Addition of cariogenic pathogens to complex oral microflora drives significant changes in biofilm compositions and functionalities.

BACKGROUND: Dental caries is a microbe and sugar-mediated biofilm-dependent oral disease. Of particular significance, a virulent type of dental caries, known as severe early childhood caries (S-ECC), is characterized by the synergistic polymicrobial interaction between the cariogenic bacterium, Streptococcus mutans, and an opportunistic fungal pathogen, Candida albicans. Although cross-sectional studies reveal their important roles in caries development, these exhibit limitations in determining the significance of these microbial interactions in the pathogenesis of the disease. Thus, it remain

Demonstration of N,N-Dimethyldithiocarbamate as a Copper-Dependent Antibiotic against Multiple Upper Respiratory Tract Pathogens.

Transition metals are necessary cofactors and structural elements in living systems. Exposure to high concentrations of biologically important transition metals, such as zinc and copper, results in cell toxicity. At the infection site, the immune system deploys metal sorbent proteins (e.g., lactoferrin and calprotectin) to starve pathogens of necessary metals (such as iron), while phagocytes expose engulfed pathogens to high levels of other metals, such as copper and zinc. The opportunistic pathogen Streptococcus pneumoniae (the pneumococcus) encounters macrophages during initial and protracte

Antifungal Activity against Filamentous Fungi of Ts1, a Multifunctional Toxin from Tityus serrulatus Scorpion Venom.

Antimicrobial peptides (AMPs) are ubiquitous and multipotent components of the innate immune defense arsenal used by both prokaryotic and eukaryotic organisms. The search for new AMPs has increased in recent years, due to the growing development of microbial resistance to therapeutical drugs. In this work, we evaluate the effects of Tityus serrulatus venom (Tsv), its fractions and its major toxin Ts1, a beta-neurotoxin, on fungi growth. The fractions were obtained by ion-exchange chromatography of Tsv. The growth inhibition of 11 pathogenic and non-pathogenic filamentous fungi (Aspergillus fum

Cholesterol biosynthesis from lanosterol. Molecular cloning, tissue distribution, expression, chromosomal localization, and regulation of rat 7-dehydrocholesterol reductase, a Smith-Lemli-Opitz syndrome-related protein.

The cDNA encoding the 471-amino acid rat 7-dehydrocholesterol reductase (DHCR), an enzyme that has been implicated in both cholesterol biosynthesis and developmental abnormalities (e.g. Smith-Lemli-Opitz syndrome) in mammals, has been cloned and sequenced, and the primary structure of the enzyme has been deduced. The DHCR gene was mapped to chromosome 8q2.1 by fluorescence in situ hybridization. Rat DHCR, calculated molecular mass of 54.15-kDa polypeptide, shares a close amino acid identity with mouse and human DHCRs (96 and 87%, respectively) as compared with its other related proteins (e.g.

Case Report: a hypermetabolic mediastinal mass (SUVmax 16.7) mimicking malignancy with spontaneous resolution: an inflammatory myofibroblastic proliferation and diagnostic pitfall.

BACKGROUND: Hypermetabolic mediastinal masses on 2-deoxy-2-[18F]fluoro-D-glucose ([18F]FDG) positron emission tomography/computed tomography (PET/CT) are presumed malignant until proven otherwise. However, benign inflammatory and organizing processes may exhibit equally intense metabolic activity, posing a diagnostic challenge-particularly when the lesion abuts major vascular structures and limits safe tissue sampling. CASE SUMMARY: A 55-year-old male smoker presented with cough, right-sided chest pain, low-grade fever, and mild dysphagia. Contrast-enhanced CT identified an ill-defined mass i

Biochar-earthworm-plant interactions regulate soil organic carbon stabilization and Cd immobilization in vegetable and corn cropping system.

