A Novel Ligand of Toll-like Receptor 4 From the Sheath of Wuchereria bancrofti Microfilaria Induces Proinflammatory Response in Macrophages.

BACKGROUND: Lymphatic filariasis, frequently caused from Wuchereria bancrofti infection, is endemic in several parts of the globe and responsible for human health problems and socioeconomic loss to a large extent. Inflammatory consequences originating from host-parasite interaction play a major role in the disease pathology and allied complications. The identity of the key mediator of this process is yet unknown in filarial research. METHODS: Microfilarial protein (MfP) was isolated from the sheath of W. bancrofti microfilariae through ultrafiltration, followed by chromatographic separation. …

Randomised controlled clinical trial of increased dose and frequency of albendazole and ivermectin on Wuchereria bancrofti microfilarial clearance in northern Malawi.

BACKGROUND: In Africa, albendazole and ivermectin are currently used in combination for annual mass drug administration (MDA) for lymphatic filariasis (LF) elimination. Rapid and sustained clearance is desirable for public health impact and elimination of LF. Increasing the dose and/or frequency of albendazole and ivermectin treatment may be more effective in clearing microfilariae than standard MDA. METHODS: We conducted a randomised controlled open label trial in northern Malawi comparing three modified treatment groups to standard dosage of ivermectin and albendazole in adults with confirm

Immunoepidemiology of Wuchereria bancrofti infection in two East African communities: antibodies to the microfilarial sheath and their role in regulating host microfilaraemia.

The response pattern of specific antibodies to the microfilarial sheath (sheath-Ab) of the mosquito-borne filarial parasite Wuchereria bancrofti was investigated in individuals from two East African communities with different levels of endemicity. Individuals from both communities presented a strong inverse relationship between positivity for sheath-Ab and being positive for microfilariae (mf) and circulating filarial antigens (CFA). The prevalence of sheath-Ab positivity was highest in young individuals, but peaked at a younger age in the high (1-14 years) than the low (15-19 years) endemicit…

Comparison of blood feeding response and infection of Aedes aegypti to Wuchereria bancrofti using animal membranes and direct host contact.

Comparison of an artificial, whole-blood membrane feeding procedure was performed by feeding Aedes aegypti (Liverpool strain) on the blood of patients infected with Wuchereria bancrofti microfilariae with the use of 3 types of membranes produced from chicken and mouse skin and swine intestine. Direct feeding of Ae. aegypti on the skin of infected human patients served as control. For all 3 types of membranes, mosquito survival, infection, and number of infective-stage larvae per mosquito did not differ significantly from the control. However, the blood feeding response between swine intestine …

Localisation and distribution of Wuchereria bancrofti antigens recognised by antisera from tropical pulmonary eosinophilia and from individuals with intestinal helminths.

Serological analyses of sera from patients with a typical picture of filarial tropical pulmonary eosinophilia (TPE) and sera from patients from a region non-endemic for filariasis harbouring intestinal helminths, as Ascaris lumbricoides and Strongyloids stercoralis, revealed equally high titers of IgG4, usually considered diagnostic for filariasis. Ultrathin sections of adult worms of W. bancrofti embedded in the hydrophilic resin L.R. White were incubated with sera from patients with a typical picture of filarial tropical pulmonary eosinophilia (TPE) and sera from patients of a region that wa

Development and standardization of a rapid, PCR-based method for the detection of Wuchereria bancrofti in mosquitoes, for xenomonitoring the human prevalence of bancroftian filariasis.

PCR has recently been studied as a promising tool for monitoring the progress of efforts to eliminate lymphatic filariasis. PCR can be used to test concurrently at least 30 pools, with as many as 40 mosquitoes in each pool, for the presence of filarial larvae. The SspI PCR assay for the detection of Wuchereria bancrofti DNA in pools of mosquitoes has been used since 1994 in a variety of laboratories worldwide. During that time, the original assay has been modified in these different laboratories and no standardized assay currently exists. In an effort to standardize and improve the assay, a me…

A simple deterministic model for host-parasite relationship in Wuchereria bancrofti infection & its relevance to parasite regulation in human host.

