Decellularization of camel corneas for tissue engineering applications.

PURPOSE: The similarity in collagen fiber diameter between the camel cornea (CC) and the human cornea, along with its relatively large size, positions the CC as a promising candidate for tissue engineering applications. This study aimed to evaluate the effectiveness of various decellularization methods on the CC, followed by assessing protein content similarities between the CC and the human cornea, highlighting its potential for human-related tissue engineering research. METHODS: Camel corneas were extracted from freshly isolated camel eyeballs and subjected to multiple decellularization pro

Do vasoactive medications impact donor hearts clinical outcomes in pediatric heart transplantation?

OBJECTIVES: There is limited data examining donor vasopressor and/or inotrope medications (vasoactives) on pediatric orthotopic heart transplant (OHT) outcomes. We aim to evaluate the effects of vasoactives on pediatric OHT outcomes. METHODS: The United Network for Organ Sharing database was retrospectively reviewed from January 2000 to March 2018 for donor hearts. Exclusion criteria included multiorgan transplants and recipient age >18. Donors receiving vasoactives at the time of procurement were compared to donors not on vasoactives, including the number of vasoactives and the type. End-poi

The impact of machine perfusion of the heart on warm ischemia time and organ yield in donation after circulatory death.

Successful normothermic machine perfusion of heart allografts (MPH) has led to rapid growth in transplantation of donation after circulatory death (DCD) heart allografts but has introduced complexity in the procurement process. This study examines the impact of MPH use in DCD procurements on warm ischemia time (WIT) and organ yield. DCD procurements from 2019 to 2020 were identified using the OPTN database. Procurements with and without the use of MPH were compared using propensity score matching. Observed to expected (O:E) yield ratios were calculated, where the expected values were obtained

Photobiomodulation of a flowable matrix in a human skin ex vivo model demonstrates energy-based enhancement of engraftment integration and remodeling.

The use of dermal substitutes to treat skin defects such as ulcers has shown promising results, suggesting a potential role for skin substitutes for treating acute and chronic wounds. One of the main drawbacks with the use of dermal substitutes is the length of time from engraftment to graft take, plus the risk of contamination and failure due to this prolonged integration. Therefore, the use of adjuvant energy-based therapeutic modalities to augment and accelerate the rate of biointegration by dermal substitute engraftments is a desirable outcome. The photobiomodulation (PBM) therapy modulate

Relative impact of donor type on renal allograft survival in black and white recipients.

Renal allograft survival of organs from living-related donors (LRDs) is superior to that of organs from cadaveric donors (CAD). Allograft survival rates are lower in black recipients of both CAD and LRD transplants compared with recipients of other races. It is not known whether the superior donor effect of LRD varies between races. We investigated the graft survival outcome of first renal transplants performed in the United States in 1986 and 1989 to determine whether the donor effect differs between black and white recipients. A measurement of "donor type effect" is introduced. There were 7,

The pulp plasty: a composite graft for complete syndactyly fingertip separations.

Composite grafts of skin and subcutaneous fat harvested from the glabrous non-weight-bearing areas of the foot were used to graft 34 fingertips after separation of 23 complete syndactyly webs in 13 patients. Simple complete syndactylies accounted for 17% and complex complete syndactylies accounted for 83%; synonychia was encountered in 70%. Follow-up averaged 13.9 months (minimum, 2 months, maximum 33 months). All patients had 100% take of the composite graft. Subjective gradings were 94% good, 6% fair, and there were no poor results. This technique provides a relatively normal contour and sat

Pediatric tissue and organ transplantation in Florida.

Fifty to 100 children receive transplanted kidneys, hearts, livers, or bone marrow in Florida each year and many more bone allografts or other tissues (skin, cornea). Children are in the minority of the total solid organ transplantation but those with successful transplants are strong proponents of the procedure. Many (liver or heart failure) would have died without transplantation; others (kidney failure) would have lived but been tied to dialysis for life. The success rate varies with the organ or tissue transplanted. Some children return to a completely normal life without the need for immu

Expression of IL4Rα and IL13Rα1 are associated with poor prognosis of soft-tissue sarcoma of the extremities, superficial trunk, and retroperitoneum.

BACKGROUND: IL4Rα and IL13Rα1 are constituents of the type II IL4 receptor. Recently, IL4Rα and IL13Rα1 were reported to have roles in cancer progression and suggested as potential prognostic markers. However, studies on IL4Rα and IL13Rα1 in soft-tissue sarcomas have been limited. METHODS: This study investigated the immunohistochemical expression of IL4Rα and IL13Rα1 in 89 soft-tissue sarcomas of the extremities, superficial trunk, and retroperitoneum. Immunohistochemical staining for IL4Rα and IL13Rα1 were scored according to a combination of staining intensity and staining area in tissue m

Brain-dead kidney donor: selection, care, and administration.

Over four and a half years a district general hospital provided 34 cadaveric kidneys for transplantation. All brain-dead patients were regarded as potential donors, flow charts being used to maintain circulation and urine formation and facilitate administration. With this system the time lapse between diagnosis of brain death and removal of kidneys ranged from three to six hours and ischaemia was minimised. It is concluded that adoption of the system by other hospitals of comparable size would result in enough good-quality kidneys to satisfy present needs, thus reducing the initial high failur

Tricellular Adipose Regeneration Units in pGelMA Hydrogel Synergistically Enhance Fat Graft Survival through Vascularization and ECM Remodeling.

