in neurodegenerative diseases such as Parkinson's disease (PD) is a longstanding challenge. Here, a novel midbrain-hindbrain-assembloid model that recapitulates the spread of α-synuclein pathology observed in PD patients, akin to Braak's hypothesis, is presented. Initially, the presence α-synuclein pathology is demonstrated in the hindbrain organoids. Subsequently, sophisticated tissue engineering methods are employed to create midbrain-hindbrain assembloids. These assembloids allow investigation and description of the spreading of α-synuc

Knockout of Rab27b exacerbates neuropathology in alpha-synuclein mouse models.

Parkinson's Disease (PD) and other synucleinopathies are characterized by the formation of inclusions comprised of alpha-synuclein (αsyn) among other proteins, but the mechanisms by which these inclusions form and cause toxicity are not well understood. We have previously reported that the small GTPase Rab27b modulates autophagic-lysosomal function in neurons and supports lysosomal degradation of αsyn across multiple αsyn cellular models. Knockout (KO) and knockdown of Rab27b damage lysosomal degradative capacity and exacerbate αsyn pathology, while Rab27b overexpression is conversely protecti

α-Synuclein Drives SNARE-Dependent Tubular Remodeling of Vesicles.

α-Synuclein (α-syn) disrupts synaptic vesicle architecture in Parkinson's disease (PD), yet the underlying mechanisms remain unclear. Here, we identify a previously unrecognized phenomenon in which α-syn (≥20 μM) rapidly induces SNARE-dependent tubular protrusions on highly curved small unilamellar vesicles (∼45 nm in diameter). This process requires functional v- and t-SNAREs, as neither v-SNARE nor t-SNARE alone, nor predocked SNARE complexes or CDV-treated SNARE liposomes, support tubulation. Notably, the familial PD-associated mutations A30P and E46K enhance tubule formation, whereas A53T

DOPAnization of α-synuclein exacerbates dopaminergic neurodegeneration in the substantia nigra.

Parkinson's disease (PD) is characterized by the selective and progressive loss of dopaminergic (DAergic) neurons in the substantia nigra (SN). Although abnormal forms and aggregates of the α-synuclein (αSyn) protein are considered to cause PD, the underlying mechanisms of the preferential death of dopaminergic neurons are largely unknown. We recently reported that a tyrosine hydroxylase post-translationally converted a tyrosine residue of αSyn to dihydroxyphenylalanine (DOPA), termed DOPAnization, in the DAergic neurons of PD patients, which facilitated the formation of αSyn oligomers and inc

Negatively charged α-synuclein condensate modulates partitioning of molecules.

α-Synuclein (αSyn) aggregation via liquid-liquid phase separation (LLPS) has recently emerged as a crucial mechanism underlying amyloid fibril formation implicated in Parkinson's disease. However, comprehensive investigation of the physicochemical properties of αSyn condensate remains incomplete. Here, we demonstrate that αSyn condensates exhibit a highly negative electrostatic potential, revealed by preferential enrichment of positively charged fluorophores or fluorophore-labeled αSyn. We further demonstrate that αSyn LLPS is regulated by a balance between self-association and electrostatic r

Aggregation and Propagation of α-Synuclein in Parkinson's Disease: A Bibliometric Perspective.

INTRODUCTION: Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by the aggregation and propagation of alpha-synuclein (α-syn), processes that contribute to neuronal dysfunction and cell death. Although substantial progress has been made in understanding α-syn pathology, a comprehensive bibliometric evaluation of global trends in this field remains lacking. This study aims to systematically map the research landscape surrounding α-syn aggregation and propagation in PD, offering insights into its molecular mechanisms and clinical relevance. METHODS: A bibliometr

Alpha-synuclein amyloids catalyze the degradation of ATP and other nucleotides.

Intracellular accumulation of alpha-synuclein amyloids is a main pathological hallmark in a subgroup of human neurodegenerative diseases called synucleinopathies. Cell death of energy-deprived dopaminergic neurons causes decreased dopamine levels, which underly many of the neurological symptoms in the most prevalent synucleinopathy, Parkinson's disease. Amyloid-mediated toxicity can proceed via gain-of-function through diverse pathways. In this work, we report that alpha-synuclein amyloids can degrade adenosine triphosphate in a catalytic fashion, producing adenosine diphosphate and adenosine

Pathophysiological Significance of α-Synuclein in Sympathetic Nerves: In Vivo Observations.

BACKGROUND AND OBJECTIVES: Lewy body diseases (LBDs) such as Parkinson disease (PD) feature increased deposition of α-synuclein (α-syn) in cutaneous sympathetic noradrenergic nerves. The pathophysiologic significance of sympathetic intraneuronal α-syn is unclear. We reviewed data about immunoreactive α-syn, tyrosine hydroxylase (TH, a marker of catecholaminergic fibers), and the sympathetic neurotransmitter norepinephrine (NE) in skin biopsies from control participants and patients with PD, the related LBD pure autonomic failure (PAF), the non-LBD synucleinopathy multiple system atrophy (MSA),

Toward alpha-synuclein seed amplification assay in clinical practice.