A deterministic immigration-death model, which reflects the population dynamics of W. bancrofti in human host has been applied to study the relationship between vector and human infections. Application of the model showed that the rate of acquisition and loss of human infection were approximately equal (L = 0.130 and M = 0.129). The relationship of infective resting density (IRD) in vector population with maximum intensity (Imax) of infections and microfilaria prevalence (MFP) in human population were examined by using the least squares polynomial regressions. The fifth order polynomial regres

Uptake and development of Wuchereria bancrofti in Aedes aegypti and Haitian Culex quinquefasciatus that were fed on a monkey with low-density microfilaremia.

Colonized mosquitoes of Culex quinquefasciatus (Haitian strain) and Aedes aegypti (Liverpool strain) were blood fed on a patas monkey (Erythrocebus patas) that had been experimentally infected with the Haitian strain of Wuchereria bancrofti and harbored a consistently low microfilaremia (1-3 mf per 20 mm3). Both species ingested more than twice the expected number of microfilariae (mf), i.e. 1.9 and 0.77 mf per mosquito, respectively. However, at 10-16 hours post ingestion only 4.2% of the mf had migrated from the blood meal in Cx. quinquefasciatus versus 20.7% in Ae. aegypti. Subsequently, on…

Studies on bancroftian filariasis in Liberia, West Africa. V. The influence of treatment with diethylcarbamazine and vector control on the transmission of Wuchereria bancrofti.

The transmission intensity of bancroftian filariasis was investigated during a study on filariasis control carried out in eight villages of the savanna area in the hinterland of Liberia. Although the microfilarial reservoir had been reduced considerably in the human population of two villages by mass treatment with the filaricide drug diethylcarbamazine the transmission of Wuchereria bancrofti was continued without any distinct reduction. The transmission however decreased distinctly or was even interrupted when residual spraying with DDT was used in two other villages for vector control. The …

Lymphatic Filariasis Elimination Status: Wuchereria bancrofti Infections in Human Populations after Five Effective Rounds of Mass Drug Administration in Zambia.

Lymphatic filariasis (LF), also commonly known as elephantiasis, is a neglected tropical disease (NTD) caused by filarial parasites. The disease is transmitted via a bite from infected mosquitoes. The bites of these infected mosquitoes deposit filarial parasites, Wuchereria or Brugia, whose predilection site is the lymphatic system. The damage to the lymph system causes swelling in the legs, arms, and genitalia. A mapping survey conducted between 2003 and 2011 determined LF as being endemic in Zambia in 96 out of 116 districts. Elimination of LF is known to be possible by stopping the spread o…

Identification and biochemical characterization of macrophage migration inhibitory factor-2 (MIF-2) homologue of human lymphatic filarial parasite, Wuchereria bancrofti.

Homologues of human macrophage migration inhibitory factor (hMIF) have been reported from vertebrates, invertebrates and prokaryotes, as well as plants. Filarial parasites produce two homologues of hMIF viz., MIF-1 and MIF-2, which play important role in the host immune modulation. Earlier, we have characterized MIF-1 (Wba-mif-1) from Wuchereria bancrofti, the major causal organism of human lymphatic filariasis. Here, we are reporting the molecular and biochemical characterization of MIF-2 from this parasite (Wba-mif-2). The complete Wba-mif-2 gene and its cDNA were amplified, cloned and seque…

Functional analysis of genetic polymorphism in Wuchereria bancrofti glutathione S-transferase antioxidant gene: impact on protein structure and enzyme catalysis.

Wuchereria bancrofti glutathione S-transferase (Wb-GST) is referred as a promising chemotherapeutic target for lymphatic filariasis. GST represents the major class of detoxifying enzymes of the tissue dwelling parasitic helminths. Though many inhibition studies were carried out for Wb-GST, understanding its genetic distribution in parasite population is necessary to develop ideal inhibitor. Our genetic polymorphic studies exposed the existence of three variant Wb-GST alleles in the four endemic regions of India. Moreover, it also revealed the variability in the distribution of Wb-GST alleles i…

Circulating fibrosis markers, eosinophil cationic protein and eosinophil protein X in patients with Wuchereria bancrofti infection: association with clinical status.

