Recombinant vaccinia vector-based vaccine (Tiantan) boosting a novel HBV subunit vaccine induced more robust and lasting immunity in rhesus macaques.

This study explored several prime-boost strategies in rhesus macaques using various novel hepatitis B virus (HBV) vaccines that showed promise as prophylactic and therapeutic approaches in our previous study using in a mouse model. The tested vaccines included an HBV particle subunit (HBSS1) vaccine and the recombinant vaccinia (RVJSS1) or adenoviral (rAdSS1) vector-based vaccines containing S (1-223aa) and PreS1 (21-47aa). The strength and maintenance of humoral activity (IgG and neutralizing antibodies) and cellular immunity (interferon-γ production assessed by IFN-γ enzyme-linked immunosorb

Engineering a dimeric porcine deltacoronavirus S-RBD subunit vaccine for enhanced immunogenicity and neutralizing antibody responses.

BACKGROUND: Porcine deltacoronavirus (PDCoV) is an emerging swine enteric coronavirus that has caused global epidemics and substantial economic losses, posing a potential risk of cross-species transmission, including to humans. However, commercial vaccines are currently scarce. The receptor binding domain (RBD) of the spike (S) protein is a key target for inducing neutralizing antibodies, but the RBD monomer typically exhibits low molecular weight and weak immunogenicity. This study aimed to develop a tandem dimeric RBD subunit vaccine and evaluate its immunogenicity and protective efficacy in

Intranasal delivery of a polymeric nanoparticle subunit vaccine for the induction of protective immunity against respiratory syncytial virus.

Respiratory syncytial virus (RSV) is the leading cause of lower respiratory tract infection in children under the age of 5, worldwide. Maternal vaccination and monoclonal antibodies provide only temporary protection for young children. As antibody wanes, young children remain at risk for severe RSV disease, creating an unmet need for a direct childhood vaccine. We developed an intranasal (IN) subunit vaccine, comprised of a biotinylated nanoparticle and RSV prefusion protein DS-Cav1 (PreF-bNP) with a flexible "plug-n-play" design that enables dose optimization of antigen and adjuvants. In a pr

Development of a cross-protective subunit cocktail vaccine against diverse serotypes of Glaesserella parasuis infection in pigs.

Glässer's disease, caused by Glaesserella parasuis (GPS), causes substantial economic losses in the swine industry worldwide. This situation is largely attributable to the poor cross-protective efficacy of currently available vaccines. In this study, a multi-antigen cocktail subunit vaccine against GPS infection was developed. In silico antigenicity analysis, the immunoprotective efficacy, and antibody titers of 14 GPS antigens were assessed in a murine model. Finally, VacJ, PtsG, and GAPDH were identified as the component of the cocktail vaccine, with Gel-01 as the optimal adjuvant. Immunizat

Development of a genetically engineered multivalent subunit vaccine against "struck", "braxy", lamb dysentery, and enterotoxemia in sheep.

The development of genetically engineered multivalent subunit vaccines is needed to provide protection against infections caused by Clostridium septicum and multiple types of Clostridium perfringens. In this study, a pET-32a(+) plasmid encoding the C. septicum α-toxin gene and the C. perfringens β-toxin gene was constructed; a positive clone was designated pET-32a-α-β1. This plasmid, along with pET-32a-α-β2-ε-previously engineered to express a C. perfringens α-β2-ε trivalent recombinant chimeric fusion protein-was transformed into Escherichia coli DE3 for recombinant protein expression after i

Porcine genetic markers of immunocompetence shape the immune response to a classical swine fever virus subunit vaccine.

Breeding programs aimed at enhancing robustness and disease resistance in pigs represent a complementary strategy to biosecurity and vaccination measures. The objective of the present study was to validate a panel of genetic biomarkers and immunity phenotypes as indicators of the immune response to the E2-CD154 subunit vaccine against classical swine fever virus (CSFV). A total of 120 eight-week-old Duroc pigs were genotyped using a panel of six genetic markers, previously described as associated with innate and adaptive immunity traits. Additionally, total plasmatic IgG concentrations and SOX

Immune efficacy evaluation of a PEDV spike protein-derived multi-epitope subunit vaccine candidate in mice.