A dual strategy for carbon (C) sequestration and Cd remediation was developed by leveraging the synergistic effects of biochar and earthworms on soil organic C transformation and Cd bioavailability. However, the combined capacity of these amendments to simultaneously enhance soil organic C storage and reduce crop Cd uptake, particularly through the regulation of dynamic organic C pools, remains insufficiently understood. A field experiment was conducted to evaluate the individual and combined effects of biochar (600 g plot⁻¹) and earthworms (Amynthas aspergillum; 50 worms plot⁻¹) on soil organ

Antibiofilm efficacy of 1,064-nm neodymium-doped yttrium aluminum garnet laser on Trichophyton rubrum and Candida albicans: in vitro study.

Fungal biofilms are the most important virulence factors of onychomycosis. However, traditional antifungal therapies lack antibiofilm efficacy. Although neodymium-doped yttrium aluminum garnet (Nd:YAG) laser is one of the most common laser therapies used for onychomycosis, its usage is restricted to aesthetic endpoints. Therefore, this study aimed to assess the antibiofilm activity of Nd:YAG against Trichophyton rubrum (NCCP 22456) and Candida albicans (NCCP 32703) biofilms. Biofilms were subjected to Nd:YAG (fluence 200 mJ/cm2, pulse duration, 0.1 ms; spot size, 1.5 mm; frequency, 30 Hz) for

The Cdc42 and Rac1 GTPases are required for capillary lumen formation in three-dimensional extracellular matrices.

Here we show a requirement for the Cdc42 and Rac1 GTPases in endothelial cell (EC) morphogenesis in three-dimensional extracellular matrices. Cdc42 and Rac1 specifically regulate EC intracellular vacuole and lumen formation in both collagen and fibrin matrices. Clostridium difficile toxin B (which blocks all three Rho GTPases) completely inhibited the ability of ECs to form both vacuoles and lumens, whereas C3 transferase, a selective inhibitor of Rho, did not. Expression of either dominant-negative (N17) or constitutively active (V12) Cdc42 using recombinant adenoviruses dramatically inhibite

A combined experimental and computational strategy to define protein interaction networks for peptide recognition modules.

Peptide recognition modules mediate many protein-protein interactions critical for the assembly of macromolecular complexes. Complete genome sequences have revealed thousands of these domains, requiring improved methods for identifying their physiologically relevant binding partners. We have developed a strategy combining computational prediction of interactions from phage-display ligand consensus sequences with large-scale two-hybrid physical interaction tests. Application to yeast SH3 domains generated a phage-display network containing 394 interactions among 206 proteins and a two-hybrid ne

A novel role for protein kinase Kin2 in regulating HAC1 mRNA translocation, splicing, and translation.

A signaling network called the unfolded protein response (UPR) resolves the protein-folding defects in the endoplasmic reticulum (ER) from yeasts to humans. In the yeast Saccharomyces cerevisiae, the UPR activation involves (i) aggregation of the ER-resident kinase/RNase Ire1 to form an Ire1 focus, (ii) targeting HAC1 pre-mRNA toward the Ire1 focus that cleaves out an inhibitory intron from the mRNA, and (iii) translation of Hac1 protein from the spliced mRNA. Targeting HAC1 mRNA to the Ire1 focus requires a cis-acting bipartite element (3'BE) located at the 3' untranslated leader. Here, we re

Amino- and carboxy-terminal domains of the yeast Rab escort protein are both required for binding of Ypt small G proteins.

The Rab escort protein (REP) is an essential component of the heterotrimeric enzyme Rab geranylgeranyl transferase that modifies the carboxy-terminal cysteines of the Ras-like small G proteins belonging to the Rab/Ypt family. Deletions in the human CHM locus, encoding one of the two REPs known in humans, result in a retinal degenerative syndrome called choroideremia. The only known yeast homologue of the choroideremia gene product is encoded by an essential gene called MRS6. Besides three structurally conserved regions (SCRs) previously detected in the amino-terminal half of REPs and RabGDIs,

In vitro and in vivo activity of a novel antifungal small molecule against Candida infections.

