DNA-AuNP networks on cell membranes as a protective barrier to inhibit viral attachment, entry and budding.

Viral infections have caused numerous diseases and deaths worldwide. Due to the emergence of new viruses and frequent virus variation, conve … efore, our results suggest that the DNA-AuNP networks have not only prophylactic effects to inhibit virus attachment and entry, but also therapeutic effects to inhibit viral budding and cell-to-cell spread. More importantly, this proof-of-principle study provides a pathway for the development of a universal, broad-spectrum antiviral therapy.

Intestinal Tropism of an Infectious Bronchitis Virus Isolate Not Explained by Spike Protein Binding Specificity.

An infectious bronchitis virus (IBV) with an unusual enteric tropism (CalEnt) was isolated from a California broiler flock exhibiting runting-stunting syndrome. IBV was detected in the small intestine, but not in the respiratory tract or kidney. During virus isolation in embryos, it did not replicate in chorioallantoic membrane (CAM) but could be recovered from intestines. Its S1 protein showed 93% amino acid sequence identity to a California variant isolated in 1999 (Cal99). Intestinal lesions were reproduced following ocular/nasal inoculation of specific-pathogen-free chickens, but respirato

Pathological lesions and viral localization of influenza A (H5N1) virus in experimentally infected Chinese rhesus macaques: implications for pathogenesis and viral transmission.

Chinese rhesus macaques infected with influenza virus A/Tiger/Harbin/01/2002 (H5N1) developed acute interstitial pneumonia with diffuse alveolar damage. The results of virus isolation, reverse transcriptase polymerase chain reaction, immunohistochemistry, and in situ hybridization showed that the lung was the major target organ of the H5N1 virus infection. No virus was detected in the extrapulmonary organs. The results of immunohistochemistry and in situ hybridization also showed that pneumocytes and macrophages of the lower airway, not the ciliary epithelium of the trachea and bronchi, were t

A solid-phase enzyme-linked assay for influenza virus receptor-binding activity.

Receptor-binding properties of influenza viruses are usually characterized by the ability of viruses to interact with more or less defined sialic acid-containing carbohydrates, glycoproteins, or glycolipids assayed by haemagglutination (HA) or HA inhibition (HAI) tests. To overcome some drawbacks of these tests a solid-phase enzyme linked assay analogous to sandwich ELISA was developed. The virus is adsorbed specifically to the well of plastic microtitre plates coated with fetuin, and the binding of horseradish peroxidase (HRP)labelled sialylglycoproteins (SGPs) by the solid phase-attached vir

The DNA segregation mechanism of Epstein-Barr virus nuclear antigen 1.

Latent Epstein-Barr virus (EBV) genomes are maintained in human cells as low copy number episomes that are thought to be partitioned by attachment to the cellular mitotic chromosomes through the viral EBNA1 protein. We have identified a human protein, EBP2, which interacts with the EBNA1 sequences that govern EBV partitioning. Here we show that, in mitosis, EBP2 localizes to the condensed cellular chromosomes producing a staining pattern that is indistinguishable from that of EBNA1. The localization of EBNA1 proteins with mutations in the EBP2 binding region was also examined. An EBNA1 mutant

[Polymelia in a female calf - a case report with surgical removal].

The female German Holstein calf presented here was born with a complete additional forelimb. The forelimb's attachment point was between the shoulder blades. This is referred to as notomelia, a form of polymelia. On the 15th day of the calf's life, the additional limb was removed surgically. The surgery and wound healing progressed in a satisfactory manner. The cytogenetic tests did not reveal any chromosomal abnormalities. Genetic testing showed no indication of presence of a chimera that could be the cause of the anomaly. Antigens of the Schmallenberg virus were not detected, an antibody tit

A Sialylated Voltage-Dependent Ca2+ Channel Binds Hemagglutinin and Mediates Influenza A Virus Entry into Mammalian Cells.

Influenza A virus (IAV) infection is initiated by the attachment of the viral glycoprotein hemagglutinin (HA) to sialic acid on the host cell surface. However, the sialic acid-containing receptor crucial for IAV infection has remained unidentified. Here, we show that HA binds to the voltage-dependent Ca2+ channel Cav1.2 to trigger intracellular Ca2+ oscillations and subsequent IAV entry and replication. IAV entry was inhibited by Ca2+ channel blockers (CCBs) or by knockdown of Cav1.2. The CCB diltiazem also inhibited virus replication in vivo. Reintroduction of wild-type but not the glycosylat

Transmission of Crimean-Congo haemorrhagic fever virus from experimentally infected sheep to Hyalomma truncatum ticks.

Crimean-Congo haemorrhagic fever (CCHF) virus was inoculated into West African sheep that were simultaneously infested with adult Hyalomma truncatum ticks. Certain sheep developed a viraemia and antibodies, indicating virus infection and replication; however, the length and magnitude of the viraemia and serological responses corresponded to the animals' immunological status. Tick attachment and feeding was not influenced by sheep infection. CCHF virus infection was acquired by 11-33% of female and 0-60% of male ticks. Infection in the ticks did not influence their feeding success, as judged by

CLDN3 inhibits rotavirus attachment by targeting residue 74 of VP7.

Rotavirus (RV) VP8* peptide-induced CLDN3 mislocalization supports the hypothesis that CLDN3 negatively regulates viral binding, while the molecular basis of this inhibitory function remains unresolved. To counteract the CLDN3 defense strategies, RV infection indeed disrupts its localization to the plasma membrane. We also found that RV infection could reduce its protein levels in both in vitro and animal models. Knockdown or knockout of CLDN3 effectively promotes RV binding and entry. Further, we found that CLDN3 EC1 loop could interact with the N-terminal domain of VP7 and structural studies

Synthetic polymeric inhibitors of influenza virus receptor-binding activity suppress virus replication.