Porcine epidemic diarrhea virus (PEDV)-induced piglet diarrhea remains one of the most threatening infectious diseases in the swine industry across many countries. Although commercial vaccines are available, their efficacy in preventing the incidence and prevalence of porcine epidemic diarrhea (PED) remains unsatisfactory. Here we developed three subunit vaccine candidates based on four antigenic domains of the PEDV spike (S) protein expressed in E. coli: core neutralizing epitope (COE), S1 subunit domain A (S1A), and more conservative antigenic epitopes heptad repeat 1 (HR1) and heptad repeat

In silico design of a VacA-based multi-epitope Subunit Vaccine Candidate for immunoprotection against Helicobacter pylori.

Helicobacter pylori, a globally prevalent gastric pathogen associated with gastric carcinoma, ulcers of the bowel, and persistent gastritis where the vacuolating cytotoxin A (vacA), a major virulence factor, that promotes cellular damage and immune evasion. Despite increasing antibiotic resistance, no licensed vaccine exists for effective prevention or treatment. In present study, we implemented an all-encompassing immunoinformatic strategy in developing a novel vacA- driven multi-epitope subunit vaccine (MESV). Utilising the IEDB database, cytotoxic as well as helper T-cell epitopes were dete

Robust immunoreaction induced by a subunit vaccine of PEDV spike protein based on GEM surface-display system.

The global porcine epidemic diarrhea (PED) pandemic underscores the urgent need for safe, effective, and readily deployable subunit vaccines against porcine epidemic diarrhea virus (PEDV). In this study, we developed a subunit vaccine by displaying the PEDV spike (S) protein on Gram-positive enhancer matrix (GEM) particles via a protein anchor (PA), resulting in stable particulate antigen complexes designated GEM-S. The immunogenicity of GEM-S was evaluated in mice. Immune responses in lymph nodes (LNs), spleen, and serum were assessed using reverse transcription-quantitative polymerase chain

Recombinant fusion subunit vaccine provides enhanced immune protection against Cryptocaryon irritans infection in grouper.

Cryptocaryon irritans causes severe losses in marine aquaculture, yet effective vaccines remain limited. In this study, we developed a novel fusion vaccine composed of an immobilization antigen (IA8) and the 26S proteasome, and evaluated its immunogenicity and protective efficacy in Epinephelus coioides. The recombinant fusion protein was successfully expressed in Escherichia. coli and exhibited strong immunoreactivity. Following parasite challenge, the fusion vaccine conferred the highest protection, reducing parasite abundance by 81.2%, compared with 44.6% and 22.6% for IA8 and 26S proteasom

A novel subunit vaccine based on glycoprotein H and glycoprotein L fusion against pseudorabies virus infection.

Pseudorabies (PR) is a highly infectious disease caused by pseudorabies virus (PRV), which leads to significant economic losses. Vaccination remains the most effective method to control PRV infection. To develop an efficient DIVA (differentiation of infected from vaccinated animals) vaccine against PRV, this study expressed the envelope glycoprotein H (gH), glycoprotein L (gL), and a recombinant gH-gL fusion protein in a mammalian cell expression system and evaluated their protective efficacy in a mouse model. Immunization with gH, gL, or gH-gL induced high levels of specific antibodies capabl

Construction and immunogenicity evaluation of a tuberculosis subunit vaccine based on fusion protein AP2 and novel adjuvant ASM.

Tuberculosis is a severe global respiratory infectious disease caused by infection with Mycobacterium tuberculosis (M. tb). Bacillus Calmette-Guérin (BCG) is currently the only approved vaccine for TB prevention; however, its protective efficacy against tuberculosis in adults is limited. To overcome the limitations of BCG, recombinant plasmids were constructed and expressed using antigens Rv2074 and Rv2642. Subsequently, a full-length gene synthesis strategy incorporating a linker was adopted to construct the fusion antigen AP2 and express its corresponding protein. Peripheral blood samples fr

Multi-tope subunit vaccine for human anthrax through pan-genome-based reverse vaccinology approach.

