Cyclic nucleotide phosphodiesterase activity in the quail oviduct: hormonal regulation and involvement in estrogen-induced growth.

A previous study has shown that cAMP was involved in estrogen-activated growth in the quail oviduct. The present study was undertaken to investigate the hormonal regulation of 3',5'-cyclic nucleotide phosphodiesterase activity in the oviduct. Tamoxifen, an antiestrogen compound, and 3-isobutyl-1-methylxanthine, an inhibitor of phosphodiesterase, were also used to determine the relationship between estradiol-induced cell proliferation and cAMP phosphodiesterase activity. Cyclic nucleotide phosphodiesterase was almost completely restricted to the cytosolic fraction (108,000 X g supernatant) of t

Human hematoxylin-stainable protein of keratohyalin granules origin. I. Extraction and purification.

A human hematoxylin-stainable protein (HSP) was extracted from the massive stratum corneum of epidermal cysts. This protein was purified in two steps; first, through preparative isoelectric focusing, and, second, by affinity column chromatography bound with the specific, monoclonal antibody to keratohyalin granules (Ted-H-1). In sodium dodecyl sulfate-polyacrylamide gel electrophoresis, the purified HSP consisted of two proteins (70 and 62 kilodaltons [kd]), but only the 62-kd protein was detected in the 15,000 g supernatant fraction of the extract using the immunoblotting technique. The amino

Comparative studies on insulin-like growth factor II and insulin processing by vascular endothelial cells.

Processing of circulating polypeptide hormones by vascular endothelial cells may be critical to hormone transport from the vascular lumen to tissue sites of action. The comparative processing of insulin-like growth factor II (IGF II) and insulin by cultured bovine aortic endothelial cells was examined. These cells possess high-affinity receptors for IGF II and insulin, as shown by competitive-binding studies. At 37 degrees C, internalization determined by both resistance to an acid wash and electron microscopy was rapid with 50-70% of bound IGF II and insulin internalized at 60 min. Subsequent

A possible mechanism of induction and translocation into blood stream of rat alkaline phosphatase activity by bile duct ligation.

We investigated the effects of bile duct ligation on alkaline phosphatase (ALP) activities in liver, calvarium, duodenum, and ileum in rats and its possible mechanism of action. ALP isozyme activities in the ligated rats were significantly elevated in the liver and duodenum, while those in the ileum and calvarium were markedly decreased. The ALP isozyme activity elevated by the ligation was obviously suppressed by prior administration of indomethacin, an inhibitor of prostaglandin synthesis. Moreover, phorbol ester also elevated the ALP activity as well as the phosphatase level in the ligated

Measurement of individual aflatoxin exposure among people having different risk to primary hepatocellular carcinoma.

The establishment of the carcinogenic role of aflatoxins has been impeded by the lack of suitable tests to measure individual exposure for long-term studies. It was shown that the use of immuno-concentration followed by high pressure liquid chromatography (HPLC) or immunoassay can regularly detect aflatoxins down to pg/ml in fluids including urine. Aflatoxin M1 (AFM1) in free form was identified as the major metabolite in urine suitable for use as an approximate dosimetric indicator of recent exposure to aflatoxin B1 (AFB1). A panel of monoclonal antibodies of IgG class were developed in the m

Structural similarities in the kappa light chains of human rheumatoid factor paraproteins and serum immunoglobulins bearing a cross-reactive idiotype.

The monoclonal antibody (MoAb) 17.109 recognizes a cross-reactive idiotype (CRI) associated with the light chains of Waldenstrom's macroglobulins with rheumatoid factor (RF) activity. The MoAb also reacts with a proportion of IgM-RF molecules from the sera of rheumatoid arthritis and primary Sjogren's syndrome patients, and from the sera of seropositive normal human subjects. In the present experiments, we used affinity chromatography to purify the 17.109 CRI-positive immunoglobulin from serum and have analyzed the isolated material by Western blotting. The purified 17.109 CRI-positive materia

Immunofluorescence study on the organization of actin in astroglial cells in primary cultures.

Actin antibodies were purified by affinity chromatography from the serum of rabbits immunized with actin isolated from bovine skeletal myofibrils. By the indirect immunofluorescence technique the pattern of organization of actin was studied in cultured astroglial cells prepared from cerebral hemispheres of newborn rats. Labelling with monospecifc actin antibody revealed that the flat epitheloid astrocytes contained bundles of actin fibres arranged in characteristic patterns. Actin fibres disaggregated in round cells produced by trypsinization and reorganized when cells returned to their elonga

Biosynthetic labeling of insulin receptor: studies of subunits in cultured human IM-9 lymphocytes.

We have identified the subunits of the insulin receptor in cultured human lymphocytes (IM-9 line) by biosynthetic labeling with [35S]methionine and specific precipitation with autoantibodies against the insulin receptor. IM-9 lymphocytes were cultured with [35S]methionine and extracted with Triton X-100. Insulin receptors were concentrated and purified 20-fold by chromatography of the cell extract on wheat germ agglutinin-agarose, and then specifically precipitated by receptor antibodies after addition of a second antibody. Analysis of the immunoprecipitates by sodium dodecyl sulfate/polyacryl

Identification and validation nucleolin as a target of curcumol in nasopharyngeal carcinoma cells.

UNLABELLED: Identification of the specific protein target(s) of a drug is a critical step in unraveling its mechanisms of action (MOA) in many natural products. Curcumol, isolated from well known Chinese medicinal plant Curcuma zedoary, has been shown to possess multiple biological activities. It can inhibit nasopharyngeal carcinoma (NPC) proliferation and induce apoptosis, but its target protein(s) in NPC cells remains unclear. In this study, we employed a mass spectrometry-based chemical proteomics approach reveal the possible protein targets of curcumol in NPC cells. Cellular thermal shift

Evidence that partially desialylated variants of human chorionic gonadotropin (hCG) are the factors in crude hCG that inhibit the response to thyrotropin in human thyroid membranes.