A new approach to anti-influenza chemotherapy is based on the development of synthetic inhibitors of virus attachment to host cells. These inhibitors are prepared by anchoring the minimum receptor determinant of influenza virus, sialic acid, to polymeric or liposomal carriers. In this study, a series of poly(acrylic acid-co-acrylamides) and dextrans bearing pendant glycylamidobenzylsialoside groups were synthesized and evaluated for their binding to a panel of influenza A and B virus strains and for their ability to inhibit virus infectivity in cell culture. Significant type-, subtype-, and st…

Peptides developed against receptor binding sites of the E glycoprotein neutralize tick-borne encephalitis virus.

Infection caused by tick-borne encephalitis virus (TBEV) often manifests with meningitis or encephalitis. Domain III (DIII) of the envelope (E) glycoprotein on the TBEV surface facilitates virion attachment to the cell receptor and initiates cell entry. As a result, this research focused on the DIII to develop antiviral molecules that could prevent cell entry. We first identified two receptor-binding sites (RBS, 300SGLTYTMCDK309 and 317APTDSGHDTVVMEVTFSGTKPCR339) on DIII, that are involved in binding to human brain microvascular endothelial cells (hBMECs). Then we sought to isolate peptides fr

GGCX promotes Eurasian avian-like H1N1 swine influenza virus adaption to interspecies receptor binding.

The Eurasian avian-like (EA) H1N1 swine influenza virus (SIV) possesses the capacity to instigate the next influenza pandemic, owing to its heightened affinity for the human-type α-2,6 sialic acid (SA) receptor. Nevertheless, the molecular mechanisms underlying the switch in receptor binding preferences of EA H1N1 SIV remain elusive. In this study, we conduct a comprehensive genome-wide CRISPR/Cas9 knockout screen utilizing EA H1N1 SIV in porcine kidney cells. Knocking out the enzyme gamma glutamyl carboxylase (GGCX) reduces virus replication in vitro and in vivo by inhibiting the carboxylatio

Determining the Importance of Carbohydrate-Based Structures in Murine Norovirus Binding to Commensal Bacteria.

Norovirus-bacterial interactions influence viral replication and immune responses, yet the molecular details that mediate binding of these viruses to commensal bacteria are unknown. Studies with other enteric viruses have revealed that LPS and other lipid/carbohydrate structures facilitate virus-bacterial interactions, and it has also been shown that human noroviruses (HuNoVs) can interact with histo-blood group antigen (HBGA)-like compounds on the surface of bacterial cells. Based on these findings, this study hypothesized that carbohydrate-based compounds were the ligands that facilitated bi

Unraveling the Epidemiology, Geographical Distribution, and Genomic Evolution of Potentially Lethal Coronaviruses (SARS, MERS, and SARS CoV-2).

SARS CoV appeared in 2003 in China, transmitted from bats to humans via eating infected animals. It affected 8,096 humans with a death rate of 11% affecting 21 countries. The receptor binding domain (RBD) in S protein of this virus gets attached with the ACE2 receptors present on human cells. MERS CoV was first reported in 2012 in Middle East, originated from bat and transmitted to humans through camels. MERS CoV has a fatality rate of 35% and last case reported was in 2017 making a total of 1,879 cases worldwide. DPP4 expressed on human cells is the main attaching site for RBD in S protein of

Analysis of chromatin attachment and partitioning functions of bovine papillomavirus type 1 E2 protein.

Recent studies have suggested that the tethering of viral genomes to host cell chromosomes could provide one of the ways to achieve their nuclear retention and partitioning during extrachromosomal maintenance in dividing cells. The data we present here provide firm evidence that the partitioning of the bovine papillomavirus type 1 (BPV1) genome is dependent on the chromatin attachment process mediated by viral E2 protein and its multiple binding sites. On the other hand, the attachment of E2 and the E2-mediated tethering of reporter plasmids to host chromosomes are not necessarily sufficient f

Completely conserved VP2 residue K140 of KREMEN1-dependent enteroviruses is critical for virus-receptor interactions and viral infection.

UNLABELLED: The KREMEN1 (KRM1) protein is a cellular receptor for multiple enteroviruses that cause hand, foot, and mouth disease (HFMD), including coxsackievirus CVA2, CVA3, CVA4, CVA5, CVA6, CVA10, and CVA12. The molecular basis for the broad recognition of these viruses by the KRM1 receptor remains unclear. Here, we report the indispensable role of the completely conserved VP2 capsid protein residue K140 (designated K2140) in mediating receptor recognition and infection by CVA10 and other KRM1-dependent enteroviruses. Residue K2140 not only facilitates receptor recognition, cell attachment,

Salvianolic acid A inhibits pseudorabies virus infection by directly inactivating the virus particle.

BACKGROUND: Pseudorabies virus (PRV), a member of the family Herpesviridae, is responsible for significant economic losses in the pig industry and has recently been associated with human viral encephalitis, leading to severe neurological symptoms post-recovery. Despite the widespread impact of PRV, there are currently no approved effective drugs for treating PRV-related diseases in humans or pigs. Therefore, the exploration and discovery of safe and effective drugs for the prevention and treatment of PRV infection is of paramount importance. PURPOSE: The objective of this study is to screen a
