DNA variants affecting chromatin structure are key to the genetic architecture of obsessive compulsive disorder.

BACKGROUND: Obsessive Compulsive Disorder (OCD) affects 2 to 3% of the population and is marked by intrusive thoughts and repetitive behaviours. OCD is increasingly recognized as a polygenic disorder involving gene-environment interactions, with genetic risk largely driven by common variants and a smaller contribution from rare pathogenic variation. Despite these insights, the functional roles of implicated genes remain unclear. Recent genomic studies suggest that chromatin dysregulation contributes to the pathogenesis of OCD. RESULTS: We analysed the recently published large-scale genome-wid

Nuclear proteome analysis suggests spatial and temporal compartmentalization of metabolic enzymes during Trypanosoma cruzi cell cycle.

Trypanosoma cruzi, the etiological agent of Chagas Disease (CD), represents a significant public health concern and serves as a valuable model for investigating the cell cycle in early-diverging eukaryotes. Its unique cellular features, including the absence of chromosome condensation and the coordination of nuclear division with specialized organelles, provide insights into non-canonical regulatory mechanisms, and underscore the pivotal role of the nucleus in maintaining genomic integrity and regulating fundamental processes such as DNA replication and gene expression. To investigate nuclear d

Retinoic Acid Receptor-related Orphan Receptor α Drives Glucose Reprogramming and Mitochondrial Rescue Mitigate Subarachnoid Hemorrhage-Induced Early Brain Injury.

AIMS: Subarachnoid hemorrhage (SAH) is a devastating cerebrovascular event characterized by early brain injury (EBI) within 72 h that is driven by oxidative stress, mitochondrial dysfunction, and metabolic collapse. The retinoic acid receptor-related orphan receptor alpha (RORα) is a nuclear receptor implicated in metabolic and inflammatory regulation, but it has not been studied in SAH. We aimed to determine whether RORα confers neuroprotection after SAH and to elucidate its underlying mechanisms. METHODS AND RESULTS: We used mouse SAH models and primary cortical neurons to assess the RORα e

DNA Methylation and Fetal Programming of Cardiovascular Disease: From Congenital Heart Diseases to Adult Cardiovascular Dysfunction.

DNA methylation, the most extensively studied epigenetic mechanism, acts as a critical interface between maternal environmental influences and fetal cardiovascular development. During embryogenesis, tightly orchestrated methylation remodeling regulates the transcriptional networks required for cardiogenesis, including chamber septation, valve formation, and myocardial maturation. Disruption of these methylation patterns contributes to congenital heart disease (CHD), with distinct methylation signatures identified in tetralogy of Fallot, double-outlet right ventricle, bicuspid aortic valve, and

Global epigenetic immunohistochemical markers reveal developmental connections of distinct cell types during fetal development.

PURPOSE: The prenatal development of the human retina is a complex process orchestrated by several epigenetic mechanisms. This study investigated some global epigenetic markers (histone H3, histone H4, 5-methylcytosine, 5-hydroxymethylcytosine) during the fetal development of the human retina in 34 human eyes from 11 to 38 weeks of gestation. METHODS: Immunohistochemical staining was performed to assess the distribution and intensity of the selected epigenetic markers across developmental stages. For the general retinal staining intensity, a score from 0 to 3 was used, whereas for the percent

Peripheral Blood DNA Methylation Changes Precede Lymphoma Diagnosis in Primary Sjögren's Disease.

OBJECTIVE: Primary Sjögren's disease (SjD) is a systemic autoimmune disease associated with an increased risk of lymphoma. The molecular mechanisms underlying lymphomagenesis remain poorly understood, and sensitive biomarkers for early identification of patients at high risk of developing lymphoma are lacking. This study investigated whether DNA methylation patterns in blood are associated with subsequent development of lymphoma in patients with SjD. METHODS: Peripheral blood DNA samples collected from patients with SjD between 1998 and 2013 were analyzed using the Illumina HumanMethylation45

Neuron-specific WDR5 epigenetically upregulates ARID5B to impair GABAergic synaptic transmission and promotes epileptogenesis.

Rationale: Temporal lobe epilepsy (TLE) is a prevalent, drug-resistant neurological disorder that causes severe disability, highlighting the need to identify novel therapeutic targets. Emerging evidence reveals widespread transcriptional dysregulation during epileptogenesis, in which multiple dysregulated genes functionally contribute to disease progression. However, the epigenetic basis of these transcriptomic changes remains poorly characterized. Methods: We established experimental epilepsy models and systematically investigated the remodeling of major histone methylation modifications duri

Epigenetic Regulation of Uterine Smooth Muscle Tumors: Histone Modifications in Uterine Fibroids and Leiomyosarcoma.