INTRODUCTION: Seed amplification assays (SAAs) demonstrate remarkable diagnostic performance in alpha-synucleinopathies. However, existing protocols lack accessibility in routine laboratories, mainly due to the requirement for in-house production of recombinant alpha-synuclein (aSyn). This study proposes a cerebrospinal fluid (CSF) aSyn-SAA protocol using solely commercial reagents to facilitate its clinical implementation. METHODS: Routine clinical care CSF samples from 126 patients, comprising 47 with Lewy body diseases (LBD) (41 with dementia with Lewy bodies, six with Parkinson's disease)

Lipidic folding pathway of α-Synuclein via a toxic oligomer.

Aggregation intermediates play a pivotal role in the assembly of amyloid fibrils, which are central to the pathogenesis of neurodegenerative diseases. The structures of filamentous intermediates and mature fibrils are now efficiently determined by single-particle cryo-electron microscopy. By contrast, smaller pre-fibrillar α-Synuclein (αS) oligomers, crucial for initiating amyloidogenesis, remain largely uncharacterized. We report an atomic-resolution structural characterization of a toxic pre-fibrillar aggregation intermediate (I1) on pathway to the formation of lipidic fibrils, which incorpo

Rab27b Promotes Lysosomal Function and Alpha-Synuclein Clearance in Neurons.

Alpha-synuclein (αsyn) is the key pathogenic protein implicated in synucleinopathies including Parkinson's disease (PD) and dementia with Lewy bodies (DLB). In these diseases, αsyn is thought to spread between cells where it accumulates and induces pathology; however, mechanisms that drive its propagation or aggregation are poorly understood. We have previously reported that the small GTPase Rab27b is elevated in human PD and DLB and that it can mediate the autophagic clearance and toxicity of αsyn in a paracrine αsyn cell culture model. Here, we expanded our previous work and characterized th

Liganded magnetic nanoparticles for magnetic resonance imaging of α-synuclein.

Aggregation of the protein α-synuclein (α-syn) is the histopathological hallmark of neurodegenerative diseases such as Parkinson's disease (PD), dementia with Lewy bodies (DLB), and multiple system atrophy (MSA), which are collectively known as synucleinopathies. Currently, patients with synucleinopathies are diagnosed by physical examination and medical history, often at advanced stages of disease. Because synucleinopathies are associated with α-syn aggregates, and α-syn aggregation often precedes onset of symptoms, detecting α-syn aggregates would be a valuable early diagnostic for patients

Misfolded α-synuclein co-occurrence with Alzheimer's disease proteinopathy.

INTRODUCTION: Multi-etiology dementia necessitates in vivo markers of copathologies including misfolded α ${{\alpha}}$ -synuclein (syn). We measured misfolded syn aggregates (syn-seeds) via qualitative seed amplification assays (synSAA) and examined relationships with markers of Alzheimer's disease (AD). METHODS: Cerebrospinal fluid (CSF) was obtained from 420 participants in two AD risk cohorts (35% male; 91% cognitively unimpaired; mean [standard deviation] age, 65.42 [7.78] years; education, 16.17 [2.23]) years). synSAA results were compared to phosphorylated tau (T), amyloid beta (A), an

α-Synuclein Pathology in Synucleinopathies: Mechanisms, Biomarkers, and Therapeutic Challenges.

Parkinson's disease and related synucleinopathies, including dementia with Lewy bodies and multiple system atrophy, are characterised by the pathological aggregation of the α-synuclein (aSyn) protein in neuronal and glial cells, leading to cellular dysfunction and neurodegeneration. This review synthesizes knowledge of aSyn biology, including its structure, aggregation mechanisms, cellular interactions, and systemic influences. We highlight the structural diversity of aSyn aggregates, ranging from oligomers to fibrils, their strain-like properties, and their prion-like propagation. While the r

The Aggregation Continuum of α-Synuclein and Its Relevance to Brain Aging.

α-Synuclein aggregation in synucleinopathies involves abnormal accumulation of α-synuclein protein in neurons. This aggregation process generates oligomers, protofibrils, and fibrils, disrupting cellular function and contributing to the progression of neurodegeneration observed in Parkinson's disease, multiple system atrophy, and dementia with Lewy bodies. Typically, these aggregates range in size from tens to hundreds of nanometers to a few micrometers. The intermediate-sized aggregation species, so-called oligomers, have been implicated in the neurotoxicity observed in Parkinson's disease. S

Differential microglial responses to structurally distinct alpha-synuclein polymorphs.

Synucleinopathies are age-related neurological disorders which include dementia with Lewy bodies (DLB), Parkinson's disease (PD), and multiple system atrophy (MSA). A hallmark of these diseases is the pathological accumulation of α-synuclein aggregates, along with sustained neuroinflammatory responses. Recent studies have demonstrated the existence of structurally distinct α-synuclein aggregates in this group of the diseases. While the correlation between specific forms of α-synuclein and distinct pathological characteristics has been extensively studied, their relationship to neuroinflammatio
