Visualizing the antivascular effect of bortezomib on the hypoxic tumor microenvironment.

BACKGROUND: Although physical inactivity may lead to increasing obesity prevalence, research on anthropometric variables changes based on physical activity (PA) in children and adolescents is limited. PA decreases with age, while sedentary behavior increases. The study aimed to examine differences in objectively measured sedentary time, light-intensity physical activity (LPA), and moderate-to-vigorous intensity physical activity (MVPA) between children and adolescents, and the differences in the percentiles of anthropometric variables between physically active and inactive groups according to

Influence of the tumor microenvironment on genetic mutations in thyroid carcinoma.

In contrast to cancers with high immunotherapy responsiveness, such as lung cancer and melanoma, thyroid carcinoma (THCA) immunotherapy remains investigational. To establish a theoretical foundation for THCA immunotherapy, we investigated the association between genetic mutations and tumor microenvironment (TME) by analyzing RNA-sequencing data and somatic mutation profiles from 571 THCA samples in The Cancer Genome Atlas (TCGA) database. The ESTIMATE algorithm was first applied to calculate ImmuneScores and StromalScores. Samples were subsequently stratified into immune-high and immune-low gr…

Emerging role of microRNAs as modulators of the tumor immune microenvironment.

The advent of immune checkpoint inhibitors (ICIs) has transformed the treatment landscape of several malignancies, but durable clinical benefits are only observed in a subset of patients. Primary and acquired resistance to immunotherapy, largely driven by an immunosuppressive tumor immune microenvironment (TIME), remains among the most critical hurdles to overcome. Amid the molecular regulators orchestrating immune evasion, tumor-derived microRNAs (miRNAs) have emerged as pivotal players. These small non-coding RNAs reshape the TIME through diverse mechanisms, including impairment of antigen p

Neutrophils in glioblastoma: orchestrators of the tumor microenvironment and immune evasion.

Glioblastoma (GBM) is the most aggressive primary brain tumor in adults and remains refractory to current therapies. Beyond profound immunosuppression, GBM is characterized by a complex tumor microenvironment (TME) in which neutrophils have emerged as critical yet understudied regulators of tumor progression and immune evasion. Tumor-associated neutrophils (TANs) display marked functional plasticity, acquiring pro-tumor or anti-tumor phenotypes depending on microenvironmental cues. GBM recruits and reprograms infiltrating neutrophils through chemokine-driven trafficking, hypoxia, and tumor-der…

Cancer and Immune Cells: A Metabolic Battle in the Tumor Microenvironment.

Immunometabolism has emerged as a critical field that links cellular energy regulation with immune function, including in the context of cancer. The tumor microenvironment (TME) is characterized by hypoxia, acidosis, and nutrient competition, all of which promote metabolic reprogramming in cancer cells while suppressing immune surveillance. Antitumor immune cells such as cytotoxic T lymphocytes, natural killer (NK) cells, and M1 macrophages rely on glycolysis and oxidative phosphorylation to sustain effector functions. In contrast, tumor-promoting cells, including regulatory T cells, myeloid-d…

Metabolic licensing and restriction of innate immunity in the tumor microenvironment.

Tumor progression is driven by metabolic remodeling that generates a microenvironment characterized by nutrient deprivation, hypoxia, lactat … the level of individual pathways or individual cell types, obscuring shared principles by which the tumor microenvironment controls innate immunity. Here, we use the concepts of metabolic licensing and metabolic restriction to describe how metabolic capacity and environmental constraints interact to shape innate immune function. Metabolic licensing refers to context-dep …

The role of spatial transcriptomics in exploring the tumor microenvironment in neuroblastoma.

Neuroblastoma is the most common extracranial solid tumor in children and exhibits significant variability in clinical outcomes. Our current understanding of the biology of neuroblastoma has been limited by an incomplete understanding of the complex interaction between tumor cells and the host cells within the tumor microenvironment (TME). The TME is composed of a rich ecosystem of immune and stromal cells, which may play a role in tumor establishment, growth, treatment resistance, and recurrence. Single-cell analysis platforms have been employed to characterize the tumor and non-tumor cells o…

Impact of the tumor microenvironment on survival in anaplastic thyroid carcinoma.

Anaplastic thyroid carcinoma (ATC) is a highly aggressive malignancy characterized by nearly 100% mortality and significant therapeutic challenges. This study investigated the tumour microenvironment (TME) in ATC, focusing on immune checkpoint molecules (PD-L1/PD-1), tumour-associated macrophages (TAMs), and E-cadherin expression. A retrospective cohort of 22 ATC patients treated at a tertiary care centre in India, between January 2017 and August 2022, was analysed. Immunohistochemical evaluation revealed PD-L1 expression in 68.2% of cases, with a median tumour proportion score (TPS) of 50. PD

Establishment of tumor microenvironment following bisphenol A exposure in the testis.

Although detrimental roles of bisphenol A (BPA) in xenoestrogen target organs, testis and epididymis, and male fertility are well-documented, disruption of the immune privilege system in the male reproductive tract following BPA exposure remains poorly understood. Therefore, this study aimed to explore the precise mechanisms of BPA in interfering immune privilege in the testis on RNA sequencing results. CD-1 male mice were daily treated no-observed-adverse-effect (NOAEL, 5 mg BPA/kg BW) and lowest-observed-adverse-effects (LOAEL, 50 mg BPA/kg BW) of BPA by oral gavage for 6 weeks. Following th

Spatially confined sub-tumor microenvironments in pancreatic cancer.

