A three-dimensional vocal fold posturing model based on muscle mechanics and magnetic resonance imaging of a canine larynx.

In this work, a high-fidelity three-dimensional continuum model of the canine laryngeal framework was developed for simulating laryngeal posturing. By building each muscle and cartilage from magnetic resonance imaging (MRI), the model is highly realistic in anatomy. The muscle mechanics is modeled using the finite-element method. The model was tested by simulating vocal fold postures under systematic activations of individual as well as groups of laryngeal muscles, and it accurately predicted vocal fold posturing parameters reported from in vivo canine larynges. As a demonstration of its appli

Sensory feedback independent pre-song vocalizations correlate with time to song initiation.

The song of the adult male zebra finch is a well-studied example of a learned motor sequence. Song bouts begin with a variable number of introductory notes (INs) before actual song production. Previous studies have shown that INs progress from a variable initial state to a stereotyped final state before each song. This progression is thought to represent motor preparation, but the underlying mechanisms remain poorly understood. Here, we assessed the role of sensory feedback in the progression of INs to song. We found that the mean number of INs before song and the progression of INs to song we

Identity and novelty in the avian syrinx.

In its most basic conception, a novelty is simply something new. However, when many previously proposed evolutionary novelties have been illuminated by genetic, developmental, and fossil data, they have refined and narrowed our concept of biological "newness." For example, they show that these novelties can occur at one or multiple levels of biological organization. Here, we review the identity of structures in the avian vocal organ, the syrinx, and bring together developmental data on airway patterning, structural data from across tetrapods, and mathematical modeling to assess what is novel.

Evaluating alpha-synuclein proteinopathy and consequences for birdsong in zebra finch basal ganglia area X.

Lewy body pathology is a major hallmark of Parkinson's Disease (PD) and other dementias. The process of Lewy body formation is largely driven by the aggregation of alpha-synuclein (αsyn), an abundant presynaptic chaperone protein that has been shown to propagate between neurons when misfolded. Preclinical animal models inducing αsyn aggregation in the brain have demonstrated a range of behavioral consequences, though studies connecting behavioral changes to specific features of pathology are lacking. Considering vocal impairment manifests early in individuals with PD and fails to resolve upon

Audiovisual non-verbal dynamic faces elicit converging fMRI and ERP responses.

In an everyday social interaction we automatically integrate another's facial movements and vocalizations, be they linguistic or otherwise. This requires audiovisual integration of a continual barrage of sensory input-a phenomenon previously well-studied with human audiovisual speech, but not with non-verbal vocalizations. Using both fMRI and ERPs, we assessed neural activity to viewing and listening to an animated female face producing non-verbal, human vocalizations (i.e. coughing, sneezing) under audio-only (AUD), visual-only (VIS) and audiovisual (AV) stimulus conditions, alternating with

The role of auditory experience in the formation of neural circuits underlying vocal learning in zebra finches.

The initial establishment of topographic mapping within developing neural circuits is thought to be shaped by innate mechanisms and is primarily independent of experience. Additional refinement within topographic maps leads to precise matching between presynaptic and postsynaptic neurons and is thought to depend on experiential factors during specific sensitive periods in the animal's development. In male zebra finches, axonal projections of the cortical lateral magnocellular nucleus of the anterior neostriatum (lMAN) are critically important for vocal learning. Overall patterns of topographic

Preservation of function and histologic appearance in the injured glottis with topical mitomycin-C.

OBJECTIVE: To evaluate the functional and histological effects of a single application of topical mitomycin-C after laser injury in the posterior canine glottis. STUDY DESIGN: A prospective, randomized study of 16 canines. METHODS: A supersaturated (1%) solution of topical mitomycin-C was applied to a unilateral, laser-induced injury near the cricoarytenoid joint in eight dogs. The mitomycin-soaked pledget was placed immediately after induction of the injury and was left in contact with exposed cartilage for 3 minutes. The opposite side was not injured to provide an internal control. In eigh

Intravenous mesenchymal stem cell therapy after recurrent laryngeal nerve injury: a preliminary study.

OBJECTIVES/HYPOTHESIS: Intravenous administration of mesenchymal stem cells (MSCs) has been recently shown to enhance functional recovery after stroke and spinal cord injury. The therapeutic properties of MSCs are attributed to their secretion of a variety of potent antiinflammatory and neurotrophic factors. We hypothesize that intravenous administration of MSCs after recurrent laryngeal nerve (RLN) injury in the rat may enhance functional recovery. STUDY DESIGN: Animal Research. METHODS: Twelve 250-gram Sprague-Dawley rats underwent a controlled crush injury to the left RLN. After confirmin

Characterization of raised phonation in an evoked rabbit phonation model.

OBJECTIVES/HYPOTHESIS: Our laboratory has developed an in vivo rabbit model to investigate the effects of phonation on expression and turnover of the vocal fold extracellular matrix. As a logical outgrowth of this research to include phonotrauma in the present study, we investigated the hypothesis that an increase in airflow rate delivered to the glottis produces a change in glottal configuration and an increase in mean phonation intensity. STUDY DESIGN: Prospective animal study. METHODS: Six New Zealand white breeder rabbits weighing 3 to 5 kg were used in this study. A rigid endoscope and

Neuropathic pain modeling: Focus on synaptic and ion channel mechanisms.

Animal models of pain consist of modeling a pain-like state and measuring the consequent behavior. The first animal models of neuropathic pain (NP) were developed in rodents with a total lesion of the sciatic nerve. Later, other models targeting central or peripheral branches of nerves were developed to identify novel mechanisms that contribute to persistent pain conditions in NP. Objective assessment of pain in these different animal models represents a significant challenge for pre-clinical research. Multiple behavioral approaches are used to investigate and to validate pain phenotypes inclu

Functional electrical stimulation of laryngeal adductor muscle restores mobility of vocal fold and improves voice sounds in cats with unilateral laryngeal paralysis.