Autologous fat transplantation (AFT) remains limited by low graft survival rates due to insufficient vascularization and extracellular matrix (ECM) support. Here, we developed an adipose tissue regeneration unit (ATRU) system combining three key cellular components (adipose-derived stem cells, ADSCs; human umbilical vein endothelial cells, HUVECs; and fibroblasts, FBs) within a porous gelatin methacryloyl (pGelMA) hydrogel microenvironment. Through controlled coculture in microwell plates, this system generated functional microtissues, demonstrating synergistic effects: ADSCs provided adipogen

Females transplanted with ovaries subjected to hypoxic preconditioning show impair of ovarian function.

BACKGROUND: Cryopreservation of the ovarian tissue has shown promising results. However, there remain controversial issues such as the short half-life of grafts. In this aspect, there are some evidences that preconditioning the ovarian tissue before transplantation is beneficial. OBJECTIVE: To determine the effect of hypoxic preconditioning in vitro on ovarian tissue prior to transplantation. METHODS: Eighteen female adult Wistar rats, were sorted into three experimental groups. Ovaries were maintained in DMEM low glucose serum free at 37°C with 5% CO2, at atmospheric oxigen concentration (n

[Expanding the grafts pool for pediatric liver transplantation with segmental grafts from living donor and cadaveric reduced size or split grafts].

AIM: To compare short and long-term results of pediatric liver transplantation (LT), utilizing segmental grafts from living donors (LD) vs. cadaveric (CAD) reduced size or split grafts. METHODS: A retrospective analysis of a single center experience (1993-2000), comparing the surgical outcome, the graft function and the survival rates between these groups. RESULTS: Of 195 LTs in pediatric recipients (age < 18), 48 (25%) were with LD grafts and 47 (24%) with CAD grafts (reduced size, n = 27, or split, n = 20). The mean age and weight of the LD recipients were 1.8 +/- 3 yrs and 9.0 +/- 8.1 kg

Indirect 3D bioprinting and characterization of alginate scaffolds for potential nerve tissue engineering applications.

Low-concentration hydrogels have favorable properties for many cell functions in tissue engineering but are considerably limited from a scaffold fabrication point of view due to poor three-dimensional (3D) printability. Here, we developed an indirect-bioprinting process for alginate scaffolds and characterized the potential of these scaffolds for nerve tissue engineering applications. The indirect-bioprinting process involves (1) printing a sacrificial framework from gelatin, (2) impregnating the framework with low-concentration alginate, and (3) removing the gelatin framework by an incubation

Multiple organ donation: its impact on the recovery of cadaver kidneys.

Cyclosporin A not only has improved renal allograft survival but it also has caused a renewed interest in cardiac and hepatic transplantation. The South Texas Organ Bank was sensitive to the need for multiple organ donors and also concerned that the recovery of multiple organs could have a negative impact on the quantity or quality of kidneys recovered. From July 1983 through March 1985 cadaveric kidneys were obtained from 43 renal donors and multiple vital organs were obtained from 11 additional donors. There was no statistical difference between the renal and multiple organ donors in the inc

Normothermic Regional Perfusion in Controlled Donation After Circulatory Death: Growing Evidence for Liver Transplantation.

Normothermic regional perfusion (NRP) has emerged as a pivotal strategy in controlled donation after circulatory death (cDCD) liver transplantation, mitigating ischemia-reperfusion injury and improving graft outcomes. Compared to super rapid recovery (SRR), NRP significantly reduces rates of early allograft dysfunction, primary nonfunction, and biliary non-anastomotic stricture (NAS), with biliary complications reported in 5%-16% and NAS as low as 0%-2%. Outcomes from cDCD-NRP grafts are increasingly comparable to those of donation after brain death (DBD). Viability assessment during NRP remai

Reinforcing Global Oversight of Organ Transplantation: Activity and Outcome Monitoring Through the Development of Registries.

Establishing transparency and oversight of organ transplantation by regulatory agencies is of paramount importance to assure ethical, legal, and clinically robust transplantation practices. Registries reporting activity and outcome data of the donor and recipient, including donor source (living or deceased), must be developed for each transplant and should be a mandatory requirement to achieve accreditation to perform transplant surgeries. Collected data for the living organ donor must include the nationality, the nature of their relationship with the recipient, and the complications encounter

The living kidney donor profile index fails to discriminate allograft survival: implications for its use in kidney paired donation programs.

The inclusion of blood group- and human leukocyte antigen-compatible donor and recipient pairs (CPs) in kidney paired donation (KPD) programs is a novel strategy to increase living donor (LD) transplantation. Transplantation from a donor with a better Living Donor Kidney Profile Index (LKDPI) may encourage CP participation in KPD programs. We undertook parallel analyses using data from the Scientific Registry of Transplant Recipients and the Australia and New Zealand Dialysis and Transplant Registry to determine whether the LKDPI discriminates death-censored graft survival (DCGS) between LDs.

Assessing Donor-Recipient Size Mismatch in Pediatric Heart Transplantation: Lessons Learned From Over 7,500 Transplants.

BACKGROUND: To date, no studies have identified an optimal metric to match donor-recipient (D-R) pairs in pediatric heart transplantation (HT). OBJECTIVES: This study sought to identify size mismatch metrics that predicted graft survival post-HT. METHODS: D-R pairs undergoing HT in Pediatric Heart Transplant Society database from 1993 to 2021 were included. Effects of size mismatch by height, weight, body mass index, body surface area, predicted heart mass, and total cardiac volume (TCV) on 1- and 5-year graft survival and morbidity outcomes (rejection and cardiac allograft vasculopathy) wer