Uterine smooth muscle tumors (USMTs) represent a diverse group of neoplasms arising from the myometrium, ranging from benign uterine fibroids (leiomyomas) to highly aggressive uterine leiomyosarcoma. While genetic alterations contribute to tumor development, growing evidence highlights the crucial role of epigenetic regulation in shaping tumor behavior. Among these mechanisms, histone modification has emerged as a key regulator of chromatin structure and gene expression. Histone modifications, including acetylation, methylation, phosphorylation, ubiquitination, ADP-ribosylation, and SUMOylatio

Ovarian H3K27ac remodeling is associated with impaired follicular development in laying hens with fatty liver hemorrhagic syndrome.

Fatty liver hemorrhagic syndrome (FLHS) is a metabolic disease of laying hens that reduces egg production and is accompanied by reproductive impairment, but the ovarian regulatory mechanisms that connect nutritional stress to follicular dysfunction remain unclear. This study examined whether active chromatin remodeling in the ovary is associated with FLHS induced by a high-energy, low-protein (HELP) diet. Hy-Line Brown hens were assigned to a basal diet or HELP diet, and ovarian tissue was collected for histone H3 lysine 27 acetylation (H3K27ac) chromatin immunoprecipitation sequencing and RNA

Epigenetics, bacterial metabolites and type 2 diabetes: a new view of gut-genome interactions.

Type 2 diabetes (T2D) is traditionally viewed as a disorder of glucose homeostasis, yet growing evidence suggests that its pathogenesis is deeply influenced by interactions between the gut microbiota and host gene regulation. Bacterial metabolites have emerged as key molecular mediators capable of linking environmental exposures, metabolism and epigenetic control of gene expression. These insights are reshaping current understanding of how microbial signals contribute to metabolic disease. This review examines the emerging role of bacterial metabolites as modulators of host epigenetic landscap

Epigenetic Plasticity in Triple-Negative Breast Cancer: Mechanisms of Therapy Resistance, Biomarkers, and Therapeutic Vulnerabilities.

Triple-negative breast cancer (TNBC) is an aggressive and clinically heterogeneous breast cancer subtype characterized by the absence of estrogen receptor, progesterone receptor, and HER2 overexpression, limited targeted treatment options, early relapse, and frequent development of therapy resistance. Although TNBC often shows initial sensitivity to chemotherapy, durable responses are commonly undermined by the emergence of adaptive resistant cell states rather than solely by fixed genetic mutations. This review synthesizes the role of epigenetic plasticity as a central mechanism that enables

Characterization of Transcriptional, Epigenetic, and Phenotypic Plasticity and Discovery of Biomarkers in Acute and Chronic Murine Schistosomiasis Infection.

Parasites can induce changes in their hosts, favoring the success of the infection and its development at each stage of their life cycle. The host minimizes the effects of the parasite's presence through its defense system, balancing the parasite-host relationship. The intricate parasite-host relationship provides physiological, immunological, and molecular cues that suggest interaction and mutual regulation of the transcriptome and epigenome, promoting phenotypic plasticity and survival in a changing environment. There has been a growing interest in the epigenetic mechanisms of Schistosoma ma

The Tumor Multi-Omic Landscape of Endometrial Cancers Developed on a Background of Adiposity.

Background: High body mass index (BMI) is a causal risk factor for endometrial cancer, but the tumor molecular mechanisms affected by adiposity remain poorly understood. Here, we characterize the tumor multi-omic landscape of endometrial cancers that have developed on a background of lifelong germline genetic liability to elevated BMI. Methods: We built a polygenic score (PGS) for BMI in women using data on independent, genome-wide significant variants associated with adult BMI in 434,794 women. We performed germline (blood) genotype quality control and imputation on data from 354 endometrial

DNA methylation predicts lung function and pulmonary exacerbation in sputum samples from patients with cystic fibrosis.

BACKGROUND: Lung disease is variable among patients with cystic fibrosis (CF) and depends on genetic and environmental factors. To better understand the molecular determinants of lung disease variability, we carried out an epigenome-wide association study (EWAS) in sputum samples from patients with CF. METHODS: We profiled 64 sputum samples using Human Methylation EPIC BeadChips and assessed the correlation between DNA methylation levels and four clinical traits: lung function (FEV1pp), lung function variation (FEV1pp slope), presence and number of pulmonary exacerbations. Sputum samples were
