Vaginal neutrophils eliminate sperm by trogocytosis.

STUDY QUESTION: What is the vaginal polymorphonuclear (PMN) spermicidal mechanism to reduce the excess of sperm? SUMMARY ANSWER: We show that PMNs are very efficient at killing sperm by a trogocytosis-dependent spermicidal activity independent of neutrophil extracellular traps (NETs). WHAT IS KNOWN ALREADY: Trogocytosis has been described as an active membrane exchange between immune cells with a regulatory purpose. Recently, trogocytosis has been reported as a mechanism which PMNs use to kill tumour cells or Trichomonas vaginalis. STUDY DESIGN, SIZE, DURATION: We used in vivo murine models…

Intermittent Hypoxia-Hyperoxia Training Improves Cognitive Function and Decreases Circulating Biomarkers of Alzheimer's Disease in Patients with Mild Cognitive Impairment: A Pilot Study.

Источник пометил публикацию как отозванную

Alzheimer's disease (AD) affects not only the central nervous system, but also peripheral blood cells including neutrophils and platelets, which actively participate in pathogenesis of AD through a vicious cycle between platelets aggregation and production of excessive amyloid beta (Aβ). Platelets adhesion on amyloid plaques also increases the risk of cerebral microcirculation disorders. Moreover, activated platelets release soluble adhesion molecules that cause migration, adhesion/activation of neutrophils and formation of neutrophil extracellular traps (NETs), which may damage blood brain ba…

Advances and Challenges in 3D Bioprinting of Cartilage Organoids: From Material Innovation to Functional Regeneration.

Articular cartilage injury and degenerative disorders are major contributors to joint dysfunction. Because cartilage is avascular and aneural, its intrinsic healing capacity is extremely limited, which continues to present a major clinical challenge. Cartilage organoids, defined as three-dimensional constructs that recapitulate key structural, cellular, and functional features of native cartilage tissue, including cell-cell and cell-matrix interactions, zonal organization, and responsiveness to biochemical and mechanical cues, have emerged as promising platforms for cartilage regeneration and

Combined Insults of a MASH Diet and Alcohol Binges Activate Intercellular Communication and Neutrophil Recruitment via the NLRP3-IL-1β Axis in the Liver.

Binge drinking in obese patients positively correlates with accelerated liver damage and liver-related death. However, the underlying mechanism and the effect of alcohol use on the progression of metabolic-dysfunction-associated steatotic liver disease (MASLD) remain unexplored. Here, we show that short-term feeding of a metabolic-dysfunction-associated steatohepatitis (MASH) diet plus daily acute alcohol binges for three days induce liver injury and activation of the NLRP3 inflammasome. We identify that a MASH diet plus acute alcohol binges promote liver inflammation via increased infiltratio

The role of TLR9 on Leishmania amazonensis infection and its influence on intranasal LaAg vaccine efficacy.

Leishmania (L.) amazonensis is one of the etiological agents of cutaneous leishmaniasis (CL) in Brazil. Currently, there is no vaccine appro … ction. In addition, neutrophils from WT and TLR9-/- mice had similar capacity to produce neutrophil extracellular traps (NETs) in response to L. amazonensis. L. amazonensis did not activate dendritic cells from WT and TLR9-/- mice, analysed by MHCII and CD86 expression. However, in vivo, TLR9-/- mice were slightly more susceptible to L. amazonensis infection than WT …

Oxygen consumption rates and oxygen concentration in molt-4 cells and their mtDNA depleted (rho0) mutants.

Respiratory deficient cell lines are being increasingly used to elucidate the role of mitochondria and to understand the pathophysiology of mitochondrial genetic disease. We have investigated the oxygen consumption rates and oxygen concentration in wild-type (WT) and mitochondrial DNA (mtDNA) depleted (rho(0)) Molt-4 cells. Wild-type Molt-4 cells have moderate oxygen consumption rates, which were significantly reduced in the rho(0) cells. PCMB (p-chloromercurobenzoate) inhibited the oxygen consumption rates in both WT and rho(0) cells, whereas potassium cyanide decreased the oxygen consumption

Placental endothelial ferroptosis induced by neutrophil NETosis leads to intrauterine growth restriction in dengue virus infection.

Studies on dengue virus (DENV) infection during pregnancy show that symptomatic dengue is associated with adverse fetal outcomes, but the mechanisms remain unclear. Our previous research indicated that neutrophils contribute to intrauterine growth restriction (IUGR) by damaging placental vasculature, yet the molecular mechanisms underlying this microvascular injury remain poorly defined. To address this, this study examined placental microvascular ferroptosis by bulk and single-cell transcriptomic reanalyses, immunofluorescence staining, and Western blotting in DENV-2-infected E18.5 placentas

TAOK1 Protects against Renal Ischemia-Reperfusion Injury in Aged Mice through the STK3/BNIP3 Mitophagy Pathway.

The elderly suffer from a higher incidence and greater severity of acute kidney injury (AKI), yet the underlying molecular mechanisms remain poorly understood. This study dissects the possible role of thousand and one amino acid protein kinase 1 (TAOK1) in age-related AKI, specifically examining its regulation of mitophagy. Aged (23~24 months) C57BL/6 mice were subjected to renal ischemia-reperfusion injury (IRI) and TAOK1 was overexpressed in these mice. Renal function, histopathology, inflammation, neutrophil extracellular trap (NET) formation, and fibrosis were assessed. The involvement of

Evaluation of Rhinella marina Extracts on the Effector Functions of Human Neutrophils.