Candida is the most common fungal pathogen of humans worldwide and has become a major clinical problem because of the growing number of immunocompromised patients, who are susceptible to infection. Moreover, the number of available antifungals is limited, and antifungal-resistant Candida strains are emerging. New and effective antifungals are therefore urgently needed. Here, we discovered a small molecule with activity against Candida spp. both in vitro and in vivo. We screened a library of 50,240 small molecules for inhibitors of yeast-to-hypha transition, a major virulence attribute of Candi

Ferrichrome: bacterial function, synthetic mimics, and biomedical applications.

Iron is an essential micronutrient for most living organisms, but its bioavailability is limited. To facilitate iron uptake, many bacteria and fungi secrete siderophores, which are small, high-affinity iron-chelating molecules. Siderophores play a key role in microbial competition and immune evasion, and additionally serve as an entry point for bacteriophages and toxins. Ferrichrome, a well-studied fungal hydroxamate siderophore, is utilized as an iron source by numerous bacterial species, including human-associated and pathogenic bacteria. Ferrichrome is a cyclic hexapeptide composed of three

From traditional remedy to in vitro evidence: phytochemical composition and skin-related pharmacological activity of birch witches' broom (Taphrina betulina).

BACKGROUND: Birch witches' broom is an abnormal plant structure caused by infection of birch trees with the dimorphic fungus Taphrina betulina Rostr., (1883). Although classified as a disease, witches' broom has traditionally been used in folk medicine to treat skin disorders. This suggests that fungal infection may induce metabolic reprogramming in host tissues, leading to the production and accumulation of biologically active secondary metabolites. We hypothesized that infection-driven metabolic changes in witches' broom tissue could generate compounds capable of modulating keratinocyte viab

Candidozyma auris (formerly Candida auris): resistant, long lasting, and everywhere.

BACKGROUND: Invasive fungal diseases represent a significant global health concern, with Candidozyma auris (formerly Candida auris) emerging as a major healthcare-associated pathogen. Its multidrug resistance, environmental persistence, prolonged skin colonization, and efficient nosocomial transmission have driven sustained outbreaks and endemicity worldwide, and recent taxonomic changes have further complicated surveillance and diagnostics. OBJECTIVES: This narrative review summarizes current evidence on the taxonomy, epidemiology, clinical impact, antifungal resistance, transmission, and in

Temperature and genetic background drive mobilization of diverse transposable elements in a global human fungal pathogen.

Transposable elements (TEs) are key agents of genome evolution across all domains of life. These mobile genetic elements can cause mutations through transposition or by promoting structural rearrangements. Stress conditions can amplify TE mobility, either by impairing TE suppression mechanisms or through stress-induced interactions between transcription factors and TE sequences, offering a route for rapid genetic change. As such, TEs represent an important source of adaptability within populations. To investigate the interplay between environmental stress and eukaryotic TE dynamics relevant to

Invasive aspergillosis in autoimmune inflammatory rheumatic diseases: epidemiology, risk factors, diagnosis, management and challenges.

BACKGROUND: Invasive aspergillosis (IA) is a life-threatening opportunistic fungal infection caused by Aspergillus species. In recent years, IA appears to have become more frequently reported among patients with autoimmune inflammatory rheumatic diseases (AIIRD), likely reflecting the broader use of immunosuppressive therapies, with incidence in high-risk AIIRD subgroups reported to reach approximately 6.7% in selected cohorts. OBJECTIVE: This review aims to summarize the current evidence on the epidemiology, susceptibility mechanisms, risk factors, clinical presentation, diagnosis, and manag

Role of nanoclusters in the design and development of novel strategies for diagnosis and treatment of infectious diseases.

Despite major advances in sanitation, vaccination, and antimicrobial chemotherapy, infectious diseases continue to pose a huge threat to global health. The ever-increasing occurrences of antimicrobial resistance, emerging pathogens, and persistent diagnostic delays further exacerbate this situation. Over the years, nanotechnology has generated a lot of material platforms to address these failures, and in this study, we critically examine nanoclusters-ultrasmall, atomically precise assemblies (<2 nm cores) with discrete electronic states, as an under-recognized but potentially transformative cl