Intratumoral heterogeneity is a critical frontier in understanding how the tumor microenvironment (TME) propels malignant progression. Here, we deconvolute the human pancreatic TME through large-scale integration of histology-guided regional multiOMICs with clinical data and patient-derived preclinical models. We discover "subTMEs," histologically definable tissue states anchored in fibroblast plasticity, with regional relationships to tumor immunity, subtypes, differentiation, and treatment response. "Reactive" subTMEs rich in complex but functionally coordinated fibroblast communities were i…

The Role of Tumor Microenvironment in Multiple Myeloma Development and Progression.

Multiple myeloma (MM) is a hematologic cancer characterized by clonal proliferation of plasma cells in the bone marrow (BM). The progression, from the early stages of the disease as monoclonal gammopathy of undetermined significance (MGUS) and smoldering multiple myeloma (SMM) to MM and occasionally extramedullary disease, is drastically affected by the tumor microenvironment (TME). Soluble factors and direct cell-cell interactions regulate MM plasma cell trafficking and homing to the BM niche. Mesenchymal stromal cells, osteoclasts, osteoblasts, myeloid and lymphoid cells present in the BM cr…

[The effect of the microenvironment of head and neck cancers on tumor progression].

In the last 20 years the incidence and mortality of head and neck squamous cell carcinomas showed an increasing rate in Hungary. In our work we examined the microenvironment of head and neck cancers localized in different anatomical regions. Clinical evidence shows that the prognosis of hypopharyngeal tumors is poorer than that of head and neck cancers in other anatomical locations. We investigated if tumor size or vascularity correlates with the biological behavior of these tumors. The results showed that the tumor size of laryngeal cancers in T2 stage were significantly larger than that of h

PTPRN2-mediated silencing of tumor-intrinsic MHC-II promotes immune-excluded tumor microenvironment in colorectal cancer.

Colorectal cancer (CRC) frequently exhibits an immune-excluded tumor microenvironment that limits the efficacy of PD-1 blockade. Through systematic profiling of 106 protein tyrosine phosphatases (PTPs), we identify three PTP-based molecular subtypes, among which the immune-excluded Cluster 2 is highly resistant to immunotherapy. Integrative analyses across patient cohorts highlight PTPRN2 as a prominent PTP enriched in immune-excluded tumors and strongly associated with anti-PD-1 non-response. Functional studies demonstrate that PTPRN2 promotes CRC cell proliferation and invasion, and more imp…

Mechanism of liposome arsenic trioxide regulating tumor microenvironment and sensitizing tumor immune response after radiofrequency ablation.

OBJECTIVE: We hypothesized that preoperative administration of liposome arsenic trioxide (LATO) would downregulate hypoxia-inducible factor-1α (HIF-1α) in residual tumors following incomplete radiofrequency ablation (iRFA), thereby modulating the tumor microenvironment, suppressing residual tumor proliferation and angiogenesis, and consequently enhancing the antitumor efficacy of programmed death-ligand 1 (PD-L1) blockade. METHODS: Using a murine Hepa1-6 hepatocellular carcinoma model subjected to incomplete radiofrequency iRFA, cohorts were stratified based on preoperative LATO administratio…

Single-Cell Glycomics of the Pancreatic Tumor Microenvironment: Technologies, Glyco-Immune Checkpoints, and Tumor-Immune Communication.

In this Review, we summarize recent conceptual and technological advances in single-cell glycomics, with a particular focus on emerging strategies that enable glycan-resolved analysis of tumor-immune interactions. We highlight single-cell glycan and RNA sequencing (scGR-seq), a methodology that converts glycan information into amplifiable nucleic acid signals, enabling simultaneous profiling of transcriptomes and glycomes at the single-cell level. In parallel, we introduce GlycoChat, a computational framework designed to systematically infer and visualize glycan-lectin interaction networks wit

Nitric Oxide-Producing Multiple Functional Nanoparticle Remodeling Tumor Microenvironment for Synergistic Photodynamic Immunotherapy against Hypoxic Tumor.

The treatment of pancreatic cancer faces significant challenges due to connective tissue hyperplasia and severe hypoxia. Unlike oxygen-dependent Type II photosensitizers, Type I photosensitizers can produce a substantial amount of reactive oxygen species, even under hypoxic conditions, making them more suitable for photodynamic therapy of pancreatic cancer. However, the dense extracellular matrix of pancreatic cancer limits the penetration efficiency of photosensitizers, and the presence of immunosuppressive cells in the tumor microenvironment reduces the therapeutic effect. To address these c…

Deciphering T-cell exhaustion in the tumor microenvironment: paving the way for innovative solid tumor therapies.

In solid tumors, the tumor microenvironment (TME) is a complex mix of tumor, immune, stromal cells, fibroblasts, and the extracellular matrix. Cytotoxic T lymphocytes (CTLs) constitute a fraction of immune cells that may infiltrate into the TME. The primary function of these T-cells is to detect and eliminate tumor cells. However, due to the immunosuppressive factors present in the TME primarily mediated by Myeloid-Derived Suppressor Cells (MDSCs), Tumor associated macrophages (TAMs), Cancer Associated Fibroblasts (CAFs) as well as the tumor cells themselves, T-cells fail to differentiate into…
