Repair of ionizing radiation-induced DNA damage and risk of second cancer in childhood cancer survivors.

The study's purpose was to assess whether individuals who developed a second malignant neoplasm (SMN) after treatment for a first malignant neoplasm (FMN) had a lower ability to repair DNA double-strand breaks (DSBs) using a bioassay with γH2AX intensity as a surrogate endpoint. In a case-control study nested in a cohort of childhood cancer survivors, lymphoblastoid cell lines (LCLs) were established from blood samples collected from 94 cases (SMN) and 94 matched controls (FMN). LCLs were irradiated with ionizing radiation (2 and 5 Gy) and γH2AX intensities measured 1, 3, 5 and 24h post-irradi

Carcinoma showing thymus-like elements (CASTLE) with amyloid deposition in the parotid gland.

Carcinoma showing thymus-like elements (CASTLE) is a rare tumor that commonly occurs in the thyroid gland. Extrathyroidal CASTLE is rarer, and only 11 cases of CASTLE of major salivary glands have been reported to date. We report the first case of amyloid deposition in parotid CASTLE. A 63-year-old man presented with a slowly growing mass in the left parotid region. Computed tomography revealed an approximately 28 × 23 mm mass lesion in the left parotid gland, and squamous cell carcinoma was suspected on biopsy. The patient underwent a parotidectomy with neck dissection. Morphologically, the t

[Epithelial tumors with thymus differentiation of the thyroid gland and the neck].

Epithelial tumors with thymus-like differentiation occurring in the thyroid gland and the neck, delineate four entities with distinct histological features and different behaviour. The spindle epithelial tumors with thymus-like differentiation are potentially malignant tumors occurring in the thyroid gland of young patients. They are highly cellular with pattern of spindle cells and tubulopapillary formations and mucinous glands. Carcinomas showing thymus-like differentiation of the thyroid gland are histologically identical to some thymic carcinomas. The ectopic hamartomatous thymoma is stric

Insights into Thyroid and Salivary Gland Cytopathology: Highlights from a 45th European Congress of Cytology Slide Seminar.

INTRODUCTION: Thyroid and salivary gland cytopathology frequently present diagnostic challenges due to complex presentations, overlapping features between benign and malignant conditions, particularly with gray-zone entities and rare pathologies. To address these issues, the 45th European Congress of Cytology (ECC) held a slide seminar focused on challenging cases. This article reviews key findings from the 6 cases discussed, emphasizing the importance of a comprehensive diagnostic approach. The objective of this article was to illustrate the diagnostic challenges of rare thyroid and salivary

Spindle epithelial tumor with thymus-like differentiation: a case report with cytologic, histologic, immunohistologic, and ultrastructural findings.

Spindle epithelial tumor with thymus-like differentiation (SETTLE) is a rare and distinctive low-grade neoplasm of thymic or related branchial pouch differentiation. The tumor usually presents in the thyroid or lateral neck of children and adolescents and could mimic spindle-cell carcinoma, synovial sarcoma, or malignant teratoma. We report the clinical, cytologic, histologic, immunohistochemical, and ultrastructural features of a SETTLE present for 10 years in a 15-year-old boy. The fine-needle aspirate, initially interpreted as synovial sarcoma, contained numerous clusters of bland spindle c

Carcinoma showing thymus-like elements of the thyroid gland: report of three cases including one case with breast cancer history.

Carcinoma showing thymus-like elements (CASTLE) is a rare malignant tumor of the thyroid or adjacent neck soft tissues, whose histogenesis is still debated. It may resemble other primary or metastatic poorly differentiated tumors histologically and the differential diagnosis is crucial for CASTLE has a better prognosis. However, CASTLE as a second primary tumor has not been reported in the literature. We report three cases of thyroid CASTLE, including a unique tumor following breast-conserving surgery for early-stage breast invasive carcinoma. There were two female and one male. All three tumo

[Advances in the research on immunopathogenesis of myasthenia gravis].

Microscopic structure and immunologic function of the thymus gland are outlined, with an emphasis on its role in the process of T and B lymphocytes maturation. Other types of thymic cells are also described, including epithelial and myoid cells that presumably have a central role in initiation of an autoimmunologic process leading to myasthenic symptoms development. A detailed account is presented of issues concerning pathophysiology of myasthenia, the structure of nicotinic acetylcholine receptor (AchR) at neuromuscular junctions, and mechanisms underlying its destruction by autoantibodies. The thymus. Pediatric surgical aspects.

Since the original description of thymic death in an infant 400 years ago, the thymus has been recognized as an important structure to practitioners caring for infants and children. The source of many cysts, masses, and tumors in the neck and mediastinum, the thymus gland merits the pediatric surgeon's attention. The thymus is clearly an important lymphoid organ, the removal of which may be therapeutic in MG, but congenital absence leads to profound cell-mediated immunodeficiency. The immunologic sequelae of its neonatal extirpation remains obscure. It is apparent that further research is need

Cervical thymic cancer infiltrating the trachea and thyroid.

