Isolation and detection of Fasciola hepatica DNA in Lymnaea viatrix from formalin-fixed and paraffin-embedded tissues through multiplex-PCR.

Detection of Fasciola hepatica infection in Lymnaea viatrix through analysis of histological cuts is based upon morphological characters of the parasite during the intra-mollusk phase of parasitism. At this stage, trematode forms are very similar and, thus, very difficult to differentiate. Specific detection may also be impaired by the presence of other helminthes in the mollusk. Histological samples are usually fixed in formalin, embedded in paraffin, sectioned and HE stained. In the current study, a method for the extraction of DNA from formalin-fixed, paraffin-embedded tissues was standardi

Workshop summary: food safety: meat- and fish-borne zoonoses.

In this workshop, J.P. Dubey (USA), Th. Hiepe (Germany), and P. Deplazes (Switzerland), were invited speakers. The main areas covered were toxoplasmosis, microsporidiosis, trematode infections, taeniosis/cysticercosis and trichinellosis. The public health and economic impact of meat- and fish-borne parasitic zoonoses is considerable in terms of morbidity and even mortality in humans as well as in losses due to reduced productivity in animals and condemnation of parasitised meat and fish. In this context, the increasing demands of consumers for meat and fish free of pathogens and chemical resid

The role of the thymus in the eosinophil response of rats infected with Fasciola hepatica.

The blood eosinophil response of normal and congenitally athymic nude rats was studied following infection with the parasitic trematode Fasciola hepatica. A marked eosinophilia was evident irrespective of the presence or absence of a thymus in the infected rats, suggesting that thymus-dependent mechanisms are not essential for the induction of eosinophilia in the rat.

A Preliminary Study of a Lettuce-Based Edible Vaccine Expressing the Cysteine Proteinase of Fasciola hepatica for Fasciolosis Control in Livestock.

Oral vaccination with edible vaccines is one of the most promising approaches in modern vaccinology. Edible vaccines are an alternative to conventional vaccines, which are typically delivered by injection. Here, freeze-dried transgenic lettuce expressing the cysteine proteinase of the trematode Fasciola hepatica (CPFhW) was used to orally vaccinate cattle and sheep against fasciolosis, which is the most important trematode disease due to the parasite's global distribution, wide spectrum of host species and significant economic losses of farmers. In the study, goals such as reducing the intensi

Inhibition of lactate dehydrogenase isoenzyme associated with anaerobic respiration in schistosomiasis intermediate host snails.

Lactate dehydrogenase isoenzyme (LD5) which is associated with anaerobic respiration was inhibited to a certain degree in Biomphalaria alexandrina snails, the intermediate host for Schistosoma mansoni. Urea and thiourea were used as inhibitors. The effect of LD5 inhibition on the mortality rate of infected Biomphalaria alexandrina snails and on the susceptibility of the snails to the trematode infection was also studied.

A Contribution Toward Understanding Trematode Diversity in Freshwater Snails of the Southwestern Kathmandu Valley, Nepal.

BACKGROUND: Freshwater snails serve as intermediate hosts for trematode parasites and play a key role in transmitting various parasitic diseases. Snail-borne trematodiases pose important public and veterinary health risks in South Asia, where freshwater snails sustain transmission in agricultural and periurban waters. However, data from the Kathmandu Valley remain limited and fragmented across habitats and seasons. OBJECTIVES: This study aimed to determine the diversity and distribution of cercarial larvae in freshwater snail species from the southwestern part of the Kathmandu Valley, Nepal.

Invasive snails, parasite spillback, and potential parasite spillover drive parasitic diseases of Hippopotamus amphibius in artificial lakes of Zimbabwe.

BACKGROUND: Humans impose a significant pressure on large herbivore populations, such as hippopotami, through hunting, poaching, and habitat destruction. Anthropogenic pressures can also occur indirectly, such as artificial lake creation and the subsequent introduction of invasive species that alter the ecosystem. These events can lead to drastic changes in parasite diversity and transmission, but generally receive little scientific attention. RESULTS: In order to document and identify trematode parasites of the common hippopotamus (Hippopotamus amphibius) in artificial water systems of Zimba

Stable isotopes unravel the feeding mode-trophic position relationship in trematode parasites.

Stable isotopes have been sporadically used over the last two decades to characterise host-parasite trophic relationships. The main reason for this scarcity is the lack of an obvious pattern in the ratio of nitrogen stable isotope values (δ15 N) of parasites in comparison to their host tissues, which would be key to understand any host-parasite system dynamics. To circumvent this, we focused on a single snail host, Zeacumantus subcarinatus, and three of its trematode parasites. We used stable isotopes to investigate each host-trematode trophic relationship and shed light on the mechanisms util

EXPERIMENTAL INFECTIONS WITH EUHAPLORCHIS CALIFORNIENSIS AND A SMALL CYATHOCOTYLID INCREASE CONSPICUOUS BEHAVIORS IN CALIFORNIA KILLIFISH (FUNDULUS PARVIPINNIS).

Some parasites manipulate their host's phenotype to enhance predation rates by the next host in the parasite's life cycle. Our understanding of this parasite-increased trophic transmission is often stymied by study-design challenges. A recurring difficulty has been obtaining uninfected hosts with a coevolutionary history with the parasites, and conducting experimental infections that mimic natural processes. In 1996, Lafferty and Morris provided what has become a classic example of parasite-increased trophic transmission; they reported a positive association between the intensity of a brain-in

Induced expression of CYP2A5 in inflamed trematode-infested mouse liver.