Anthrax is a zoonotic disease caused by Bacillus anthracis and poses a serious threat to public health. Frequent anthrax outbreaks in wild animals and livestock in the tropical regions lead to mortality and substantial economic losses. Farm workers, veterinarians, and individuals in close contact with infected animals are at risk of contracting the infection. In this study, a multi-tope vaccine for humans was designed through a pan-genome-based reverse vaccinology approach. We predicted twelve proteins that can elicit both B-cell and T-cell-mediated immunity, from which we identified eighteen

Designing of a conserved subunit multiepitope vaccine candidate against Francisella tularensis Schu S4 using immunoinformatics.

Francisella tularensis Schu S4 is the most virulent strain responsible for tularemia, a highly infectious zoonotic disease. Despite its potential as a pathogen of concern, no effective vaccine has been developed for this pathogen. In this study, two multi-epitope subunit vaccine candidates were designed targeting the virulence factors DacD and DsbA to reduce disease severity caused by F. tularensis Schu S4. Using a comprehensive immunoinformatics approach, the most efficient B- and T-cell epitopes were identified from conserved sequences and linked with adjuvants and linkers to generate two va

Designing a broad-spectrum multi-epitope subunit vaccine against leptospirosis using immunoinformatics and structural approaches.

INTRODUCTION: Leptospirosis, caused by Leptospira interrogans, is a neglected zoonotic disease that poses a significant global health risk to both humans and animals. The rise of antimicrobial resistance and the inefficacy of existing vaccines highlight the urgent need for new preventive strategies. METHODS: An immunoinformatics approach was employed to design a multi-epitope subunit vaccine (MESV) against leptospirosis. B-cell, cytotoxic T lymphocyte (CTL), and helper T lymphocyte (HTL) epitopes were selected from five key Leptospira proteins. These epitopes were fused with a heparin-binding

Engineered Low-Endotoxin Bacterial Biomimetic Vesicles for Enhanced Oral Dual-Antigen Subunit Vaccine Delivery.

Subunit vaccines are promising for disease prevention because of their safety and cost-effectiveness. However, their efficacy is limited by low immunogenicity and gastrointestinal degradation after oral administration. To address this issue, low-endotoxin Salmonella choleraesuis strain SC-L3 was engineered via lipid A modification to generate bacterial biomimetic vesicles (BBVs) with reduced endotoxin activity. BBVs were functionalized using ClyA-embedded SpyCatcher and Streptococcus protein G for dual antigen coupling, and further coated with chitosan oligosaccharides (COS) to enhance mucosal

Comparative analysis of the efficacy of vaccines using structural protein subunits of the severe fever with thrombocytopenia syndrome virus.

The severe fever with thrombocytopenia syndrome virus (SFTSV) represents a significant emerging health threat as a tick-borne pathogen that causes SFTS, with mortality rates ranging between 10 and 30%. Despite the considerable risk presented by SFTSV, an effective vaccine has yet to be developed. Our study assessed the efficacy of recombinant protein vaccines, focusing on the purified nucleocapsid protein (NP) and surface glycoproteins (Gn and Gc), against SFTSV in both singular and combined formulations. Individual vaccinations with NP or Gn subunits yielded partial protection in type I inter

Preparation and characterization of curdlan-chitosan conjugate nanoparticles as mucosal adjuvants for intranasal influenza H1N1 subunit vaccine.

Intranasal vaccination offers crucial protection against influenza virus pandemics. However, antigens, especially subunit antigens, often fail to induce effective immune responses without the help of immune adjuvants. Our research has demonstrated that a polyelectrolyte complex, composed of curdlan sulfate/O-(2-hydroxyl) propyl-3-trimethyl ammonium chitosan chloride (CS/O-HTCC), effectively triggers both mucosal and systemic immune responses when administrated intranasal. In this study, stable nanoparticles formed by curdlan-O-HTCC conjugate (CO NP) were prepared and characterized. Furthermore