The patient was a 59-year-old female with cervical thymic cancer infiltrating the trachea and right lobe of the thyroid, with associated metastasis to a cervical lymph node. The tumor was completely resected along with the infiltrated organs, in addition to dissection of the cervical and mediastinal lymph nodes, total thymectomy and tracheoplasty. Histologically the tumor consisted of lymphoepithelioma-like carcinoma and mucoepidermoid carcinoma. Postoperative radiotherapy for the neck and upper mediastinum was performed at a total dose of 44 Gy. The patient is alive without recurrence 8 month

The World Health Organization Reporting System for Lymph Node, Spleen, and Thymus Cytopathology: Part 1 - Lymph Node.

BACKGROUND: Fine-needle aspiration biopsy (FNAB) of lymph nodes is a widely used method for evaluating lymphadenopathy. FNAB offers general advantages of rapid turnaround time, low cost and minimal morbidity, and more specific advantages in various clinical situations, such as deeply located lymph nodes or patients with significant comorbidities. The FNAB sample can be utilized for a wide range of ancillary tests, including microbiological studies, immunocytochemistry for primary and metastatic neoplasms and flow cytometry immunophenotyping in cases of lymphoid-rich samples, where there is a s

Immunohistochemical study of a non-invasive canine thymoma: a case report.

We describe a case of canine thymoma in an 11-year-old pure-bred female Spanish Mastiff. On post-mortem examination a big mass was found in the thoracic cavity. Histologically the neoplastic parenchyma showed great structural variability; the principal findings included the presence of tumour cells forming cords or sheets, cystic formations, Hassall-corpuscle like squamous formations, and small or intermediate calibre vessels encircled by areas with eosinophilic material and lymphocytes. In order to determine the histogenesis of the thymic neoplasia we used immunohistochemical techniques speci

A case of large "thymic granuloma". Neoplasm of T-zone histiocyte.

An unusual case of classic thymic granuloma is reported. A 16-year-old boy presented with a huge mediastinal mass and the superior vena caval syndrome. Irradiation and combination chemotherapy induced complete remission, despite several episodes of lymph node recurrence. Examination of the first biopsy specimen was considered to indicate a histiocytic tumor or malignant form of eosinophilic granuloma, because of the diffuse proliferation of histiocytes and marked infiltration of eosinophils. Late involvement in the lymph node appeared to have the same histologic characteristics as the mediasti

Ultrahigh-performance liquid chromatography-high-resolution mass spectrometry-based plasma metabolomics study of thymoma and thymic hyperplasia.

RATIONALE: Thymoma is a rare malignant tumor but it is the most common primary tumor of the anterior mediastinum. The current imaging methods for thymoma screening suffer from false positive rate problems, and thymoma pathogenesis remains elusive. Study of thymoma metabolic characteristics could provide clues for improving the diagnosis and understanding the pathogenesis of thymoma. METHODS: Metabolic profiling of plasma from thymoma and thymic hyperplasia patients was performed using ultrahigh-performance liquid chromatography combined with high-resolution mass spectrometry in both positive

Immunohistochemical Characterization of Proliferative Lesions in the Thymus of Aging CD-1 Mice From Two Studies on the RITA Database, With Special Reference to the Perivascular Space.

Thymic lymphoid hyperplasia is a common age-related finding, which occurs particularly in female CD-1 mice. The main differential diagnoses are malignant lymphoma and thymoma. A systematic investigation of control groups from two carcinogenicity studies was performed including measurements of thymic size, and the immunohistochemistry (IHC) markers pan-Cytokeratin (pan-CK) for thymic epithelial cells; CD3 and CD45R/B220 for T and B lymphocytes, respectively; CD31 for endothelial cells; and F4/80 for macrophages. Thymoma can be differentiated by increased numbers of proliferating epithelial cell

Recent thymic emigrants as the bridge between thymoma and autoimmune diseases.

Recent thymic emigrants (RTEs) are naïve T cells that egress the thymus following intrathymic development. This continuous process, including self-renewal, is crucial to establish and maintain human immune function. Several biomarkers can identify RTEs, but none of them is specific. Additional methods to detect and study RTEs phenotypically and functionally revealed alterations in RTEs in various adverse health conditions, including autoimmune diseases, systemic disorders and thymic abnormalities such as thymoma. Often associated with autoimmune disease, thymoma is the only tumor that can gene

The thymus: a comprehensive review.

BACKGROUND: Nonspecific abdominal pain is the commonest cause of a patient presenting to a doctor with abdominal pain of less than one week's duration. The differential diagnosis of NSAP is not always easy due to many similarities in the clinical presentation at onset and many cases may be misdiagnosed in the initial situation. To the Authors' knowledge, the diagnostic accuracy of history-taking is rarely considered in NSAP, and therefore the aim of the present study was to investigate the contribution of history-taking to correctly diagnosing NSAP in the clinical situation. PATIENTS AND METH