Trematode infections have long been associated with specific types of cancer. We investigated the ability of the liver fluke Fasciola hepatica to alter host enzymes in a manner that might provide insight into the phenomenon of biologically associated cancers. Our data demonstrate an increased activity of the CYP2A5 isozyme in male mouse liver infected with F.hepatica. Induction of this enzyme was further assessed immunohistochemically. The infection affected CYP2A5 distribution in hepatic tissue. Inflammation and proliferation in liver tissue were observed at the same time that CYP2A5 activity

The prevalence and distribution of Alaria alata, a potential zoonotic parasite, in foxes in Ireland.

The digenean trematode Alaria alata, an intestinal parasite of wild canids is widely distributed in Europe. The recent finding of the mesocercarial life cycle stage in the paratenic wild boar host suggests that it may potentially infect humans Mohl et al. (Parasitol Res 105:1-15, 2009). Over 500 foxes were examined during a wildlife survey for zoonotic diseases in 2009 and 2010. The prevalence of A. alata ranged from 21% to 26% in 2009 and 2010, and the intensity of infection varied, with the majority of foxes having between one and ten trematodes, but a small number of animals had parasitic b

Human helminth infections above latitude 60°N: reports published 2001-2024.

This article surveys reports of human helminth infection from geographical regions above latitude 60°N published in the period 2001–2024. We take a global approach encompassing the Americas and Eurasia. The helminth genera thus described herein include nematode (Trichinella, Toxocara, Anisakis, Pseudoterranova), cestode (Echinococcus, Dibothriocephalus) and trematode (Opisthorchis, Trichobilharzia). The primary reports identified infections principally by serology (community-based or individual, including imported cases) and outbreaks. There were also articles reporting national data compiled

The incidence, pathogenesis and treatment of helminth infections in rhesus monkeys (Macaca mulatta).

In a survey of 259 newly-imported rhesus monkeys, 5 species of nematode parasites were commonly found. Of these 5 species, Oesophagostomum spp. and Strongyloides fulleborni were implicated in the deaths of 3 monkeys. 5 other nematode species, 2 cestode species and 1 trematode species were also found; their pathogenicity is discussed. The results of treatment of parasitic infections with 6 different drugs is reported. Thiabendazole at an initial dose of 100 or 133 mg/kg and repeated at 50 mg/kg was found to be the most effective treatment against Oesophagostomum spp.

Effects of dietary intake of garlic on intestinal trematodes.

The current strategy for the control of helminth infections relies on chemotherapy. However, resistance appearance is promoting the necessity of developing new drugs against trematodes. Herein, potential trematocidal effects of garlic (Allium sativum) are investigated in the context of intestinal foodborne trematodes, employing the Echinostoma caproni-mouse model. Daily administration of dietary doses of garlic was conducted in three groups of mice: (i) before infection (prophylaxis), (ii) after infection (therapeutic) and (iii) both, before and after infection (continuous). A fourth group of

Genetic identification of gastrointestinal parasites in the world's most endangered ungulate, the hirola (Beatragus hunteri).

BACKGROUND: The hirola (Beatragus hunteri) is the world's most critically endangered antelope. Its parasitic fauna has never been genetically characterised, raising questions about its possible role in the species' population decline. This study aimed to assess the presence of gastrointestinal parasites in hirolas and their transmission pathways. METHODS: Thirty-one hirola faecal samples were examined using coprological methods. The identification of trematode and nematode species was performed by PCR amplification of the internal transcribed spacer 2 (ITS-2) and 18S ribosomal RNA (18S rRNA)

DNA barcoding of Iranian radicine freshwater snails begins to untangle the taxonomy and phylogeography of intermediate hosts of schistosomiasis and fasciolosis from the Middle East and across Central Asia.

In the Middle East radicine snails are of considerable medical and veterinary importance acting as vectors of trematodes. In Iran, such snails are responsible for the transmission of the zoonotic trematodes Schistosoma turkestanicum and Fasciola gigantica. Historically, Radix gedrosiana has been incriminated as an important intermediate host for both trematodes, however, controversy remains over the snail's true taxonomic status. This species has been determined using morphological characters that has resulted in erroneous identification of species, affecting understanding of population biolog

Temporal dynamics of trematode intermediate snail host environmental DNA in small water body habitats.

Environmental DNA (eDNA) surveying has potential to become a powerful tool for sustainable parasite control. As trematode parasites require an intermediate snail host that is often aquatic or amphibious to fulfil their lifecycle, water-based eDNA analyses can be used to screen habitats for the presence of snail hosts and identify trematode infection risk areas. The aim of this study was to identify climatic and environmental factors associated with the detection of Galba truncatula eDNA. Fourteen potential G. truncatula habitats on two farms were surveyed over a 9-month period, with eDNA detec

Revealing cryptic parasite diversity in a definitive host: echinostomes in muskrats.

Many trematode groups have a long history of systematic revision, which can make parasite identification a difficult task. The trematode parasites of muskrats are no exception. Here, we highlight the systematic issues associated with trematodes of muskrats (Ondatra zibethicus). Then, we demonstrate the utility of using both morphological and molecular tools to identify these parasites. Morphological examinations of specimens from muskrats (n = 63) first suggested that at least 4 genera were present including Echinostoma, Wardius, Quinqueserialis, and Notocotylus. For the latter 3 groups, the 2