Previous studies have revealed that pure unmodified hCG (hCGp) has little potency to inhibit the binding of bovine TSH (bTSH) to human thyroid membranes and to either stimulate adenylate cyclase or inhibit TSH-stimulated adenylate cyclase therein. On the other hand, preparations of crude hCG (hCGc) as well as enzymatically desialylated hCGp (asialo-hCGp) are relatively potent inhibitors of bTSH binding (TBI activity) and bTSH-stimulated adenylate cyclase activity in human thyroid membrane preparations. In the present studies we have sought to identify and characterize the inhibitory moieties i

The acyl-CoA dehydrogenation deficiencies. Recent advances in the enzymic characterization and understanding of the metabolic and pathophysiological disturbances in patients with acyl-CoA dehydrogenation deficiencies.

Acyl-CoA dehydrogenation deficiencies are defined as disorders of the metabolism of branched chain and straight chain acyl-CoA esters and of glutaryl-CoA. The acyl-CoA dehydrogenation process is comprised of three enzymes, i.e. acyl-CoA dehydrogenase (isovaleryl-CoA, isobutyryl-CoA/2-Me-butyryl-CoA, short-chain acyl-CoA, general (medium-chain) acyl-CoA, long-chain acyl-CoA or glutaryl-CoA), electron transfer flavoprotein (ETF) and electron transfer flavoprotein dehydrogenase (ETF DH). Patients with isovaleryl-CoA dehydrogenase deficiency, glutaryl-CoA dehydrogenase deficiency and general (medi

Molecular features of the serological IgG repertoire elicited by egg-based, cell-based, or recombinant haemagglutinin-based seasonal influenza vaccines: a comparative, prospective, observational cohort study.

BACKGROUND: Egg-based inactivated quadrivalent seasonal influenza vaccine (eIIV4), cell culture-based inactivated quadrivalent seasonal influenza vaccine (ccIIV4), and recombinant haemagglutinin (HA)-based quadrivalent seasonal influenza vaccine (RIV4) have been licensed for use in the USA. In this study, we used antigen-specific serum proteomics analysis to assess how the molecular composition and qualities of the serological antibody repertoires differ after seasonal influenza immunisation by each of the three vaccines and how different vaccination platforms affect the HA binding affinity an

Mineralocorticosteroid receptor of the chick intestine. Oligomeric structure and transformation.

The binding of [3H]aldosterone in the chick intestine cytosol was analyzed in terms of affinity and specificity. In this tissue, aldosterone binds to the mineralocorticosteroid receptor, with a high affinity (Kd approximately 0.3 nM) and low capacity (approximately 50 fmol/mg protein), and to the glucocorticosteroid receptor. The selective labeling of the mineralocorticosteroid receptor was achieved by incubating the cytosol with [3H]aldosterone in the presence of RU 486. This synthetic steroid completely inhibited the binding of [3H]aldosterone to the glucocorticosteroid receptor and did not

[A case of leprechaunism with disorders of insulin-like growth factor-I(IGF-I)/somatomedin C(SMC) binding protein and its receptor].

Leprechaunism is a rare genetic disorder characterized by physical abnormalities, intrauterine and postnatal growth retardation, poorly developed subcutaneous fat and muscle at birth, and early death. This patient, who was a 1.5 year-old female with typical clinical features of leprechaunism, had relatively high levels of plasma GH and IGF-I/SMC but no glucose intolerance or insulin resistance. Studies were undertaken to elucidate (1) the differences among some kinds of methods for IGF-I/SMC measurement, (2) the distribution patterns of IGF-I/SMC between two kinds of its binding protein (SMBP)

Human salivary extracellular vesicles: a comparative study of isolation techniques.

Saliva offers a valuable source of analyte for extracellular vesicle (EV) -based biomarker research, however, due to the complex composition of saliva, the isolation of salivary EVs remains technically challenging. The aim of our study is to systematically compare three salivary EV isolation techniques by assessing the obtained EV sample quality and quantity, and to highlight differences between the three methods with respect to costs, isolation time, and potential for parallel sample handling (i.e. sample throughput). For the enrichment of EVs we used three EV isolation techniques with saliva

Structure-based design and biophysical characterization of the linker sequence that associate to the VH-VL domain interface of the single-chain Fv antibody to suppress oligomerization propensity.

A single-chain antibody is an artificial small size antibody fragment whose variable regions are connected by a flexible short peptide linker. The scFv antibody is considered an attractive molecule for constructing advanced therapeutic antibodies, such as CAR-T cells and multi-specific antibodies. However, the application of scFv antibodies in advanced drugs is limited by their propensity to oligomerize. Due to weak interactions between the VH and VL domains, scFvs exist in an equilibrium between open and closed states, leading to inter-chain VH-VL interactions that form dimers, trimers, and a

Surrogate-matrix calibration for quantitative GC-IMS headspace analysis of urine.

Quantitative analysis of volatile organic compounds (VOCs) in biological matrices remains challenging due to matrix-dependent headspace partitioning, the presence of endogenous analytes, and the intrinsically non-linear response of certain detectors. In this context, static headspace gas chromatography-ion mobility spectrometry (HS-GC-IMS) offers high sensitivity for VOC profiling, but these combined effects complicate calibration and quantitative interpretation. In this work, a practical framework for quantitative VOC analysis in urine using HS-GC-IMS is presented, in which calibration is per