Functional electrical stimulation (FES) has been proposed as a potential treatment for restoring motor functions of denervated motor systems. We investigated whether FES of paralyzed laryngeal adductor muscles could restore adduction to the vocal folds. In addition, we studied the effect of stimulated vocal fold adduction on the intensity and overall quality of voice production. We recorded movement of the vocal fold, electromyographic activity of muscles recruited for vocalization, and sound production in unanesthetized decerebrate cats during FES of the paralyzed thyroarytenoid (TA) muscle.

Vocal state change through laryngeal development.

Across vertebrates, progressive changes in vocal behavior during postnatal development are typically attributed solely to developing neural circuits. How the changing body influences vocal development remains unknown. Here we show that state changes in the contact vocalizations of infant marmoset monkeys, which transition from noisy, low frequency cries to tonal, higher pitched vocalizations in adults, are caused partially by laryngeal development. Combining analyses of natural vocalizations, motorized excised larynx experiments, tensile material tests and high-speed imaging, we show that voca

Development and Application of Automated Vocal Fold Tracking Software in a Rat Surgical Model.

OBJECTIVE: The rat is a widely used model for studying vocal fold (VF) function after recurrent laryngeal nerve injury, but common techniques for evaluating rat VF motion remain subjective and imprecise. To address this, we developed a software package, called RatVocalTracker1.0 (RVT1.0), to quantify VF motion and tested it on rats with iatrogenic unilateral vocal fold paralysis (VFP). METHODS: A deep neural network was trained to identify the positions of the VFs and arytenoid cartilages (ACs) in transoral laryngoscope videos of the rat glottis. Software was developed to estimate glottic mid

Non linear behavior of vocal fold vibration in an experimental model of asymmetric larynx: role of coupling between the two folds.

One of the main factors involved in the laryngeal dynamic's non-linearity is the coupling between the two vocal folds. Evaluation of the different coupling elements requires various investigations. The present work evaluate the non-linearity effects in clinical situations such as unilateral laryngeal paralysis using Lyapunov's coefficients. It also demonstrate the coupling effects on excised larynx made experimentally asymmetrical, using a specially designed device to measure vibrations from each vocal fold (opto-reflectometer).

Comparison of glottal flow rate characteristics based on experimental and computational data.

In a recent computational model, Farahani and Zhang [J. Acoust. Soc. Am. 136, EL369-EL375 (2014)] concluded that intraglottal vortices did not affect the closing mechanism of the folds. In order to determine the validity of any model that addresses the issue of vortex significance, it is important that the results of the computational model are comparable to experimental results. The results of Farahani and Zhang's model are inconsistent with data published for experimental models, which may challenge the validity of their conclusions.

Novel experimental rabbit model of anterior glottic web formation.

CONCLUSION: The rabbit model of anterior glottic web (AGW) formation using the laryngofissure technique resulted in reproducible and stable AGW formation that may facilitate research into this area. OBJECTIVE: To introduce and validate a novel experimental animal model of AGW formation using the rabbit. METHODS: The inner larynges of eight New Zealand white rabbits were exposed through the laryngofissure technique. The mucosa of the bilateral true vocal fold was stripped off using the bevel of a needle tip. On the basis of the laryngoscopic findings at 8 weeks postoperatively, the extent of

Ultrastructure of multinuclear giant cells of a human vocal cord Teflon granuloma.

The ultrastructural characteristics of multinuclear foreign-body giant cells (MCG) in a human vocal cord Teflon granuloma are described. The cells were found to contain varying numbers of Teflon particles within their cytoplasm. The particles, rounded or oval in shape, were surrounded by a rather electron-dense membrane. Numerous lysosomal structures were discerned within the cells. The fine structure of MCG in granulomas induced by foreign materials other than Teflon has already been described in man as well as in several experimental animals, and the Teflon MCG resembled these cells in many

Significance of vitamin A to brain function, behavior and learning.

Retinoid acid, the bioactive metabolite of vitamin A, is a potent signaling molecule in the brains of growing and adult animals, regulates numerous gene products, and modulates neurogenesis, neuronal survival and synaptic plasticity. Vitamin A deficiency (VAD) is a global health problem, yet our knowledge of its effects on behavior and learning is still emerging. Here we review studies that have implicated retinoids in learning and memory deficits of post-embryonic and adult rodent and songbird models. Dietary vitamin A supplementation improves learning and memory in VAD rodents and can amelio

Region-specific immunological response of the different laryngeal compartments: significance of larynx-associated lymphoid tissue.

The occurrence of primary extranodal marginal-zone B-cell lymphoma [of the mucosa-associated lymphoid tissue (MALT) type] has only been described in the supraglottic region, implying that preexistent organized lymphoid tissue is present at that site only. To date, studies have not established clearly whether organized MALT shows a site-related distribution in the larynx. The supraglottic region of the false vocal folds and the subglottis from 87 unselected cadavers with no known history of nasal, oral, pharyngeal, laryngeal, tracheal, or esophageal disease were examined for the presence of org

Animal Models of Tourette Syndrome-From Proliferation to Standardization.

Tourette syndrome (TS) is a childhood onset disorder characterized by motor and vocal tics and associated with multiple comorbid symptoms. Over the last decade, the accumulation of findings from TS patients and the emergence of new technologies have led to the development of novel animal models with high construct validity. In addition, animal models which were previously associated with other disorders were recently attributed to TS. The proliferation of TS animal models has accelerated TS research and provided a better understanding of the mechanism underlying the disorder. This newfound suc