Neutrophils play a central role in the innate immune response. However, in inflammatory and autoimmune diseases, the exacerbated activation of these cells can result in excessive release of reactive oxygen species (ROS), proteases, and inflammatory mediators, contributing to tissue damage. In this context, natural compounds derived from amphibian venoms have attracted interest due to their pharmacological and immunomodulatory potential. This study aimed to evaluate in vitro the effects of the crude methanolic extract (CME) of Rhinella marina on the effector functions of human neutrophils. For

Advances in the Diagnosis and Management of Allergic Bronchopulmonary Aspergillosis/Mycosis.

Allergic bronchopulmonary aspergillosis/mycosis (ABPA/ABPM) is an immunologically mediated pulmonary disorder triggered by the colonization of filamentous fungi in the airways of patients with asthma or cystic fibrosis, leading to type I and III hypersensitivity responses, eosinophilic mucus plug formation, and central bronchiectasis. Over the past decade, advances in basic and clinical research have substantially reshaped the understanding and management of this disease. Although Aspergillus species, particularly Aspergillus fumigatus, remain the predominant causative fungi, epidemiological s

Interface N-glycans drive myeloperoxidase dimerization in vitro.

Myeloperoxidase (MPO) is a highly glycosylated heme oxidoreductase that, together with lactoperoxidase (LPO) and eosinophil peroxidase (EPO), contributes to host defence through the generation of (pseudo)hypohalous acids. Among the human heme peroxidases, mature MPO forms a unique covalently linked homodimer. Dimerization enhances stability and is essential for some biological functions, including chromatin disruption during neutrophil extracellular trap formation. Thus, we hypothesized that MPO dimerization must reflect a tightly regulated step in MPO biosynthesis and is driven by specific st

Targeting the plasticity of intestinal neutrophils: bidirectional regulation strategies by natural products.

Historically, neutrophils have been regarded primarily as pro-inflammatory cells, yet recent advancements have revealed their phenotypic heterogeneity and functional plasticity with versatile immunophenotypes. Distinct subpopulations of neutrophils exhibit a functional duality, not only initiating and amplifying inflammation, but also actively promoting tissue restoration in diseases, such as inflammatory bowel disease (IBD), colorectal cancer (CRC) and intestinal infections. They contribute to the formation of a dynamic immune microenvironment in concert with the intestinal microbiota, epithe

Screening and Targeting the HMGB1/RAGE Axis via Proteomics to Alleviate Inflammatory Responses in a Pseudomonas aeruginosa-Induced Rat Model of VAP.

PURPOSE: Ventilator-associated pneumonia (VAP) is a common and serious complication in mechanically ventilated patients, with Pseudomonas aeruginosa (PA) being one of the most frequently encountered pathogens. Dysregulated protein expression is closely associated with the progression of VAP; therefore, identifying and modulating potential protein targets are crucial for developing novel therapeutic strategies. METHODS: Quantitative proteomics was employed to analyze protein expression changes in a PA-induced rat model of VAP. Bioinformatics analysis was performed to identify key signaling pat

Gestational diabetes: from pathogenesis to therapeutic intervention.

Gestational diabetes mellitus (GDM), affecting up to 15% of pregnancies worldwide, is a complex metabolic disorder arising from the interaction of pregnancy-induced insulin resistance and inadequate pancreatic β-cell compensation. This review synthesizes current advances in understanding the pathogenesis and therapeutic strategies of GDM. Mechanistically, GDM develops through a multifactorial network involving placental hormone–driven insulin resistance, impaired β-cell adaptive remodeling, genetic susceptibility, and epigenetic modifications that alter key metabolic regulators such as INSR an

Paradigm shift in hydrogen medicine: Target molecules and catalytic mechanisms elucidated at the atomic level.

Molecular hydrogen (H2) has been shown to exert antioxidant and anti-inflammatory effects across a broad spectrum of disease models and clinical trials; however, its direct reaction rate with hydroxyl radicals (•OH) in aqueous solution is exceedingly slow, and the underlying mechanism of action has remained unresolved for many years as the "hydrogen paradox". The present review delineates a paradigm shift that resolves this enigma from three distinct perspectives. First, oxidized Fe-porphyrin "hematin [Fe(III)-OH]" has been identified as a redox-related biosensor for H2, reducing •OH selective

Reduced ALDH2 Activity and Cardiovascular Carbonyl Stress: From Reactive Aldehyde Networks to Coronary Microvascular Obstruction.

Primary percutaneous coronary intervention restores epicardial flow after ST-segment elevation myocardial infarction, but tissue perfusion may remain impaired. Coronary microvascular dysfunction, angiographic no-reflow, cardiac magnetic resonance-defined microvascular obstruction, and intramyocardial hemorrhage are overlapping but non-equivalent phenotypes. This review examines reduced mitochondrial aldehyde dehydrogenase 2 (ALDH2) activity as a susceptibility amplifier rather than an established independent cause of these outcomes. Reperfusion generates reactive oxygen species that initiate l

Circulating cell-free DNA as a biomarker in systemic lupus erythematosus: A narrative review.

BACKGROUND: Systemic lupus erythematosus (SLE) is a multifactorial, clinically heterogenous autoimmune disease. Reliable biomarkers are essential for early diagnosis, disease activity monitoring, flare prediction, and guiding therapeutic decisions. Biomarkers currently used in clinical practice - such as inflammatory markers, complement levels, and anti-double-stranded DNA (anti-dsDNA) antibodies - are valuable but lack sufficient sensitivity and specificity for predicting flares and disease progression. Circulating cell-free DNA (cfDNA) has emerged as a promising biomarker candidate. OBJECTI