We measured the concentrations of several circulating fibrosis markers (type I collagen I, type III procollagen, hyaluronan) and eosinophil granule proteins (ECP and EPX) in lymphatic filariosis patients to investigate their relationship with clinical, parasitological and immunological data. This study was conducted in Polynesian patients with various stages of the disease (acute lymphangitis, chyluria, hydrocoele, elephantiasis), a closely related microbial lymphangitis and endemic controls. We observed modifications of the different markers in this pathology. Serum type I collagen and PIIINP

Molecular characterization of a calcium binding translationally controlled tumor protein homologue from the filarial parasites Brugia malayi and Wuchereria bancrofti.

We have cloned homologues of the mammalian translationally controlled tumor protein (TCTP) from the human filarial parasites Wuchereria bancrofti and Brugia malayi. TCTP genes from B. malayi and W. bancrofti were expressed in a T7 promoter vector as histidine tagged fusion proteins. Both the recombinant B. malayi TCTP (rBm-TCTP) and recombinant W. bancrofti TCTP (rWb-TCTP) have a molecular mass of approximately 28 kDa with the histidine tag. Sequence analyses showed that there is a 98% similarity between the two filarial TCTPs at amino acid levels and are immunologically cross-reactive. Analys…

Specificity of surface molecules of adult Brugia parasites: cross-reactivity with antibody from Wuchereria, Onchocerca and other human filarial infections.

The extent of structural and immunological similarity between surface antigens of three species of Brugia filarial parasites was studied by lodogen-mediated surface labelling of adult worms of B. malayi, B. pahangi and B. timori. The close homology and cross-reactivity between these antigens reported in previous surface labelling studies with Bolton-Hunter reagent, was verified in this system. The surface antigens of adult B. pahangi are also recognised by antibody from patients with Wuchereria bancrofti, the major human lymphatic filariae, and by antibody to Loa loa and Mansonella perstans. Immediate and delayed hypersensitivity skin test responses to the Dirofilaria immitis filarial skin test (Sawada) antigen in Wuchereria bancrofti filariasis.

In order to assess the immunodiagnostic utility of FST skin test antigen purified from Dirofilaria immitis, skin test responses were analyzed in 177 inhabitants of a South Pacific island hyperendemic for Wuchereria bancrofti filariasis. Although the distribution of immediate hypersensitivity skin test responses was similar to those found in other filarial endemic populations, individual immediate skin test reactions lacked the sensitivity and specificity to identify those with microfilaremia or those with any clinical or parasitologic evidence of filarial disease. Delayed hypersensitivity type…

Development of a specimen-sparing multichannel bead assay to detect antiparasite IgG4 for the diagnosis of Schistosoma and Wuchereria infections on the coast of Kenya.

To better delineate the impact of parasitic coinfection in coastal Kenya, we developed a novel specimen-sparing bead assay using multiplex flow immunoassay (MFI) technology to simultaneously measure serum or plasma immunoglobulin G4 (IgG4) against Brugia malayi antigen (BMA) and Schistosoma haematobium soluble worm antigen (SWAP). Properties of the bead assay were estimated by latent class analysis using data from S. haematobium egg counts/filarial rapid diagnostic cards (RDTs), parasite-specific enzyme-linked immunosorbent assays (ELISAs), and the multichannel IgG4 assay. For schistosomiasis,

Effects of midgut bacteria and two protease inhibitors on the reproductive potential and midgut enzymes of Culex pipiens infected with Wuchereria bancrofti.

Laboratory investigations were carried out to asses the effect of some proteases inhibitors on the reproductive potential of Culex pipiens females resulted from larvae treated with different protease inhibitors. The fecundity and engorgement of symbiotic and aposymbiotic C. pipiens females were significantly reduced. The blood meal digestion period increased significantly. On the other hand, enzyme band with molecular weight of 40 KDa which may be cysteine protease was detected in untreated symbiotic and aposymbiotic female midguts. The results may explain that the absence of this enzyme bands
